First Evidence Of Plague (Yersinia Pestis) In Nebraska Is Found In Panhandle Predators, Dallas Virchow, Wayne L. Kramer, Charles S. Brown, Scott E. Hygnstrom, Alan M. Barnes

Transactions of the Nebraska Academy of Sciences and Affiliated Societies

In September, 1992, plague (Yersinia pestis) antibody was found in the blood of a coyote (Canis latrans) (1:128 titer) and a badger (Taxidea taxus) (1:2048 titer) taken near a suspect black-tailed prairie dog (Cynomys ludovicianus) town in Sioux County. However, a 1989-1991 survey of 96 coyotes taken from nine Nebraska Panhandle counties demonstrated no plague antibodies. Passive hemagglutination tests of Nobuto blood-sampling paper proved useful to quickly survey a predator population that is distributed across a broad geographic area.

Phosphorylation Of The Rex Protein Of Human T-Cell Leukemia Virus Type 1, Yoshifumi Adachi, Terry D. Copeland, Chiaki Takahashi, Tetsuya Nosaka, Aftab Ahmed, Stephen Oroszlan, Masakazu Hatanaka

Pharmacy Faculty Articles and Research

Rex protein, the posttranscriptional regulator of human T-cell leukemia virus type I (HTLV-I), is required for the control of viral structural protein expression and virus replication. Rex is a phosphoprotein found predominantly in the cell nucleolus, whose function is thought to be regulated by its nucleolar localization and phosphorylation. Therefore, we investigated the in vivo phosphorylation of Rex protein in more detail. Phosphorylation of Rex occurred in all HTLV-I-infected cell lines examined in vivo, primarily at serine residues and to a very small extent at threonine residues. Treatment of cells with 12-O-tetradecanoylphorbol-13-acetate (TPA) led to significant but transient enhancement of …

Effects Of Bacterial Lipopolysaccharide On Plasma Corticosterone Concentrations And Body Temperatures Of New Zealand Rabbits (Oryctolagus Cuniculus), Phillip Keith Wilkins, Stanley N. David, Lawrence W. Hinck

Journal of the Arkansas Academy of Science

Twelve New Zealand rabbits were injected with Salmonella typhosa endotoxin, 10 ng/kg b.w. ,via an auricular marginal vein and the effects of the pyrogen on the rectal temperatures and plasma corticosterone concentrations of these animals were observed. Our data showed significant increases of the core temperatures from the normal 39.3 +/- 0.18 to 40.9 +/- 0.43 C (p < 0.001). Radioimmunoassay results of the plasma corticosterone levels were 5.76 +/- 3.7 ug/100 ml in the pre-injection blood samples and 9.02 +/- 3.7 ug/100 ml in the plasmas obtained from the animals, one hour after the pyrogen was administered. The increase of corticosterone was significant (p

Purification Of Core-Binding Factor, A Protein That Binds The Conserved Core Site In Murine Leukemia Virus Enhancers., Shuwen W. Wang, Nancy A. Speck Jan 1992

Purification Of Core-Binding Factor, A Protein That Binds The Conserved Core Site In Murine Leukemia Virus Enhancers., Shuwen W. Wang, Nancy A. Speck

Dartmouth Scholarship

The Moloney murine leukemia virus causes thymic leukemias when injected into newborn mice. A major genetic determinant of the thymic disease specificity of the Moloney virus genetically maps to two protein binding sites in the Moloney virus enhancer, the leukemia virus factor b site and the adjacent core site. Point mutations introduced into either of these sites significantly shifts the disease specificity of the Moloney virus from thymic leukemia to erythroleukemia (N. A. Speck, B. Renjifo, E. Golemis, T. Frederickson, J. Hartley, and N. Hopkins, Genes Dev. 4:233-242, 1990). We have purified several polypeptides that bind to the core site …

Trypanosome Infection Rates In Tsetse Flies (Diptera: Glossinidae) And Cattle During Tsetse Control Operations In The Kagera River Region Of Rwanda, Steve Mihok, Leonard H. Otieno, Christopher S. Tarimo Jan 1992

Trypanosome Infection Rates In Tsetse Flies (Diptera: Glossinidae) And Cattle During Tsetse Control Operations In The Kagera River Region Of Rwanda, Steve Mihok, Leonard H. Otieno, Christopher S. Tarimo

All Peer-Reviewed Publications

Trypanosome infections were monitored in three species of tsetse fly (Glossina pallidipes Austen, G. morsitans centralis Machado, and G. brevipalpis News-tead) at four locations in the Kagera River region of Rwanda from May 1989 to September 1990. Two of the four areas (Mpanga Ranch and Bukora Ranch) were subjected to tsetse fly suppression operations with odour-baited traps. Proboscis infections of the Trypanosoma congolense and T. vivax types accounted for roughly equal numbers of the 207 mature infections detected (3.8%). Variation in infection rates was area-specific rather than tsetse species-specific. Order of magnitude differences in tsetse fly densities among areas were …

Studies On Tsetse Midgut Factors That Induce Differentiation Of Bloodstream Trypanosoma Brucei Brucei In Vitro, M. O. Imbuga, E. O. Osir, V. L. Labongo, N. Darji, L. H. Otieno Jan 1992

Studies On Tsetse Midgut Factors That Induce Differentiation Of Bloodstream Trypanosoma Brucei Brucei In Vitro, M. O. Imbuga, E. O. Osir, V. L. Labongo, N. Darji, L. H. Otieno

All Peer-Reviewed Publications

An in vitro system for studying the transformation of bloodstream forms of Trypanosoma brucei brucei into procylic (midgut) forms is described. In this system, transformation of the parasites was stimulated by Glossina morsitans morsitans midgut homogenates at 27° C but not at 4° C. The transformation-stimulating capacity was irreversibly destroyed by heating the midgut homogenates at 60° C for 1 h. A correlation was established between the transformation activity of the midgut homogenates and trypsin activity. The protease inhibitors (soybean trypsin inhibitor and N-p-tosyl-l-lysine-chloromethyl-ketone) inhibited trypsin activity and completely blocked the transformation of the parasites. Furthermore, the midgut homogenates could …

Influence Of D(+)-Glucosamine On Infection Rates And Parasite Loads In Tsetse Flies (Glossina Spp.) Infected With Trypanosoma Brucei, Steve Mihok, Leonard H. Otieno, Nafisa Darji, David Munyinyi Jan 1992

Influence Of D(+)-Glucosamine On Infection Rates And Parasite Loads In Tsetse Flies (Glossina Spp.) Infected With Trypanosoma Brucei, Steve Mihok, Leonard H. Otieno, Nafisa Darji, David Munyinyi

All Peer-Reviewed Publications

Teneral Glossina morsitans centralis, G. m. morsitans and G. pallidipes were infected with three different clones of Trypanosoma brucei in blood containing d(+)-glucosamine, an inhibitor of tsetse midgut lectin. On average, 5 days of d(+)-glucosamine treatment tripled infection rates, without affecting the proportion of infections that matured. Total infection rates were equal in males and females, but twice as many infections matured in males. Counts of parasites in the guts and salivary glands of 277 flies revealed order of magnitude differences among flies, with females consistently having 2-3-times as many parasites as males. Parasite numbers varied in a sex-specific manner …

Laboratory And Field Observations On Anti-Tick Properties Of The Plant Gynandropsis Gynandra (L.) Brig, M. M. Malonza, O. O. Dipeolu, A. O. Amoo, S. M. Hassan

All Peer-Reviewed Publications

A shrubby plant, abundant in east Kenya, Gynandropsis gynandra (L.) Brig., was shown to exhibit repellent and acaricidal properties to larvae, nymphs and adult Rhipicephalus appendiculatus and Amblyomma variegatum ticks. All stages of ticks avoided the leaves of the plant and a high percentage of the ticks which were continuously exposed to its leaves died; surviving ticks were weak and inactive but regained activity when exposed to fresh air. The effectiveness of the plant's leaves as a repellent and acaricide was most pronounced on nymphs and least pronounced on adults. Field investigations indicated that ticks were not found up to …

The Ability Of Simian Virus 40 Large T Antigen To Immortalize Primary Mouse Embryo Fibroblasts Cosegregates With Its Ability To Bind To P53., Jiyue Y. Zhu, Marina Abate, Philip W. Rice, Charles N. Cole Dec 1991

The Ability Of Simian Virus 40 Large T Antigen To Immortalize Primary Mouse Embryo Fibroblasts Cosegregates With Its Ability To Bind To P53., Jiyue Y. Zhu, Marina Abate, Philip W. Rice, Charles N. Cole

Dartmouth Scholarship

The large T antigen encoded by simian virus 40 (SV40) plays essential roles in the infection of permissive cells, leading to production of progeny virions, and in the infection of nonpermissive cells, leading to malignant transformation. Primary mouse embryo fibroblasts (MEFs) are nonpermissive for SV40, and infection by wild-type SV40 leads to immortalization and transformation of a small percentage of infected cells. We examined the ability of an extensive set of mutants whose lesions affect SV40 large T antigen to immortalize MEFs. We found that immortalization activity was retained by all mutants whose lesions are located upstream of codon 346. …

Predation Of Free-Living Engorged Female Rhipicephalus Appendiculatus, Esther N. Mwangi, Robin M. Newson, Godwin P. Kaaya

All Peer-Reviewed Publications

In experiments done over a period of 1 1/2 years using engorged female Rhipicephalus appendiculatus tethered in a grass plot, 42% predation was observed in long grass (40-60 cm), and 36% in short grass (6-10 cm). Deaths due to environmental factors were 4.8% and 6.8% in long and short grass, respectively. Six groups of animals were confirmed to be predators of the ticks, namely: ants, spiders, rodents, birds, lizards and shrews. The implications of these results in making tick population models, and the possibility of using predators in integrated tick-control packages are discussed. © 1991 Elsevier Science Publishers B.V.

Arkansas Cooperative Beef Bull Performance Test 1991, M. W. Eaton, A. E. Brown Jr., J. A. Hornsby, R. W. Parham, J. D. Shockey, W. C. Loe

Arkansas Agricultural Experiment Station Research Series

Performance testing of beef bulls was initiated to improve productivity of Arkansas beef cattle through breeding. This cooperative testing program provides cattlemen with reliable information for identifying animals with superior breeding value for the traits of production measured in the test. By using the information as a basis for herd sire selection, a breeder can greatly increase the chances of obtaining a bull that will sire rapid-gaining, more efficient, highquality calves. Such calves can increase profits for both the breeder and the feeder. This testing program provides information that is useful to financial institutes in arranging their lending programs and …

Risk Assessment For Developmental Toxicity: Airborne Occupational Exposure To Ethanol And Iodine, Donald R. Mattison Jun 1991

Risk Assessment For Developmental Toxicity: Airborne Occupational Exposure To Ethanol And Iodine, Donald R. Mattison

RISK: Health, Safety & Environment (1990-2002)

Dean Mattison explains hazard identification, hazard characterization and exposure characterization as furnishing a foundation for Risk assessment generally. He then illustrates their application in assessing the fetal Risk posed by two common substances. Ultimately, he argues that only after Risks have been so examined can women of child bearing age (or anyone) decide what if any measures are appropriate to avoid them.

Fertile Women May Now Apply: Fetal Protection Policies After Johnson Controls, Barbara Ruhe Grumet

RISK: Health, Safety & Environment (1990-2002)

In its recent interpretation of Title VII of the Civil Rights Act, the U.S. Supreme Court leaves little room for permissible occupational sex discrimination. However, its decision has wider implications. Here, Professor Grumet takes a look at some of them from both a legal and a social perspective, including matters such as employees' reproductive freedom and employers' potential liability for a variety of possible injuries to employees' offspring.

A Flow Cytometric Analysis Of Immunocyte Populations In Peripheral Blood And Pleural Effusions Of Patients With Cancer, Mandy L. Bohn Jun 1991

A Flow Cytometric Analysis Of Immunocyte Populations In Peripheral Blood And Pleural Effusions Of Patients With Cancer, Mandy L. Bohn

Loma Linda University Electronic Theses, Dissertations & Projects

Two-color flow cytometric analysis was performed on paired samples of peripheral blood (PB) and pleural effusions (PE) of patients with metastatic malignancies. The purpose of these analyses was to test the hypothesis that there are significant differences in the distribution of immunocyte populations in the PE compared to that of the PB.

The majority of the pleural fluid immunocytes were CD3+ cells. The ratio of CD4+ to CD8+ cells was higher in the PE compared to PB. Levels of CD8+, CD19+ and CD14+ cells were not significantly different in the PE compared to the levels in the PB. CD16+CD56+ cells …

Department of Pathology and Laboratory Medicine

When defined in terms of markers for normal cell lineages, most invasive breast cancer cells correspond to the phenotype of the common luminal epithelial cell found in the terminal ductal lobular units. Luminal epithelial cells cultured from milk, which have limited proliferative potential, have now been immortalized by introducing the gene encoding simian virus 40 large tumor (T) antigen. Infection with a recombinant retrovirus proved to be 50-100 times more efficient than calcium phosphate transfection, and of the 17 cell lines isolated, only 5 passed through a crisis period as characterized by cessation of growth. When characterized by immunohistochemical staining …

Getting The Best Out Of Weaner Sheep In The Eastern Wheatbelt, Sally Revell Jan 1991

Getting The Best Out Of Weaner Sheep In The Eastern Wheatbelt, Sally Revell

Journal of the Department of Agriculture, Western Australia, Series 4

A study of the growth of Merino weaner flocks over summer and autumn in Western Australia 's eastern wheatbelt during 1989 and 1990 has identified efficient weaner management strategies, and provided a basis for future research and extension needs of this area. The average liveweight of several weaner flocks was unexpectedly low at the start of summer. Weaner flocks may need to be supplemented before the start of summer to prevent undue weight loss at this time.

Origin And Evolution Of Defective Interfering Rnas Of Tomato Bushy Stunt Virus, David A. Knorr, Thomas Jack Morris Jan 1991

Origin And Evolution Of Defective Interfering Rnas Of Tomato Bushy Stunt Virus, David A. Knorr, Thomas Jack Morris

School of Biological Sciences: Faculty Publications

Viruses with defective genomes have been identified in association with virtually every major family of viruses and have been widely utilized as tools for investigating virus functions in animal cell culture systems (Perrault, 1981). It is generally thought that defective interfering viruses (DIs) arise through deletion, rearrangement, or recombination of a competent viral genome. DIs tack the ability for independent existence relying on their parental helper viruses to supply factors required for replication, maturation, and/or encapsidation (Huang and Baltimore, 1977). The interference attributed to DIs is thought to result from competition with the helper virus for factors required in trans …

Infection Rates In Glossina Morsitans Morsitans Fed On Waterbuck And Boran Cattle Infected With Trypanosoma Congolense, Steve Mihok, Reardon O. Olubayo, David F. Wesonga

All Peer-Reviewed Publications

Teneral Glossina morsitans morsitans were fed on waterbuck (Kobus defassa) and Boran cattle (Bos indicus) infected experimentally with Trypanosoma congolense clone IL2895. Infection rates in tsetse varied from 9 to 31% when fed on cattle, and from 2 to 59% when fed on waterbuck. In waterbuck, infections were often detected through the development of parasites in tsetse at times when parasitaemia could not be detected through microscopic examination of blood. Male and female, and 1- and 2-day-old flies were equally susceptible to infection on both hosts. Infection in tsetse was associated with a 14% absolute reduction in survival during the …
