Sorghum bicolor (L.) Monech (sorghum) is cultivated as food for humans and livestock and is valued for its low input requirements. However, sorghum grain protein is deficient in the essential amino acid lysine and has poor protein digestibility because of the abundance of proline-rich kafirins that constitute >70% of proteins that form low-digestibility protein bodies. To reduce kafirin expression in the endosperm, and elicit wholesale proteome rebalancing to increase non-kafirin proteins, a single-guide RNA (sgRNA) Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR/Cas9) construct was previously used to target members of the highly-repetitive alpha-kafirin family, k1C in RTx430 sorghum. As riparian communities become increasingly degraded, restoration treatments are being implemented in streams to recover ecosystem functions. In the Jemez Mountains, beaver dam analogs (BDAs) are used to restore degraded streams by connecting the floodplain and increasing the extent of anoxic soil conditions. These environmental disturbances are expected to alter how riparian plants recruit, particularly their allocation to seed-based and vegetative regeneration strategies. We investigated whether BDAs and associated changes in soil water filled pore space (WFPS) affected riparian plant recruitment. Inland Pacific Northwest prairie is an endangered ecosystem that has been reduced by conversion to agriculture. There is growing interest in the role of soil microorganisms in prairie ecosystems, which may be crucial for restoration of the plant community through interspecies interactions within the soil near plant roots. One method for improving soil microorganism community composition is compost tea: enriched liquid compost extract. Compost tea is used in regenerative agriculture to improve crop yields and protect against pathogens, ostensibly through the activity of the added compost microbes. Soil health is foundational to sustainable agriculture, environmental resilience, and long-term food security. Yet, the adoption of soil health assessments remains limited, often perceived as too technical, costly, or difficult to implement in real-world contexts. To better understand these challenges, the first chapter presents findings from a statewide survey of 41 soil health practitioners in Nebraska, including farmers, extension educators, NRCS staff, and conservation professionals. Respondents highlighted the need for simpler, more accessible, and context-specific tools for soil health assessment that help practitioners make informed decisions aligned with their management goals. Water availability is the primary constraint for plant survival and reproduction in hyper-arid ecosystems. In such environments, plant fitness depends on an alignment between physiological traits and limited water resources. The Namib Desert of Namibia exemplifies this extreme selection pressure. Within the Namib Desert, Lithops ruschiorum, a succulent endemic to the gravel plains, exhibits habitat specificity and numerous physiological adaptations to cope with the hyper-aridity.

Effective nitrogen (N) management is vital for sustaining corn yield and reducing environmental impacts in Nebraska cropping systems. Because karyotype change commonly generates reproductive isolation between diverging species, rapid karyotype evolution, like that seen in plants, may increase the total rate of diversification. However, few studies have investigated this predicted relationship. Tests of this prediction have identified a positive correlation between the rate of karyotype change (specifically whole genome duplications) and diversification, but tests have been restricted to relatively small and young clades. Fortunately, novel macroevolutionary models have recently become available to investigate patterns of karyotype evolution in large phylogenies, providing new opportunity to investigate variation in karyotype driven diversification. The majority of durum wheat (Triticum turgidum spp. durum; AABB; 2n=4x=28) is predominantly grown in the northern Great Plains of the United States, and is used in the production of semolina, a coarse flour that makes pasta and couscous. In vineyard cropping systems, the rhizosphere microbiome arises through recruitment and filtering of the bulk soil microbiome shaped over time by soil carbon and nitrogen, climate, pH and historical legacies of vineyard management. The recruitment and filtering steps occur under the genetic influences of rootstock and scion that together drive variability in rhizosphere microbiome diversity and composition.
This study compares rhizosphere microbiomes of four Pinot Noir blocks under two rootstocks (101-14 and 420A) and four scions (777, 828, Calera, Mt. Eden). Anthropogenic factors have been the cause of the decline of tree species across the globe. From unsustainable logging and development to disease and improper management, threats to forests have taken their toll. The American chestnut (Castanea dentata) was once an integral basal tree species in Appalachian forests, but chestnut blight drove the species to functional extinction by the mid-1900s. The recent creation of genetically modified chestnuts gives hope for future restoration of the species throughout its historical range. Despite blight resistance, effective restoration will require optimal site selection for finding and planting chestnuts, which will maximize limited resources. …

Australian bush tomatoes in northern Western Australia likely have seed dispersal mechanisms that depend on processes outside of frugivory, which is unusual for this genus. Solanum tudununggae seeds are wind dispersed by the censer mechanism, meaning that seeds fall from holes in the top of the fruit when the branches move. In another species, piles of abscised and burr-like fruits have been found strewn around the bases of S. ossicruentum, possibly due to the extinction of animals that may have been a means of dispersal via epizoochory. Crown gall disease can be very detrimental to grape and wine producers around the world by reducing vine vigor and yield. It is caused by the bacteria Allorhizobium vitis (A. Vitis) carrying a tumor inducing (Ti) plasmid. The A. vitis bacteria have been found in soil and cane tissues in vineyards around the world, and infect all cultivated grapes. Microgreens are young, edible seedlings harvested upon emergence of the first true leaf. Their popularity is growing as a nutritionally dense food source that can be grown quickly in space-efficient systems. However, little is known about optimal cultivation practices and the effects of fertilization on growth and nutrition of microgreens. This study investigates the role of calcium (Ca) fertilization in optimizing the growth, yield, and nutrient content of daikon radish (Raphanus sativus) microgreens. Plants form the basis of life on Earth. However, due to the phenomenon of plant awareness disparity, a self-perpetuating cycle whereby plants are overlooked on an individual and global level ensues. Lack of plant awareness leads to a lessened emphasis on plants in sustainability and conservation as well as biology education, resulting in negative impacts on biodiversity and food security. Malaria remains one of the most devastating parasitic diseases globally, disproportionately affecting sub-Saharan Africa, where Rwanda continues to face high transmission rates and rising resistance to conventional interventions. Despite decades of control efforts, the emergence of drug-resistant Plasmodium falciparum and insecticide-resistant Anopheles mosquitoes threatens to reverse recent gains. This research responds to the urgent need for sustainable and culturally relevant alternatives by exploring the therapeutic potential of Rwandan traditional medicinal plants. These indigenous remedies, long used by local healers, offer an underexplored reservoir of bioactive compounds that may provide the basis for novel antimalarial drugs.

Greenhouses used for teaching or by hobbyist are smaller than those used for commercial production or found in the retail environment. Structural materials, covering, mechanical equipment, and control systems often are packaged as a kit made available through greenhouse suppliers. Supplier construction services may not be available, in which buyers must use local contractors to complete the installation. Greenhouses are unique structures that differ greatly from sheds or other outbuildings. Spring-fed aquatic ecosystems in the Ozark Highlands are characterized by stable water depths, clear water, and diverse aquatic vegetation (filamentous algae, submerged, floating, and emergent plants). Sudden changes in water depth can impact aquatic vegetation dynamics but the impacts of prolonged changes are poorly understood. I investigated seasonal and annual changes to aquatic vegetation abundance in a spring impoundment during and after a two-year period of low water depth that was caused by a broken outlet below an artificial dam. Water depth was significantly lower in 2020 and 2021 (low water years), compared to 2022 and 2023 (high water years). …
