D-fenfluramine (d-FEN) was once widely prescribed and was among the most effective weight loss drugs, but was withdrawn from clinical use because of reports of cardiac complications in a subset of patients. Discerning the neurobiology underlying the anorexic action of d-FEN may facilitate the development of new drugs to prevent and treat obesity. Through a combination of functional neuroanatomy, feeding, and electrophysiology studies in rodents, we show that d-FEN-induced anorexia requires activation of central nervous system melanocortin pathways. These results provide a mechanistic explanation of d-FEN's anorexic actions and indicate that drugs targeting these downstream melanocortin pathways may prove to …

Interleukin-12 (IL-12) has been used in numerous immunotherapy protocols against melanoma. However, delivery of IL-12 in the form of recombinant protein can result in severe toxicity, and gene therapy has had limited success against B16.F10 murine melanoma. The purpose of this study was to examine the effectiveness of in vivo electroporation for the delivery of plasmid DNA encoding IL-12 as an antitumor agent against B16.F10 melanoma. A self-consistent model analysis of electroporation in biological cells has been carried out based on an improved energy model. The simple energy model used in the literature is somewhat incorrect and unphysical for a variety of reasons. Our model for the pore formation energy E(r) includes a dependence on pore population and density. It also allows for variable surface tension, incorporates the effects of finite conductivity on the electrostatic correction term, and is dynamic in nature. Self-consistent calculations, based on a coupled scheme involving the Smoluchowski equation and the improved energy model, are presented. It is shown that E(r) becomes …

Water gain in the brain consequent to hyponatremia is counteracted by mechanisms that initially include a compensatory displacement of liquid from the interstitial space to cerebrospinal fluid and systemic circulation and subsequently an active reduction in cell water accomplished by extrusion of intracellular osmolytes to reach osmotic equilibrium. Potassium (K+), chloride (Cl–), amino acids, polyalcohols, and methylamines all contribute to volume regulation, with a major contribution of ions at the early phase and of organic osmolytes at the late phase of the regulatory process. Regulation of B cell activation depends on integration of signals transmitted by the B cell receptor (BCR) and a variety of co-receptors. CD72 is a B cell co-receptor that is expressed in all stages of B cell development except plasma cells. Ligation of CD72 enhances B cell growth and differentiation. Recently, the class IV semaphoring, CD100, has been identified as the natural ligand for CD72. Cytoplasmic domain of CD72 has been shown to be associated with SHP-1 leading to the proposal that the positive effects of CD72 on B cell response may result from sequestration of negative signals from BCR. …

This study was designed to demonstrate the microbiological qualities of cream pastries and puddings obtained from five different confectioneries in Van. A total of 175 samples, consisting of 100 pudding samples (sütlaç 25, keşkül 30, supangile 25, kazandibi 20) and 75 cream pastries (butter-cream 25, chocolate-cream 25, fruit-cream 25), were analysed. The drop plate technique was used to determine the numbers of total anaerobic bacteria, enterobactericeae, lactobacilli, pseudomonas and yeasts/moulds, and the spread plate technique was used to determine the total number of micrococci/staphylococci. This study was carried out to determine the hatchability characteristics of rock partridges mated at the ratio of one male to three females, one male to four females and one male to five females. The effects of various salinities and fresh water (FW) (0.4 ppt) on the growth performance, body traits and fillet composition of juvenile and young sea bass (Dicentrarchus labrax) in FW were investigated in two separate experiments in this study. In Experiment 1, following acclimation, sea bass juveniles (9.5 ± 0.5 g) were reared in 240-L fibreglass tanks (40 x 40 x 150 cm) for 90 days in five different treatments (FW, 10, 20, 30 and 40 ppt). The fish cultured in FW, 10 and 20 ppt grew better than those at 30 or 40 ppt (P < 0.05). Salinity did not affect the crude protein, lipid or ash content of the fish (P > 0.05). Yet, the dry …

In this research 30 sucuk samples were obtained from 5 meat plants with high capacity, during 3 months (January 1998-March 1998) at 15 day intervals. At the results of determination of the 30 sucuk samples, 80% of them was not shown acceptable colour development and 93.33% had a soft texture. 83.33% of the samples cut from the surface was not indicated good colour development and 50% of them has not good marble form. In this study, leptospiral antibodies were investigated by Microscopic Agglutination Test (MAT) and Enzyme Linked Immunosorbent Assay (ELISA) in order to detect leptospirosis and serotypes which cause leptospira infection in cattle raised in Kars and Ardahan. Live and sonicated antigens prepared from Leptospira hardjo and Leptospira grippotyphosa serotypes were used. Blood samples were taken from 150 cattle under 2 years of age and 840 cattle 2 years of age or older (a total of 990) which were unvaccinated against leptospirosis in 30 rural settlements. This study was performed to determine the incidence and anatomo-pathologic aspects of reproductive organ tumors in laying chickens. For this research, 10,475 16-18-month-old, commercial laying hens of the Babcock genotype were obtained from four flocks in Afyon city. They were examined right after slaughtering, and reproductive organ tumors in 804 hens (7.67%) were observed. Out of these 804 hens, leiomyoma was found in 760 hens (7.25%), oviduct adenocarcinoma in 39 hens (0.37%) and leiomyoma together with oviduct adenocarcinoma in 5 hens (0.05%) were detected. In this study, the gonadosomatic index value and flesh productivity of Scorpion Fish (Scorpaena porcus Linne., 1758) were investigated for fish caught in the Sinop region using bottom nets. With this purpose, the gonadosomatic index value of a total of 633 samples were examined. The gonadosomatic index was found to have a maximum value in June and a minimum value in September. The flesh productivity of females was determined to be 34.24 ± 0.014 and 34.47 ± 0.015 for males and 34.35 ± 0.007 as a mean. The seasonal variation and meat composition of blue crabs (Callinectes sapidus Rathbun, 1896) were investigated. Twenty (10 male, 10 female) crabs were used each season. The total weight of the samples was estimated and the ratios of crude protein, lipid, dry matter and crude ash were obtained from meat of the body and claws. Comparatively higher values of dry matter, crude ash and lipid were determined for the claw meat of males in winter. Hydrocephalus associated lesions such as diverticula, clefts and tears observed in periventricular white matter were studied pathologically in three puppies with necropsy between 1993 and 1994. Enlargement of the cranial vault was present in the puppies. The animals were blind, and their morphological alterations were similar. Marked hydrocephalus caused by mesencephalic canal malformations and obstruction was detected in all cases. Compression atrophy was seen in the nucleus caudatus, hippocampus, putamen and cerebellum in all animals. Cerebral sulci malformations were also detected in the cranial parts of the brain. This study was carried out in order to determine the species diversity, abundance, and distribution and seasonal frequency occurrence times of the eggs and larvae of Teleost fish in Izmir Bay during 1989-1990. A total of 32140 eggs and 6582 larvae were collected from plankton samples and 69 species belonging to 27 families were identified. The most abundant families included Sparidae 23.79%, Clupeidae 24.63%, Mullidae 12.22%, Serranidae 8.72% and Mugilidae 8.04% of total eggs, and Gobiidae 67.38%, Sparidae 9.43% and Blennidae 9.31% of total larvae. The cardiovascular/respiratory and clinical effects of a xylazine-ketamine combination or a butorphanol-ketamine combination for the induction of anaesthesia in dogs were compared. A total of 12 dogs (two groups 6 dogs each) were premedicated with either xylazine (group I) or butorphanol (group II) and anaesthetized with ketamine HCl and monitored at 0, 5, 15, 30 and 60 min after induction. Sedative effects in both groups started within 15 min. Respiratory rate was significantly decreased in group I (P < 0.05). Body temperature was significantly altered after 15 min in group I (P < 0.05 and P < 0.001). Haematological analyses indicated that there were no differences in pH values between the groups. It was concluded that butorphanol followed by ketamine was comparable with xylazine plus ketamine for the induction of anaesthesia in healthy dogs.

Protein bands of crude antigens of Hymenolepis nana were determined by SDS-PAGE and Western blotting. Thirty Swiss albino mice were allotted into two groups of 15 each as positive (infected with H. nana) and negative (non-infected with H. nana) groups. The natural infections of H. nana and other helminths were determined by centrifugal flotation of faeces. The goal of this study was to evaluate the abnormal spermatozoa rates and types of American Bronze turkeys under Turkish conditions. Semen was collected from toms by abdominal massage and pooled once a week for 10 weeks. After pooling, the semen samples were examined morphologically and mean abnormal rates and types were evaluated. A total abnormal spermatozoa rate of 17 ± 0.06% was determined. In this study, corpus mandibula fractures were experimentally induced on 30 rabbits and 3 different materials were used for fixation. Thirty rabbits were arranged into 3 groups and then wire, polydioxanone and mini plate and screw osteosynthesis materials were used. The fracture healing was examined and compared with clinical, radiological and histological results. From the radiological results the parameters of groups were compared with Lane and Sandhu scale system in terms of callus tissue, mineralization and remodeling. In this study, the capture efficiencies and catch rates of three different traps were compared for the blue crab Callinectes sapidus in Beymelek Lagoon, Antalya, Turkey. All gear was used simultaneously and in the same habitats. The mean catch per unit effort (CPUE) was determined to be significantly higher for hoop nets than for traps. Gear effectiveness was examined by comparing CPUE (CPUE as number of crab per trap per haul, and g per trap haul), mean weight of individual crabs, catch composition and width frequency distributions of the blue crab for each gear.

Characterisation Of An Exopolysaccharide Preventing Phage Adsorption In Lactococcus Lactis Subsp. Cremoris Ma39, Mustafa Akçeli̇k, Pinar Şanlibaba

Lactococcus lactis subsp. cremoris strain MA39 produces an extracellular polysaccharide containing rhamnose, glucose and galactose. A 16.5 kb plasmid encoding exopolysaccharide production in MA39 was determined by plasmid curing experiments. Phage adsorption assays showed that four different lactococcal phages were adsorbed to MA39-40; only the 16.5 kb plasmid cured an exopolysaccharide non-producing mutant of strain MA39, with high efficiency (93.2-98.5%) while adsorption of these phages was completely inhibited in the wild type strain MA39. These results suggest that exopolysaccharide prevents phage adsorption by masking phage receptor sites.

Thymic nurse cells are isolated thymic epithelial/thymocyte complexes described in vitro. Their in vivo existence is controversial. We examined rat thymi in order to distinguish thymic nurse cells in vivo using the zinc iodide-osmium tetroxide (ZIO) fixation/staining technique. There were mainly two types of strongly ZIO reactive cells in the thymic cortex: macrophages and small clusters of thymic epithelial cells. The latter group was composed of thymic epithelial cells and intact thymocytes forming complexes. Thus we suggest that these structures are in situ equivalents of thymic nurse cells.

A case of neonatal renal oxalosis in a 7-day-old calf is described. Clinical biochemistry revealed increased blood urea nitrogen. Gross findings included thin renal medulla and dilated renal calyces containing numerous, small, pale yellow and granular calculi. Histologically, most cortical and medullary tubules were dilated with birefringent crystals, whereas the glomeruli appeared unaffected. The crystals were morphologically consistent with calcium oxalate by the Pizzolato technique. Diffuse interstitial fibrosis and focal accumulations of lymphocytes were also noted.
