The Prairie Naturalist
Lead is toxic to all vertebrate species and ingestion of spent lead pellets from hunting is the primary method of exposure in birds. Both acute and chronic effects occur in response to lead ingestion including death, weight loss, and reduced body function, but the effect is highly variable among species. Most research has focused on lead ingestion impacts on waterfowl, but less is known about the effects of lead ingestion by upland game such as ring-necked pheasants (Phasianus colchicus). We gavage- fed zero, one, or three lead pellets to 90 (n = 30/group) captive-raised adult hen pheasants and monitored survival …

Portions of the United States are becoming increasingly dominated by annual species, with cheatgrass (Bromus tectorum L.) and medusahead (Taeniatherum caput-medusae [L.] Nevski) as the most common and well-known invaders (Bansal et al. 2014). Other invasive annual species also are present and increasing in abundance, including ventenata (Ventenata dubia [Leers] Coss.), an invasive annual grass that has been expanding within the Pacific Northwest, Great Basin, and Great Plains regions of the United States. Adult Red-Headed Woodpecker Interaction With Bullsnake After Arboreal Nest Depredation, Brittney J. Yohannes, James L. Howitz

Nest success rates often are higher among cavity-nesting birds than those that nest in open cups or on the ground. Among cavity-nesting birds, woodpeckers have some of the highest rates of nest success. A review of woodpecker nesting ecology across species documented nest success ranging from 0.42 to 1.00 with a median of 0.80 (n = 84 populations), and that predation was low, ranging from 0.00 to 0.35 with a median of 0.13 (n = 33 populations). The constrained opening to a cavity nest limits predation to relatively small animals or those able to break open cavity walls. …

Animal movement patterns are variable, with certain species primarily being diurnal and others nocturnal. Pronghorn (Antilocapra americana) are regarded as diurnal animals moving predominately during daylight hours. Anecdotal accounts, however, suggest that pronghorn move during the night but the extent, frequency, and importance of these nocturnal movement behaviors are unknown. To evaluate movements, we combined global positioning system relocation data from collared female pronghorn in the Northern Sagebrush Steppe between 2003 and 2007 with sunrise/sunset data within a geographical information system platform. The use of safe and humane methods for the capture of wild animals is imperative in wildlife research, wildlife damage management, and feral animal control. When capturing animals successfully and humanely, several items must be addressed, including animal safety, personnel safety, non-target captures, and cost (Sikes et al. 2016). Additionally, public pressures against the use of traps have led to significant changes in trapping regulations across North America and beyond (Andelt et al. 1999, Darrow and Shivik 2008) and can place research projects at risk to loss of Institutional Animal Care and Use Committee (IACUC) approval (Larkin et al. 2003). …

Animals are heterotrophic by definition, but species from many taxonomic groups are hosts to epibiota that may alter their net metabolism. We tested the degree to which snail-shell epibiota can generate net ecosystem productivity for snails and their epibiota (snail–epibiota ecosystems; SEEs) after accounting for snail respiration. We focused on 3 species from the Lavigeria snail assemblage in Africa’s Lake Tanganyika and quantified the scaling of SEE metabolism with shell size under light and dark conditions. The metabolism of snails and their epibiota shifted significantly across the size gradient. SEEs of large snails (>20 mm) were consistently autotrophic during …

Giant Canada geese (Branta canadensis maxima) were extirpated from Iowa by the early 1900s due to unregulated hunting, egg gathering, and wetland drainage in the nineteenth century (Bishop 1978). Ef- forts to reintroduce Canada geese in Iowa began in 1964 (Bishop and Howing 1972) and involved releasing flightless adults and goslings at nearly 30 sites across the state (Zenner and LaGrange 1998a). In 1972, 13 flightless pairs were released at Rice Lake Wildlife Management Area (WMA; Bishop 1978). Reproductive parameters such as conception date, and therefore parturition date, influence offspring characteristics such as sex ratio and birth mass. For example, late-conceiving reindeer (Rangifer tarandus) produced more female than male offspring compared to those who conceived early (Holand et al. 2006). Females that conceive later and therefore give birth later also can produce lighter offspring compared to those who conceive early (Schwartz et al. 1994, Holand et al. 2006). Because birth mass is positively related to adult body mass (Michel et al. 2015), conception and parturition date likely affect adult phenotype. In turn, this may influence survival because heavy …

The trumpeter swan (Cygnus buccinator) is North America’s largest waterfowl species and breeds in wetland habitats across the northern United States, Alaska and Canada. Summer observations by ornithologists, including those of John James Audubon in 1843, indicated a substantial number of trumpeter swans were breeding in North Dakota prior to European settlement. However, the species was extirpated by the late 1800s and there were no subsequent breeding records for over a century (Stewart 1975). Since 2007, 5 recent nesting records indicate that trumpeter swan has returned as a regular breeding species in North Dakota.

Laminin-α2 related Congenital Muscular Dystrophy (LAMA2-CMD) is a progressive muscle disease caused by partial or complete deficiency of laminin-211, a skeletal muscle extracellular matrix protein. In the last decade, basic science research has queried underlying disease mechanisms in existing LAMA2-CMD murine models and identified possible clinical targets and pharmacological interventions. Experimental rigor in preclinical studies is critical to efficiently and accurately quantify both negative and positive results, degree of efficiency of potential therapeutics and determine whether to move a compound forward for additional preclinical testing. Cancer-based immunotherapy has led the evolution of biologics that can stimulate immune responses towards tumor eradication. The synthesis of small to intermediate size molecules with the targeting and effector functions of mAb may represent a novel class of immunotherapeutics that may overcome the limitations of their biological counterparts.Towards this objective, B7H6 has been identified as a protein ligand localized on the cell surface of transformed tumor cells. B7H6 binds specifically to the activating receptor NKp30, constitutively expressed on all resting and active NK cells. The Dahlquist Lab investigates the global, transcriptional response of Sacchromyces cerevisiae, baker’s yeast, to the environmental stress of cold shock, using DNA microarrays for the wild type strain and strains deleted for a particular regulatory transcription factor. Gene regulatory networks (GRNs) consist of transcription factors (TF), genes, and the regulatory connections between them that control the resulting mRNA and protein expression levels. We use mathematical modeling to determine the dynamics of the GRN controlling the cold shock response to determine the relative influence of each transcription factor in the network. How primordial metabolic networks such as the reverse tricarboxylic acid (rTCA) cycle and clay mineral catalysts coevolved remains a mystery in the puzzle to understand the origin of life. While prebiotic reactions from the rTCA cycle were accomplished via photochemistry on semiconductor minerals, the synthesis of clays was demonstrated at low temperature and ambient pressure catalyzed by oxalate. Herein, the crystallization of clay minerals is catalyzed by succinate, an example of a photoproduced intermediate from central metabolism. The experiments connect the synthesis of sauconite, a model for clay minerals, to prebiotic photochemistry. We report the temperature, pH, and concentration dependence …

Phytoplankton blooms follow predictable annual cycles in the Gulf of Maine, characterized by a large spring bloom and a smaller bloom in the fall. Marine phytoplankton form the foundation of primary production in Gulf of Maine waters, and thus community changes in composition, and abundance could have cascading effects on our coastal ecosystems.

While BRAF is among the most well-established oncogenes in human cancers, more recently it has garnered attention for its role in suppressing antitumor immunity, especially in melanoma. Because tumor-infiltrating lymphocyte (TIL) density is strongly prognostic in colorectal cancer (CRC)7, we decided to investigate the connection between TIL density and the BRAF-activating V600E mutation in CRC.
We used ESTIMATE to quantify immune infiltrate in samples from the TCGA colon adenocarcinoma (COAD) dataset (n = 216). This is an algorithm that uses the gene-expression signature of 141 immune-related genes to infer the presence of immune cells in the tumor infiltrate. …

Dystrophin deficiency is the genetic basis for Duchenne muscular dystrophy (DMD), but the cellular basis of progressive myofiber death in DMD is not fully understood. Using two dystrophin-deficient mdx mouse models, we find that the mitochondrial dysfunction is among the earliest cellular deficits of mdx muscles. Mitochondria in dystrophic myofibers also respond poorly to sarcolemmal injury. These mitochondrial deficits reduce the ability of dystrophic muscle cell membranes to repair and are associated with a compensatory increase in dysferlin-mediated membrane repair proteins. Dysferlin deficit in mdx mice further compromises myofiber cell membrane repair and enhances the muscle pathology at an asymptomatic …

According to Just, various forms of specific biology (including especially cell biology) plus “general biology” are necessary for explaining adequately the origin of ethical behaviors. Social ethical behaviors, especially mutual aid and cooperative interactions with others and the environment, are essential to evolutionary advances among living creatures, ranging from humans to cells. Accordingly, theory of ethics (moral theory) should have roots in biology.

An Amphipathic Trans-Acting Phosphorothioate Rna Element Delivers An Uncharged Phosphorodiamidate Morpholino Sequence In Mdx Mouse Myotube, H. Jain, J. Boehler, D. Verthelyi, Kanneboyina Nagaraju, S. Beaucage

An efficient method for the delivery of uncharged polyA-tailed phosphorodiamidate morpholino sequences (PMO) in mammalian cells consists of employing a synthetic 8-mer amphipathic trans-acting poly-2′-O-methyluridylic thiophosphate triester element (2′-OMeUtaPS) as a transfection reagent. Unlike the dTtaPS DNA-based element, this RNA element is potent at delivering polyA-tailed PMO sequences to HeLa pLuc 705 cells or to myotube muscle cells. However, much like dTtaPS, the 2′-OMeUtaPS-mediated internalization of PMO sequences occurs through an energy-dependent mechanism; macropinocytosis appears to be the predominant endocytic pathway used for cellular uptake. Ataxia-telangiectasia (A-T) is a rare genetic disorder caused by loss of function of the ataxia-telangiectasia-mutated kinase and is characterized by a predisposition to cancer, pulmonary disease, immune deficiency and progressive degeneration of the cerebellum. As animal models do not faithfully recapitulate the neurological aspects, it remains unclear whether cerebellar degeneration is a neurodevelopmental or neurodegenerative phenotype. Epigenetics remains an exciting subdiscipline of biology, generating discoveries and insights about development. Because epigenetic phenomena can draw attention to the dynamic, interactional, and probabilistic nature of phenotype development, epigenetics research could hasten the demise of both nature-nurture debates and reductionist, genetically determinist perspectives on phenotype development. However, new data alone will not inevitably transform conceptualizations of phenotype origins, because it remains possible to assimilate epigenetic phenomena into traditional conceptual frameworks; epigenetic discoveries could even strengthen biologically determinist conclusions if traditional conceptualizations are retained. Although epigenetics will not force conceptual transformation, epigenetics research encourages the dismissal of the nature-nurture dichotomy …

Objective
To identify changes in skeletal muscle microRNA expression after endurance exercise and associate the identified microRNAs with mRNA and protein expression to disease-specific pathways in polymyositis (PM) and dermatomyositis (DM) patients.
Methods
Following a parallel clinical trial design, patients with probable PM or DM, exercising less than once a week, and on stable medication for at least one month were randomized into two groups at Karolinska University Hospital: a 12-week endurance exercise group (n = 12) or a non-exercised control group (n = 11). The loss and recovery of muscle mass and function following injury and during rehabilitation varies among individuals. While recent expression profiling studies have illustrated transcriptomic responses to muscle disuse and remodeling, how these changes contribute to the physiological responses are not clear. In this study, we quantified the effects of immobilization and subsequent rehabilitation training on muscle size and identified molecular pathways associated with muscle responsiveness in an orthopaedic patient cohort study. Years of research in software engineering have given us novel ways to reason about, test, and predict the behavior of complex software systems that contain hundreds of thousands of lines of code. Many of these techniques have been inspired by nature such as genetic algorithms, swarm intelligence, and ant colony optimization. In this paper we reverse the direction and present BioSIMP, a process that models and predicts the behavior of biological organisms to aid in the emerging field of systems biology. Calyptrate flies include about 22,000 extant species currently classified into Hippoboscoidea (tsetse, louse, and bat flies), the muscoid grade (house flies and relatives) and the Oestroidea (blow flies, bot flies, flesh flies, and relatives). Calyptrates are abundant in nearly all terrestrial ecosystems, often playing key roles as decomposers, parasites, parasitoids, vectors of pathogens, and pollinators. For oestroids, the most diverse group within calyptrates, definitive fossils have been lacking. The first unambiguous fossil of Oestroidea is described based on a specimen discovered in amber from the Dominican Republic. The specimen was identified through digital dissection by CT scans, which provided morphological …
