Agroecology incorporates ecology and agronomy to inform crop management. Industrial hemp (Cannabis sativa L.) is a prime system to investigate agroecological questions because it has only recently been authorized for commercial cultivation and its sensitivity to biotic and abiotic stressors is unresolved. Here, I investigate how some conventional management choices, including the application of fungicide, plastic mulch, and irrigation, impact the physiology, chemistry, and microbial ecology of industrial hemp. Overall, I found that industrial hemp can be both resilient and resistant to these stressors, an important development for the industry. In Chapter II, I tested the impacts of fungicide application …

Soybean cyst nematode (SCN), Heterodera glycines (HG) Ichinohe, is an endoparasitic nematode and one of the major yield-limiting pathogens of soybean (Glycine max L.) in the United States. Soybean cyst nematode alone accounts for more than $1.5 billion economic loss annually in North America. First reported in the U.S.A. in 1954 in New Hanover County, North Carolina and later in 1995 in Union County, South Dakota. Prior to this study, SCN has spread in 34 counties in the state. Each year, a variety of diseases impact the production of corn in South Dakota, though very little documentation about these diseases exists in the state. This survey documents the foliar diseases of corn and the distribution of each to create a baseline of knowledge. Cucurbit crops comprise the largest area in vegetable production in South Dakota (USDA 2022). Information on the prevalence, distribution, and fungicide sensitivity of their fungal pathogens are unknown. This study characterized the major foliar fungal species affecting cucurbits in South Dakota through a 2-year, state-wide survey. Between 2023 and 2024, 789 leaves were recovered from pumpkin, cucumber, watermelon, squash, and bottle gourd. Alternaria spp. (70.72% of leaves) Fusarium spp. (6.84%), Podosphaera xanthii (4.18%), both Alternaria spp. and Fusarium spp. (3.42%), and both Alternaria spp. and Podosphaera xanthii (1.90%), Stagonosporopsis spp. (1.27%) and other genera of fungi (4.94%) were identified using …

Roughly 50% of South Dakota cropland is under the practice of no-till, with large increases in acres of no-till in the last 20 or so years. No-till is beneficial for Great Plains states, such as South Dakota, as soil residues are withheld. Residues are shown to increase soil health through retention of moisture and increase of microorganisms, but residues can serve as a source of inoculum for fungal pathogens that can overwinter on them. Tan spot, a foliar disease of wheat caused by the ascomycetes fungus Pyrenophora tritici repentis (Ptr), poses a significant threat to wheat crops globally. The disease gained economic importance in the late 1970s due to shifts in farming practices, such as minimal tillage and planting on crop residues, providing favorable conditions for the pathogen to overwinter and cause infections. Ptr is classified into eight races based on the production of necrotrophic effectors—Ptr ToxA (necrosis-inducing), Ptr ToxB, and Ptr ToxC (chlorosis-inducing on respective susceptible and sensitive cultivars)—resulting in distinct symptoms on susceptible wheat differentials. Fusarium head blight (FHB), commonly called scab is the most devastating disease of wheat and barley caused by the ascomycete fungus, Fusarium graminearum. Pyraclostrobin, a QoI fungicide, is extensively used in the management of fungal leaf spot diseases. In this study, 215 P. tritici-repentis isolates collected from wheat fields in South Dakota were evaluated for their sensitivity to Pyraclostrobin. Of 215 isolates, 48 isolates (22%) exhibited insensitivity based on G143A mutation and EC50 values using spore germination assay. Our results indicate reduced sensitivity to insensitivity to pyraclostrobin in some isolates suggesting regular monitoring of the P. tritici-repentis population to QoI fungicides is essential to track the evolution of insensitive population. Phomopsis stem canker is a major threat to sunflower (Helianthus annuus L.) production worldwide and its prevalence has increased from 1.5% in 2001 to 61.0% in 2015 in the sunflower producing U.S. states of Colorado, Kansas, Minnesota, Nebraska, North Dakota, South Dakota, and Texas. Several Bacillus amyloliquefaciens isolates obtained from wheat residue have been studied as biological agents to control wheat diseases. Notable traits of isolates 1BA and 1D3 include growth with high salt (10% NaCl), at temperatures up to 50°C and over a wide pH range. BIOLOG Gen III plates were used to study and further characterize Bacillus amyloliquefaciens 1BA and 1D3. Both isolates showed tolerance to high salt concentration supporting previous studies. They also grew in the presence of lithium chloride, potassium tellurite, and sodium bromate. Both isolates grew at pH 5, with almost identical carbon source utilization fingerprints. Rising atmospheric carbon dioxide (CO2) may affect plant/pathogen interactions. This study focused on the effects of elevated CO2 on Root-Knot Nematode (Meloidogyne arenaria) and Tobacco Mosaic Virus (TMV) infection in genetically resistant versus susceptible tomatoes (Solanum lycopersicum). Both resistant and susceptible tomatoes were grown in chambers with either ambient CO2 or CO2 elevated to 750 ppm and infected with M. arenaria or TMV. Soybean cyst nematode (SCN) is a microscopic worm that infects and feeds from the soybean roots, hence robbing the plant of nutrients, and entry wounds may facilitate other pathogens to infect the roots. Current SCN management practices include cultural, chemical, and biological methods. Plants have evolved a complex defense system against pests and pathogens utilizing many types of receptors, signaling factors, and defense compounds to detect pathogen presence and respond effectively. Since many pathogens have evolved immunesuppressing effectors used to reduce plant resistance, plants have evolved a family of receptors that detect pathogenic effectors as a result of an evolutionary arms race. The soybean cyst nematode (SCN; Heterodera glycines, Ichinohe) is an important pathogen of soybean in South Dakota which causes significant yield losses. SCN has been found in 30 South Dakota counties as on 2017 and is estimated to cause yield loss of 1.9 Metric tons (Mt) annually. SCN has diverse crop and weed hosts as it can reproduce in several crops and weeds. Lilium grayi (Gray’s Lily), a southern Appalachian endemic species, is threatened by a Lilium-specific fungal pathogen, Pseudocercosporella inconspicua. The disease is characterized by tan lesions that can cause early senescence, while also lowering seed production and viability. This project tested for P. inconspicua conidia and accessed health at nine locations. The disease was present and ubiquitous across the range of L. grayi. Through identification of P. inconspicua conidia in the field, L. superbum (Turk’s Cap Lily) was identified as an additional host, while L. michauxii (Michaux’s Lily) was disease-free. Ochratoxin A (OTA) is a toxic metabolite produced by several species of fungal genera Penicillium and Aspergillus. The major OTA producer in temperate regions (i.e. U.S., Canada, Europe) is Penicillium verrucosum. Oat (Avena sativa), like any other cereal grain, can be contaminated with OTA when storage conditions are favorable for fungal growth. Spot blotch (SB), caused by Bipolaris sorokiniana (Sacc.) Shoem, and bacterial leaf streak (BLS), caused by Xanthomonas translucens pv. undulosa (Smith et al.), two important foliar diseases of wheat in the major production regions of the US and the world. Deployment of adequate host resistance against them depends on determining the resistance genes or quantitative trait loci (QTLs) responsible for the trait and identification of genetic markers linked to QTL that facilitate markers assisted breeding. Tan spot, caused by the ascomycete fungus, Pyrenophora tritici-repentis (Ptr), is a major foliar fungal disease of wheat worldwide. To date, Ptr isolates have been grouped into eight races based on the production of three host selective effectors (toxins), Ptr ToxA, ToxB, and ToxC, which are associated with necrosis and chlorosis symptoms on wheat differential lines. SIP470, a putative tobacco lipid transfer protein, was identified in a yeast two-hybrid screen to interact with SABP2. SABP2 is a critical role in SA-mediated signaling in tobacco and other plants. In vitro studies using purified recombinant SIP470 confirmed that it is a lipid binding protein. In an attempt to determine its role in mediating stress responses, Arabidopsis T-DNA insertion knockout lines lacking SIP470 homolog were used for the analysis. These mutant plants were defective in basal resistance against microbial pathogens. Expression of defense gene PR-1 was also delayed in these mutant plants.
