Pain And Distress Associated With Polyclonal Antibody Production: Discussion And Recommendations, The Humane Society Of The United States

ANIMAL RESEARCH

The Humane Society of the United States (HSUS) held a workshop in August 2002 in order to develop recommendations for minimizing pain and distress associated with polyclonal antibody (Pab) production. A small group of experts in the fields of antibody production, animal welfare, in vitro alternatives, and/or regulatory compliance participated in the roundtable discussion. The workshop was a scientifically based meeting, and recommendations were based on the extensive experience of the workshop participants as well as published literature regarding the relevant issues.

Interleukin-10 (IL-10) is a cytokine with anti-inflammatory activities. IL-10 knockout (KO) mice exhibit severe intestinal inflammation in the presence of normal gut flora. We hypothesized that IL-10 KO mice would also be susceptible to inflammatory periodontal disease resulting in alveolar bone loss (ABL). The purpose of this study was to compare naturally occurring ABL in IL-10 KO and wild type (WT) mice. Twelve IL-10 KO and 12 WT mice, age-(7 months) and sex-(male) matched, were used. Pain and distress caused by specific research models and techniques raise serious concerns for those in the animal welfare community as well as in the scientific community. Yet good estimates of how much animal pain and/or animal distress is caused by particular techniques or methods are not yet available. The HSUS has compiled a preliminary list of research models and techniques that cause pain and distress. Analyses by the USDA and HSUS indicate that the majority of the animals reported in Column E are used in various testing procedures, with vaccine testing prominent among them. It is argued that a concept of evaluation of animal models that is broader and more useful than validation is available. Productive generativity refers to the degree to which a model furthers understanding and leads to more-effective treatment interventions. Results of the application of this novel evaluative frame to several animal models of eating disorders show that this animal-based research has not been productive. The question of the relation between clinic and animal laboratory is discussed.

Pain and distress are core issues in the field of animal experimentation and in the controversy that surrounds it. We sought to add to the empirical base of the literature on pain and distress by examining government data on experimental procedures that caused unrelieved pain and distress (UPAD) in animals. Of the species regulated by the U.S. Department of Agriculture (USDA), most of the approximately 100,000 animals subjected to UP AD during the year analyzed (1992) were guinea pigs and hamsters. Since the early 1970s, scientists have used preference tests (tests that require animals to choose between two or more different options or environments) as a means of answering questions about animal welfare. Preference tests have been used to establish animals' preferences for common housing options such as ambient temperature, illumination and preferred types of bedding and flooring; to improve the effectiveness of devices such as loading ramps and nest boxes; and to clarify how strongly animals avoid various aspects of confinement and methods of restraint.

In his challenging article, Steneck (1997) criticized the creation of the Institutional Animal Care and Use Committee (IACUC) system established by the 1985 amendments to the Animal Welfare Act. He saw the IACUC review and approval of biomedical and behavioral research with animals as an unnecessary "reassignment" of duties from existing animal care programs to IACUC committees. We evaluated the relationship, in cynomolgus macaques (Macacafascicularis), between rank for order of blood collection with serum concentrations of 3,5,3'-triiodothyronine (T3), thyroxine (T4), free thyroxine (FT4), and serum cortisol. These relationships were determined for males and females that were housed in two room arrangements. For both room arrangements, males and females were housed separately. For room arrangement 1, macaques were housed on both sides of the animal holding room. The sides of the animal holding room were designated as side A or side B. This study evaluated candidate carriers for recombinant human bone morphogenic protein-2 (rhBMP-2) in periodontal reconstructive surgery.

Canine demineralized bone matrix (DBM), bovine inorganic crystallene bone matrix (Bio-Oss), bovine derived microfibrillar collagen matrix (Helistat), poly (D,L-lactide-coglycolide) micropaiticles (PLGA), and polylactic acid granules (Drilac) were used with rhBMP-2 (20 (µg/100 µl implant volume) in routine critical size canine supraalveolar periodontal defects. Contralateral defects in six beagle dogs were randomly assigned to receive: DBM/rhBMP-2, DBM-control, Bio-Oss/rhBMP-2, Helistat/rhBMP-2, PLGA/rhBMP-2, or Drilac/rhBMP-2, all with autologous blood except for Helistat and PLGA. In general it is accepted that oral tissues have an abundant blood supply; however, there is little information quantifying blood flow to these regions. In this study, the maxillary tissues were chosen to measure the blood flow in attached and reflected gingival tissue in adult cats using the radiolabeled microsphere method.

The objective of this study was to evaluate the efficacy of a new conservative corticotomy procedure to facilitate dental translation using fenestrations. This pilot study compared treatment and control quadrants of each individual animal's maxilla and mandible with both having the same retraction appliance and force on the 2nd premolar following extraction of the 1st molars. The pulps of mandibular molars of 15 athymic and 15 conventional rats were surgically exposed and left open to their oral flora. Each group was divided into 3 subgroups of 5 animals each. The rats were sacrificed after their pulps were exposed for 2, 4, or 8 weeks. After fixing, decalcifying, and embedding, the specimens were sectioned and stained with H and E. The best understood functional activity of the pineal gland is its diurnal production of melatonin in response to environmental lighting cues. Several enzymes of the melatonin pathway respond to daily photoperiod changes, for example hydroxyindole-O-methyltransferase (HIOMT) and serotonin N-acetyltransferase (SNAT). Increased levels of the glycolytic enzyme neuron-specific enolase (NSE) are thought to reflect increased physiological demands placed on neurons and neuroendocrine tissues. To investigate the basis of these abnormalities, the hepatic nuclear thyroid hormone receptor was extracted with 0.4 M KCl, 1.1 mM MgCl₂, 20 mM Tris/HCl, pH 7.9 from hepatocyte nuclei of normal C57BL/KsJ, heterozygous C57BL/KsJ-db m (db/m), and diabetic C57BL/KsJ-db m (db/db) mice. Studies in experimental animals have produced evidence that increasing the fat in the diet decreases the animal’s ability to restrict tumor development and growth. An increase in dietary cholesterol and cholic acid will produce hypercholesterolemia in rats. For ten months, two groups of rats (six in each group) were fed commercial rat pellets (control group) or rat pellets plus cholesterol (1%) and cholic acid (0.25%) (experimental group). A blood sample from each animal was divided into three aliquots for three separate experiments on the erythrocytes: a membrane cholesterol and phospholipid determination, a 3-0-[¹⁴C] methylglucose (30MG) transport determination, and a 2,3-diphosphoglycerate (2,3 DPG) determination. We used morphometric analysis to determine if placental structure is modified by carbon monoxide, Camm-Hartley guinea pigs were exposed throughout gestation to chronic conditions of carbon monoxide (averaging about 180 parts per million). Maternal weight, placental weight, fetal weight, and fetal length were recorded at the time of placental fixation. Using a computerized image analyzer on photomicrographs obtained through light microscopy, we measured maternal and fetal percent vessel volumes, surface areas, and vessel numbers. Maternal vessel numbers and surface area remained basically unchanged but decreased slightly. Maternal percent volume decreased 15%. However, fetal capillary number increased 35% (p < 0.01) from a control value of 2325 ± 184 per mm² (SE …

Cell mediated immune (CMI) cytotoxic reactivity of Balb/c mice against H238 cells, a Herpes simplex virus (HSV) Type 2 - induced sarcoma, was measured by the ¹²⁵IUdR release assay. The Balb/c mouse response to the growing HSV-induced tumor, treated with radiation and , was measured by survival rate, tumor growth and immune cytotoxicity of spleen and peritoneal exudate cells (PEC) as determined by ¹²⁵IUdR release assay. Endodontic therapy was evaluated as a possible control or prevention modality of radiation type dental caries in dogs. Two adult male dogs with permanent dentitions had endodontic therapy performed on alternating teeth in each jaw. The nontreated teeth served as controls.

The dogs were irradiated with 7000 rad Cobalt-60 radiation fractionated over an eight week period. The dogs were killed at 60 and 182 days post-irradiation. The teeth were removed from the jaws and immersed in methylene blue dye, sectioned, and the dye penetration pattern photographed. Diffusion of the dye into the tooth structure was quantified and statistically analyzed.

This study compared the ability of three different bone growth factor combinations with that of Ca(OH)₂ to stimulate apical closure as well as the effectiveness of Mineral Trioxide Aggregate (MTA) as an apical barrier. Fifty-six roots of second and fourth mandibular premolars of seven six- month old beagle dogs were used. Periradicular lesions were created over a four week period. This thesis describes an in vivo perfusion apparatus which can continuously deliver controlled quantities of solutions to localized, contralateral intraoral areas in albino rats. Three animals were per fused on the experimental area (the mesial sulcular area of the maxillary first molar) on one side with tap water and Trypan blue. Localization of the solution delivered and minimal histological changes due to the presence of the apparatus was demonstrated on these animals.
