Interisland Variability Of Dune Plant Community Structure On Virginia's Barrier Islands, Justin K. Shafer

The barrier islands of the Virginia Coast Reserve LTER site provide an opportunity to study interisland variability of dune plant communities. My research focused on the variation in biomass and diversity of communities among islands and dune positions. Grassy dunes of young, intermediate and old age were sampled on Smith, Hog, and Parramore Islands. Aboveground biomass was obtained from harvest plots, and roots were extracted from cores. Data were collected on depth to groundwater and total soil nitrogen. Variation in mean aboveground biomass was evident among the islands with the highest values on Hog Island, followed by Smith, then Parramore …

Sarracenia purpurea is a rare wetland plant in Virginia and a threatened species in Maryland, with two potential subspecies in the region. I utilized restriction fragments from the intron of the chalcone synthase gene to compare S. purpurea populations and determine whether the subspecies concept was supported. I performed a census of existing populations, compiled all known historical data on the species, and investigated the reasons for the species demise and predicted dates of extinction. Bloom phenology was examined to see if climate change may have influenced bloom period. Soil, vegetation, and climatic information was obtained to determine if taxonomic …

The bacterial plant pathogen Pseudomonas syringae uses a type III protein secretion system to inject type III effectors into plant cells. Primary targets of these effectors appear to be effector-triggered immunity (ETI) and pathogen-associated molecular pattern (PAMP)-triggered immunity (PTI). The type III effector HopG1 is a suppressor of ETI that is broadly conserved in bacterial plant pathogens. Here we show that HopG1 from P. syringae pv. tomato DC3000 also suppresses PTI. Interestingly, HopG1 localizes to plant mitochondria, suggesting that its suppression of innate immunity may be linked to a perturbation of mitochondrial function. Sorghum bicolor is considered to be an allelopathic crop species, producing phytotoxins such as the lipid benzoquinone sorgoleone, which likely accounts for many of the allelopathic properties of Sorghum spp. Current evidence suggests that sorgoleone biosynthesis occurs exclusively in root hair cells and involves the production of an alkylresorcinolic intermediate (5-[(Z,Z)-8',11',14'-pentadecatrienyl]resorcinol) derived from an unusual 16:3D9,12,15 fatty acyl-CoA starter unit. This led to the suggestion of the involvement of one or more alkylresorcinol synthases (ARSs), type III polyketide synthases (PKSs) that produce 5-alkylresorcinols using medium to long-chain fatty acyl-CoA starter units via iterative condensations with malonyl-CoA. Triticum mosaic virus (TriMV), the type member of the newly proposed Poacevirus genus, and Wheat streak mosaic virus (WSMV), the type member of Tritimovirus genus of the family Potyviridae, infect wheat naturally in the Great Plains and are transmitted by wheat curl mites. In this study, we examined the ability of these viruses to infect selected cereal hosts, and found several differential hosts between TriMV and WSMV. Additionally, we examined the interaction between WSMV and TriMV in three wheat cultivars at two temperature regimens (19 and 20 to 26°C), and quantified the virus concentration in single and double infections …

Very-long-chain fatty acids (VLCFAs) are essential for many aspects of plant development and necessary for the synthesis of seed storage triacylglycerols, epicuticular waxes, and sphingolipids. Identification of the acetyl-CoA carboxylase PASTICCINO3 and the 3-hydroxy acyl-CoA dehydratase PASTICCINO2 revealed that VLCFAs are important for cell proliferation and tissue patterning. Here, we show that the immunophilin PASTICCINO1 (PAS1) is also required for VLCFA synthesis. Impairment of PAS1 function results in reduction of VLCFA levels that particularly affects the composition of sphingolipids, known to be important for cell polarity in animals. Plants respond to virus infections by activation of RNA-based silencing, which limits infection at both the single-cell and system levels. Viruses encode RNA silencing suppressor proteins that interfere with this response. Wild-type Arabidopsis thaliana is immune to silencing suppressor (HC-Pro)-deficient Turnip mosaic virus, but immunity was lost in the absence of DICER-LIKE proteins DCL4 and DCL2. Systematic analysis of susceptibility and small RNA formation in Arabidopsis mutants lacking combinations of RNA-dependent RNA polymerase (RDR) and DCL proteins revealed that the vast majority of virus derived small interfering RNAs (siRNAs) were dependent on DCL4 and RDR1, although full antiviral defense …

Home gardens, “the peridomestic area belonging to the household where members plant and/or tend useful plants” (Perrault-Archambault and Coomes 2008), are found throughout the world. However, their use and importance vary from region to region. In the Peruvian Amazon, owners use home gardens for a domestic supply of foods, craft materials, medicines, condiments, and shade (Miller and Nair 2006). With this wide range in function, reflected in species content, home gardens are very biodiverse.

A total of 206 root samples were collected from ornamental plants growing in ornamental nurseries and various landscapes in Florida. Isozyme phenotypes, especially esterase (EST) and malate dehydrogenase (MDH) were the main methods used to identify the root-knot nematode species. When needed, the morphology of female perineal patterns, morphometric characters and mitochondrial DNA were used to aid in the identification. Six Meloidogyne spp., M. arenaria , M. floridensis , M. graminis , M. incognita , M. javanica and M. mayaguensis were found infecting ornamental plants in Florida. Myxomycetes, the true slime molds, are highlighted in research and teaching that emphasizes various stages of the life cycle as experimental models. Past and current phylogenetic classifications of Myxomycetes on the tree of life are presented. Life cycle stages are illustrated, described, and discussed. Simple laboratory demonstrations and experiments are described that include spore germination, spore release, and moist chamber cultures utilizing organic matter from various microhabitats. Novel compounds isolated from fruiting bodies and plasmodia of 22 myxomycete species are tabulated, some of which exhibit biological activity that function as antibiotics, antimicrobials, and are cytotoxic to cancer cells. Human-mediated nutrient amendments have widespread effects on plant communities. One of the major consequences has been the loss of species diversity under increased nutrient inputs. The loss of species can be functional group dependent with certain functional groups being more prone to decline than others. We present results from the sixth year of a long-term fertilization and litter manipulation study in an old-field grassland. We measured plant tissue chemistry (C:N ratio) to understand the role of plant physiological responses in the increase or decline of functional groups under nutrient manipulations. Shiga toxin (Stx) produced by the invasive Shigella dysenteriae serotype 1 (S. dysenteriae1) causes gastrointestinal and kidney complications. It has been assumed that Stx is released intracellularly after enterocyte invasion by S. dysenteriae1. However, there is little information about Stx distribution inside S. dysenteriae1-infected enterocytes. Here, we use intestinal epithelial T84 cells to characterize the trafficking of Stx delivered into the cytosol, in ways that mimic aspects of S. dysenteriae1 infection. We find that cytoplasmic Stx is transported into nucleoli. Stx nucleolar movement is carrier- and energy-dependent. Stx binding to the nucleoli of normal human …

We performed a comparative study of the vascular flora of a serpentine outcrop, Pine Hill, and that of a granite outcrop, Settlement Quarry, from Little Deer Isle and Deer Isle, respectively, Hancock County, Maine. We established four transects along a gradient from exposed to forested areas within each outcrop. Plants were recorded for presence and percent cover from circular plots along each transect. Soil and tissue samples were collected to examine soil-tissue elemental relations. One hundred thirty-two taxa were recorded from serpentine and 89 from granite. An epidemic of cytospora canker [Valsa melanodiscus, anamorph Cytospora umbrina] is associated with dieback and mortality of Alnus incana ssp. tenuifolia in the Southern Rocky Mountains and had begun by the late 1980s. Isolations showed that inoculum was often abundant on bark and bud surfaces even in winter, especially near diseased stems. The pathogen was occasionally isolated from internal tissues of dormant buds and asymptomatic wood and phloem. In infected stems, the pathogen was detected up to 5 cm beyond visible canker margins. The data suggest that the pathogen can cause latent infections, but the infection court …

Wildfires in the Santa Monica mountains in Southern California burn down a decent percentage of the local vegetation. However, some plants like Heteromeles Arbutifolia, are considered to be resprouters because they return a mere two years after being burnt down. It is hypothesized that the artificially browsed resprout leaves will have a lower tensile strength than both the young and adult leaves because they use a lower amount of carbohydrates per unit-leaf-area. Our research is concerned with the further exploration of leaf biomechanics, a relatively unexamined field. The purpose of this experiment was to find a correlation between environmental stress factors and tensile strength of leaves of Encelia californica. We hypothesized the beach species would have a higher tensile strength than canyon species. After evaluating data collected from both Pepperdine’s Presidents Canyon and Malibu beach we found that while the canyon species could withstand a higher maximum load of force, there was no difference between the canyon and the beach species with leaf area taken into account. MicroRNAs (miRNAs) are 21–24 nucleotide riboregulators, which selectively repress gene expression through transcript cleavage and/or translational inhibition. It was thought that most plant miRNAs act through target transcript cleavage due to the high degree of complementarity between miRNAs and their targets. However, recent studies have suggested widespread translational inhibition by miRNAs in plants. The mechanisms underlining translational inhibition by plant miRNAs are largely unknown, but existing evidence has indicated that plants and animals share some mechanistic similarity of translational inhibition. This experiment sought to establish a connection between short term growth in Salvia leucophylla found in Southern California and nutrient availability in the soil. It was hypothesized that adding artificially produced nutrient mix Miracle Grow would increase the strength of the leaves in the herb after one week of addition. Newer leaves were tested in the Instron device for tensile strength after one week, and after data analysis, it was concluded that there was no difference in tensile strength of leaves treated with fertilizer compared with leaves without fertilizer.

Southern California and the Santa Monica Mountains experience a climate similar to that of the Mediterranean basin. This means hot, dry summers, mild, wet winters, and large amounts of plant diversity. A major issue that can affect these regions is water availability and processing. Presented here is a comparison of the predawn fluorescence of adult Heteromeles arbutifolia growing on the Pepperdine Seaver campus next to the lacrosse field, and adult Heteromeles growing in Tapia canyon. The analysis of the leaf temperature measured by an IR thermometer showed an average of 9.32°C drop when measuring plants in Tapia canyon compared to those growing on the warmer Pepperdine campus. The air temperature measured by the Kestrel and IR thermometer showed Tapia canyon being an average of 7.27°C colder than predawn measurements on Pepperdine campus. This predation has become more pronounced as the climate shifts to hotter, drier, and longer summers. We hypothesized that there would be difference in photosynthetic trade-offs and capabilities between coastal and inland populations, specifically with respect to leaves. In the botanical field of study, invasive plant species can choke out native species thus making them endangered or extinct.
