Drought is one of the most limiting factors for rice (Oryza sativa L.) growth and development with vegetative and reproductive stages the most sensitive and distinct phases. During the vegetative stage, drought can cause reduction in growth and biomass accumulation. Moreover, water stress at reproductive stage can reduce yield significantly. Plants are protected against drought by three different mechanisms: drought avoidance, drought tolerance, and drought escape. An integrated approach combining physiology, breeding, and genomics could be an effective way to characterize and mitigate this problem. The mitochondrial genome (mitogenome) and plastid genome (plastome) of plants vary immensely in genome size and gene content. They have also developed several eccentric features, such as the preference for horizontal gene transfer of mitochondrial genes, the reduction of the plastome in non-photosynthetic plants, and variable amounts of RNA editing affecting both genomes. Different organismal lifestyles can partially account for the highly diverse organellar genomes across the tree of green plants. For example, endosymbiotic and parasitic lifestyles can dramatically affect the genomic architectures of plant mitochondria and plastids. The adoption of genetically engineered (GE) crops has created a demand for practical methods to mitigate pollen dispersal and gene flow. The goal of this project was to measure the ability of a narrow forest windbreak to reduce downwind pollen fluxes from switchgrass (Panicum virgatum L.), a North American grass and model biofuels feedstock. Switchgrass fields were established in two identical plots where one had a forest windbreak and the other was in an open (control) site. Switchgrass reproduction, pollen dispersal, wind speed, and wind direction were measured over two years. Daily release of switchgrass pollen peaked at 11:00-13:30 …

The extent and nature of favorable alleles contained in the collection are not well known nor is the distribution of genetic variation and how it relates to phenotypic variation. The genotyping of the entire USDA Soybean Germplasm Collection marked the beginning of a systematic exploration of genetic diversity for genetic research and breeding. Maize was domesticated from teosinte in Mexico some 7,000 to 10,000 years ago and quickly spread through the Americas. It has become one of the most important crops at a local and global level. Two types, Northern Flint corn and Southern Dent corns provided the basis of the genetic background of modern maize hybrids. The development of hybrids, first double-cross and later single-cross hybrids, along with a transition to high input farming provided huge yield increases, which have continued to improve with improving technology.

Starch composition and grain quality of rice is greatly influenced by genotype and environmental factors. The detrimental effects of high nighttime temperatures on rice yield and quality has recently become apparent, with some of the warmest average nighttime temperatures being recorded in the past few years. One of the most notable effects of this stress, an increase in grain chalk formation, correlates with a decrease in quality. Fatty acid desaturases (FADs) are enzymes that act in the chloroplast or the endoplasmic reticulum (ER) to incorporate double bonds into the acyl chains of fatty acids, and recent evidence indicates that at least one of these enzymes, FAD7, also influences plant resistance to aphids. FAD7 is an enzyme in the chloroplast that is found throughout the plant kingdom and that desaturates 16- and 18-carbon fatty acids (FAs) with two double bonds (dienoic acids) to generate FAs with three double bonds (trienoic acids). This study examined the effect of GM labeling on consumer willingness to pay (WTP) for edamame. It also investigated how people reacted to different messages and whether the order of receiving positive or negative information about GM technology impacted their willingness to pay (WTP). The study had three components: (1) a sensory test of GM and non-GM labeled products; (2) a non-hypothetical experimental auction to assess WTP for GM, non-GM and unlabeled products; and (3) a questionnaire to collect demographics and other information from the participants. Rice (Oryza sativa L.) kernel fissuring is a major concern of both rice producers and millers. Fissures are small cracks in rice kernels that increase breakage of kernels when milled, and decrease the value of processed rice. This study employed molecular gene tagging methods to fine-map a fissure resistance (FR) locus found in ‘Cybonnet’, a semidwarf tropical japonica cultivar, as well as transfer this trait to rice genotypes of taller, non-semidwarf plant height that are better adapted to some rice production systems. Increased soybean [Glycine max (L.) Merr.] commodity prices in recent years have generated interest in high-input systems to increase yield. The objective of this study was to evaluate the effects of current, high-yielding cultivars under high- and low-input systems on soybean yield and yield components. Research trials were conducted at 19 locations spanning nine states from 2012 to 2014. CYP82e4, CYP82e5, and CYP82e10 mutant alleles that minimize the conversion of nicotine to nornicotine have been introgressed into numerous existing low converting (LC) burley varieties and parental lines developed by the Kentucky-Tennessee tobacco breeding program. A backcross breeding protocol was utilized, with the objective being the creation of "e3" varieties that differed from their LC counterparts only for nornicotine and nitroso-nornicotine content. Field studies were conducted in Kentucky and Tennessee during the 2013 growing season, with 17 prospective parental lines and 20 prospective commercial varieties grown and compared to their original counterparts. Doubled haploidy (DH) is a plant breeding technique that is often utilized by plant breeders to minimize the time required to reach homozygosity in breeding lines. The first objective of this study was to compare two methods of generating DH lines in tobacco (Nicotiana tabacum L.). Inbred burley tobacco varieties TN 90LC and GR 149LC were used to produce both androgenic derived doubled haploids (ADDH) and maternally derived doubled haploids (MDDH). The research described herein, focuses on the biochemical and proteomic analysis of the maize endosperm and what influences kernel texture. Quality Protein Maize (QPM) is a hard endosperm version of the high-lysine opaque2 (o2) mutant but the genes involved in modifying the soft o2 endosperm are unknown. Pyrophosphate (PPi)-dependent fructose 6-phosphate 1-phosphotransferase (PFP) catalyzes the ATP-independent conversion of fructose 6-phosphate to fructose 1, 6-bisphosphate in glycolysis. We found a large increase in transcript and protein levels of the alpha regulatory subunit of PFP (PFPα) in QPM endosperm. In vitro enzyme assays show a significant increase in forward PFP …

Drought stress is one of the major abiotic factors affecting crop growth and limiting production worldwide. Cotton genotypes vary in drought tolerance, and the effects of drought stress on the anatomy and physiology of cotton leaves and roots have been reported. However, information on physiological and metabolic processes of leaves and flowers of modern cotton cultivars under water-deficit stress during reproductive development is not well elucidated. Soil waterlogging (WL) affects wheat production worldwide, including the southern of the U.S. Little is known about the genetics of tolerance mechanisms to WL, particularly of potentially toxic elements such as aluminum, iron and sodium. Legumes form symbiotic and pathogenic interactions with microbes. Understanding the molecular mechanisms underlying the legume-microbe interactions would help us to improve crop production in a sustainable manner. This thesis covers two independent research projects. The first project was to study the role of alternative splicing in RCT1-mediated disease resistance. RCT1 is a TIR-NBS-LRR-type plant resistance (R) gene in Medicago truncatula that confers broad-spectrum resistance to Colletotrichum trifolii, a fungal pathogen that causes anthracnose disease in Medicago. RCT1 undergoes alternative splicing at both coding and 3'-untranslated regions, thereby producing multiple transcript variants in its expression profile. …

A red clover (Trifolium pratense) population (UK), from a cross between the cultivar Kenland and a 2,4-D tolerant population (Florida), was recurrently selected for 2,4-D tolerance with evaluations after the 6th, 7th, and 8th selection cycles. All UK populations were more 2,4-D tolerant than Kenland. The 2,4-D tolerance following the 6th selection cycle was similar to the Florida population and tolerance was increased following 7 and 8 cycles of selection by removing plants showing 2,4-D injury and doubling the rate of 2,4-D used for selection.

The objective of this study was to determine dairy cattle preference amongst four species of cool-season forage grasses: eight orchardgrasses (Dactylis glomerata L.), five tall fescues [Schedonorus arundinaceus (Schreb.) Dumort.], five perennial ryegrasses (Lolium perenne L.), and six festuloliums [xFestulolium braunii (K. Richt.) A. Camus.]; 24 cultivars in total. Each grazing trial utilized four Holstein-Friesian heifers over six hours. Maturity differences were eliminated by having animals graze only vegetative material. After six grazing trials (three each in 2014 and 2015), consistent results in animal preference were not found; three of the six trials did show preference …

Mild variants of many viruses are able to protect infected plants from subsequent invasion by more severe variants of the same viruses through a process known as cross-protection. In the past, the cross-protective viral variants were commonly derived from mild field isolates that were sometimes genetically heterogeneous, providing variable levels of crossprotection. Here, we report a novel approach to rapidly generate crossprotective variants of the tomato-infecting Pepino mosaic virus (PepMV) independently of the availability of mild field isolates. Yellow spot (syn. tan spot), caused by Pyrenophora tritici-repentis, is an important foliar disease of wheat in Australia that causes losses exceeding 50 % when conditions are favourable for disease development. Although good progress has been made internationally to understand yellow spot resistance, relatively few resistance genes have been identified and mapped in Australian germplasm and only one (tsn1 on chromosome 5BL) is in general and known use in Australian breeding programs. Drought is an increasing issue that many farmers encounter especially in hot arid climates with little rainfall. High temperatures and inadequate rainfall at certain stages in crop development can have disastrous consequences to yield. NEBRASKA CORN HYBRID TESTS CROP PRODUCTION SUMMARY: According to the National Agricultural Statistics Service, there were 8.75 million acres of corn harvested in Nebraska in 2014 producing approximately 1.58 billion bushels of grain. The total average corn yield for Nebraska in 2014 was a record 181 bushels per acre (bu/a). Total corn yields from the previous 10 years are reported below.

Characterizing the impact of selection and genetic drift in the formation of heterotic groups and patterns in maize can reveal important insights into the mechanism underlying adaptation, and the relative importance of each force in defining population structure. The objectives were to characterize the role selection for hybrid performance had in defining population structure in both a reciprocal full-sib selection (RFS) program and a large collection of historically important inbred lines.

MutS Homolog 1 (MSH1) encodes a plant-specific protein that functions in mitochondria and chloroplasts. We showed previously that disruption or suppression of the MSH1 gene results in a process of developmental reprogramming that is heritable and non-genetic in subsequent generations. In Arabidopsis, this developmental reprogramming process is accompanied by striking changes in gene expression of organellar and stress response genes. This developmentally reprogrammed state, when used in crossing, results in a range of variation for plant growth potential. Here we investigate the implications of MSH1 modulation in a crop species. Percobaan ini bertujuan untuk menentukan persentase naungan dan konsentrasi pupuk organik cair yang terbaik terhadap pertumbuhan dan perkembangan tanaman anggrek Phalaenopsis hasil persilangan kultivar Sayukudion x Taida Snow. Dilaksanakan di Rumah Kaca Fakultas Pertanian Universitas Padjadjaran, Jatinangor, Sumedang, dengan ketinggian tempat 700 meter dpl, pada bulan Oktober 2011 sampai Januari 2012. Fourteen U.S plant introduction (PI) accessions of C. lanatus var. lanatus (4), C. lanatus var. citroides (5) C. colocynthis (5) and a known susceptible commercial cultivar ‘Sugar Baby’ were evaluated for resistance to the twospotted spider mite, Tetranychus urticae Koch, in a limited free-choice and free-choice bioassay under laboratory conditions. The limited free choice bioassay, involved nine Petri dish cages that held five randomly assigned leaves individually inoculated with two adult females and one adult male. Seed carbohydrate content is an important aspect in breeding for food-grade soybeans commercialized in the soyfood market. Sucrose and stachyose are the primary carbohydrates in soybean seed. Sucrose affects the quality and taste of various soyfoods such as tofu, soymilk, and natto; however, consumption of soy-based products with high stachyose concentrations can cause diarrhea and flatulence. A mutant line (V99-5089) with high-sucrose and low-stachyose has been identified.
