Phytophthora root and crown rot (PRCR) is currently the most important disease on lavender (Lavandula spp.) in the United States. The disease was first described on English lavender (L. angustifolia) in a Maryland nursery in 1991, with Phytophthora nicotianae as the causal agent. Since that time, the disease has been reported on multiple continents and as caused by several species of Phytophthora. This study examined the distribution and pathogenicity of Phytophthora species on lavender in the United States, requested lavender grower feedback regarding their production and concerns, and examined efficacy of selected management options. In 2016, a survey was conducted into the causes of twig blight on peach (Prunus persica) in five orchards across South Carolina. From this survey, isolates from the fungal genus of Cytospora (Sordariomycetes, Diaporthales) were reported on peach, for the first time, within the state. Cytospora species are known to cause oozing cankers on a wide range of woody hosts leading to dieback, loss of limbs, and reduction in yield, prompting concern for important economic crops. Prunus necrotic ringspot virus (PNRSV), prune dwarf virus (PDV), and peach latent mosaic viroid (PLMVd) are important pollen-borne pathogens that affect peach orchards in the southeastern United States. My aim with this research is to identify potential sources of virus/viroid inoculum sources in the wild and determine pathogen movement and interactions between wild Prunus spp. and cultivars of peach (P. persica). Managing weeds is one of the most challenging aspects of growing specialty crops. Weed control options are limited for specialty crops due to absence of effective herbicide options. The limited current herbicide options risk carryover and damage to subsequent crops. Bacterial wilt is another problem causing significant yield losses in southeast vegetable production. After the termination of methyl bromide due to health and environmental concerns, several other chemical fumigants gained interest, including 1,3-dichloropropene, chloropicrin and metam sodium/potassium. Background: The semi-endoparasitic reniform nematode (Rotylenchulus reniformis) is a major yield-limiting pest of multiple crops in the tropics and sub-tropics, including upland cotton (Gossypium hirsutum). Reniform-resistant cotton varieties are urgently needed, but genes that confer resistance to reniform nematode have not been identified in any species. Parasitism by reniform nematode involves significant developmental changes in plant roots, leading to the formation of multicellular feeding structures called syncytia. Here, we present de novo transcriptomes assembled from syncytial and non-syncytial cotton roots on three sampling dates across a 12-day time course. Rotylenchulus reniformis (reniform nematode) and Hoplolaimus columbus (Columbia lance nematode) are two of the main plant-pathogenic nematode species affecting cotton production in the United States. For the selection and implementation of effective management practices it is important to understand distribution patterns and population genetic structure of these two nematodes in cotton agroecosystems. In a three-year field study, the spatial distribution of reniform nematode was characterized in two commercial fields in South Carolina to determine the effects of relations with soil texture and crop rotation on reniform nematode distribution (Chapter 1). The costs and need for clean water are increasing for greenhouse, nursery, and agricultural businesses. Runoff from irrigated agriculture and specialty crops also can take a toll on the surrounding environment, because irrigation runoff water can contain high amounts of nutrients, contaminants, and plant pathogens. To assuage the pollution contributed to runoff water and reduce the overall volume of water used, nurseries are starting to reuse irrigation runoff water. The United States is the largest producer of strawberries worldwide, generating approximately $2.4 billion worth of fruit each year. Strawberry production, however, is often threatened by pathogens. One of the most destructive fungal pathogens is Botrytis cinerea, the causal agent of gray mold disease. The control of gray mold in commercial fields is largely dependent on the application of fungicides, including the dicarboximide iprodione and the hydroxyanilide fenhexamid. Phytophthora foliage blight of garden phlox (Phlox paniculata ) - caused by Phytophthora nicotianae - is a devastating disease that has been seen consistently in nursery production facilities in South Carolina. Fusarium wilt of watermelon, caused by Fusarium oxysporum f. sp. niveum , was first described in 1894 in South Carolina and Georgia and has become a limiting factor in watermelon production worldwide. Nematode glutathione S-transferases (GSTs) have been implicated in plant-nematode interactions as effector proteins with an important role in the establishment of feeding sites. Studies with root-knot nematode in Arabidopsis thaliana suggest that GSTs may protect the nematode against oxidative plant defenses and modulate plant responses to parasitism. Our objective was to study the function of a GST from reniform nematode (Rotylenchulus reniformis) in soybean. We used a virus-induced gene silencing (VIGS) system, utilizing a Bean pod mottle virus (BPMV)-based vector and a partial sequence of gsts-1 from Meloidogyne incognita to silence the putative reniform nematode homolog. Phytophthora cinnamomi is a devastating pathogen that can attack over 900 hosts. It is the most common species of Phytophthora isolated from woody ornamental crops in South Carolina but little is known about variability among isolates of P. cinnamomi that attack these plants. Therefore, 142 isolates of P. cinnamomi recovered from diseased plant samples submitted to the Clemson University Plant Problem Clinic between 1995 and 2011 were characterized for mycelium growth habit, growth rate, mefenoxam sensitivity, mating type, and sporangium morphology. Rapid blight is a disease of cool-season turfgrasses caused by Labyrinthula terrestris. Disease symptoms include water-soaked lesions, yellowing and browning of foliage, and coalescing patches of dead turf. L. terrestris belongs to a group of marine microorganisms in the kingdom Chromista (also known as Stramenopila). Morphological characteristics of Labyrinthula spp. include spindle-shaped vegetative cells that move in an ectoplasmic network. The biology and pathology of L. terrestris is not clearly understood; therefore, the ultrastructure, life cycle, and histopathology of L. terrestris were investigated. Although recycling irrigation water is beneficial to the nursery industry as it serves to decrease potable water usage and reduces the release of excess agrichemicals into the environment, it also poses the risk of becoming the source for and means of dispersing water-borne plant pathogens. This project was composed of two separate studies. In one study, the species of Phytophthora that have been found associated with diseased floriculture crops in South Carolina and four other states were characterized and identified using molecular (RFLP fingerprints and DNA sequences for ITS regions and cox I and II genes), morphological (sporangia, oogonia, antheridia, oospores, and chlamydospores), and physiological (mating behavior and cardinal temperatures) characters. In addition, sensitivity to the fungicide mefenoxam was determined. In all, 87 isolates from 63 host plants that were collected over a 13-year period (1996 through 2008) were examined. The efficacy of compost tea was evaluated and compared alone and in combination with two biofungicides Serenade Max and Sonata for control of early blight caused by the fungal pathogen Alternaria tomatophila on tomato Solanum lycopersicum (L), heirloom cultivar `Cherokee Purple'. Results of field experiments over two growing seasons indicated that compost tea provided a low level of protection against early blight. The results were variable; most likely due to dry conditions and the presence of bacterial wilt the first year and tomato spotted wilt virus the second year.
