Purpose
While well-designed drainage systems could improve crop growth and yield by mitigating waterlogging and salinity stresses, field evidence of the yield responses to changes in plant-water relations and ion concentrations in leaves is scarce. We investigated the changes in ion concentrations in leaves and plant-water relations of sunflower caused by drainage in waterlogged saline soil, and their relationships to growth and yield.
Methods
Over two growing seasons, we tested four drainage treatments: undrained, surface drains (SD; 0.1 m deep, 1.8 m apart), subsoil drains (SSD; 0.5 m deep, 4.5 m apart) and SSD + SD. All plots were inundated …

Salinity causes a detrimental impact on plant growth, particularly when the stress occurs during germination and early development stages. Barley is one of the most salt-tolerant crops; previously we mapped two quantitative trait loci (QTL) for salinity tolerance during germination on the short arm of chromosome 2H using a CM72/Gairdner doubled haploid (DH) population. Here, we narrowed down the major QTL to a region of 0.341 or 0.439 Mb containing 9 or 24 candidate genes belonging to 6 or 20 functional gene families according to barley reference genomes v1 and v3 respectively, using two DH populations of CM72/Gairdner and Skiff/CM72, …

Context: Rotations in rainfed farming systems of southwest Australia have shifted towards intensified cropping and it is necessary to reassess soilborne pathogens and plant parasitic nematodes within this context. Aims: We tested the hypothesis that these recent changes in rotations and agronomy have altered the efficacy with which rotations reduce the incidence of common root pathogens and plant parasitic nematodes. Methods: We tracked changes in common pathogen DNA in soil and the incidence and severity of crop root damage in 184 paddocks, over 6 years from 2010 to 2015, and related this to farmer practices. Loose smut (LS) disease is a serious problem that affects barley yield. Breeding of resistant cultivars and identifying new genes controlling LS has received very little attention. Therefore, it is important to understand the genetic basis of LS control in order to genetically improve LS resistance. To address this challenge, a set of 57 highly diverse barley genotypes were inoculated with Egyptian loose smut race(s) and the infected seeds/plants were evaluated in two growing seasons. Plant growth-promoting rhizobacteria (PGPRs) stimulate plant growth through their ability, to increasing the root length and growth, by asymbiotic nitrogen fixation, by producing siderophores, solubilization of mineral phosphates and mineralization of other nutrients. Lead (Pb) exposure has long been recognized as a hazard to human health. Urban garden soils often contain elevated levels of Pb, mainly from legacy sources, which is a main barrier for urban gardening. The capacity of gardeners to access, understand, and act on scientific data related to soil contamination is also variable. This synthesis paper briefly summarizes the current scientific knowledge on soil Pb in urban gardens. Our objective is to produce clear recommendations about assessing actual risks and limiting exposure. Association mapping panels represent foundational resources for understanding the genetic basis of phenotypic diversity and serve to advance plant breeding by exploring genetic variation across diverse accessions. We report the whole-genome sequencing (WGS) of 400 sorghum (Sorghum bicolor (L.) Moench) accessions from the Sorghum Association Panel (SAP) at an average coverage of 38× (25–72×), enabling the development of a high-density genomic marker set of 43 983 694 variants including single-nucleotide polymorphisms (approximately 38 million), insertions/deletions (indels) (approximately 5 million), and copy number variants (CNVs) (approximately 170 000). Herbicide performance is directly related to the amount of active ingredient that has been deposited on the plant. Hence, spray solution characteristics and application parameters are crucial in determining the efficacy of an herbicide application. To maximize the effectiveness of chemical control, methods to deliver full chemical dose must be utilized: allowing the active ingredient to be readily absorbed once added to the carrier and mitigating off-target movement and low herbicide doses. Recombination allows for the exchange of genetic material between two parents which plant breeders exploit to make new and improved varieties. This recombination is not distributed evenly across the chromosome. In crops, it mostly occurs in the euchromatic regions of the genome and even then, recombination is focused into recombination hotspots flanked by recombination cold spots. Understanding the distribution of these hotspots along with the sequence motifs associated with them may lead to methods that enable breeders to better exploit recombination in breeding.

Resistance to protoporphyrinogen IX oxidase (PPO)-inhibitors in Amaranthus palmeri and Amaranthus tuberculatus is mainly contributed by mutations in the PPO enzyme, which renders herbicide molecules ineffective. The deletion of glycine210 (ΔG210) is the most predominant PPO mutation. ΔG210-ppo2 is overexpressed in rice (Oryza sativa c. ‘Nipponbare’) and Arabidopsis thaliana (Col-0). A foliar assay was conducted on transgenic T1 rice plants with 2× dose of fomesafen (780 g ha−1), showing less injury than the non-transgenic (WT) plants. BACKGROUND
The commercialization of dicamba-resistant soybean has resulted in increased concern for off-target movement of dicamba onto sensitive vegetation. To mitigate the off-target movement through physical drift, one might consider use of rope wicks and other wiper applicators. Although wiper-type application methods have been efficacious in pasture settings, the utility of dicamba using wiper applicators in agronomic crops is not available in scientific literature. To determine the utility of roller wipers for dicamba applications in dicamba-resistant soybean, two separate experiments were conducted in the summer of 2020 and replicated in both Keiser and Fayetteville, AR, USA.

Genome targeting with CRISPR/Cas9 is a popular method for introducing mutations and creating knock-out effects. However, limited information is currently available on the mutagenesis of essential genes. This study investigated the efficiency of CRISPR/Cas9 in targeting rice essential genes: the singleton TARGET OF RAPAMYCIN (OsTOR) and the three paralogs of the Sucrose non-fermenting-1 (SNF1)-related kinase 1 (OsSnRK1α), OsSnRK1αA, OsSnRK1αB and OsSnRK1αC. Strong activity of constitutively expressed CRISPR/Cas9 was effective in creating mutations in OsTOR and OsSnRK1α genes, but inducible CRISPR/Cas9 failed to generate detectable mutations. BACKGROUND
Resistance to protoporphyrinogen oxidase (PPO)-inhibiting herbicides is endowed primarily by target-site mutations at the PPX2 gene that compromise binding of the herbicide to the catalytic domain. In Amaranthus spp. PPX2, the most prevalent target mutations are deletion of the G210 codon, and the R128G and G339A substitutions. These mutations strongly affect the dynamic of the PPO2 binding pocket, resulting in reduced affinity with the ligand. Here we investigated the likelihood of co-occurrence of the most widespread target site mutations in the same PPX2 allele.

Climate change related drought is projected to harm maize production. Water use strategies can help mitigate the impact of drought on crop yield. However, little is known about maize metabolic response to drought at different developmental stages. To shed light on this, drought conditions were applied to maize at the six-leaf stage (V6), twelve leaf stage (V12), and tassel stage (VT). V6 and VT took eight days to achieve a low stomatal conductance threshold, but V12 took 16 days. Tedera is a drought-tolerant perennial forage legume introduced in Australia in 2006. In October 2018, T15-1218 Lanza®, the world’s first tedera variety, was released by the Department of Primary Industries and Regional Development and Meat & Livestock Australia for commercial use. A key agronomic practise for the successful establishment and adoption of tedera is to have a robust herbicide package to control a range of grass and broadleaf weeds well tolerated by tedera. Heat stress is a primary constraint to Australia's barley production. In addition to impacting grain yield, it adversely affects physical grain quality (weight and plumpness) and market value. The incidence of heat stress during grain filling is rising with global warming. However, breeding for new superior heat-tolerant genotypes has been challenging due to the narrow window of sensitivity, the unpredictable nature of heat stress, and its frequent co-occurrence with drought stress. Reducing groundwater withdrawals from the Mississippi River Valley Alluvial Aquifer is imperative to sustain future irrigated cropping systems in the mid-southern USA. This research was conducted to determine the impacts of cover crops and irrigation sensor thresholds on corn (Zea mays L.) and soybean (Glycine max L.) production, water productivity, irrigation water use efficiency, and soil physical properties in the Mississippi Delta. Rotations and associated management practices in rainfed farming systems of southwest Australia have shifted towards intensified cropping. Survey data from 184 fields spanning 14 Mha of southwest Australia were used to document water use efficiency (WUE) and water-limited yield potential (WLYP) of commercial crops and crop sequences and identify biophysical variables influencing WUE. WUE achieved in commercial wheat crops was 10.7 kg grain/ha.mm. Using a boundary function Ywl = 25 × (WU − 45), farmers achieved 54% of WLYP. Climate variables affected WUE more than management and biotic variates, the highest latitude region having WUE of 9.0 kg grain/ha.mm, compared …

This series of experiments is designed to understand the development and chemistry of sickle gloss. Sickles are common in the archaeological record and have long been studied for their eponymous “sickle gloss”. There is debate as to how this gloss is formed. Five experimental sickles containing flint and novaculite blades were used to harvest high and low moisture content rye and common fescue and associated field weeds. The differences in the development of sickle gloss were examined. High moisture content plants create thick, smooth fields of polish with undulating, billowing margins at a higher developmental speed. Low moisture content plants …

The muscadine (Vitis rotundifolia syn. Muscadinia rotundifolia) is a grape species native to the southeastern United States. Muscadines are one of three grape species in subgenus Muscadinia with a chromosome number of 2n=40 (V. rotundifolia, V. munsoniana, and V. popenoei), making them genetically distinct from the European wine and table grape (Vitis vinifera) and other species in subgenus Euvitis. Rooting hardwood cuttings from muscadine vines has traditionally been considered an exceptionally difficult task. Cost-effective weed suppression is an important consideration for tomato growers. Growers often choose methods which minimize hand labor, as hand weeding can be prohibitively expensive. This project determined economic viability of high tunnel tomatoes treated with several methods of weed control, both organic and chemical. These methods included: 2-week hand weeding, 1-week hand weeding, preemergent, straw, landscape fabric, and untreated weedy control plots. These treatments were applied to randomized blocks in a high-tunnel. Weeding, planting, and harvest were all timed to determine labor and material costs of weed management strategy implementation. After harvest, marketable yield was weighed to determine revenue. …
