Current as well as past studies have provided evidence for a connection between macroeconomic/agricultural and cellular/biochemical basis of feed efficiency (FE; gain:feed) in farm animal production. Our investigation of respiratory chain activity, mitochondrial protein oxidation and protein expression was an expansion to the subcellular/molecular level. In this study we investigated the entire complex (Cx) of the electron transport chain (ETC) for differences in Cx activities in mitochondria isolated from skeletal muscle in high (HFE) and low (LFE) feed efficient swine within the same genetic line. Skeletal muscle atrophy is a hallmark of many pathologies and is associated with disease prognosis, quality of life, and mortality. Specifically, lack of contractile activity of the muscle can result in disuse-associated atrophy. Disuse-induced atrophy is a common phenomenon in intensive care unit (ICU) patients and significantly impacts patient prognosis and ability to transition of out of intensive care. Poultry nutritionists continue to identify methods to lower feed price without compromising broiler growth performance, processing yields, or animal well-being. A common approach is to reduce dietary crude protein (CP), the second most expensive component in a broiler diet. Subsequently, reducing CP will reduce the inclusion levels of intact protein sources (e.g., soybean meal [SBM]), which can be accomplished by supplementing individual amino acids (AA). This methodology not only reduces diet cost, pending if feed-grade AA are more economical, but can contribute to improving broiler health and environmental sustainability. The inherent variability and lack of standardization of in vivo assays to determine the metabolizable energy (ME) of DDGS has led to inconsistent values and limited its potential to use in broiler diets. Therefore, two experiments were conducted to characterize the content of nitrogen-corrected ME (MEn) in 11 DDGS sources obtained from various ethanol plants using a regression-based broiler assay and to attempt to develop equations to predict MEn based on its chemical composition. Over the last 60 plus years, broiler breeders have been under an incredible amount of genetic pressure. This genetic pressure comes from the continually increasing consumer demand for poultry products. The objective of this dissertation is to understand the effects of different nutritional and management practices on broiler breeder’s performance. The first series of experiments investigate the impact of broilerization on hen and early progeny performance. Broilerization is a process only used by genetic companies. With close to 5,000 captive tigers in the United States, it is vital to ensure that these tigers are receiving adequate welfare whether they are in an accredited facility, or privately owned. The goal of this study was to assess whether captive tigers at a rescue facility with experience being transported outside of the facility would respond differently to the presentation of their transport vehicle than their naïve counterparts who had only been transported within the facility. For studies 1 through 3, 20 steers of predominantly Angus breeding, were stratified by body weight (279 ± 13.2 kg), assigned randomly to 6 paddocks, and fed a low S ground corn and soybean meal supplement that did not contain any byproduct feeds (0.31% total dietary S; LS) or LS supplement with an additional 0.25% S provided from sodium sulfate (Na2SO4; 0.58% total dietary S; HS) for a 114-d growing phase. "Alzheimer’s disease (AD) is the sixth leading cause of death in the United States with approximately 5.8 million Americans currently living with AD. Due to the lack of a disease modifying treatment for AD and the aging baby boomer generation, this number is projected to grow to 13.8 million by 2050 (Gaugler et al., 2019). Primary cilia are shown to have membrane swelling, also known as ciliary bulbs. However, the role of these structures and their physiological relevance remains unknown. Here, it is reported that a ciliary bulb has extracellular vesicle (EV)‐like characteristics. The ciliary extracellular‐like vesicle (cELV) has a unique dynamic movement and can be released by mechanical fluid force. To better identify the cELV, differential multidimensional proteomic analyses are performed on the cELV. Objectives:
Sepsis is a leading cause of death in the United States. Putative targets to prevent systemic inflammatory response syndrome include antagonism of toll-like receptors 2 and 4 and CD44 receptors in vascular endothelial cells. Proteoglycan-4 is a mucinous glycoprotein that interacts with CD44 and toll-like receptor 4 resulting in a blockade of the NOD-like receptor pyrin domain-containing-3 pathway. We hypothesized that endothelial cells induced into a sepsis phenotype would have less interleukin-6 expression after recombinant human proteoglycan 4 treatment in vitro.

Background: With a recent meta-analysis indicating that 9.8% of women worldwide consume alcohol during pregnancy, understanding the effects of fetal alcohol spectrum disorder (FASD) is critical for both identifying and treating those afflicted. Traditional preclinical research investigating the behavioral effects of neurodevelopmental disorders, such as FASD, has utilized tasks designed for specific animal models. For this reason, preclinical research into FASD requires a shift into clinically relevant behavioral testing. One such method involves touch-sensitive screens integrated into an operant box to emulate clinical computer-based tasks, such as the Cambridge Neuropsychological Test Automated Battery.

Babesia microti is an apicomplexan parasite transmitted by tick bite that infects red blood cells. It is increasingly associated with transfusion-transmitted infections including fatal infection as the parasite survives blood banking procedures. The infection can range from asymptomatic to fatal. The parasite can persist in relatively asymptomatic individuals for up to two years. The host and parasite determinants of severity of infection and/or parasite clearance remain largely unknown. Numerous studies have examined relationships between adrenal glucocorticoids and the immune system’s ability to fight disease. The relationship can vary, with glucocorticoids having either stimulatory or inhibitory effects. The pathogenic bacterium Mycoplasma gallisepticum (MG) elicits an adaptive immune response in birds, in which B cells produce MG-specific antibodies to fight the infection and prevent reoccurrence. Previous research found that Eastern Bluebirds that produce the most antibodies in response to MG infection also have the highest glucocorticoid levels. Based on current guidelines for housing rodents after surgical instrumentation, rodents may be housed at temperatures that hinder surgical recovery and cause thermal stress– room temperature (21°C) being one of them. Rodents are often housed at room temperature since this temperature is easy for humans to be caring for them. However, if recovering rodents are thermally stressed, experimental results will be confounded. This paper focuses on the ability of the mimic octopus Thaumoctopus mimicus to be operantly conditioned to an auditory stimulus. The octopus is known to be the most advanced of the invertebrates and has learning abilities that are comparable to vertebrates in spite of their differences in brain structure. These animals have been shown to react to visual and tactile stimuli and can be operantly conditioned to perform behaviors to obtain a food reward. This study characterized the growth of bacteria isolated from ballast tank fluids in hexadecane, petroleum, plant, and algae-derived fuels. The study was performed to explore the capacity of ballast tank isolates to survive and grow within fuels that may be stored within ballast tanks. Results of the hexadecane analysis indicated that most isolates had higher viable cell counts in media supplemented with hexadecane. Members of Alteromonas, Pseudoalteromonas, and a single Brevundimonas species had viable cell counts that were one or several orders ofmagnitude greater than that of controls. Microglia are the macrophages of the brain and spinal cord, playing an important role in the immune response to disease states of the nervous system. This study conducts an investigation on the activity of microglia in response to low grade neuroepithelial tumors. Using mouse models and microglial markers, a qualitative and quantitative analysis of microglia activation, migration, and invasion within the brain cortex during early stages of tumor development was conducted. C1q TNF Related Protein 3 (CTRP3), is a cytokine that is primarily secreted from adipose tissue, which classifies it as an adipokine. Our previous research has shown that CTRP3 prevents alcoholic fatty liver disease (ALD) in male mice. However, even when accounting for confounding factors such as absolute and relative alcohol intake, females are more sensitive to the effects of consumption compared to male mice. Membrane fusion is key to organism homeostasis and occurs when a transport vesicle fuses with a target compartment, merging the two membranes into one while releasing the contents of the transport vesicle into the target compartment. This process is controlled by SNARE proteins. Syntaxin-17 protein plays a crucial role in the fusion of the autophagosome membrane with the lysosome membrane, allowing for degradation of misfolded intracellular components and pathogens. Forage-based ruminant diets supplemented with condensed tannins or polyphenolic-containing legumes may alter nitrogen (N) metabolism in the animal and reduce gas emissions from soil receiving excreta. The objective of the study was to determine if soil receiving urine or manure from sheep fed forage diets supplemented with condensed tannin or polyphenolic-containing legumes would decrease N gas emissions. Acute myeloid leukemia (AML) is an often devastating hematologic malignancy with 5-year overall survival lingering near 20%. Acquiring a deeper understanding of molecular underpinnings of leukemogenesis will provide a basis for developing more effective therapeutic strategies for patients with AML.
Here, we identified overexpression of hnRNP K as a recurrent abnormality in a subset (~20%) of AML patients. High levels of this RNA-binding protein associated with inferior clinical outcomes in de novo AML. A sensitive Net Energy (NE) system is needed to measure body heat production (HP) generated primarily by daily maintenance and synthesis and degradation of myofibrillar and sarcoplasmic protein. The hypothalamo-pituitary-adrenal (HPA) axis is the regulatory system for the neuroendocrine stress response within vertebrates. Within the HPA axis corticotropin releasing hormone (CRH) is a major regulator and driving hormone. A structure named the nucleus of the hippocampal commissure (NHpC) has been found to contain CRH neurons and also these neurons respond to early food deprivation stress significantly prior to the paraventricular nucleus (PVN), the major driving nucleus of the classic neuroendocrine HPA axis. Necrotic enteritis (NE) has re-emerged following restriction of antimicrobial usage and costs $6 billion every year worldwide. The primary objective of the studies was to evaluate prevention and treatment of NE using a microbiota metabolic product, secondary bile acid deoxycholic acid (DCA), in drinking water. Woody Breast (WB) myopathy causes significant economic losses to the global poultry industry and the lack of objective, non-invasive and rapid methods for its detection as well as profitable alternatives for its utilization are contributing factors. Today, the most widely used class of anthelmintic used to treat parasitism, is the macrocyclic lactone. Reproductive performance is crucial for sustained financial success in the beef cattle industry. This dissertation includes a population study that quantified the incidence of Neospora caninum infections in the central region of the United States and tested its relationship with reproductive performance in beef cattle. Trial one of that study concluded that 6.9% of open, replacement heifers (n = 1306) tested seropositive. The second trial in that project found that 9.6% of the breeding age females (n = 500) tested were seropositive for Neospora caninum; and that state in which the cattle lived and age impacted (P < 0.05) infection rate. Cattle grazing wheat pasture have the potential to gain BW exceptionally well, but excessive nitrogen intake results in increased excretion and increased greenhouse gas (GHG) emissions. Supplemental concentrates with the addition of an ionophore given to ruminants grazing wheat is a potential practice for producers to increase nitrogen efficiency while decreasing GHG emissions. Therefore, the objective of this experiment was to quantify the effects of energy (2.95 kg/d) and lasalocid (200 mg/hd/d) supplementation on nutrient intake, energy metabolism, respiratory gas fluxes, and performance of grazing cattle.
