The bacterial plant pathogen Pseudomonas syringae possesses a type III protein secretion system that delivers many virulence proteins into plant cells. A subset of these proteins (called Avr proteins) is recognized by the plant’s innate immune system and triggers defences. One defence-associated response is the hypersensitive response (HR), a programmed cell death (PCD) of plant tissue. We have previously identified HopPtoD2 as a type III secreted protein from P. s. pv. tomato DC3000. Sequence analysis revealed that an N-terminal domain shared homology with Avr- PphD and a C-terminal domain was similar to protein tyrosine phosphatases (PTPs). We demonstrated that purified …

Characterization Of Wound-Inducible Genes Encoding Enzymes For Terpenoid Biosynthesis In Medicago Truncatula, Mandy M. Cox

In addition to having numerous applications and pharmaceuticals, terpenoids are an important class of defensive compounds that can accumulate in plants after pathogen infection or injury by insects. Sequences of DNA encoding putative terpene synthases and an oxidosqualene synthase, isolated from insect-damaged Medicago truncatula leaves, were selected from an expressed sequence tag (EST) data base. The cDNA clones were used as radio-labeled probes to analyze gene expression in leaves treated with factors known to trigger a defense response in plants. Pseudomonas syringae uses a type III protein secretion system encoded by the Hrp pathogenicity island (Pai) to translocate effector proteins into plant cells. One of these effector proteins is HopPsyA. A small open reading frame (ORF), named shcA, precedes the hopPsyA gene in the Hrp Pai of P. s. syringae 61. The predicted amino acid sequence of shcA shares general characteristics with chaperones used in type III protein secretion systems of animal pathogens. A functionally non-polar deletion of shcA in P. s. syringae 61 resulted in the loss of detectable HopPsyA in supernatant fractions, consistent with ShcA acting as a …

The ability of Pseudomonas syringae pv. tomato DC3000 to parasitize tomato and Arabidopsis thaliana depends on genes activated by the HrpL alternative sigma factor. To support various functional genomic analyses of DC3000, and specifically, to identify genes involved in pathogenesis, we developed a draft sequence of DC3000 and used an iterative process involving computational and gene expression techniques to identify virulence-implicated genes downstream of HrpLresponsive promoters. Hypersensitive response and pathogenicity (Hrp) promoters are known to control genes encoding the Hrp (type III protein secretion) machinery and a few type III effector proteins in DC3000. The purpose of this study was to characterize the variation pattern that exists in the wild Musa acuminata (AA) populations. widely distributed in Malaysia. Fourteen populations of Musa acuminata, which included subspecies malaccensis, siamea and truncata were collected from different locations including Selangor, Pahang, Perak, Kelantan, Kedah and Terengganu for studies on variation within and between populations, based on morphological characters, biochemical, ploidy and genomic DNA content. Techniques of numerical taxonomy were employed to determine the variation pattern and these included use of the clustering method and principal component analysis. Pseudomonas syringae is a member of an important group of Gram-negative bacterial pathogens of plants and animals that depend on a type III secretion system to inject virulence effector proteins into host cells. In P. syringae, hrpyhrc genes encode the Hrp (type III secretion) system, and avirulence (avr) and Hrpdependent outer protein (hop) genes encode effector proteins. The plant pathogenic bacterium Pseudomonas syringae is divided into pathovars differing in host specificity, with P. syringae pv. syringae (Psy) and P. syringae pv. tomato (Pto) representing particularly divergent pathovars. P. syringae hrp/hrc genes encode a type III protein secretion system that appears to translocate Avr and Hop effector proteins into plant cells. DNA sequence analysis of the hrp/hrc regions in Psy 61, Psy B728a, and Pto DC3000 has revealed a Hrp pathogenicity island (Pai) with a tripartite mosaic structure. We present here data showing that the Avr proteins HrmA and AvrPto are secreted in culture via the native Hrp pathways from Pseudomonas syringae pathovars that produce these proteins. Moreover, their secretion is strongly affected by the temperature and pH of the culture medium. Both HrmA and AvrPto were secreted at their highest amounts when the temperature was between 18 and 22°C and when the culture medium was pH 6.0. In contrast, temperature did not affect the secretion of HrpZ. pH did affect HrpZ secretion, but not as strongly as it affected the secretion of HrmA. This finding suggests that …

The host-specific plant pathogen Pseudomonas syringae elicits the hypersensitive response (HR) in nonhost plants and secretes the HrpZ harpin in culture via the Hrp (type III) secretion system. Previous genetic evidence suggested the existence of another harpin gene in the P. syringae genome. hrpW was found in a region adjacent to the hrp cluster in P. syringae pv. tomato DC3000. hrpW encodes a 42.9-kDa protein with domains resembling harpins and pectate lyases (Pels), respectively. Cowpea is an important legume food crop that is commonly grown in Arkansas and numerous other southern states. The application of biotechnological approaches for the improvement of U.S. cowpea genotypes is currently not possible due to the lack of a regeneration and transformation system. Subterranean clover is widely recognised as the most important pasture legume species in southern Australia, having been sown over about 20 million hectares.

Together with superphosphate and trace elements, it has been a key factor in improving and maintaining soil fertility, and has resulted in dramatic increases in crop, livestock and wool production.

While the value of subterranean clover is well established, it does have some shortcomings.

Many varieties grown today are not sufficiently adapted to the environments and management systems in use. Others lack sufficient pest and disease resistance.

Three New Late-Midseason Subterranean Clovers Released For High Rainfall Pastures, Phil Nichols, Donald Nicholas

Three new subterranean clovers - Denmark, Gou/bum and Leura - have been released in 1992 by the National Subterranean Clover Improvement Program. These subterranean clovers are black-seeded, have low oestrogen levels and improved disease resistance. Their release offers the potential for substantial improvements in pasture productivity in areas of southern Australia that have long growing seasons.

This article outlines some of the testing procedures and subsequent selection of these varieties and describes their characteristics and potential role in Western Australia.

The introduction of improved pasture legumes into the cereal growing areas of southern Australia changed the face of our agricultural systems.

Subterranean clover was the first pasture legume to have a major impact, and the first varieties were available commercially in the 1930s. Since then, many varieties have been released, leading to the most recent release of Denmark, Gou/bum and Leura in 1992.

The development of burr, barrel and murex medics also had an impact on the pasture production of less acid heavy soils. Yellow serradella is still being developed for the more acid sandy soils.

Viruses with defective genomes have been identified in association with virtually every major family of viruses and have been widely utilized as tools for investigating virus functions in animal cell culture systems (Perrault, 1981). It is generally thought that defective interfering viruses (DIs) arise through deletion, rearrangement, or recombination of a competent viral genome. DIs tack the ability for independent existence relying on their parental helper viruses to supply factors required for replication, maturation, and/or encapsidation (Huang and Baltimore, 1977). The early use of field peas in Western Australia was as a green manure crop or to feed stock be grazing the mature crop. Varieties which produced a large bulk of green material were sown. There was no control over insects or diseases.

In 1985, the Feseral Government recognised that to realize the potential of grain legumes grown throughout Australia, more research was needed.
