Carboxylic Acids And Light Interact To Affect Nanoceria Stability And Dissolution In Acidic Aqueous Environments, Matthew L. Hancock, Eric A. Grulke, Robert Yokel
Carboxylic Acids And Light Interact To Affect Nanoceria Stability And Dissolution In Acidic Aqueous Environments, Matthew L. Hancock, Eric A. Grulke, Robert Yokel
Nanobiotechnology Center Faculty Publications
Cerium atoms on the surfaces of nanoceria (i.e., cerium oxide in the form of nanoparticles) can store or release oxygen, cycling between Ce3+ and Ce4+; therefore, they can cause or relieve oxidative stress within living systems. Nanoceria dissolution occurs in acidic environments. Nanoceria stabilization is a known problem even during its synthesis; in fact, a carboxylic acid, namely citric acid, is used in many synthesis protocols. Citric acid adsorbs onto nanoceria surfaces, limiting particle formation and creating stable dispersions with extended shelf life. To better understand factors influencing the fate of nanoceria, its dissolution and stabilization have …
Gnetin C For Chemoprevention And Therapy Of Prostate Cancer, Gisella Campanelli
Gnetin C For Chemoprevention And Therapy Of Prostate Cancer, Gisella Campanelli
Selected Full-Text Dissertations 2020-
Prostate cancer is common among aging men. Despite current management, it continues to be associated with a high mortality rate. A need, therefore, exists for improved treatment approaches that target every stage of the disease. In this study, we investigated the efficacy of Gnetin C, a resveratrol dimer found abundantly in melinjo (Gnetum gnemon) seeds, across three different treatment modalities. The first involved a Gnetin C-supplemented diet as a chemopreventive strategy in a high-risk transgenic mouse model which overexpresses MTA1 on a background of Pten heterozygosity (R26MTA1; Pten+/f). The second involved the administration of Gnetin C by intraperitoneal (i.p.) injection …
Utilization Of Physiological Based Pharmacokinetic Modeling To Predict Systemic Exposure And Guide Dosing Decisions For Therapeutic Drug Monitoring Of Beta-Lactams In Critically Ill Patients, Hui Wei
Selected Full-Text Dissertations 2020-
Sepsis leads to a continuous pathophysiologic process that dysregulates host response which potentially causes significant organ damage and even death, especially respiratory, renal and cardiac failure. Changes in the clearance and volume of distribution are typically result in pharmacokinetic variabilities and often make it challenging for clinicians to appropriately dose beta-lactam antibiotics. Therapeutic drug monitoring (TDM) is necessary to further optimize beta-lactam pharmacokinetic/pharmacodynamic (PK/PD) parameters to improve clinical outcomes and reduce therapeutic failure.
Cefepime (CEF) and meropenem (MEM) primarily excreted by the kidney, and altered the kidney function of the ICU patients would affect the clearance of the medications. On …

Health Sciences
Introduction: Multimodal management of rectal cancer, combining neoadjuvant Radio chemotherapy followed by total Mesorectal therapy (TME), has been shown to improve locoregional control. However, the histological response is variable independently of the initial tumor stage. Some recent data suggest the usefulness of the lymphocyte-monocyte ratio (LMR), neutrophil-lymphocyte ratio (NLR), and platelet-lymphocyte ratio (PLR) in predicting the histological response to neoadjuvant therapy. This study aimed to investigate the correlation between the complete histological response (pCR) after neoadjuvant radio chemotherapy and biological parameters (LMR, NLR and PLR) in rectal cancer patients.
PATIENTS AND METHODS: This retrospective study was conducted in three Moroccan …

Theses, Dissertations and Capstones
Disulfiram (Antabuse®) is an oral prescription drug used to treat alcohol abuse disorder (AUD). Disulfiram (DSF) and its metabolites act on the body by inhibiting multiple enzymes involved in alcohol metabolism. This study aims to understand the pharmacology of disulfiram as an antibacterial medication. Previous studies on DSF showed antibiotic activity against multidrug-resistant Staphylococcus aureus strains (MRSA). The initial aim of this study was to decipher the mechanisms involved in inhibiting bacterial growth by DSF, which was done by performing biochemical studies. The studies revealed that DSF had antagonistic effects on redox buffering and energy production. Another aim of this …

Cancer Immunotherapy: A Breakthrough In Patient Care, Abdou-Samad Kone, Nour El Imane Issam Salah, Amina Ghouzlani, Abdallah Badou
Health Sciences
Cancer is one of the world's leading causes of death. Conventional therapies, such as surgical resection, radiotherapy, and chemotherapy, have improved patient management and have shown overwhelming progress. However, the incidence and mortality rates among cancer patients are still extremely high, at approximately 20,5 % and 10,6 % for both sexes, respectively. This may be due to the heterogeneity of tumors, their special microenvironment, and the development of resistance to certain conventional therapies. The concept of immunotherapy provided a new perspective on patient treatment. The interaction of tumor cells with their microenvironment depends on different factors and conditions that promote …

Influences Of Host Cytokine Signaling On Kshv Infection In Tonsil-Derived B Lymphocytes, Nedaa Alomari
Pharmaceutical Sciences (PhD) Dissertations
Kaposi sarcoma-associated herpesvirus (KSHV) is the etiologic agent of kaposi sarcoma. It is also associated with the B cell lymphoproliferative disorders, primary effusion lymphoma (PEL), multicentric Castleman disease (MCD) and KSHV inflammatory cytokine syndrome (KICS). Existing research reveals that KSHV has developed numerous strategies to alter host cytokine signaling activities to facilitate viral persistence and escape host immune surveillance.
Despite three decades of research, there are still many questions in this field awaiting further investigation. This dissertation will investigate host cytokines as factors influencing the establishment of KSHV infection in the human tonsil. We have identified and characterized different cytokines …

Hippocampal Versus Cortical Deletion Of Cholinergic Receptor Muscarinic 1 In Mice Differentially Affects Post-Translational Modifications And Supramolecular Assembly Of Respiratory Chain-Associated Proteins, Mitochondrial Ultrastructure, And Respiration: Implications In Alzheimer's Disease, Mohammad Golam Sabbir, Mamiko Swanson, Robert C. Speth, Benedict C. Albensi
HPD Articles
In a previous retrospective study using postmortem human brain tissues, we demonstrated that loss of Cholinergic Receptor Muscarinic 1 (CHRM1) in the temporal cortex of a subset of Alzheimer's patients was associated with poor survival, whereas similar loss in the hippocampus showed no such association. Mitochondrial dysfunction underlies Alzheimer's pathogenesis. Therefore, to investigate the mechanistic basis of our findings, we evaluated cortical mitochondrial phenotypes in Chrm1 knockout (Chrm1) mice. Cortical Chrm1 loss resulted in reduced respiration, reduced supramolecular assembly of respiratory protein complexes, and caused mitochondrial ultrastructural abnormalities. These mouse-based findings mechanistically linked cortical CHRM1 loss with poor survival of …
Elucidating The Impact Of Western Diet On Bone Marrow Cytokine Signatures: Understanding Poor Outcomes In Susceptibility To Infection And Progression Of Cancer, Lahari Kondeti
Theses, Dissertations and Capstones
Among other disease outcomes, obesity is associated with 1) reduction in long-lasting immune protection and 2) acquisition of cancer. Given our interest in hematopoietic stem and progenitor cell (HSPC) dynamics, we questioned if similar mechanisms in the bone marrow microenvironment contribute to obesity-induced HSPC alterations responsible for 1) decreased immune protection and 2) transformation of HSPCs in the pathogenesis of hematologic malignancies. The negative effects of obesity on immune protection are most notably established in influenza models. Influenza infection susceptibility is increased and influenza vaccine-induced immunity wains in obese individuals. Ongoing studies aim to define mechanisms for waning immunity at …

Prg4/Cd44/Pp2a Signaling Axis: A Significant Regulator Of Gout Arthritis, Sandy Elsayed
Prg4/Cd44/Pp2a Signaling Axis: A Significant Regulator Of Gout Arthritis, Sandy Elsayed
Pharmaceutical Sciences (PhD) Dissertations
Introduction: Gout is a common chronic arthritis caused by monosodium urate (MSU) crystal deposition in joints leading to activation of synovial resident macrophages and downstream production of interleukin-1b (IL-1b). Xanthine oxidase (XO) catalyzes purines into uric acid (UA) and reactive oxygen species (ROS) that activate NLRP3 inflammasome. Protein phosphatase-2A (PP2A) may be involved in regulating inflammatory pathways in macrophages. Proteoglycan 4 (PRG4) and its receptor CD44 and its transducer PP2A signaling axis play a biologically significant role in regulating urate crystal inflammation.
Methods: Monocytes and macrophages were primed with TLR2 agonist and stimulated with MSU crystals. Fingolimod (PP2A …

Assembly Of Synaptic Protein-Dna Complexes: Critical Role Of Non-Specific Interactions, Sridhar Vemulapalli, Mohtadin Hashemi, Anatoly B. Kolomeisky, Yuri L. Lyubchenko
Journal Articles: Pharmaceutical Sciences
The synaptic protein-DNA complexes, formed by specialized proteins that bridge two or more distant sites on DNA, are critically involved in various genetic processes. However, the molecular mechanism by which the protein searches for these sites and how it brings them together is not well understood. Our previous studies directly visualized search pathways used by SfiI, and we identified two pathways, DNA threading and site-bound transfer pathways, specific to the site-search process for synaptic DNA-protein systems. To investigate the molecular mechanism behind these site-search pathways, we assembled complexes of SfiI with various DNA substrates corresponding to different transient states and …
Amyloid Cascade Hypothesis For Alzheimer’S Disease. Does It Work Under Physiological Conditions?, Yuri L. Lyubchenko
Amyloid Cascade Hypothesis For Alzheimer’S Disease. Does It Work Under Physiological Conditions?, Yuri L. Lyubchenko
Journal Articles: Pharmaceutical Sciences
Plaques in the brain consisting of proteins are a hallmark of diseases like Alzheimer’s disease (AD) and Parkinson’s disease (PD). Such aggregates can be assembled spontaneously by specialized proteins such as amyloid beta (Ab) proteins in the case of AD. Numerous in vitro studies made a foundation for the Amyloid Cascade Hypothesis (ACH), according to which the misfolding of proteins leads to their self-assembly into toxic oligomers along with the formation of amyloid fibrils assembled as plaques in the brain. Notably physiological concentration of Ab proteins in the brain is in the low nanomolar concentration, so no spontaneous aggregation of …
