Mitochondrial genomes (mitogenomes) of flowering plants are well known for their extreme diversity in size, structure, gene content, and rates of sequence evolution and recombination. In contrast, little is known about mitogenomic diversity and evolution within gymnosperms. Only a single complete genome sequence is available, from the cycad Cycas taitungensis, while limited information is available for the one draft sequence, from Norway spruce (Picea abies). To examine mitogenomic evolution in gymnosperms, we generated complete genome sequences for the ginkgo tree (Ginkgo biloba) and a gnetophyte (Welwitschia mirabilis). Background: Plant defense traits require resources and energy that plants may otherwise use for growth and reproduction. In order to most efficiently protect plant tissues from herbivory, one widely accepted assumption of the optimal defense hypothesis states that plants protect tissues most relevant to fitness. Reproductive organs directly determining plant fitness, including flowers and immature fruit, as well as young, productive leaf tissue thus should be particularly well-defended. To test this hypothesis, we quantified the cyanogenic potential (HCNp)—a direct, chemical defense—systemically expressed in vegetative and reproductive organs in lima bean (Phaseolus lunatus), and we tested susceptibility of these …

The interaction between lipid oxidation products and bovine sarcoplasmic (SP) and myofibrillar protein (MP) homogenates in the presence of green tea was investigated. To monitor the effect of green tea on lipid oxidation, aldehydes were measured while effect on protein was monitored via changes in myoglobin, thiols, and tryptophan fluorescence over nine days of refrigerated storage. The presence of SP and MP decreased free aldehydes in the buffers. The SP bound more aldehydes than MP. The tea compounds exhibited more favorable binding energies than aldehydes near histidine 64 close to the heme moiety of myoglobin. Addition of tea lowered tryptophan …

The bacterial pathogen Pseudomonas syringae depends on effector proteins secreted by its type III secretion system for the pathogenesis of plants. The majority of these effector proteins are known suppressors of immunity, but their plant targets remain elusive. Using Arabidopsis thaliana as a model host, we report that the HopE1 effector uses the host calcium sensor, calmodulin (CaM), as a co-factor to target the microtubule- associated protein 65 (MAP65), an important component of the microtubule network. HopE1 interacted with MAP65 in a CaMdependent manner, resulting in MAP65-GFP dissociation from microtubules. Transgenic Arabidopsis expressing HopE1 had reduced secretion of the immunity …

Corn (Zea mays L.) N use is of continued interest due to agronomic performance and environmental issues. This 2-yr study evaluated era hybrid response to fertilizer nitrogen (FN) rate in a factorial arrangement of one popular hybrid per five decades (1960–2000 eras) and five N rates (0–224 kg N ha–1). An additional hybrid per era was grown at 168 kg N ha–1. Hybrid productivity and nitrogen use efficiency (NUE) increased across the eras, but not between the 1980 and 1990 eras. The objective of the trial was to compare foliar fungicides applied to three growth stages of corn for grey leaf spot (GLS) and southern rust (SR) efficacy. Irrigated corn was grown based on Nebraska Extension irrigation recommendations at the South Central Ag Lab near Clay Center, NE. Soils were a silt loam with 6.7 pH and 1.8% OM and the previous crop was soybean. Reduced tillage was performed to the field prior to planting. Corn (DKC 65-79 RIB, tolerant to gray leaf spot) was planted at approximately 34,000 seed/A on 26 May. Eight treatments were arranged in a randomized complete …

Some plant-parasitic nematodes occur in every [sunflower] production field. Nematode populations likely include multiple species, although not all species cause notable disease. Symptoms depend on the species, the population density, and the crop's interaction with environmental factors. Several nematode species are known to cause disease in sunflower, although little research has focused on their impact or management in the crop. The highest population densities of plant-parasitic nematodes often occur in isolated areas that are randomly distributed in fields, as do the most severe plant symptoms. Symptoms are not necessarily diagnostic for nematodes, and the problem may be misdiagnosed. The most …

Sweet sorghum [Sorghum bicolor (L.) Moench] has potential for bioenergy. Itis adapted to a variety of U.S. locations and the extracted juice can be directly fermented into ethanol. However, little research on fungal stalk rots, diseases that pose serious constraints for yield and quality of juice and biomass, has been reported. A greenhouse bioassay was designed to assess charcoal rot (Macrophomina phaseolina) and Fusarium stalk rot (Fusarium thapsinum) in plants at maturity, the developmental stage at which these diseases are manifested. Tidal freshwater wetlands (TFW), which lie at the interface of saltwater and freshwater ecosystems, are predicted to experience moderate salinity increases due to sea level rise. Increases in salinity generally suppress CH4 production, but it is uncertain to what extent elevated salinity will affect CH4 cycling in TFW. It is also unknown whether CH4 production will resume when freshwater conditions return. The ability to produce CH4 is limited to a monophyletic group of the Euryarchaeota phylum called methanogens (MG), who are limited to a small number of substrates (e.g., acetate, H2, and formate) produced from the breakdown of fermentation products. …

Pathogen exposure to sublethal doses of fungicides may result in mutations that may represent an important and largely overlooked mechanism of introducing new genetic variation into strictly clonal populations, including acquisition of fungicide resistance. We tested this hypothesis using the clonal plant pathogen, Sclerotinia sclerotiorum. Nine susceptible isolates were exposed independently to five commercial fungicides with different modes of action: boscalid (respiration inhibitor), iprodione (unclear mode of action), thiophanate methyl (inhibition of microtubulin synthesis) and azoxystrobin and pyraclostrobin (quinone outside inhibitors). Pseudomonas syringae pv. tomato DC3000 (PtoDC3000) is an extracellular model plant pathogen, yet its potential to produce secreted effectors that manipulate the apoplast has been under investigated. Here we identified 131 candidate small, secreted, non-annotated proteins from the PtoDC3000 genome, most of which are common to Pseudomonas species and potentially expressed during apoplastic colonization. We produced 43 of these proteins through a custom-made gateway-compatible expression system for extracellular bacterial proteins, and screened them for their ability to inhibit the secreted immune protease C14 of tomato using competitive activity-based protein profiling. Nicotinic acetylcholine receptors (nAChRs) are targets for developing new drugs to treat severe pain, nicotine addiction, Alzheimer disease, epilepsy, etc. α-Conotoxins are biologically and chemically diverse. With 12–19 residues and two disulfides, they can be specifically selected for different nAChRs. Acetylcholine-binding proteins from Aplysia californica (Ac-AChBP) are homologous to the ligand-binding domains of nAChRs and pharmacologically similar. X-ray structures of the α-conotoxin in complex with Ac-AChBP in addition to computer modeling have helped to determine the binding site of the important residues of α-conotoxin and its affinity for nAChR subtypes. In parasitic plants, the reduction in plastid genome (plastome) size and content is driven predominantly by the loss of photosynthetic genes. The first completed mitochondrial genomes (mitogenomes) from parasitic mistletoes also exhibit significant degradation, but the generality of this observation for other parasitic plants is unclear. We sequenced the complete mitogenome and plastome of the hemiparasite Castilleja paramensis (Orobanchaceae) and compared them with additional holoparasitic, hemiparasitic and nonparasitic species from Orobanchaceae. Comparative mitogenomic analysis revealed minimal gene loss among the seven Orobanchaceae species, indicating the retention of typical mitochondrial function among Orobanchaceae species. Soybean (Glycine max) is a major plant source of protein and oil and produces important secondary metabolites beneficial for human health. As a tool for gene function discovery and improvement of this important crop, a mutant population was generated using fast neutron irradiation. Visual screening of mutagenized seeds identified a mutant line, designated MO12, which produced brown seeds as opposed to the yellow seeds produced by the unmodified Williams 82 parental cultivar. The new essential oil-Endemic and sub endemic plants in Outer Mongolian Gobi were isolated by hydro distillation from the aerial parts of Brachanthemum gobicum Krasch, B. mongolicum Krach, B. mongolorum Grub., Asterothamnus centrali-asiaticus Novopork, A. molliusculus Novopork., Ajania trifida (Turcz) Tzvel, A. achilleoides (Turcz) Tzvel., Ferula ferulaeoides (Stend) Eug. Kor., Artemisia davajamczii Darijima., A. caespitosa Ldb, A. subchrysolepis Filat, A. schischkinii Krasch, A.intricata Franch, A.ordasica Krasch, A.gobica (Krasch) Grub., A. sphalerocephala Krasch that grown in wild in Outer Mongolian Gobi. Background: Sorghum is an important C4 crop which relies on applied Nitrogen fertilizers (N) for optimal yields, of which substantial amounts are lost into the atmosphere. Understanding the genetic variation of sorghum in response to limited nitrogen supply is important for elucidating the underlying genetic mechanisms of nitrogen utilization.

Results: A bi-parental mapping population consisting of 131 recombinant inbred lines (RILs) was used to map quantitative trait loci (QTLs) influencing different agronomic traits evaluated under normal N (100 kg.ha−1 fertilizer) and low N (0 kg.ha−1 fertilizer) conditions. Reduced prolamin (zein) accumulation and defective endoplasmic reticulum (ER) body formation occurs in maize opaque endosperm mutants opaque2 (o2), floury2 (fl2), defective endosperm*B30 (DeB30), and Mucronate (Mc), whereas other opaque mutants such as opaque1 (o1) and floury1 (fl1) are normal in these regards. This suggests that other factors contribute to kernel texture. A liquid chromatography approach coupled with tandem mass spectrometry (LC-MS/MS) proteomics was used to compare non-zein proteins of nearly isogenic opaque endosperm mutants. It is critical to evaluate conservation practices that protect soil and water resources from climate change in the Midwestern United States, a region that produces one-quarter of the world’s soybeans and one-third of the world’s maize. An over-winter cover crop in a maize–soybean rotation offers multiple potential benefits that can reduce the impacts of higher temperatures and more variable rainfall; some of the anticipated changes for the Midwest. Erwinia tracheiphila is one of the most economically important pathogens of cucumbers, melons, squashes, pumpkins, and gourds in the northeastern and midwestern United States, yet its molecular pathology remains uninvestigated. Here, we report the first draft genome sequence of an E. tracheiphila strain isolated from an infected wild gourd (Cucurbita pepo subsp. texana) plant. The genome assembly consists of 7 contigs and includes a putative plasmid and at least 20 phage and prophage elements.

Cytoplasmic male sterility (CMS) has consistently been associated with the expression of mitochondrial open reading frames (ORFs) that arise from genomic rearrangements. Spontaneous fertility reversion in CMS has been observed in several cases, but a clear understanding of fertility reversion controlled by nuclear genetic influences has been lacking. Here, we identified spontaneous fertile revertant lines for Brassica juncea CMS cytoplasm in which the mitochondrial genome has undergone substoichiometric shifting (SSS) to suppress ORF220 copy number. Climate is the major factor affecting crop production; therefore, various agro-meteorological indicators have been frequently used to evaluate the impact of climate on crop production. In this study, we examined the temporal variations of agrometeorological indicators (growing degree days, total precipitation, dry spells and drought indices) during 1960-2014 and their impact on soybean yields in East Arkansas. Results show an increasing trend in growing degree days (GDDs) and dry spells, though the total precipitation during the soybean growing season remained nearly unchanged during the study period. Generally, GDDs and dry spells show a strong correlation with yields. We also evaluated …

In western North America, many low-elevation, dry forest types historically experienced frequent, low-severity fires. However, European settlement and fire suppression policies have contributed to over a century of fire exclusion, substantially altering forest structure and composition. There is considerable interest in restoring fire resilient characteristics to these forests through fuel reduction treatments. One limitation of current research on the impacts of fuel treatments is treatment longevity, as few studies have been able to quantify long-term responses to commonly applied treatments. Coast redwood (Sequoia sempervirens D. Don) depends on foliar water uptake from fog and rain to maintain water balance during the dry summer season. As fog and rain decline across the redwood region, understanding how foliar uptake capacity responds to climate has become increasingly important. This study investigated how three peripheral leaf traits that influence foliar water uptake, visible wax coverage, stomatal density, and guard cell length, vary across climatic gradients and how those traits affect uptake across 14 sites in California. An infectious cDNA clone of Triticum mosaic virus (TriMV) (genus Poacevirus; family Potyviridae) was used to establish three independent lineages in wheat to examine intra-host population diversity levels within protein 1 (P1) and coat protein (CP) cistrons over time. Genetic variation was assessed at passages 9, 18 and 24 by single-strand conformation polymorphism, followed by nucleotide sequencing. The founding P1 region genotype was retained at high frequencies in most lineage/passage populations, while the founding CP genotype disappeared after passage 18 in two lineages.We found that rare TriMV genotypes were present only transiently and lineages followed independent evolutionary trajectories, …
