Genetic Diversity In An Invasive Clonal Plant? A Historical And Contemporary Perspective, Elliot D. Weidow Aug 2018

Genetic Diversity In An Invasive Clonal Plant? A Historical And Contemporary Perspective, Elliot D. Weidow

LSU New Orleans Theses and Dissertations

Introduced populations of Eichhornia crassipes (Pontederiaceae) possess extremely low levels of genetic diversity due to severe bottleneck events and clonal reproduction. While populations elsewhere have been well studied, North American populations of E. crassipes remain understudied. We used Amplified Fragment Length Polymorphism markers to assess genetic diversity and population structure in North American E. crassipes populations. Patterns of diversity over the past fifty years were analyzed using herbarium specimens. Furthermore, we sampled populations across the Gulf Coast of the United States throughout a year to determine contemporary genetic diversity and assess potential seasonal effects. Genetic diversity was found to be …

Dna Barcoding Evidence For The North American Presence Of Alfalfa Cyst Nematode, Heterodera Medicaginis, Tom Powers, Andrea Skantar, Timothy Harris, Rebecca Higgins, Peter Mullin, Saad Hafez, Zafar Handoo, Tim Todd, Kirsten S. Powers

Department of Plant Pathology: Faculty Publications

Specimens of Heterodera have been collected from alfalfa fields in Kearny County, Kansas and Carbon County, Montana. DNA barcoding with the COI mitochondrial gene indicate that the species is not Heterodera glycines, soybean cyst nematode, H. schachtii, sugar beet cyst nematode, or H. trifolii, clover cyst nematode. Maximum likelihood phylogenetic trees show that the alfalfa specimens form a sister clade most closely related to H. glycines, with a 4.7% mean pairwise sequence divergence across the 862 nucleotides of the COI marker. Morphological analyses of juveniles and cysts conform to the measurements of H. medicaginis, the alfalfa cyst nematode originally described …

Tof-Sims Analysis Of Plant Seed Interactions With Plant Growth-Promoting Bacteria, Tammy Pheuphong, Rachel Komorek, Xiao-Ying Yu

STAR Program Research Presentations

The use of time-of-flight secondary ion mass spectrometry (ToF-SIMS) in plant biology is a relatively unexplored and quickly developing field. The majority of research in plant SIMS involves the application of ToF-SIMS to study dried wood tissues, and only a handful of studies apply SIMS on plant stems, roots, and/or seeds. Our project provides a brief description and review of previous work using SIMS on plant stems, roots, and/or seeds, along with an emphasis on the sample preparation in each study. Additionally, the use of Brachypodium distachyon (Brachypodium) as a model system for research on grasses has also become more …

Evaluation Of Glyphosate And Ppo-Inhibiting Herbicide Tank-Mixtures To Manage Glyphosate Resistance In Soybean, Jesaelen Gizotti De Moraes

Department of Agronomy and Horticulture: Dissertations, Theses, and Student Research

Protoporphyrinogen oxidase (PPO)-inhibiting herbicides in combination with glyphosate for postemergence (POST) applications is one of the primary alternatives to manage glyphosate-resistant weeds and the only effective POST chemical option in conventional and glyphosate-tolerant soybean to control glyphosate and ALS-inhibiting resistant weeds. Antagonistic interactions have been reported between many different herbicide modes of action and optimal droplet size may be affected by tank-mixtures of different herbicides. Additionally, the impact of adjuvants on the factors aforementioned as well as on physical properties needs to be thoroughly investigate to maximize herbicide efficacy. Therefore, the objectives of this research were to: 1) conduct greenhouse …

Ecological Drivers Of Plant Community Composition In A Patterned Fen At Seney National Wildlife Refuge, Michigan, Alex Graeff

All NMU Master's Theses

Regional variation in geomorphology, vegetation, fen landforms, and water chemistry create a variety of unique peatlands across the Northern Hemisphere. In the Great Lakes region, patterned fens have been extensively studied in northern Minnesota, but largely ignored in Michigan. The purpose of this study was to describe vegetation, landforms, and water chemistry in a patterned fen at Seney National Wildlife Refuge. Percent cover of plant species and environmental variables were measured at 298 relevé style plots across 6 landform types: triangular swamp forests, featureless water tracks, peripheral water tracks, strings, flarks, and ponded sedge lawns. Additionally, several morphometric parameters were …

Identification And Characterization Of Salinity Tolerance Genes By Activation Tagging In Arabidopsis, Jawaher Alkahtani Aug 2018

Identification And Characterization Of Salinity Tolerance Genes By Activation Tagging In Arabidopsis, Jawaher Alkahtani

Graduate Theses and Dissertations

Salinity often affects irrigated areas in arid and semi-arid regions of the world. The existence and accumulation of soluble salts in the soil layers limit the growth of crops essential for our food. Salt stress dramatically affects plant growth, plant development, as well as crop yield. Arabidopsis thaliana is the plant model that provides a comprehensive knowledge of plant development, genetics and physiology, and response to abiotic stresses such as salinity. The redundancy of genes due to duplication, even in the simple model genome of Arabidopsis, limits the value of knockout (KO) mutagenesis to provide complete information on gene function. …

Effect Of Allelic Variation In Rht Loci On Plant Height And Grain Yield Of Soft Red Winter Wheat (Triticum Aestivum L.), Habib Hayat Aug 2018

Effect Of Allelic Variation In Rht Loci On Plant Height And Grain Yield Of Soft Red Winter Wheat (Triticum Aestivum L.), Habib Hayat

Graduate Theses and Dissertations

Plant height in wheat (Triticum aestivum L.) is controlled in large part by two major Rht genes, Rht-B1 and Rht-D1, which pleiotropically impact lodging and grain yield. Prior to the Green Revolution, wheat varieties contained only ‘wild-type’ Rht alleles (Rht-B1a and Rht-D1a) and were tall and prone to lodging. Introgression of a semi-dominant mutation at either of these two loci (Rht-B1b or Rht-D1b) results in a semi-dwarf phenotype and reduced plant height. When combined (Rht-B1b and Rht-D1b) an extremely short double-dwarf phenotype is observed. The objective of this study was to determine the impact of allelic variation in Rht-B1 and …

Association Mapping And Genomic Selection For Yield And Agronomic Traits In Soft Winter Wheat, Dennis Bulusan Lozada Aug 2018

Association Mapping And Genomic Selection For Yield And Agronomic Traits In Soft Winter Wheat, Dennis Bulusan Lozada

Graduate Theses and Dissertations

Tools such as genome-wide association study (GWAS) and genomic selection (GS) have expedited the development of crops with improved genetic potential. While GWAS aims to identify significant markers associated with a trait of interest, the goal of GS is to utilize all marker effects to predict the performance of new breeding lines prior to testing. A GWAS for grain yield (GY), yield components, and agronomic traits was conducted using a diverse panel of 239 soft winter wheat (SWW) lines evaluated in eight site-years in Arkansas and Oklahoma. Broad sense heritability of GY (H2=0.48) was moderate compared to other traits including …

The environmental fate and potential impacts of nanopesticides on agroecosystems under realistic agricultural conditions are poorly understood. As a result, the benefits and risks of these novel formulations compared to the conventional products are currently unclear. Here, we examined the effects of repeated realistic exposures of the Cu(OH)2 nanopesticide, Kocide 3000, on simulated agricultural pastureland in an outdoor mesocosm experiment over 1 year. The Kocide applications were performed alongside three different mineral fertilization levels (Ambient, Low, and High) to assess the environmental impacts of this nanopesticide under low-input or conventional farming scenarios. The effects of Kocide over time were …

Cost Effective Strategy For The Synthesis Of Azo Dyes By Using Cashew Nut Shell Liquid (Cnsl), Mathias Yohana, Monica M. Ndoile, Quintino A. Mgani

Tanzania Journal of Science

The synthesis and dyeing properties of two types of azo dyes are reported in this paper. The two types of dyes were obtained from coupling of amino cardanol with unsaturated anacardic acid [ACUSA (Aminocardanol unsaturated anacardic acid)] and saturated anacardic acid [ACSA (Aminocardanol saturated anacardic acid)] separately. Both, anacardic acid and aminocardanol were obtained from CNSL as their source material. ACSA dyes were solid and dark brown while ACUSA were gelatinous and dark red. The performance of both ACUSA and ACSA dyes was moderate on textile fabrics with varying cotton content. The dyeing efficiency of the ACUSA was better than …

Semisynthesis Of A Mellein-Type 3,4-Dihydroisocoumarin From Cashew Nut Shell Liquid (Cnsl), Clarence A. Mgina, Fatma Kadir

Tanzania Journal of Science

Mellein-type 3,4-dihydroisocoumarin 39 (i.e., 8-methoxy-3-tridecyl-3,4-dihydroisocoumarin or 8-methoxy-3-tridecylisochroman-1-one), and its precursors, namely, methyl 2-methoxy-6-pentadecylbenzoate (37), methyl 2-methoxy-6-pentadecanoylbenzoate (38) and (E)-methyl 2-methoxy-6-(pentadec-1-enyl)benzoate (19) were synthesized from anacardic acid (12) as a starting material obtained from Cashew Nut Shell Liquid (CNSL) in an overall yield of 78%. The transformation of 12 to 39 involved protection of the reactive phenolic and carboxylic acid groups of compound 12 through methylation followed by hydrogenation so as to saturate the mono-, di- and tri-unsaturated C15 chains of anacardic acid (12). Subsequent benzylic oxidation and reduction of the keto functional group with concomitant dehydration of the alcohol led …

Utilizing Random Regression Models For Genomic Prediction Of A Longitudinal Trait Derived From High‐Throughput Phenotyping, Malachy T. Campbell, Harkamal Walia, Gota Morota

Department of Agronomy and Horticulture: Faculty Publications

The accessibility of high‐throughput phenotyping platforms in both the greenhouse and field, as well as the relatively low cost of unmanned aerial vehicles, has provided researchers with an effective means to characterize large populations throughout the growing season. These longitudinal phenotypes can provide important insight into plant development and responses to the environment. Despite the growing use of these new phenotyping approaches in plant breeding, the use of genomic prediction models for longitudinal phenotypes is limited in major crop species. The objective of this study was to demonstrate the utility of random regression (RR) models using Legendre polynomials for genomic …

Identification Of Genomic Regions Contributing To Protein Accumulation In Wheat Under Well-Watered And Water Deficit Growth Conditions, Ibrahim S. Elbasyoni, Sabah M. Morsy, Raghuprakash Kastoori Ramamurthy, Atef M. Nassar Jul 2018

Identification Of Genomic Regions Contributing To Protein Accumulation In Wheat Under Well-Watered And Water Deficit Growth Conditions, Ibrahim S. Elbasyoni, Sabah M. Morsy, Raghuprakash Kastoori Ramamurthy, Atef M. Nassar

Department of Agronomy and Horticulture: Faculty Publications

Sustaining wheat production under low-input conditions through development and identifying genotypes with enhanced nutritional quality are two current concerns of wheat breeders. Wheat grain total protein content, to no small extent, determines the economic and nutritive value of wheat. Therefore, the objectives of this study are to identify accessions with high and low grain protein content (GPC) under well-watered and water-deficit growth conditions and to locate genomic regions that contribute to GPC accumulation. Spring wheat grains obtained from 2111 accessions that were grown under well-watered and water-deficit conditions were assessed for GPC using near-infrared spectroscopy (NIR). Results indicated significant influences …

Leaf Micromorphology Aids Taxonomic Delineation Within The Hypervariable Genus Metrosideros (Myrtaceae) On O'Ahu, Gary L. Sur, Richard Keating, Elizabeth Stacy

Life Sciences Faculty Research

Variation in leaf micromorphology can aid delimitation of taxonomically difficult groups. The woody genus Metrosideros (Myrtaceae) dominates Hawai'i's native forests and comprises striking, predominantly infraspecific, macromorphological variation, including many forms unrecognized in current taxonomic treatments. On taxonomically rich O'ahu, 10 taxa (varieties or unnamed morphotypes of M. polymorpha and other species) occur in a predictable sequence with overlapping ranges from low to high elevation along the numerous leeward ridges of the Ko'olau Range. We used scanning electron and light microscopy to examine mature, nonsenescing sun leaves from each of these 10 taxa. Parametric and nonparametric tests were used to compare …

The Effect Of Kalanchoe Pinnata On The Cell Migration Of Astrocytomas, Jorge Rosa Jun 2018

The Effect Of Kalanchoe Pinnata On The Cell Migration Of Astrocytomas, Jorge Rosa

The International Student Science Fair 2018

Astrocytomas, from cell line 1321N1,are cancerous astrocytes. These are a type of glial cell in the brain which is responsible for maintaining synaptic contact and the signaling capacities among neurons. They also represent a fundamental element in controlling blood flow and defining brain micro structure. Due to the fact that this variant is cancerous, a condition commonly described as an abnormal cell cycle and uncontrolled reproduction of cells, they have the ability to metastasize.

In the metastatic process of cancer cells the initial phase is know as tumor invasion. This stage consists mainly of the cells migrating away from the …

Lipid Transport Required To Make Lipids Of Photosynthetic Membranes, Evan Labrant, Allison C. Barnes, Rebecca Roston Jun 2018

Lipid Transport Required To Make Lipids Of Photosynthetic Membranes, Evan Labrant, Allison C. Barnes, Rebecca Roston

Department of Biochemistry: Faculty Publications

Photosynthetic membranes provide much of the usable energy for life on earth. To produce photosynthetic membrane lipids, multiple transport steps are required, including fatty acid export from the chloroplast stroma to the endoplasmic reticulum, and lipid transport from the endoplasmic reticulum to the chloroplast envelope membranes. Transport of hydrophobic molecules through aqueous space is energetically unfavorable and must be catalyzed by dedicated enzymes, frequently on specialized membrane structures. Here, we review photosynthetic membrane lipid transport to the chloroplast in the context of photosynthetic membrane lipid synthesis. We independently consider the identity of transported lipids, the proteinaceous transport components, and membrane …

The mechanisms controlling the transcription of gene sets in specific regions of a plant embryo shortly after fertilization remain unknown. Previously, we showed that G564 mRNA, encoding a protein of unknown function, accumulates to high levels in the giant suspensor of both Scarlet Runner Bean (SRB) and Common Bean embryos, and a cis-regulatory module containing three unique DNA sequences, designated as the 10-bp, Region 2, and Fifth motifs, is required for G564 suspensor-specific transcription [Henry KF, et al. (2015) Plant Mol Biol 88:207–217; Kawashima T, et al. (2009) Proc Natl Acad Sci USA 106:3627–3632]. We tested the hypothesis that …

Nonpolar Residues In The Presumptive Pore‐Lining Helix Of Mechanosensitive Channel Msl10 Influence Channel Behavior And Establish A Nonconducting Function, Grigory Maksaev, Jennette K. Shoots, Simran Ohri, Elizabeth S. Haswell

Biology Faculty Research

Mechanosensitive (MS) ion channels provide a universal mechanism for sensing and responding to increased membrane tension. MscS‐like (MSL) 10 is a relatively well‐studied MS ion channel from Arabidopsis thaliana that is implicated in cell death signaling. The relationship between the amino acid sequence of MSL10 and its conductance, gating tension, and opening and closing kinetics remains unstudied. Here, we identify several nonpolar residues in the presumptive pore‐lining transmembrane helix of MSL10 (TM6) that contribute to these basic channel properties. F553 and I554 are essential for wild type channel conductance and the stability of the open state. G556, a glycine residue …

Isolating And Identifying Fungal Endophytes From Roots Of Rare Orchid Cypripedium Arietinum In Grand Sable Dunes, Michigan, Kari Farkas-Lasich

All NMU Master's Theses

Recently the population of Cypripedium arietinum in Grand Sable Dunes, Michigan was estimated at more than 3.5 million plants, greater than 90% of the world’s known population. There is little research on this species’ root associated fungal endophytes. This new population estimate allows the ethical, destructive sampling required to shed light on root associated symbionts and further successful preservation and restoration of the species. Samples were collected of current year’s roots from 75 C. arietinum orchids at 75 locations within 4 jack pine forests in the Grand Sable Dunes, Pictured Rocks National Lakeshore, Michigan. Fungal endophyte cultures were obtained from …

Pipecolic acid (Pip), a non-proteinaceous product of lysine catabolism, is an important regulator of immunity in plants and humans alike. In plants, Pip accumulates upon pathogen infection and has been associated with systemic acquired resistance (SAR). However, the molecular mechanisms underlying Pip-mediated signaling and its relationship to other known SAR inducers remain unknown. We show that in plants, Pip confers SAR by increasing levels of the free radicals, nitric oxide (NO), and reactive oxygen species (ROS), which act upstream of glycerol-3-phosphate (G3P). Plants defective in NO, ROS, G3P, or salicylic acid (SA) biosynthesis accumulate reduced Pip in their distal uninfected …
