Exposure to cannabinoids during critical development periods has increased with epileptic children being commonly prescribed CBD for seizures and pregnant women taking it recreationally. Many studies have been done on the possible benefits and drawbacks of cannabinoid exposure on the human brain, but not much is known about how it can affect the developing brain long-term. Based on current guidelines for housing rodents after surgical instrumentation, rodents may be housed at temperatures that hinder surgical recovery and cause thermal stress– room temperature (21°C) being one of them. Rodents are often housed at room temperature since this temperature is easy for humans to be caring for them. However, if recovering rodents are thermally stressed, experimental results will be confounded. This paper focuses on the ability of the mimic octopus Thaumoctopus mimicus to be operantly conditioned to an auditory stimulus. The octopus is known to be the most advanced of the invertebrates and has learning abilities that are comparable to vertebrates in spite of their differences in brain structure. These animals have been shown to react to visual and tactile stimuli and can be operantly conditioned to perform behaviors to obtain a food reward. Microglia are the macrophages of the brain and spinal cord, playing an important role in the immune response to disease states of the nervous system. This study conducts an investigation on the activity of microglia in response to low grade neuroepithelial tumors. Using mouse models and microglial markers, a qualitative and quantitative analysis of microglia activation, migration, and invasion within the brain cortex during early stages of tumor development was conducted. C1q TNF Related Protein 3 (CTRP3), is a cytokine that is primarily secreted from adipose tissue, which classifies it as an adipokine. Our previous research has shown that CTRP3 prevents alcoholic fatty liver disease (ALD) in male mice. However, even when accounting for confounding factors such as absolute and relative alcohol intake, females are more sensitive to the effects of consumption compared to male mice. Forage-based ruminant diets supplemented with condensed tannins or polyphenolic-containing legumes may alter nitrogen (N) metabolism in the animal and reduce gas emissions from soil receiving excreta. The objective of the study was to determine if soil receiving urine or manure from sheep fed forage diets supplemented with condensed tannin or polyphenolic-containing legumes would decrease N gas emissions. Acute myeloid leukemia (AML) is an often devastating hematologic malignancy with 5-year overall survival lingering near 20%. Acquiring a deeper understanding of molecular underpinnings of leukemogenesis will provide a basis for developing more effective therapeutic strategies for patients with AML.
Here, we identified overexpression of hnRNP K as a recurrent abnormality in a subset (~20%) of AML patients. High levels of this RNA-binding protein associated with inferior clinical outcomes in de novo AML. Thus, to evaluate its putative oncogenic capacity in myeloid disease, we overexpressed hnRNP K in murine hematopoietic stem and progenitor cells isolated from fetal liver cells (FLCs). …

The modern broiler is growing at a rapid rate generating tremendous amounts of heat. A sensitive Net Energy (NE) system is needed to measure body heat production (HP) generated primarily by daily maintenance and synthesis and degradation of myofibrillar and sarcoplasmic protein. The first two chapters present evaluation of the classic way to calculate NE versus a new methodology; the Arkansas NE (Ark NE) system, with birds from two genetic lines fed diets with different AA content or different ME content in two different environmental temperatures.

The hypothalamo-pituitary-adrenal (HPA) axis is the regulatory system for the neuroendocrine stress response within vertebrates. Within the HPA axis corticotropin releasing hormone (CRH) is a major regulator and driving hormone. A structure named the nucleus of the hippocampal commissure (NHpC) has been found to contain CRH neurons and also these neurons respond to early food deprivation stress significantly prior to the paraventricular nucleus (PVN), the major driving nucleus of the classic neuroendocrine HPA axis. Necrotic enteritis (NE) has re-emerged following restriction of antimicrobial usage and costs $6 billion every year worldwide. The primary objective of the studies was to evaluate prevention and treatment of NE using a microbiota metabolic product, secondary bile acid deoxycholic acid (DCA), in drinking water. Woody Breast (WB) myopathy causes significant economic losses to the global poultry industry and the lack of objective, non-invasive and rapid methods for its detection as well as profitable alternatives for its utilization are contributing factors. The first area aimed to determine if there are carcass conformation changes that can be used to identify WB characteristics in commercial broilers using image analysis. Internal parasitism inevitability prompts economic loss in beef cattle production by decreasing growth performance and reproductive traits. Today, the most widely used class of anthelmintic used to treat parasitism, is the macrocyclic lactone. Many studies have conflicting results on the efficacy of macrocyclic lactones (ML) efficacy against internal parasitism. Reproductive performance is crucial for sustained financial success in the beef cattle industry. This dissertation includes a population study that quantified the incidence of Neospora caninum infections in the central region of the United States and tested its relationship with reproductive performance in beef cattle. Trial one of that study concluded that 6.9% of open, replacement heifers (n = 1306) tested seropositive. The second trial in that project found that 9.6% of the breeding age females (n = 500) tested were seropositive for Neospora caninum; and that state in which the cattle lived and age impacted (P < 0.05) infection rate. Breed composition, number of farm dogs on the ranch, and use of total mixed rations were not associated (P > 0.1) with seropositive …

Cattle grazing wheat pasture have the potential to gain BW exceptionally well, but excessive nitrogen intake results in increased excretion and increased greenhouse gas (GHG) emissions. Supplemental concentrates with the addition of an ionophore given to ruminants grazing wheat is a potential practice for producers to increase nitrogen efficiency while decreasing GHG emissions. Therefore, the objective of this experiment was to quantify the effects of energy (2.95 kg/d) and lasalocid (200 mg/hd/d) supplementation on nutrient intake, energy metabolism, respiratory gas fluxes, and performance of grazing cattle. The developing brain is susceptible to extensive neurotoxicity following exposure to sedative/anesthetic drugs (SADs). Every year hundreds of thousands of children around the world are exposed to SADs with no viable non-neurotoxic agents approved for clinical use. Allopregnanolone (AlloP) has well-established sedative effects in adults and neonates. AlloP and many SADs produce sedation/anesthesia through allosteric modulation of GABAA receptors, which is one of two principal mechanisms behind SAD-induced neurotoxicity. Mist netting is the most popular field technique used to estimate avian species abundance. It can, however, provide a biased estimate of species abundance as not all birds are equally likely to be captured. This is partially due to net avoidance, the purposeful evasion of mist nets by birds. Most benthic organisms living in the intertidal zone have planktonic larvae that reside temporarily in the water column before settling in their adult habitats. Larvae aggregate in offshore larval pools, and transport horizontally and vertically in the water to remain in the nearshore and during their pelagic life. While some horizontal transport of larvae can be attributed to advection, behavioral responses, like vertical swimming and buoyancy control, allow larvae to position themselves at depths where flow direction can be exploited. Gout is a chronic arthritis caused by the deposition of poorly soluble monosodium urate monohydrate (MSU) crystals in peripheral joints. Resident macrophages initiate inflammation in response to MSU mediated by NF-κB nuclear translocation and NLRP3 inflammasome activation. We investigated the role of CD44, a transmembrane receptor, in mediating MSU phagocytosis by macrophages. We used an antibody that sheds the extracellular domain (ECD) of CD44 to study the role of the receptor and its associated protein phosphatase 2A (PP2A) in macrophage activation. This investigation will collect data to assist in determining if elevated aquatic phosphate levels affects the metamorphosis rate of larval western barred tiger salamanders (Ambystoma mavortium). Monoammonium phosphate fertilizers are being used on crops in Nebraska (NDA, 2017). This area lines up with the area the western barred tiger salamanders are disappearing from (Damme, 2018). Monoammonium phosphate is made up of nitrogen and phosphate. Repeat-associated non-AUG-initiated translation of expanded CGG repeats (CGG RAN) from the FMR1 5′-leader produces toxic proteins that contribute to neurodegeneration in fragile X-associated tremor/ataxia syndrome. Here we describe how unexpanded CGG repeats and their translation play conserved roles in regulating fragile X protein (FMRP) synthesis. In neurons, CGG RAN acts as an inhibitory upstream open reading frame to suppress basal FMRP production. Activation of mGluR5 receptors enhances FMRP synthesis. This enhancement requires both the CGG repeat and CGG RAN initiation sites. Using non-cleaving antisense oligonucleotides (ASOs), we selectively blocked CGG RAN. Many types of neurons act as multimodal signaling centers. Yet, we have only limited insight into the regulation and functional consequences of neuronal co-transmission. For example, dopamine (DA) neurons, whose degeneration causes motor deficits characteristic of Parkinson’s Diseases (PD), communicate with all their targets by DA but only a selective subset of their targets using GABA, Glutamate, and the secreted cell signaling protein Sonic Hedgehog (Shh). In a cellular model, we demonstrate that the non-receptor protein tyrosine kinase 2 beta (PTK2B) plays a critical role in mouse cultured beige adipocyte differentiation. CRISPR/Cas9-mediated knock-out of Ptk2b results in non-differentiating white adipocytes and differentiated beige adipocytes with significantly reduced thermogenic gene and protein expression, enlarged lipid droplet size, and altered mitochondrial respiration. Together, these data in a cell culture system provide evidence for a role of PTK2B in the differentiation of mouse beige adipocytes.

Drug-resistant epilepsy (DRE) is a common clinical sequela of developmental cortical malformations such as polymicrogyria. Unfortunately, much remains unknown about the aberrant GABA-mediated circuit alterations that underlie DRE's onset and persistence in this context. To address this knowledge gap, we utilized the transcranial freeze lesion model in optogenetic mice lines (Somatostatin (SST)-Cre or Parvalbumin (PV)-Cre x floxed channelrhodopsin-2) to dissect features of the SST, PV, and pyramidal neuron microcircuit that are potentially associated with DRE. Objective: Novel drug 419 was examined to see the effect it has in vivo mice and rats on alcohol consumption, nicotine locomotor sensitization, and conditioned place preference (CPP) models regarding behavioral tests on dopamine transporter activity.
Methods: Mice and rats were used to see how they react to the drug 419 and control vehicle, in each of the models. The animals were assessed to pre- and post- drug administration of novel drug 419. We examined each model to see the association between how drug 419 will help with treating drug abuse.

Traumatic Brain Injury (TBI) is defined as an impact to the head, penetration of the skull, or rapid deceleration of the skull, resulting in an alteration of brain function or neurological deficit. Cognitive deficits are common following TBI and often go unresolved due to a lack of effective treatments. These deficits often perseverate into the chronic post injury phase, so the development of rehabilitative strategies is imperative. Glaucoma is a build-up of pressure in the eye that leads to the gradual loss of sight. The intention of this study was to investigate how glaucoma affects intrinsically photosensitive Retinal Ganglion Cells (ipRGCs). These cells contain melanopsin, a photopigment, that enables the cells to react to light. Four-month-old control and five-month-old glaucoma mice retinas were dissected, stained using primary and secondary antibodies, then traced using ImageJ’s Simple Neurite Tracer. There were no significant differences in soma size, total dendritic length, dendritic field size, and dendritic field diameter between control and glaucoma ipRGCs. The commercial production of pasture and free-range eggs in the United States is expanding rapidly. According to the United States Department of Agriculture Agricultural Management Assistance, in May of 2019 there were 2.6M pasture hens and 4.5M free-range hens representing 1.9% of the nation’s laying flock. There is little industry and/or academic experience with this style of production in the United States. Programs such as the European Layer Training Initiative (ELTI), which emerged in 2019, have tried to fill this void but participation from the US was low.
