Cannabis sativa L. (hemp) develops plants with either male or female flowers, and growers of hemp greatly prefer female flowers which bear the glandular trichomes that contain cannabinoids. Feminized (all female) seeds are highly desired, which are produced by crossing a female plant with a masculinized female plant. Masculinization is achieved through the inhibition of ethylene and/or addition of gibberellins before flower initiation in female plants. The hemp industry uses silver thiosulfate (STS) to masculinize hemp, but spraying silver poses environmental concerns. This study compared STS to three other ethylene-inhibiting agents: aminoethoxyvinylglycine (AVG), cobalt nitrate (CBN), and 1-methylcyclopropene (1-MCP). Treatments …

Understanding the genetic basis of complex traits is a fundamental goal of evolutionary genetics. Yet, the genetics controlling complex traits in many important species such as hemp (Cannabis sativa) remain poorly investigated. Because hemp’s change in legal status with the 2014 and 2018 U.S. Federal Farm Bills, interest in the genetics controlling its numerous agriculturally important traits has steadily increased. To better understand the genetics of agriculturally important traits in hemp, we developed an F2 population by crossing two phenotypically distinct hemp cultivars (Carmagnola and USO31). Fatty Acid Desaturase 7 (FAD7) is a chloroplast-localized enzyme that alters the fatty acid content of photosynthetic membranes, and that negatively regulates plant defenses against aphids. Previous studies in the model organism Arabidopsis (Arabidopsis thaliana) have shown that loss-of-function mutations in FAD7 decrease population growth of the green peach aphid (GPA; Myzus persicae Sulzer). This study further characterized the effects of a fad7 null mutant on aphids, and investigated the role of reactive oxygen species (ROS), including singlet oxygen (1O2), in plant responses to aphid resistance in fad7 as well as in wild type plants and a mutant with heightened …

In 2017, twelve Quality Protein Popcorn (QPP) inbred lines were developed and selected as premier dent by popcorn crosses fit for hybridization and testing. These QPP inbred lines were derived from specific Quality Protein dent Maize (QPM) by ConAgra Brands® popcorn line crosses to produce high lysine, vitreous popcorn lines capable of near-equal popping characteristics compared to the original popcorn parents. The QPP hybridization project commenced in the summer of 2018 utilizing these 12 inbred QPP lines and crossing them in a full diallel. Plants can sense the change of 1 oC in their growth environment and thus global climate change has a great impact on plant growth and development. The phenomenon that warm non-stress temperatures promote stem and petiole elongation, as well as leaf hyponastic growth, is collectively known as thermomorphogenesis. While it is known that the basic helix-loop-helix (bHLH) transcription factor PIF4 is highly inducible by temperature elevations and controls thermomorphogenesis in dicots (e.g., Arabidopsis thaliana), the molecular mechanism underlying thermomorphogenetic growth in monocots is not clear. Waterlogging can be detrimental to soybean [Glycine max (L.) Merr.] growth and development, with effects ranging from chlorosis and stunting to yield loss and plant death. Soybean responses to, and the effects of, waterlogging are dependent on the growth stage of the plant at the initiation of waterlogging. In the model dicotyledonous plant Arabidopsis thaliana, the basic helix-loop-helix (bHLH) transcription factor Phytochrome-Interacting Factor 4 (AtPIF4) plays a central role in regulating the ambient temperature-regulated hypocotyl growth response. Warm temperatures induce AtPIF4 transcription and the accumulated AtPIF4 promotes thermomorphogenesis by enhancing the expression of a group of growth-related genes. However, whether this thermo-induced growth response is universal, and more importantly, whether there is a conserved thermomorphogenetic mechanism shared in dicots is largely unknown. Although genome-sequence assemblies are available for a growing number of plant species, gene-expression responses to stimuli have been cataloged for only a subset of these species. Many genes show altered transcription patterns in response to abiotic stresses. However, orthologous genes in related species often exhibit different responses to a given stress. Accordingly, data on the regulation of gene expression in one species are not reliable predictors of orthologous gene responses in a related species. Here, we trained a supervised classification model to identify genes that transcriptionally respond to cold stress. A model trained with only features calculated directly from genome …

The primary goal of the Arkansas Cotton Variety Test is to provide unbiased data regarding the agronomic performance of cotton varieties and advanced breeding lines in the major cotton-growing areas of Arkansas. This information helps seed companies establish marketing strategies and assists producers in choosing varieties to plant. These annual evaluations will then facilitate the inclusion of new, improved genetic material in Arkansas cotton production. Sphingolipids are a vital component of plant cellular endomembranes and carry out multiple functional and regulatory roles. Different sphingolipid species confer rigidity to the membrane structure, facilitate trafficking of secretory proteins, and initiate programmed cell death. Although the regulation of the sphingolipid pathway is yet to be uncovered, increasing evidence has pointed to orosomucoid proteins (ORMs) playing a major regulatory role and potentially interacting with a number of components in the pathway, including both enzymes and sphingolipids. Upland cotton (Gossypium hirsutum L.) is an economically important multi-purpose crop cultivated globally for fibre, seed oil and protein. Cottonseed oil also is naturally rich in vitamin E components (collectively known as tocochromanols), with a- and c-tocopherols comprising nearly all of the vitamin E components. By contrast, cottonseeds have little or no tocotrienols, tocochromanols with a wide range of health benefits. Here, we generated transgenic cotton lines expressing the barley (Hordeum vulgare) homogentisate geranylgeranyl transferase coding sequence under the control of the Brassica napus seed-specific promoter, napin. The uses of vegetable oils are determined by functional properties arising from their chemical composition. Soybean oil was previously used in margarines and baked foods after partial hydrogenation to achieve heat and oxidative stability. This process, however, generates trans fats that are now excluded from food use because of cardiovascular health risks. Also present in soybean oil are the anti-oxidant tocopherols, with α- tocopherol (vitamin E) typically present as a minor component compared to γ-tocopherol. Genetic improvement of the fatty acid profile and tocopherol profile is an attractive solution to increase the functional and health qualities of soybean oil. The …

Objectives: The purpose of this data set is to capture the complete diurnal (i.e., daily) transcriptome of fully expanded third leaves from the C4 panacoid grasses sorghum (Sorghum bicolor), maize (Zea mays), and foxtail millet (Setaria italica) with RNA-seq transcriptome profiling. These data are the cornerstone of a larger project that examined the conservation and divergence of gene expression networks within these crop plants. The lipid matrix in cell membranes is a dynamic, bidimensional array of amphipathic molecules exhibiting mesomorphism, which contributes to the membrane fluidity changes in response to temperature fluctuation. As sessile organisms, plants must rapidly and accurately respond to environmental thermal variations. However, mechanisms underlying temperature perception in plants are poorly understood. We studied the thermal plasticity of membrane fluidity using three fluorescent probes across a temperature range of -5 to 41 °C in isolated microsomal fraction (MF), vacuolar membrane (VM), and plasma membrane (PM) vesicles from Arabidopsis plants. Aragoa, comprising 19 high-altitude North Andean species, is one of three genera in the Plantagineae (Plantaginaceae, Lamiales), along with Littorella and Plantago. Based primarily on plastid data and nuclear ITS, Aragoa is sister to a clade of Littorella + Plantago, but Plantagineae relationships have yet to be assessed using multigene datasets from the nuclear and mitochondrial genomes. Here, complete mitogenomes were assembled for two species of Aragoa (A. abietina and A. cleefii). Background: Biological mutagens (such as transposon) with sequences inserted, play a crucial role to link observed phenotype and genotype in reverse genetic studies. For this reason, accurate and efficient software tools for identifying insertion sites based on the analysis of sequencing reads are desired. Results: We developed a bioinformatics tool, a Finder, to identify genome-wide Insertions in Mutagenesis (named as “InMut-Finder”), based on target sequences and flanking sequences from long reads, such as Oxford Nanopore Sequencing. Selection for yield at high planting density has reshaped the leaf canopy of maize, improving photosynthetic productivity in high density settings. Further optimization of canopy architecture may be possible. However, measuring leaf angles, the widely studied component trait of leaf canopy architecture, by hand is a labor and time intensive process. Here, we use multiple, calibrated, 2D images to reconstruct the 3D geometry of individual sorghum plants using a voxel carving based algorithm. Automatic skeletonization and segmentation of these 3D geometries enable quan- tification of the angle of each leaf for each plant. The resulting measurements are both heritable and …

Background: Grapevine reproductive development has direct implications on yield. It also impacts on berry and wine quality by affecting traits like seedlessness, berry and bunch size, cluster compactness and berry skin to pulp ratio. Seasonal fluctuations in yield, fruit composition and wine attributes, which are largely driven by climatic factors, are major challenges for worldwide table grape and wine industry. Protein Prenylation is the addition of lipids to select proteins that play a key role during the development of plants. There are three enzymes that play a role in protein prenylation: protein farnesyltransferase (PFT), protein geranylgeranyl-transferase-I (PGGT), and Rab geranylgeranyltransferase (Rab-GGT). However, there is a protein called PPAL that has a similar alpha subunit of PFT, PGGT, and RAB-GGT but its biochemical function is unknown. Pollen cross-contamination has been a major problem for maize breeders. Various mechanical methods applied to avoid the contaminations are ineffective. The genetic factors related to maize fertilization can be used to develop an effective method to prevent pollen contamination. Pollen rejection ability controlled by Teosinte crossing barrier 1 (Tcb1) is such a genetic system. Silks possessing dominant Tcb1-s reject pollen possessing the recessive allele (tcb1). Successful fertilization occurs when Tcb1-s pollen falls upon tcb1 silks. The efficacy of dominant Tcb1-s was, however, reduced when repeatedly backcross with maize inbred lines, which suggests that there are modifiers to Tcb1-s. To find …

Soybean leaf-associated gemycircularvirus-1 (SlaGemV-1) is a novel mycovirus discovered through the metagenomic sequencing of soybean leaves which is capable of inducing hypovirulence in the highly pathogenic fungus Sclerotinia sclerotiorum. RNASeq analysis techniques were used to determine the transcriptional changes caused by the infection of virus in S. sclerotiorum, as well as the transcriptional changes in Glycine max caused by the colonization of hypovirulent, SlaGemV-1-infected S. sclerotiorum. RNASeq results indicate that viral infection leading to hypovirulence may attenuate expression of genes relating to cell wall synthesis, microtubule formation, and metabolism of steroids and natural antibiotics. Establishing genetically diverse communities that can adapt to dynamic selective pressures is crucial in ecological restoration. However, the genetic structure of native plant species used for restoration is often poorly understood. Pseudoroegneria spicata, is a keystone species of the Inland Northwestern US that has become a staple in commercially available restoration seed mixes. It is abundant in remnant prairies of the endangered Palouse Prairie ecoregion, which is characterized by rolling hills of deep loess soil. Less than 1% of native Palouse Prairie is left, due to agricultural conversion, with remnants highly fragmented and isolated. P. spicata is also common in …

Hybrid rice (Oryza sativa L.) breeding offers a significant opportunity to enhance rice production, and the cultivation of a male sterile line is the most important factor in the success of cross-breeding. One of the key elements of hybrid rice production is to develop a restorer line that is assigned as the male parent. The restorer lines provide viable pollen for fertilization of the male sterile plant due to the presence of a restorer gene (Rf) in their genomes.
