Viral determinants and mechanisms involved in extension of host range of monocot-infecting viruses are poorly understood. Viral coat proteins (CP) serve many functions in almost every aspect of the virus life cycle. The role of the Cterminal region of Wheat streak mosaic virus (WSMV) CP in virus biology was examined by mutating six negatively charged aspartic acid residues at positions 216, 289, 290, 326, 333, and 334. All of these amino acid residues are dispensable for virion assembly, and aspartic acid residues at positions 216, 333, and 334 are expendable for normal infection of wheat and maize. First Report Of Curvularia Inaequalis And Bipolaris Spicifera Causing Leaf Blight Of Buffalograss In Nebraska, B. S. Amaradasa, K. Amundsen

During the summer of 2011, foliar blight was observed on buffalograss (Buchloë dactyloides (Nutt.) Engelm.) lawns in Lincoln and Waverly, Nebraska. Disease symptoms were common when buffalograss was growing above 30°C and in drought conditions. Disease symptoms began as dark brown oblong leaf spots, followed by leaf tip dieback and eventual blighting of entire tillers. Leaf infections would progress into patches of thinning turf. Diseased leaf pieces were rinsed in distilled water and placed on 1.5% water agar. Two mitosporic fungal species having conidial morphology of Curvularia and Bipolaris were isolated. Patch burn‐grazing is a rangeland management strategy that exploits the attraction of grazing animals to recently burned areas in order to achieve management objectives. When fire is applied to a landscape in a patchy manner, leaving some patches unburned, the resulting grazing animal activity, forage utilization, and animal impact are patchily distributed within that landscape as well. Areas that have been recently burned tend to be characterized by the highest levels of grazing animal activity while areas that have gone the longest without burning tend to be characterized by the lowest levels of grazing animal activity. Salt tolerance of ten sorghum (Sorghum bicolor L. Moench) varieties (‘1790E’, ‘BTx642’, ‘Desert Maize’, ‘Macia’, ‘RTx430’, ‘Schrock’, ‘Shallu’, ‘Tx2783’, ‘Tx7078’, and ‘Wheatland’) was evaluated in two greenhouse experiments. In the first experiment, sorghum were sown in substrates moistened with either nutrient solution (no addition of salts, control) at electrical conductivity (EC) of 1.2 dS·m-1 or salt solution at EC 5, 10 or 17 dS·m-1. Seedling emergence percentage decreased in all varieties only at EC of 17 dS·m-1 compared to the control. Seedling emergence percentage of sorghum ‘Macia’ and ‘1790E’ irrigated with salt solution at EC of 17 dS·m-1 decreased by …

Soybean [Glycine max (L.) Merr.] is an important oilseed crop which produces about 30 %of the world’s edible vegetable oil. The quality of soybean oil is determined by its fatty acid composition. Soybean oil high in oleic and low in linolenic fatty acids is desirable for human consumption and other uses. Interest in soybean [Glycine max (L.) Merr.] isoflavones has increased in recent years owing to numerous reported health benefits. Consequently, quantitative trait loci (QTL) detection for marker-assisted breeding for isoflavones is being examined for genetic gains. This study sought to detect QTL for soybean isoflavones in a population of 274 recombinant inbred lines derived from a cross between ‘Essex’ and ‘Williams 82’ that were subdivided and tested by maturity (early, mid, and late). Iron (Fe) and copper (Cu) homeostasis are tightly linked across biology. In previous work, Fe deficiency interacted with Cu-regulated genes and stimulated Cu accumulation. The C940-fe (fefe) Fe-uptake mutant of melon (Cucumis melo) was characterized, and the fefe mutant was used to test whether Cu deficiency could stimulate Fe uptake. Wild-type and fefe mutant transcriptomes were determined by RNA-seq under Fe and Cu deficiency. FeFe-regulated genes included core Fe uptake, metal homeostasis, and transcription factor genes. Triacylglycerol (TAG) metabolism is a key aspect of intracellular lipid homeostasis in yeast and mammals, but its role in vegetative tissues of plants remains poorly defined. We previously reported that PHOSPHOLIPID:DIACYLGLYCEROL ACYLTRANSFERASE1 (PDAT1) is crucial for diverting fatty acids (FAs) from membrane lipid synthesis to TAG and thereby protecting against FA-induced cell death in leaves. Here, we show that overexpression of PDAT1 enhances the turnover of FAs in leaf lipids. Using the trigalactosyldiacylglycerol1-1 (tgd1-1) mutant, which displays substantially enhanced PDAT1- mediated TAG synthesis, we demonstrate that disruption of SUGAR-DEPENDENT1 (SDP1) TAG lipase or PEROXISOMAL TRANSPORTER1 (PXA1) severely decreases …

The Middle Eocene Climatic Optimum (MECO; ~ 40 million years ago [Ma]) is one of the most prominent transient global warming events in the Paleogene. Although the event is well documented in geochemical and isotopic proxy records at many locations, the marine biotic response to the MECO remains poorly constrained. We present new high-resolution, quantitative records of siliceous microplankton assemblages from the MECO interval of Ocean Drilling Program (ODP) Site 1051 in the subtropical western North Atlantic Ocean, which are interpreted in the context of published foraminiferal and bulk carbonate stable isotope (δ18O and δ13C) records. …

Goss’s bacterial wilt and blight, caused by Clavibacter michiganensis subsp. nebraskensis (Cmn), has reemerged as an important disease of Zea mays (corn) in the U.S. Midwest. Results from a 2011 multistate survey indicated that Setaria spp. (foxtail) were often present in corn fields with a history of Cmn. The objective of this research was to determine if Setaria spp. that are common in the Midwest are susceptible to infection by Cmn. Ficotylus laselvae n. sp. was recovered from under the bracts of figs (syconia) of Ficus colubrinae from La Selva, Costa Rica, during a survey of nematode rainforest biodiversity and is described herein. This is only the second report of an association between the nematode suborder Tylenchina and the sycones of figs. Previous reports of most nematode associates of the sycones of figs have been from the lumen and involved transmission by female fig wasp pollinators (Agaonidae) during pollination/oviposition (e.g., Schistonchus and Parasitodiplogaster spp.). Viral coat proteins function in virion assembly and virus biology in a tightly coordinated manner with a role for virtually every amino acid. In this study, we demonstrated that the coat protein (CP) of Wheat streak mosaic virus (WSMV; genus Tritimovirus, family Potyviridae) is unusually tolerant of extensive deletions, with continued virion assembly and/or systemic infection found after extensive deletions are made. Phytophthora ramorum is a recently emerged plant pathogen and causal agent of one of the most destructive and devastating diseases currently affecting US horticulture and forests (Rizzo et al. 2002, 2005). This oomycete pathogen was discovered in Marin County, California, in the mid-1990s, causing sudden oak death on coast live oak (Quercus agrifolia) and tanoak (Notholithocarpus densiflorus) and simultaneously discovered in Europe causing foliar blight on Rhododendron and Viburnum (Rizzo et al. 2002; Werres et al. 2001). Most managed turfgrass species require frequent inputs to maintain an acceptable level of quality. Among these inputs, nitrogen (N) fertilization is usually the most limiting in terms of growth and development. However, N fertilization is also linked to non-point source (NPS) pollution. White clover (WC) is known for its ability to provide N when mixed into stands of turfgrass, and does not pose a threat for NPS pollution. The incidences of suspected Boron (B) deficiency have increased recently in Kentucky tobacco fields, potentially due to recent changes in management practices. The symptoms observed in the field include; hollow stalk, stunted growth, deformed or no bud formation, small slits on the lower leaf midrib and uncontrollable breaking of the midrib approximately two inches from the stalk. B is a micronutrient tobacco needs in minute amounts, however excessive additions of B could cause toxicity. Common bean (Phaseolus vulgaris L.) is the most important grain legume for human consumption and has a role in sustainable agriculture owing to its ability to fix atmospheric nitrogen. We assembled 473 Mb of the 587-Mb genome and genetically anchored 98% of this sequence in 11 chromosome-scale pseudomolecules. We compared the genome for the common bean against the soybean genome to find changes in soybean resulting from polyploidy. Using resequencing of 60 wild individuals and 100 landraces from the genetically differentiated Mesoamerican and Andean gene pools, we confirmed 2 independent domestications from genetic pools that diverged before human colonization. …

Sorghum lines were bred for reduced lignin for cellulosic bioenergy uses, through the incorporation of brown midrib (bmr)6 or -12 into two backgrounds (RTx430 and Wheatland) as either single or doublemutant lines. When these lines were assessed for resistance to Fusarium thapsinum stalk rot, a cause of lodging, they were as resistant to F. thapsinum as the near-isogenic wild type. Peduncles of newly identified bmr lines from an ethyl-methanesulfonate-mutagenized population, inoculated with F. thapsinum, were as resistant as the wild-type line, BTx623. One bmr line (1107) had significantly smaller mean lesion lengths than BTx623, suggesting that a mutation …

The blast fungus Magnaporthe oryzae threatens global food security through the widespread destruction of cultivated rice. Foliar infection requires a specialized cell called an appressorium that generates turgor to force a thin penetration hypha through the rice cuticle and into the underlying epidermal cells, where the fungus grows for the first days of infection as a symptomless biotroph. Understanding what controls biotrophic growth could open new avenues for developing sustainable blast intervention programs. ALS-tolerant grain sorghum cultivars are expected to be available for farmers within the next few years. Predictions of weed emergence can be used by practitioners to schedule POST weed management operations. Common sunflower seed from Kansas was used at six Midwestern U.S. sites to examine the variability that 16 climates had on common sunflower emergence. Nonlinear mixed effects models, using a flexible sigmoidal Weibull function that included thermal time, hydrothermal time, and a modified hydrothermal time (with accumulation starting from January 1 of each year), were developed to describe the emergence data. Background: Advances in genotyping technology, such as genotyping by sequencing (GBS), are making genomic prediction more attractive to reduce breeding cycle times and costs associated with phenotyping. Genomic prediction and selection has been studied in several crop species, but no reports exist in soybean. Soybean seeds contain high levels of oil and protein, and are the important sources of vegetable oil and plant protein for human consumption and livestock feed. Increased seed yield, oil and protein contents are the main objectives of soybean breeding. The objectives of this study were to identify and validate quantitative trait loci (QTLs) associated with seed yield, oil and protein contents in two recombinant inbred line populations, and to evaluate the consistency of QTLs across different environments, studies and genetic backgrounds. False smut caused by Cercospora seminalis is an important disease of buffalograss (Buchloe dactyloides) affecting seed production. The pathogen prevents normal caryopsis development and causes considerable yield loss and reduced seed germination. The current taxonomic placement of the false-smut causal pathogen in the genus Cercospora is incorrect based on its morphological characteristics and DNA phylogeny. Glycosyl inositol phosphorylceramide (GIPC) sphingolipids are a major class of lipids in fungi, protozoans, and plants. GIPCs are abundant in the plasma membrane in plants, comprising around a quarter of the total lipids in these membranes. Plant GIPCs contain unique glycan decorations that include a conserved glucuronic acid (GlcA) residue and various additional sugars; however, no proteins responsible for glycosylating GIPCs have been identified to date. Here, we show that the Arabidopsis thaliana protein INOSITOL PHOSPHORYLCERAMIDE GLUCURONOSYLTRANSFERASE1 (IPUT1) transfers GlcA from UDP-GlcA to GIPCs. Background: Sorghum is an important cereal crop, which requires large quantities of nitrogen fertilizer for achieving commercial yields. Identification of the genes responsible for low-N tolerance in sorghum will facilitate understanding of the molecular mechanisms of low-N tolerance, and also facilitate the genetic improvement of sorghum through marker-assisted selection or gene transformation. In this study we compared the transcriptomes of root tissues from seven sorghum genotypes having differential response to low-N stress.

Glyphosate-resistant giant ragweed is one of the most competitive weeds of agronomic crops in the United States. Early emergence and rapid growth rate makes giant ragweed a competitive weed early in the season and reduces crop yields. Therefore, early spring control of giant ragweed using a preplant herbicide is critical. Glufosinate is an alternative POST herbicide for weed control in glufosinate-resistant soybean. Co-infection of wheat (Triticum aestivum L.) by Wheat streak mosaic virus (WSMV, a Tritimovirus) and Triticum mosaic virus (TriMV, a Poacevirus) of the family Potyviridae causes synergistic interaction. In this study, the effects of the synergistic interaction between WSMV and TriMV on endogenous and virus-derived small interfering RNAs (vsiRNAs) were examined in susceptible (‘Arapahoe’) and temperature-sensitive resistant (‘Mace’) wheat cultivars at 18 degrees C and 27 degrees C.
