Heterologous Expression Of Arabidopsis Thaliana Purple Acid Phosphatase Gene (Atpap15) In Crops For Phytoremediation Of Sites Contaminated With Excess Phosphorus, Jane Jeruto Bartonjo

AtPAP15 is one of the purple acid phosphatases expressed by Arabidopsis thaliana that has been extensively studied. Purified AtPAP15 has been shown to exhibit both phytase and phosphomonoesterase activities in acidic pH with maximal activity at pH 4.5. AtPAP15 is a phosphorus starvation inducible (PSI) gene that is expressed highly during phosphorus deficient conditions. In the current study, AtPAP15 was overexpressed in Nicotiana tabaccum under cauliflower mosaic virus (CaMV35S) constitutive promoter. After PCR confirmation of the gene, plants were transferred to the greenhouse and allowed to grow in pots. The flowering plant family Samydaceae was recently reinstated to include 14 genera and about 256 species of tropical trees and shrubs. Preliminary analyses of relationships in the family, however, indicated that the largest genus in the family, Casearia, is not monophyletic and that several smaller groups are probably misplaced. In order to affirm or refute those hypotheses, additional DNA sequence data with broader taxon sampling from the Old World were gathered for phylogenetic analysis. The effect of rainfall between urea application and flood establishment on N loss and grain yield of rice (Oryza sativa L.) has not been studied. The first research objective was to compare the effects of simulated rainfall amounts and N-(n-butyl) thiophosphoric triamide (NBPT) urease inhibitor rate on NH3 volatilization and rice growth. Three field experiments were conducted and NH3 volatilization was measured in two experiments for 11 days after urea application (DAU) in semi-open chambers. Invasive species present a threat to native communities and their introduction and expansion can alter community structure and dynamics. Multiple approaches can be employed for invasive species management including prevention and detection. In this study, microhabitat assessments were conducted on colonies of five species of invasive plants, Alliaria petiolata (M. Bieb.) Cavara & Grande, Lespedeza cuneata (Dum. Cours.) G. Fungal plant pathogens are persistent and global food security threats. To invade their hosts they often form highly specialized infection structures, known as appressoria. The cAMP/ PKA- and MAP kinase-signaling cascades have been functionally delineated as positive-acting pathways required for appressorium development. Negative-acting regulatory pathways that block appressorial development are not known. Here, we present the first detailed evidence that the conserved Target of Rapamycin (TOR) signaling pathway is a powerful inhibitor of appressorium formation by the rice blast fungus Magnaporthe oryzae. Introduction: Cell viability and metabolism depend on cytoplasmic water and solute content, and organisms have evolved mechanisms to sense changes in cell water content, solute concentrations, cell volume, and/or turgor. This Perspective addresses the response to osmotic challenge in land plants and describes their special dependence on cellular water status for growth and development. Understanding how plants cope with water limitation may allow us to mitigate the agricultural effects of drought, a critical limitation on global crop productivity that is likely to increase in severity as the climate changes (Long and Ort, 2010). Chiococceae are a monophyletic assemblage of morphologically very diverse groups of plants ranging in habit from subshrubs to shrubs to tall trees exhibiting an astonishing variation in shapes and sizes of corolla, and kinds of fruits and seeds. They are primarily distributed in the Neotropics but also occur in the West Pacific islands; thus exhibiting amphi-Pacific tropical disjunction. This study addresses the phylogenetic relationships and biogeography of the Chiococceae using molecular DNA sequence data, and presents novel data on the tribal and generic delimitations, intergeneric relationships, and the origin and dispersal of this group.

For the most part, the flowers of the 2015 VNPS Wildflower of the Year, Clethra alnifolia (Sweet Pepperbush), are unremarkable. Five separate sepals, 5 sepa rate petals, 10 stamens in 2 whorls, and a 3-carpellate superior ovary—an organization that can only be considered prosaic among the dicots. One floral feature, however, stands out: the anthers in the open flowers are upside-down! (See Figure 1A.) Further, these upside-down anthers open by pores (Figures 1B, 1C) rather than longitudinal slits, as in most flowering plants. Despite long-standing interest in elevational-diversity gradients, little is known about the processes that cause changes in the compositional variation of communities (β-diversity) across elevations. Recent studies have suggested that β-diversity gradients are driven by variation in species pools, rather than by variation in the strength of local community assembly mechanisms such as dispersal limitation, environmental filtering, or local biotic interactions. However, tests of this hypothesis have been limited to very small spatial scales that limit inferences about how the relative importance of assembly mechanisms may change across spatial scales. In recent years, the Santa Monica Mountains (SMM) have been plagued by frequent wildfires which threaten the native chaparral species. Nonsprouting chaparral species are completely killed by a fire, but their seeds germinate in response to fire cues. Facultative sprouters both resprout after a wildfire and release seeds that germinate post-fire. This project is based on data collected since 1986 at a biological preserve adjacent to the Malibu campus of Pepperdine University with an average fire return interval of 7.5 years. Understanding the relationship between photosynthesis, net primary productivity and growth in forest ecosystems is key to understanding how these ecosystems will respond to global anthropogenic change, yet the linkages among these components are rarely explored in detail. We provide the first comprehensive description of the productivity, respiration and carbon allocation of contrasting lowland Amazonian forests spanning gradients in seasonal water deficit and soil fertility. Using the largest data set assembled to date, ten sites in three countries all studied with a standardized methodology, we find that (i) gross primary productivity (GPP) has a simple relationship with seasonal water deficit, but …

Tan spot and Stagonospora nodorum blotch (SNB), often occurring together, are two economically significant diseases of wheat in the Northern Great Plains of the United States. They are caused by the fungi Pyrenophora tritici-repentis and Parastagonospora nodorum, respectively, both of which produce multiple necrotrophic effectors (NE) to cause disease. In this work, 120 hard red winter wheat (HRWW) cultivars or elite lines, mostly from the United States, were evaluated in the greenhouse for their reactions to the two diseases as well as NE produced by the two pathogens. Spring fires in tallgrass prairie can create environmental conditions conducive to plant growth in the subsequent growing season. Following fires, burned areas can also be attractive to grazing animals such as bison (Bison bison). Sustained grazing activity within recently burned areas can alter vegetation structure relative to nearby landscape patches that haven’t burned recently. In 2007, we collected data on bison grazing activity, vegetation structure, and the growth and reproduction of a perennial forb, Arnoglossum plantagineum, in Oklahoma tallgrass prairie. Posttranslational modification of proteins by small ubiquitin-like modifier (SUMO) is required for survival of virtually all eukaryotic organisms. Attachment of SUMO to target proteins is catalyzed by SUMO E2 conjugase. All haploid or diploid eukaryotes studied to date possess a single indispensable SUMO conjugase. We report here the unanticipated isolation of a Chlamydomonas reinhardtii (mutant5 [mut5]), in which the previously identified SUMO conjugase gene C. reinhardtii ubiquitin-conjugating enzyme9 (CrUBC9) is deleted. This surprising mutant is viable and unexpectedly displays a pattern of protein SUMOylation at 25°C that is essentially identical to wild-type cells. However, unlike wild-type …

The cell wall consists of cellulose microfibrils embedded within a matrix of hemicellulose and pectin. Cellulose microfibrils are synthesized at the plasma membrane, whereas matrix polysaccharides are synthesized in the Golgi apparatus and secreted. The trafficking of vesicles containing cell wall components is thought to depend on actin-myosin. Here, we implicate microtubules in this process through studies of the kinesin-4 family member, Fragile Fiber1 (FRA1). In an fra1-5 knockout mutant, the expansion rate of the inflorescence stem is halved compared with the wild type along with the thickness of both primary and secondary cell walls.   1. The leaf area-to-sapwood area ratio (LA:SA) is a key plant trait that links photosynthesis to transpiration. The pipe model theory states that the sapwood cross-sectional area of a stem or branch at any point should scale isometrically with the area of leaves distal to that point. Optimization theory further suggests that LA:SA should decrease toward drier climates. Although acclimation of LA:SA to climate has been reported within species, much less is known about the scaling of this trait with climate among species.

Agrobacterium tumefaciens is able to infect a diverse array of plants and causes crown gall disease. Typically these bacteria attach to plant roots and transform the plant cells to induce tumors. The mechanism of this attachment in the infection process is not yet fully understood. Using wild type Arabidopsis thaliana, Columbia-0, and several Arabidopsis mutant lines as a binding target, we screened for A. thaliana mutants with altered adhesion.

The objective of the trial was to compare industry fungicide programs with the microbial Regalia Rx plus fungicide tank mixtures for gray leaf spot (GLS) efficacy. Corn was grown under normal, irrigated agronomic practices at the South Central Ag Lab near Clay Center, NE. Soils were a silt loam with 6.7 pH and 1.8 % organic matter. Reduced tillage was performed to the field prior to planting. Corn (DKC 64-83 RIB, moderately susceptible to GLS) was planted at approximately 34,000 seed/A on 20 May. Auxin is the primary hormone responsible for plant growth and development. Regulation of auxin-response genes occurs through transcriptional activators (auxin response factors called ARFs) which bind to auxin response elements (AuxREs). Some auxin-responsive genes encode aminocyclopropane-1-carboxylic acid synthase (ACS) enzymes which regulate the production of the plant hormone ethylene. The major research objective was to evaluate transcriptional cross talk between auxin and ethylene. Both AuxREs and ethylene response elements were found in several ACS and ARF genes, suggesting cross talk between the two hormones at the transcriptional level. Freshwater Silica-Scaled Heterotrophic Protista: Heliozoa, Thaumatomonad Flagellates, Amoebae, And Bicosoecids, From The Lake Itasca Region, Minnesota, Daniel E. Wujek

Forty-nine plankton samples were collected from the Lake Itasca Region, Minnesota over a period sporadically covering the summers of 1980, 1981 and 1987. A total of 22 freshwater heterotrophic siliceous-scaled species were observed: 18 heliozoa, two thaumatomonad flagellates, one bicosoecid, and one testate amoeba. Scale identifications were based on transmission electron microscopy. New records for North America include two heliozoans and one thaumatomonad flagellate. Five heliozoa taxa and one thaumatomonad flagellate are new records for the U.S. Wujek DE. Freshwater silica-scaled heterotrophic protista: heliozoa, thaumatomonad flagellates, amoebae, and bicosoecids, from the Lake Itasca Region, Minnesota.

Fire suppression has nearly eliminated fire as a disturbance in temperate deciduous forests. Lack of fire is transforming these ecosystems through a positive feedback loop termed mesophication: cool, damp, shady conditions become continually more favorable for a few mesophytic species, while deteriorating for diverse array of heliophytic ones. Disturbances caused by urbanization fragment and degrade remnant forests. In urban settings, human management (or lack thereof) is often a dominate driver of succession. Repeated applications of fungicides with a single mode of action are believed to select for pre-existing resistant strains in a pathogen population, while the impact of sub-lethal doses of such fungicides on sensitive members of the population is unknown. In this study, in vitro evidence is presented that continuous exposure of Monilinia fructicola mycelium to some fungicides can induce genetic change in form of transposon transposition. Downy mildew (Plasmopara viticola) and botrytis bunch rot (Botrytis cinerea) are important diseases in the highlands of Santa Catarina State, a relatively new wine-growing region in Brazil. Seeds of members of the genus Cuphea accumulate medium-chain fatty acids (MCFAs; 8:0–14:0). MCFA- and palmitic acid- (16:0) rich vegetable oils have received attention for jet fuel production, given their similarity in chain length to Jet A fuel hydrocarbons. Studies were conducted to test genes, including those from Cuphea, for their ability to confer jet fuel-type fatty acid accumulation in seed oil of the emerging biofuel crop Camelina sativa.
