Gene transfer systems for phytopathogenic corynebacteria have not been reported previously. In this paper we describe a conjugative 46-megadalton plasmid (pDG101) found in Corynebacterium flaccumfaciens subsp. oortii COIOI that mediates resistance to arsenite, arsenate, and antimony(III). Transfer of the plasmid from CO101 to four other strains from the C. flaccumfaciens group occurred between cells immobilized on nitrocellulose filters or on agar surfaces. Transconjugant strains expressed the same levels of metal resistance as the donor strain and were able to act as donor strains in subsequent matings. Clavibacter: A New Genus Containing Some Phytopathogenic Coryneform Bacteria, Including Clavibacter Xyli Subsp. Xyli Sp. Nov., Subsp. Nov. And Clavibacter Xyli Subsp. Cynodontis Subsp. Nov., Pathogens That Cause Ratoon Stunting Disease Of Sugarcane And Bermudagrass Stunting Disease, Michael J. Davis, A. Graves Gillaspie Jr, Anne K. Vidaver, Russell W. Harris

A total of 23 strains of coryneform bacteria that cause ratoon stunting disease of sugarcane and Bermudagrass stunting disease were examined. These included 17 sugarcane strains from Florida, Louisiana, South Africa, Brazil, and Japan and six Bermudagrass strains from Florida and Taiwan. The sugarcane and Bermudagrass strains contained 2,4-diaminobutyric acid, rhamnose, and fucose in their cell walls, suggesting a relationship to some other phytopathogenic coryneform bacteria and related organisms with similar cell wall compositions. The cellular protein patterns of the sugarcane and Bermudagrass strains after polyacrylamide gel electrophoresis resembled the patterns of Corynebacterium michiganense subspecies more than the patterns of …

RNAs of soil-borne wheat mosaic virus (SBWMV) from virions 281 nm, 138 nm and 92 nm long (designated here by relative lengths as 1∙0L, 0∙SL and 0∙35L, respectively), were isolated and purified by three cycles of sucrose density gradient centrifugation. Infectivity assays with these RNAs proved the bipartite nature of SBWMV, the combination of 1∙0L and either 0∙5L or 0∙35L RNAs being required for infection and for multiplication of progeny viruses. The 0∙5L RNA underwent deletion mutation, producing smaller variants with various sizes, of which 0∙4L and 0∙35L RNAs were confirmed to be functional in combination with 1∙0L RNA. The …

Viruses interact genetically in various ways with bacterial and animal hosts, e.g. by lysogeny in bacteriophage and by incorporating a DNA copy of retrovirus RNA into the host genome. These phenomena lead to mutations, to the transfer of host genes by viruses, and to latent viruses being transmitted from generation to generation essentially as host genes. In contrast, there are no known examples of the incorporation of viral genomes into plant DNA.

The wild-type (WT) isolate of soil-borne wheat mosaic virus has two species of rodshaped virions 281 nm and 138 nm in length, which are designated as virions 1∙0L and 0∙5L, respectively. We reported previously that virions shorter than 0∙5L arose in assay plants inoculated with separated and recombined 1∙0L RNA and 0∙5L RNA and suggested that these short virions were caused by deletion mutation of 0∙5L RNA. Muiridin, a spore-specific protein of the fungus Botryodiplodia theobromae, comprises about 25% of the mature pycnidiospore protein. It has an apparent molecular weight of 16,000 to 17,000 and is rich in glutamine, asparagine, and arginink. Muiridin is synthesized in developing spores via a precursor with an apparent molecular welght of 24,000. Two other polypeptides present in young developing spores with apparent molecular weights of 18,000 and 15,000 are immunologically related to muiridin. We propose a pathway for muiridin synthesis. Roots and soils from western Nebraska fields of native and planted grasslands, and winter wheat of varied fallow-wheat cultivation duration, were evaluated for vesicular-arbuscular (VA) mycorrhizal root infection and spore numbers and types. Increased cultivation decreased percentage mycorrhizal infection in wheat and reduced spore numbers of Glomus fasciculatus, the dominant VA mycorrhizal fungus in these soils. Spore numbers of other VA mycorrhizal fungi did not change significantly with cultivation although mean numbers of G. mosseae increased with continued wheat production. Tire genus Nagelus (Thorne and Malek, 1968) Siddiqi, 1979 is modified and a new species from Alaska is described. The combination of scanning electron microscopy and light microscopy permits the characterization of Nagelus spp. as having a broadly oval face pattern, no longitudinal striations on the lip region, deirids surrounded by six incisures, and an irregularly tapering tail with a large hyaline region. We previously reported that isolation of symbiotic Chloreila-like algae from the Florida strain of Hydra viridis induced replication of a virus (designated HVCV-1) in the algae. We now report that isolation ofsymbiotic Chlorella-like algae from four other sources of green hydra and one source ofthe protozoan Paramecium bursaria also induced virus synthesis. Algae from one of these hydra contained a virus identical to HVCV-1 (based on its rate of sedimentation, buoyant density, reaction to HVCV-1 antiserum, and DNA restriction fragments) whereas algae from the other three hydra contained. another similar, but distinct, virus (designated HVCV-2). Influence Of Vesicular-Arbuscular Mycorrhizae On Water Movement Through Bouteloua Gracilis (H.B.K.) Lag Ex Steud, Michael F. Allen

Root growth characteristics and water transport were compared between non-mycorrhizal Bouteloua gracilis and vesicular-arbuscular mycorrhizal Bouteloua gracilis infected with Glomus fasciculatus. Mycorrhizal plants did not have significantly different leaf area or root length from non-mycorrhizal plants, hut did have significantly fewer and shorter root hairs. Mycorrhizal plants had 50% lower leaf resistance with no change in leaf or root water potentials; thus transpiration was increased 100% with a 50% reduction in whole-plant, soil-to-root and root-to-leaf resistance. The plant pathogenic corynebacteria are a fascinating group of bacteria that produce effects as diverse as any in the microbial world, ranging from wilts to abnormal growths. Several new pathogens have been discovered since this group was reviewed by Jensen (75), Starr (157), and Lelliott (96). Most subsequent reviews have dealt with taxonomy. The areas covered in this review necessarily reflect my biases and limitations. However, this review attempts to be comprehensive in some areas, and it calls attention to other areas that would benefit from more detailed investigation.

Non-mycorrhizal and vesicular-arbuscular mycorrhizal Bouteloua gracilis infected with Glomus fasciculatus were grown in defined media containing different phosphate sources and compared with respect to phosphate content and form, biomass, chlorophyll concentration, and root phosphatase activity. The phosphate sources were sodium monobasic phosphate, a mixture of sodium monobasic phosphate and calcium phytate, and calcium phytate. Inositol and inositol plus calcium were added to the sodium phosphate medium as additional treatments. Mycorrhizal infection was highest in roots of plants grown in the presence of phytate (75%). Lower root infection levels were noted in plants from the sodium phosphate (19%) and mixed phosphate …

Corn stocks showing virus-induced aberrant ratio (AR) at the “A” locus were found to have recessive alleles at the R and/or C loci. Since by the known pedigree these loci should be homozygous dominant, the results suggest an inactivation of maize genes by a mechanism as yet unknown. The presence of recessive alleles at these additional loci can explain the segregation ratios obtained in these particular stocks.

In the past, tillage research on plant diseases was concerned primarily with practices that buried plant residues in single-crop production systems. The burial of plant debris to destroy pathogens is an ancient agricultural prac­ tice (34). Each crop was tested as a single entity, and most crop rotations were based on one crop per year. The rangeland grass, Bouteloua gracilis was inoculated with its mycorrhizal symbiont, Glomus fasciculatus, to determine the influence of vesicular-arhuscular mycorrhizae on water status, stomatal behaviour and photosynthesis as well as gross plant morphology, biomass and phosphorus content. Mycorrhizal infection increased transpiration rates by over 100% with 50 to 70% lower leaf resistances to water vapour diffusion. Leaf xylem pressure was not different between mycorrhizal and non-mycorrhizal plants indicating that whole-plant resistance to water transport was reduced by more than 50%. Photosynthetic rates under saturating light conditions increased 68% with infection as a consequence of a 33% reduction in stomatal …

In vitro translation of the three single-stranded RNAs transcribed in vitro by bacteriophage φ6 RNA polymerase revealed that the large RNA codes for phage proteins P1, P2, P4, and P7, the medium RNA codes for P3, P6, and P10, and the small RNA codes for P5, P8, and P9.

The RNA polymerase in the nucleocapsid of seudomonas phaseolicola bacteriophage φ6 transcribed large, medium, and small single-stranded RNA from the viral double-stranded RNA genome by a semiconservative (displacement) mechanism. Approximately 23%, 63%, and 65% of the nucleocapsid particles in the assay mixture synthesized at least one round of large, medium, and small single-stranded RNA molecules, respectively. Some of these particles reinitiated synthesis such that an average of 1.5 large, 33 medium, and 24 small singlestranded RNAs were synthesized from each double-stranded RNA.

The RNAs of brome mosaic (BMV), barley stripe mosaic (BSMV), and tobacco mosaic (TMV) viruses were inactivated by reaction with buffered glutaraldehyde. Glutaraldehyde did not fix 4% BMV-RNA, 20% t-RNA, 5% polyadenylic acid, or 5% adenosine monophosphate into water-insoluble precipitates, or gels, in distilled water or in low or high ionic strength buffers nor did it change their ultraviolet (UV) spectra. Two SDS- and phenolpurified commercial yeast RNA preparations from different sources gave UV spectra typical of pure RNA, but could not be freed of a contaminant that reacted with glutaraldehyde by forming a precipitate. Bacteriocin-like substances were commonly produced by slow-growing Rhizobium japonicum and cowpea rhizobia on an L-arabinose medium. Antagonism between strains of R. japonicum was not detected in vitro; however, such strains were often sensitive to some bacteriocins produced by cowpea rhizobia. Inhibitory zones (2 to 8 mm from colony margins), produced by 58 of 66 R. japonicum test strains, were reproducibly detected with Corynebacterium nebraskense as an indicator. Quantitative production was not related to symbiotic properties of effective strains, since nine noninfective strains and one ineffective strain produced bacteriocin. Purification schemes were developed which yielded 0.7 to 1.0 mg of alfalfa latent virus (ALV) and pea streak virus (PSV) and 0.1 to 0.3 mg of red clover vein mosaic virus (RCVMV) per gram pea cullivar Lincoln plant tissue (excluding roots). The freezing of the tissue and the use of an appropriate extraction buffer were crucial. Virus from sap was precipitated by 6%(w/v) polyethylene glycol (PEG, MW 6,000) and concentrated by two cycles of differential centrifugation. Root segments and root-soil cores (6.5-cm diameter) from fields and nurseries of winter wheat and sorghum were tested for N2 fixation by using the acetylene reduction assay. Wheat samples (~1,200) from 109 sites generally had low or no activity (0 to 3.1 nmol of C2H4 produced per h per g [dry weight] of root segments), even after 24 h of incubation. Alfalfa latent virus (ALV), a new member of the carlavirus group, was isolated From alfalfa (Medicago sarivo) clones from rbe University of Nebraska experimental plots and from the Farmers' fields. The pea aphid, Acvrihosiphon pisum, transmitted this virus to M. sativa, Pisum sativum, and Viria faba. but not to Trifolium pratense. The sedimentation coefficient of virion I of soil-borne wheat mosaic virus was estimated to be 219S, the same, within error, as the sedimentation coefficient of the dimer of virion II, estimated to be 218S. The monomer of virion II sedimented at 177S, and was 138 nm long. Another strain of the virus had a virion II (designated lIb) that was 92 nm long and sedimented at 159S. The two virus strains coexist in some fields.

Nonionic detergent treatments released a nucleocapsid from the enveloped bacteriophage φ6. The nucleocapsid sedimented at nearly the same rate as the whole phage in sucrose density gradients, but the buoyant density in Cs2SO4 changed from 1.22 g/cm3 for the whole phage to 1.33 g/cm3 for the nucleocapsid. The detergent completely removed the lipid and 5 of the 10 proteins from the phage. Surface labeling of the phage and nucleocapsid with 125I revealed that protein P3 was on the outer surface of the whole phage and P8 was on the surface of the nucleocapsid. Both …

The last review on bacteriophages and bacteriocins of phytopathogenic bacteria was that of Okabe & Goto (98). All but one (21) of the subsequent reviews (II, 12, 60, 94, 104) have dealt primarily with viruses and bacteriocins of bacteria other than phytopathogens. The most recent microbiological compendia that in­ clude bacteriophages and bacteriocins are of limited value for phytobacteriolo­ gists because the authors generally overlook or ignore such agents (29, 47, 77, 119). Systematic sampling of waterways and irrigation runoff from agricultural lands in the North Platte Project of Nebraska in July and August of 1972–1974 demonstrated that phytopathogenic organisms were disseminated. The organisms monitored included the bean common blight bacterium Xanthomonas phaseoli, the bean white mold fungus Whetzelinia sclerotiorum and various nematodes. Although many types of nematodes often were recovered from irrigation water, Heterodera sp. cysts which cause significant disease problems in the valley were found infrequently. Patterns of movement of the bacterial and fungal organisms were correlated with previous or current season infection of bean plants. The short-term survival of X. …
