Soil fauna play critical roles in various ecosystem functions and services, but empirical data measuring their impact on dung pat decomposition and subsequent nutrient cycling into rangeland soils are limited. The objective of this study was to quantify the effect of soil fauna, using dung beetle as an indicator, on dung decomposition and subsequent translocation of dung nutrients into grassland soil over time. A field experiment was conducted early in the summer season and late in the summer season of 2014 and 2015. Traditional agricultural research and extension relies on replicated field experiments, on-farm trials, and demonstration plots to evaluate and adapt agronomic technologies that aim to increase productivity, reduce risk, and protect the environment for a given biophysical and socio-economic context. To date, these efforts lack a generic and robust spatial framework for ex-ante assessment that: (i) provides strategic insight to guide decisions about the number and location of testing sites, (ii) define the target domain for scaling-out a given technology or technology package, and (iii) estimate potential impact from widespread adoption of the technology(ies) being evaluated. Sedges (Carex spp. Linneaus. [Cyperaceae]) are a major component of Midwestern U.S. tallgrass prairie and prairie wetland plant communities, yet they are often lacking in restoration projects because they can be difficult to germinate. Cold-wet stratification and perigynia removal have been shown to increase germination in some Carex species. The germination response of 3 native species, plains oval or shortbeak sedge (C. brevior (Dewey) Mack.), field oval or troublesome sedge (C. molesta Mack.), and prairie or Bicknell’s sedge (C. bicknellii Britton.), was tested to cold-wet stratification (28d) and perigynia removal in stored seed (10 months). Coastal dune vegetation plays a key role in dune formation and stabilization through sediment trapping and erosion control. To restore degraded dunes, revegetation of dune building species is critical. Planting density has been found to effect growth of marsh species, with closer plantings alleviating stress through facilitation. As coastal dunes are high stress environments, it is expected that dune species may also exhibit facilitative interactions based on the Stress Gradient Hypothesis. Forest communities are in a constant state of change. Disturbance events can alter the physical landscape and create conditions favorable to some species while negatively impacting others. Fire has been a natural, reoccurring source of disturbance in Pacific Northwest forests. Over the past centuries the fire paradigm has changed in favor of fire suppression. Forest roads permit access provide greater access but further fracture forest community’s continuity. The 2012 Table Mountain Fire and road influence have altered the plant community’s seed rain and vegetation. Plant virus genome replication and movement is dependent on host resources and factors. However, plants respond to virus infection through several mechanisms, such as autophagy, ubiquitination, mRNA decay and gene silencing, that target viral components. Viral factors work in synchrony with pro-viral host factors during the infection cycle and are targeted by antiviral responses. Accordingly, establishment of virus infection is genetically determined by the availability of the pro-viral factors necessary for genome replication and movement, and by the balance between plant defence and viral suppression of defence responses. Short Tandem Target Mimic (STTM) is a commonly technique used for functional studies of a number of genes in several plant model systems. However, very little is known about application of STTM technique in tree species. In this study, STTM was applied to knock down microRNA396 (miR396) in transgenic poplar trees for the first time. STTM396 expression resulted in dramatic decrease in miR396 expression levels leading to taller plants with larger leaves and larger leaf cell size. Boreal peatlands store approximately one third of the earth’s terrestrial carbon, locked away in currently waterlogged and frozen conditions. Peatlands of boreal and arctic ecosystems are affected increasingly by shifting hydrology caused by climate change. The consequences of these relatively rapid ecosystem changes on carbon cycling between the landscape and the atmosphere could provide an amplifying feedback to climate warming. Alternatively, the advancement of terrestrial vegetation into once waterlogged soils could uptake carbon as a sink. Broomcorn millet (Panicum miliaceum L.) is the most water-efficient cereal and one of the earliest domesticated plants. Here we report its high-quality, chromosome-scale genome assembly using a combination of short-read sequencing, single-molecule real-time sequencing, Hi-C, and a high-density genetic map. Phylogenetic analyses reveal two sets of homologous chromosomes that may have merged ~5.6 million years ago, both of which exhibit strong synteny with other grass species. Broomcorn millet contains 55,930 proteincoding genes and 339 microRNA genes. We find Paniceae-specific expansion in several subfamilies of the BTB (broad complex/tramtrack/bric-a-brac) subunit of ubiquitin E3 ligases, suggesting enhanced regulation of protein dynamics …

Identification of the nitrate sources that adversely impact groundwater quality is a necessary first step in the control of this major worldwide pollutant. The impact of nitrate leachate from urea-ammonium nitrate (UAN) (50% urea-N, 25% ammonium-N, 25% nitrate-N) fertilizer, whose use has increased dramatically in the last three decades largely because it can be applied through sprinkler irrigation systems to corn in all growth stages, is investigated. Heterogeneity has emerged as a fundamental principle for grassland management and the importance of environmental heterogeneity for biological diversity has raised questions about the appropriateness of grassland practices that seek to promote uniform grassland structure and composition. Principles of uniformity in grassland management reflect a utilitarian target of “managing for the middle” by minimizing both overgrazing and underutilization while avoiding or preventing fire and other disturbances that consume aboveground biomass. Screening is a strategy for detecting undesirable change prior to manifestation of symptoms or adverse effects. Although the well-recognized utility of screening makes it commonplace in medicine, it has yet to be implemented in ecosystem management. To study chlorophyll development time and overall photosynthetic development in C3 and C4 leaves, seeds were germinated in complete darkness and achlorophyllous leaves were then allowed to develop in lighted conditions. Corn (Zea mays, C4), sorghum (Sorghum bicolor, C4), green bean (Phaseolus vulgaris, C3), broad bean (Vicia faba, C3), and wheat (Triticum aestivum, C3) were investigated for the first ten days of sunlight exposure. Chlorophyll concentration, chlorophyll fluorescence, and CO2 gas exchange measurements were conducted daily on the first leaf that emerged after the embryonic leaves of each plant. The research project focused on evaluating 3 different factors in creating a plant hormone lab module for Principles of Biology II lab. The main hormone of focus was brassinosteroid. In the model plant Arabidopsis thaliana, brassinosteroids promote cell elongation, cell division, root growth, and seed germination. Three goals were established to create the most efficient lab module possible. First, mutant (bes1,bri1) root length was assessed without addition of brassinolide (BL), a synthetic brassinosteroid. Unequal sex ratios are widespread in dioecious plants and understanding their cause is important to understanding fundamental aspects of their population dynamics, and yet what causes biased sex ratios in plants is still poorly understood. Nitrogen provided to crops through mineralization is an important factor in N management guidelines. Understanding of the interactive effects of soil and weather conditions on N mineralization needs to be improved. Climate change is altering ecosystems on a global scale, creating novel ecological scenarios with which plant species must cope. Factors such as altered precipitation and fire regimes and non-native plant invasion may negatively affect native plant species, while interactions between these stressors could magnify their impacts. Plants have evolved a complex defense system against pests and pathogens utilizing many types of receptors, signaling factors, and defense compounds to detect pathogen presence and respond effectively. Since many pathogens have evolved immunesuppressing effectors used to reduce plant resistance, plants have evolved a family of receptors that detect pathogenic effectors as a result of an evolutionary arms race. These receptors contain Nucleotide-Binding Site and Leucine-Rich Repeat domains and are called NBS-LRR or NLR proteins. Water deficit is one of the major constraints on plant growth and development, causing reduction of crop productivity. To minimize water loss, among many adaptation strategies, plants close their stomata to reduce transpiration. The stomatal closure is regulated by light, internal CO2 concentration, and plant hormones, mainly abscisic acid. Tropical forests cycle one third of Earth’s carbon, yet we are still unsure how tropical vegetation will respond to climate warming. Tropical biomes experience a smaller temperature margin compared to other systems, possibly making them less capable of thermal adjustments. In addition, thermal responses of vegetation have been identified as one of the areas of greatest uncertainty for global carbon models. This dissertation works to quantify tropical forest photosynthetic responses to temperature as well as assessing physiological thermal acclimation of four tropical species. BACKGROUND: The increasing popularity of pulse-width modulation (PWM) sprayers requires that application interaction effects on spray pattern uniformity be completely understood to maintain a uniform overlap of spray, thereby reducing crop injury potential andmaximizing coverage on target pests. Cuscuta is a genus of nearly 200 obligate stem parasites with a nearly cosmopolitan distribution and considerable agricultural and ecological significance. Dodder seeds are considered “unspecialized”, with no morphological adaptations towards particular dispersal vectors; however, the seed coat anatomy has recently suggested an adaptation to endozoochory. This is the first attempt to provide a genus-wide overview of the diversity in morphology and anatomy of Cuscuta seeds, together with an assessment of the water gap and exploration of various form-function relationships. Virginia Mallow (Sida hermaphrodita) is a perennial herb of the Malvaceae family that is native to riparian habitats in northeastern North America. Throughout most of its geographical distribution however, it is considered threatened and only two populations are known from Canada. Some algal viruses have coding sequences for proteins with structural and functional characteristics of pore modules of complex K+ channels. Here we exploit the structural diversity among these channel orthologs to discover new basic principles of structure/function correlates in K+ channels. The analysis of three similar K+ channels with ≤ 86 amino acids (AA) shows that one channel (Kmpv1) generates an ohmic conductance in HEK293 cells while the other two (KmpvSP1, KmpvPL1) exhibit typical features of canonical Kir channels. Best practices in laboratory culture management often include cryopreservation of microbiota, but this can be challenging with some virus particles. By preserving viral isolates researchers can mitigate genetic drift and laboratory-induced selection, thereby maintaining genetically consistent strains between experiments. To this end, we developed a method to cryopreserve the model, green-alga infecting virus, Paramecium bursaria Chlorella virus 1 (PBCV-1). We explored cryotolerance of the infectivity of this virus particle, whereby freezing without cryoprotectants was found to maintain the highest infectivity (~2.5%). Accurate estimation of the nitrogen (N) spatial distribution of rice (Oryza sativa L.) is imperative when it is sought to maintain regional and global carbon balances. We systematically evaluated the normalized differences of the soil and plant analysis development (SPAD) index (the normalized difference SPAD indexes, NDSIs) between the upper (the first and second leaves from the top), and lower (the third and fourth leaves from the top) leaves of Japonica rice.
