Little Bluestem (Schizachyrium scoparium) is a North American perennial grass that is able to tolerate high heat and droughts. This is possible by its extensive underground root network that taps deep into the ground for resources. Within the same soil systems are fungal endophytes, which are symbionts that live within plant tissues including roots. Soil microbial communities are increasingly recognized as an important factor in sustainable agriculture. Arbuscular mycorrhizal fungi (AMF), are key players in these communities, known to benefit their plant host in a number of ways, such as increased nutrient uptake. Plants can partner with multiple AMF species; however, little is known about how interactions between multispecies communities of AMF impact plant growth and yield. Epichloë is a well-known genus of vertically transmitted fungal endophytes in many species of grasses. Asimina triloba, commonly known as the pawpaw, is a unique and historically significant native fruit tree of the eastern United States. This research paper explores the taxonomy, botanical description, distribution, habitat requirements, ecological interactions, economic importance, and reproductive biology of A. triloba. The species is distinguished by its tropical-flavored fruits, adaptation to temperate climates, and reliance on clonal reproduction and specific insect pollinators. Secondary compounds produced by terrestrial invasive plants can cross terrestrial-aquatic boundaries via senesced leaves, leading to altered patterns of aquatic biodiversity and ecosystem functioning. Prior attempts to examine the impacts of invasive leaves on wetland communities have primarily employed the use of leaf leachates and focal taxa inhabiting relatively low trophic positions, and additional studies are needed to examine impacts on higher-order consumers and potential additive/synergistic effects of leachates and leaf decomposition. Global warming is gradually affecting the local climate causing pollinators and plants to change their emergence and flowering times. This can potentially cause a phenology mismatch between pollinator activity and open flowers leading to limitation in seed production in plants. However, other factors such as intense predation or competition can also shift phenology. Eastern Michigan University (EMU) owns an 86-acre property in northwest Michigan called the Parsons Center for Arts and Sciences. No botanical research has been done at the Center. A comprehensive plant species list will provide important resources for EMU students, all Parsons users (e.g., artists and community members), and county biologists. This research aims to collect and identify as many plant species at the site as possible and create two online, publicly available databases of the plant list and collection. Such a collection will bring awareness to the Parsons Center and add to its botanical value.

Mounting evidence of pollinator decline and its repercussions on agriculture and plant diversity means that scientists must work to understand the details of plant-pollinator interactions. Though there are many facets to the relationship between plants and pollinators, one major mediator of this relationship is scent. Hybridization is a common problem in conservation. It can be difficult to identify hybrid individuals but doing so is often necessary in order to make conservation recommendations. For example, botanical gardens and seed banks want to identify if individuals are hybrids of two or more species to determine if offspring could be used for conservation or restoration, and to analyze the risk of possible extinction via introgression. Our project looks at how well the program STRUCTURE can detect and cluster hybrids, their parent species, and other pure species, which we created using simulations. Most flowers are occupied by diverse communities of microbes, which can alter floral attractiveness to key pollinators and floral fitness by interacting with floral traits. Different species of flower often acquire unique communities of microbes. Consequently, pollinators are frequently forced to choose between two flowers with distinct floral microbiomes while foraging. However, it is currently unknown whether pollinators have an inherent preference for flowers containing one floral microbiome over another, and whether microbial preference is modified by variation in floral traits. Arbuscular mycorrhizal fungi (AMF) associate with 80% of vascular plant roots and are an important part of nutrient exchange. Our research quantified the relationship between (AMF) and native Pasture Thistle (Cirsium discolor) and invasive Canada Thistle (Cirsium arvense). We hypothesized that Pasture Thistle would have a stronger relationship with AMF than Canada Thistle because of the degraded mutualism hypothesis, which states that invasive species have reduced relationships with AMF. For our experiment, we grew inoculated and non-inoculated plants at 10 different levels of soil phosphorus concentrations. Pasture Thistle and Canada Thistle were both facultatively dependent upon …

The evolution of sex chromosomes is a ubiquitous and essential diversifying mechanism in eukaryotes. Yet early stages of sex-chromosome differentiation are poorly understood. The subdioecious octoploid, Fragaria virginiana, provides a rare opportunity to study the beginnings of sex chromosome evolution due to its young, polymorphic ZW sex system. Females are ZW and males/hermaphrodites are ZZ. F. virginiana has a small sex-determining region (SDR) located on the female W chromosome that has undergone three transposition events between the strawberry's four subgenomes, resulting in three SDR haplotypes: alpha (the result of a first transposition and closest to a pre-sex chromosome ancestral …

For perennial plants, the optimal timing of flowering is a tradeoff between survivorship and fecundity, a classic example of bet-hedging. For many monocarpic perennials, there is an advantage in delaying flowering until the plant reaches a larger size and is thus capable of a higher fecundity. However, in delaying flowering, there is a risk that the plant will die before reproducing at all. Here, we investigate this fitness trade-off in the imperiled dune thistle, Cirsium pitcheri. Protein prenylation, a common lipid post-translational modification, is required for growth and development in eukaryotes. Rab geranylgeranylation involves the addition of one or two 20-carbon geranylgeranyl moieties to Rab-GTPase target proteins, which regulate intracellular vesicle trafficking. The reaction is carried out by heterodimeric Rab geranylgeranyltransferase (Rab-GGT), which is composed of two associated α- and β-subunits, with the assistance of an additional protein called Rab escort protein (REP). Loss of function of the Rab-GGT α subunit RGTA1 has not been reported in any plant. While knockout of either of the two β subunits RGTB1 or RGTB2 results in …

As global change persists, changes in resource availability can influence plant-microbe interactions. To understand how resource availability can influence these interactions and species diversity, I focused this research on how varying nitrogen (N) levels affect root endophyte communities in the plant species Ammophila breviligulata, an ecosystem engineer in the dune system. I analyzed the relationship between nitrogen addition and microbial community composition across 60 plots treated with three nitrogen addition levels (control, low, and high) in a long-term experimental field site in the Lake Michigan dunes. Prenylation is a type of post-translational modification in which a 15- or 20-carbon lipid is added to the carboxyl (C) terminus of the protein. Arabidopsis thaliana contains the PROTEIN PRENYLTRANSFERASE ALPHA SUBUNIT-LIKE (PPAL) gene, which encodes a protein with homology to the α-subunits of the three known prenylation enzymes, PFT, PGGT, and Rab-GGT. We previously identified two mutations in PPAL, one of which is ppal-1, which contains a T-DNA insertion in the fourth intron. Protein prenylation is a post-translational process where lipids are added to carboxyl end groups of amino acids, which allows proteins to function properly in the eukaryotic cell. The job of prenylation is to target certain proteins to specific membranes and promote desirable protein-protein interactions. In our study we used reverse genetics techniques to investigate the function of protein prenylation in plant development. Protein Prenylation is the addition of lipids to select proteins that play a key role during the development of plants. There are three enzymes that play a role in protein prenylation: protein farnesyltransferase (PFT), protein geranylgeranyl-transferase-I (PGGT), and Rab geranylgeranyltransferase (Rab-GGT). However, there is a protein called PPAL that has a similar alpha subunit of PFT, PGGT, and RAB-GGT but its biochemical function is unknown. Protein prenylation is a post-translational modification that involves the addition of lipid groups to the carboxy end of a target protein and is necessary for protein activity. Prenylation has important roles in the cell: targeting and localizing proteins to subcellular compartments and promoting protein-protein interactions. As a highly conserved process in eukaryotes, prenylation is especially important for plant development. Iron is an essential element required for plants to carry out metabolic functions such as photosynthesis, heme biosynthesis, and chlorophyll biosynthesis. Within Arabidopsis thaliana, eight ferric reduction oxidase (FRO) genes function in iron uptake and homeostasis with tissue specific expression. However, little else is known regarding the biological role of FROs. Recent studies identify the FRO gene family as particularly responsive to the green leaf plant derived volatile (GLV) cis-3-hexenyl acetate (z3HAC). Unusual monoenoic fatty acids (UMFA’s) and specialized metabolites called anacardic acids (AnAc) are produced in glandular trichomes of Pelargonium ´ hortorum (geranium). The UMFA’s, 16:1∆11 and 18:1∆13 are precursors for the synthesis of unsaturated AnAc 22:1n5and 24:1n5 that contribute to pest resistance in geraniums. UMFAs and their derived AnAc metabolites not only provide a useful biological marker that differentiates the biosynthetic pathway for unusual mononenes from the common fatty acids (i.e. stearic, palmitic, oleic, linoleic and linolenic) but also have industrial, medical and agricultural applications. Counter to what some people think, urban areas can be biodiversity hotspots. Maintaining this biodiversity can be challenging, since exotic shrubs and vines block sunlight and threaten native plant regeneration. Since 2007, the Louisville Olmsted Parks Conservancy (LOPC) has spent $2 million on invasive plant management in Cherokee Park. Before the project began, long-term transects were established by the LOPC to collect baseline presence/absence data on 11 invasive plant species. In 2014, I revisited these transects and documented presence/absence data on the entire plant community.
