The purpose of this study was to evaluate the effects of the resorbable membrane Guidor™ and the osteogenic protein hOP-1 on hard tissue healing following periradicular surgery in cats.

Surgical endodontic therapy was performed on the maxillary canine teeth of twelve cats. After orthograde root canal therapy, flap reflection, osteotomy, resection of the root end, root end cavity preparation and the placement of a root end filling, eight of the surgical sites were treated with Guidor™ and eight with hOP-1. The remaining eight sites received no further treatment and served as controls.

For over thirty years now, research has been carried out to isolate and purify bone morphogenetic protein (BMP), a substance which has been shown to induce heterotopic bone formation in various animal species. Recent advances in the fields of developmental biology, molecular biology and genetics, have shown that the BMPs not only are responsible for postfetal bone induction (including normal bone remodeling, healing and repair), but are also critical during embryogenesis - not only in regards to the skeletal system, but quite possibly in the morphogenesis and pattern formation of other tissues and organs as well.

Periodontal regeneration includes the formation of both alveolar bone and cementum. Studies reporting histologic evidence of healing following periodontal reconstructive therapy were reviewed to determine whether regeneration of these structures improves with increasing healing interval, and whether they regenerate independently or whether regeneration of one structure is dependent upon regeneration of the other. Additionally, the review addresses whether specific reconstructive modalities predictably yield increased amounts of regeneration. Differences in regenerative potential between humans, nonhuman primates, and canines were also examined. This study evaluated candidate carriers for recombinant human bone morphogenic protein-2 (rhBMP-2) in periodontal reconstructive surgery.

Canine demineralized bone matrix (DBM), bovine inorganic crystallene bone matrix (Bio-Oss), bovine derived microfibrillar collagen matrix (Helistat), poly (D,L-lactide-coglycolide) micropaiticles (PLGA), and polylactic acid granules (Drilac) were used with rhBMP-2 (20 (µg/100 µl implant volume) in routine critical size canine supraalveolar periodontal defects. Contralateral defects in six beagle dogs were randomly assigned to receive: DBM/rhBMP-2, DBM-control, Bio-Oss/rhBMP-2, Helistat/rhBMP-2, PLGA/rhBMP-2, or Drilac/rhBMP-2, all with autologous blood except for Helistat and PLGA. The aim of this investigation was to characterize peri-implant tissue breakdown following ligature-induced plaque accumulation around osseointegrated implants in the rhesus monkey. Two hydroxyapatite-coated cylindrical implants were inserted bilaterally in the second premolar and first molar region of both mandible and maxilla in four non-human primates (Macaca mulatto). The implants were left submerged to osseointegrate for one year. Eight weeks after healing abutment connection, a two-month plaque control program was initiated. Maxillary sinus osteotomy is sometimes indicated for transantral surgical approaches to palatal roots of maxillary molars. A search of the literature showed an absence of any report on the healing potential of such defects. The purpose of this study was to determine histologically the reparative process of the nasal sinus of dogs following small and large surgical antral perforations. Blood flow to the maxilla and mandible may vary due to differences in vascular supply and in physiologic, metabolic and pathologic conditions. In general it is accepted that oral tissues have an abundant blood supply; however, there is little information quantifying blood flow to these regions. In this study, the maxillary tissues were chosen to measure the blood flow in attached and reflected gingival tissue in adult cats using the radiolabeled microsphere method.

Animal models are frequently consulted for histometric analysis of reconstructive periodontal therapy. Statistical analysis of induced lesion defects and surgically created defects from our laboratory suggest defect size homogeneity is critical for the model to discriminate treatment effects. This study characterizes natural disease defects in beagle dogs. A study model has been designed to evaluate the effectiveness of a new fenestration corticotomy procedure in two non-human primates Macaca fasicularis.

The objective of this study was to evaluate the efficacy of a new conservative corticotomy procedure to facilitate dental translation using fenestrations. The pathogenesis of periapical lesions is the interaction of the oral flora with the existing host defenses. The role of bacteria has been established in endodontic disease but the contribution of the host defenses in periapical lesion formation is less clear. The purpose of this study was to compare periapical lesion development in immunosuppressed rats with that in normal rats. Thirty-five, eight-week old Sprague-Dawley rats were equally divided into seven groups. Groups 1, 2, and 3 were given weekly injections of Cytoxan, an immunosuppressive agent. Recently we demonstrated that human dentin contained relatively high amounts of transforming growth factor beta (TGF-ß) and proposed that dentin TGF-ß originated from odontoblasts. Because of the suggested role of TGF-ß in mediating cellular interactions in dental cell development by paracrine mechanisms, we studied the expression of TGF-ß message in developing teeth by in situ hybridization. 5 µm serial sections of neonatal sheep mandibular third molars that had been fixed in glutaraldehyde and paraffin-embedded were used for in situ analysis. Two new techniques for quantification of forces generated by an expansion screw apparatus placed between pairs of cylindrical endosseous titanium implants are described, along with preliminary data gained from their use in Macaca rhesus monkeys. Strain gages were bonded to the expansion apparatus and were calibrated either directly by micro load cell, or indirectly by Instron Machine to reflect the applied force.

The pulps of mandibular molars of 15 athymic and 15 conventional rats were surgically exposed and left open to their oral flora. Each group was divided into 3 subgroups of 5 animals each. The rats were sacrificed after their pulps were exposed for 2, 4, or 8 weeks. After fixing, decalcifying, and embedding, the specimens were sectioned and stained with H and E. They were then examined under a microscopic grid and quantified by percentages of surface areas of bone, connective tissue, bone marrow, intrabony spaces, periapical lesions, and numbers of osteoclasts. Previous studies indicate that root resorption is a frequent sequela to significant amounts of periodontal repair following experimental regenerative periodontal surgical procedures in animal models. To study if root resorption can be reduced or prevented, periodontal repair was evaluated in large horizontal circumferential defects in the dog in the absence of influence from the oral environment. This study was undertaken to test the hypothesis that a corticotomised tooth moves as a tooth-bone unit when under orthodontic force. A non-human primate model was designed to test the hypothesis. The non-human primate selected was Macaca fasicularis. Three adult males with erupted third molars were chosen so that extrapolation could be made to the adult human. Polyoxyinethylene (Delrin) is a polyacetyl homopolymer which has been used successfully in many orthopedic joint replacements. To determine histologic host response to Delrin as a temporomandibular joint condylar replacement condyles were removed from ten fully mature macaca fasicularis monkeys. The condyles were replaced with a total prosthetic Delrin condyle attached to a titanium mesh. Specimens obtained at one, two, and six months indicated total acceptance of the Delrin, remodeling of the glenoid fossa and formation of bone between the middle cranial fossa and the roof of the glenoid fossa. The purpose of this study was to investigate the effects of mandibular expansion by orthopaedic and surgical procedures in the growing rat mandible.

Sixty-four 35 day old Sprague Dawley Harlan rats were divided into four groups of 16 rats each and categorized as the control, orthopaedic expansion, surgery without graft and surgery with graft respectively. Prefabricated appliances were used for orthopaedic expansion of the mandibles and maintaining surgical expansion at the symphysis in the respective groups. Expansion of the maxilla at the midpalatal suture has had much investigation and is a recognized option in orthodontic therapy. Lateral expansions of the mandible through surgical separation at the symphysis have been done in animal studies. A combination of these procedures has not yet been done together.

The purpose of this study was to expand both the maxilla and the mandible of a baboon simultaneously while maintaining occlusion. The amount of expansion and stability was studied using photographs, models, cephalograms, and tetracycline bone labels using U.V. microscopy.

Mechanical expansion of the maxilla, at the midline suture, is a widely accepted orthodontic procedure used to gain arch width in certain types of maxillary deficient arches.

In animal research on the mandible, techniques using surgical separation at the symphysis with immediate expansion up to 12 mm, fixation with a metal splint, and autogenous bone transplanted into the expanded area, have been reported to be successful.

For sometime, expansion of the maxillary arch, by means of surgical and mechanical procedures, have been acknowledged with minimal attention directed toward the mandible. In this pilot study a surgical technique was employed in separating the mandible, A-P, at the symphysis. The interosseous void, approximately 10 mm., was then filled with autogenous bone marrow collected from the greater trochanter and the crest of the ilium.

Preparations used in this study were obtained from an investigation which had undertaken the following:

Durapatite, a hydroxylapatite ceramic was implanted in 24 mature Beagle dogs to study its maintenance effect on the alveolar bone of the mandible.

After dividing the dogs into two groups, a fresh socket and a healed socket group, the posterior teeth were removed and alveolectomies were performed. Eight ceramic implants were then placed in the right mandible of each animal in each group. The healed group received the implants five weeks after tooth removal and the fresh socket group received implants immediately after extraction.

The periapical tissue reaction to three concentrations of endotoxin solutions and to a sterile saline control solution was examined in six adult cats. The maxillary and mandibular cuspids were isolated and the pulps extirpated with barbed broaches. The four solutions (1, 10, 100 μg/ml endotoxin, and saline) were then injected into the four prepared teeth for each cat and the access cavities sealed with IRM.

It has been shown previously that a root surface which has been scaled, root planed and demineralized by topical application of citric acid in vitro, is dominated by exposed collagen fibrils. The purpose of this study was to examine the mechanism of healing and possible reattachment of soft periodontal connective tissue to the root planed and subsequently acid treated root surfaces and to determine whether new collagen fibrils produced in the connective tissue could attach to these exposed fibrils.

For the past decade, evidence has accumulated which suggests that the resistance of the teeth to dental decay is in part under systemic control. It was shown that the resistance is accomplished through the optimal operation of a dentinal fluid transport mechanism which is believed to be hormonally controlled by the hypothalamic-parotid gland endocrine axis. Although circumstantial evidence for such an endocrine function was strong, the isolation of a parotid hormone which stimulates the dentinal fluid transport mechanism remained to be accomplished.

Endodontic therapy was evaluated as a possible control or prevention modality of radiation type dental caries in dogs. Two adult male dogs with permanent dentitions had endodontic therapy performed on alternating teeth in each jaw. The nontreated teeth served as controls.

The dogs were irradiated with 7000 rad Cobalt-60 radiation fractionated over an eight week period. The dogs were killed at 60 and 182 days post-irradiation. The teeth were removed from the jaws and immersed in methylene blue dye, sectioned, and the dye penetration pattern photographed. Diffusion of the dye into the tooth structure was quantified and statistically analyzed.
