Identification Of Qtl Modifying The Activity Of The Tcb1-S Locus And Characterization And Sequencing Of Two Plutonium-Beryllium Induced Reduced Gametophyte Transmission Mutants In Maize, Merritt Bryer Burch

This thesis is split into two independent projects both involving the male gametophyte generation of maize. The first project looks at how pollen interacts with the female gametophyte to reduce its transmission in cross-incompatible reactions controlled by the unilateral cross-incompatibility system, teosinte crossing barrier 1. The second project explores two plutonium-beryllium induced male gametophyte mutants and attempts to uncover their genetic basis.

Populations of invasive species are often subjected to novel selective forces in the form of anthropogenic control agents in their introduced ranges. These control agents, applied unevenly among populations within a species’ new range, can send invasive populations on drastically different evolutionary and ecological trajectories. In these studies, we aimed to see if different histories of chemical herbicide treatment are differentially influencing the genetic diversity, structure, and performance of populations of invasive watermilfoil (Myriophyllum spicatum and M. spicatum x M. sibiricum hybrids) from waterbodies in the state of Michigan. PREMISE OF THE STUDY: Few genetic markers provide phylogenetic information in closely related species of Isoëtes (Isoëtaceae). We describe the development of primers for several putative low- copy nuclear markers to resolve the phylogeny of Isoëtes, particularly in the southeastern United States.

METHODS AND RESULTS: We identified regions of interest in Isoëtes transcriptomes based on low- copy genes in other plants. Primers were designed for these regions and tested with 16 taxa of Isoëtes and one species of Lycopodium. This is a compilation of 19 research trial reports from six land-grant universities in the Midwestern United States. Crops include green beans, cantaloupe, cucumber, ornamental corn, pepper, potato, pumpkin, butternut squash, sweet corn, tomato, and watermelon. Several crops were evaluated in high tunnels or hoophouses. Most trials evaluated different cultivars or varieties. One report addressed specific growing practices: plant spacing for sweet corn.

This trial was conducted to evaluate varieties for their performance as a summer-harvested crop in soil-based high tunnel production systems. Pumpkins are grown on 5,000 acres in Indiana with an average yield of 6.75 tons per acre. Successful pumpkin production requires the use of varieties that yield well and produce pumpkins of the size, shape, color, and quality demanded by the market. Genetic resistance to the fungal disease powdery mildew is present in some varieties. This trial was designed to evaluate performance of pumpkin varieties in northern Indiana with and without treatment for powdery mildew. The trial included fourteen orange jack-o-lantern size pumpkins, one small or 'pie' pumpkin and one specialty yellow pumpkin.

Despite this, sunflowers have been forced onto marginal lands often on semi-arid and non-fertile soil, due to disease pressures and economics making it critical to produce more resilient lines and varieties to withstand these new stresses. The NLG (Novel Leaf Gene) is a recently discovered gene from aspen (Populus tremula x alba). Previous research indicates that NLG impacts leaf shape. Our preliminary analyses indicate that NLG may function as a membrane bound protein with possible localization in plasma membrane and/or nucleus. Sequence and phylogenetic analyses in Arabidopsis thaliana and Populus trichocarpa identified four homologous genes in Arabidopsis and five in Populus. Gene expression analysis was done using online tools and indicated that NLG is expressed in multiple tissue types but predominately in stem and flowers. Epigenetics is the study of heritable changes in phenotypes that are not the result of changes in DNA sequence. Examples of epigenetic affecters include methylation changes, chromatin modifications, transcription factors, and RNA-based changes. The molecular mechanisms behind epigenetic changes are not fully understood. Canalization is the buffering of gene expression against environmental changes over time, while habituation is semi-stable expression change over time due to selection. Malaria is one of the deadliest diseases in human history. Nearly half of the world’s population, is at the risk in 106 countries. Only in 2016, this disease killed about 445,000 people, 72% of them being children under age five. It also accounts for US $12 billion dollars of direct costs in Africa alone.
