Autosomal dominant polycystic kidney disease (ADPKD) is an inherited genetic disorder that results in progressive renal cyst formation with ultimate loss of renal function and other systemic disorders. These systemic disorders include abnormalities in cardiovascular, portal, pancreatic and gastrointestinal systems. ADPKD is considered to be among the ciliopathy diseases due to the association with abnormal primary cilia function. In order to understand the full course of primary cilia and its association with ADPKD, the structure, functions and role of primary cilia have been meticulously investigated. As a result, the focus on primary cilia has emerged to support the vital roles …
Rhodopsin Trafficking And Retinal Function, Nicholas Joseph Reish
Rhodopsin Trafficking And Retinal Function, Nicholas Joseph Reish
All ETDs from UAB
The health and maintenance of rod photoreceptors depends on the high-fidelity vectorial transport of rhodopsin from its site of synthesis in the inner segment to its final resting place in the outer segment disks. Multiple molecular interactions play a role in this process, including the interaction of the small GTPase rab11a with rhodopsin that was first described in vertebrates in amphibians. We found that the interaction of the small GTPase rab11a and rhodopsin is conserved in mammalian retina. This interaction is direct, is abrogated by human disease-causing truncation mutations in rhodopsin, and does not depend on the nucleotide binding status …
Gabaergic Dysfunction In Huntington Disease, Sarah Elizabeth Dougherty
Gabaergic Dysfunction In Huntington Disease, Sarah Elizabeth Dougherty
All ETDs from UAB
GABAergic neurons are responsible for regulation of neuronal signaling through inhibition. Alterations in the delicate balance of excitation and inhibition have been observed in many neurological disorders, including neurodegenerative diseases. Huntington disease (HD) is an autosomal dominant neurodegenerative disorder that is characterized by motor, psychiatric, and cognitive disturbances. As the hallmark movement pathology of HD is hyperkinesis it is intriguing to postulate that alterations in inhibition, and by extension within GABAergic cell networks, could be part of the pathogenesis of this disorder. In order to elucidate the role of GABAergic cell populations in HD we have used behavioral paradigms, molecular …

Saha Cardiovascular Research Center Faculty Publications
Studies suggest that inflammation impairs reverse cholesterol transport (RCT). We investigated whether serum amyloid A (SAA) contributes to this impairment using an established macrophage-to-feces RCT model. Wild-type (WT) mice and mice deficient in SAA1.1 and SAA2.1 (SAAKO) were injected intraperitoneally with 3H-cholesterol-labeled J774 macrophages 4 hr after administration of LPS or buffered saline. 3H-cholesterol in plasma 4 hr after macrophage injection was significantly reduced in both WT and SAAKO mice injected with LPS, but this was not associated with a reduced capacity of serum from LPS-injected mice to promote macrophage cholesterol efflux in vitro. Hepatic accumulation of 3 …
Thc-Mediated Induction Of Δfosb And Its Modulation Of Cb1r Signaling And Adaptation, Lazenka Matthew
Thc-Mediated Induction Of Δfosb And Its Modulation Of Cb1r Signaling And Adaptation, Lazenka Matthew
Theses and Dissertations
The main psychoactive and therapeutic effects of Δ9-tetrahydrocannabinol (THC) are mediated through cannabinoid type 1 receptors (CB1Rs). The therapeutic uses of THC are mitigated by the development of tolerance to these therapeutic effects, whereas tolerance does not readily develop to some of the side-effects of THC, like motor impairment and reward. The development of tolerance occurs through adaptations at CB1Rs, which include desensitization (G-protein uncoupling) and downregulation (receptor degradation). Brain region-dependent differences in THC-mediated adaptations are proposed to explain the differences in tolerance to various THC-mediated effects. These studies focused on whether ΔFosB, a stable transcription factor, could regulate CB1R …

The Journal of Genomes and Exomes is a new, peer-reviewed, open-access, online publication whose scope comprises reporting of high quality genome, exome, and gene panel sequences with attendant, detailed phenotypes. The intent of this journal is to facilitate comparisons between genome, exome and gene panel sequencing studies in order to assist significance testing of the genotypephenotype associations, particularly those in uncommon genetic diseases. While there is yet to be a consensus regarding these classifications, the definition of an empiric set is helpful in understanding error models. Herein we have suggested structured templates for submissions and the rationale for the data …
Effect Of Blood Collection Practices On Emergency Department Blood Specimen Rejection Rates, Barbara K. Vernoski
Effect Of Blood Collection Practices On Emergency Department Blood Specimen Rejection Rates, Barbara K. Vernoski
UNF Graduate Theses and Dissertations
The practice of obtaining blood as part of the placement of a new peripheral venous access device (p-VAD) is a frequent practice in the emergency department (ED). Of the concerns related to this practice is the possibility of laboratory specimen rejection due to p-VAD catheter size, use of the wrong collection device, and the absence of a standardized collection process. The objective of this study, therefore, was to examine the effect of the use of evidence-based venipuncture and p-VAD blood collection protocols on the rejection rate of blood specimens drawn by staff in the adult areas of an urban academic …

Universal health care is provided under the Federal Unified Health System (Sistema Único de Saúde) but, as in many other middle and low income countries, access to medical genetics services is limited in rural and remote regions of the country. Since there is no formally recognized Genetic Counseling profession, genetic counseling is provided by physicians, trained either in medical genetics or a related clinical discipline. A comprehensive medical genetics program has been established in Monte Santo, an inland …
The Characterisation Of Pax3 Expressant Cells In Adult Peripheral Nerve [Journal Article], Judith Blake, Melanie Ziman
The Characterisation Of Pax3 Expressant Cells In Adult Peripheral Nerve [Journal Article], Judith Blake, Melanie Ziman
Research outputs 2013
Pax3 has numerous integral functions in embryonic tissue morphogenesis and knowledge of its complex function in cells of adult tissue continues to unfold. Across a variety of adult tissue lineages, the role of Pax3 is principally linked to maintenance of the tissue’s resident stem/progenitor cell population. In adult peripheral nerves, Pax3 is reported to be expressed in nonmyelinating Schwann cells, however, little is known about the purpose of this expression. Based on the evidence of the role of Pax3 in other adult tissue stem and progenitor cells, it was hypothesised that the cells in adult peripheral nerve that express Pax3 …
Impedance Changes In Biceps Brachii Due To Isometric Contractions And Muscle Fatigue Using Electrical Impedance Myography (Eim), Travis Orth
College of Graduate Studies: Theses & Dissertations
Electrical impedance myography (EIM) is a non-invasive technique used to evaluate neuromuscular conditions by using a quantitative parameter called impedance (Z). It relies upon the application and measurement of high-frequency, low-intensity electrical current imbedded over a localized muscle to determine its opposition to current flow. In brevity, impedance determines the resistance of muscle fibers due to changes in muscle composition. The objective of this thesis was to establish a relationship between muscles at rest and during isometric contractions at various force levels(25%, 50%, and 100% of maximum voluntary contraction) of the biceps brachii over a multifrequency spectrum. Impedance measurements due …

To investigate inductive activity of formaldehyde (FA) on the expression of inducible nitric oxide synthase (iNOS) and level of nitric oxide (NO) in the liver of rats. Materials and methods: Sixteen male Wistar albino rats of the same age (3 months old) were divided into 2 experimental groups, with 8 rats as the control and 8 rats formaldehyde-treated (FAt). The FAt group received intraperitoneal injection of 10 mg/kg FA for 10 days. Activity of iNOS was shown with immunohistochemical stain kits, and the concentration of NO level was measured by spectrophotometric method in the liver tissue. Results: The FAt group …

In this study, the curative effect of valsartan on hypertension based on its effect on visfatin levels and lipid profiles was investigated. Materials and methods: Forty patients aged 38-60 with hypertension were included in the study. In order to determine initial visfatin levels and lipid profiles, blood samples were taken from the patients and all were started on 80-mg valsartan tablets. All patients were monitored for 12 weeks. Results: By using multivariate regression analysis, it was determined that visfatin levels affected systolic blood pressure (SBP) (P = 0.012, beta; -0.43, 95% CI: -0.092 to -0.012). After the patients received the …
