Australian Temperate Strawberry Breeding Program (Western Australia) Selection Of New And Advanced Short Day Breeding Lines And Cultivars In Western Australia - 2005/2006, Michaela Antoine, Dennis Phillips

Horticulture research reports

Sixty-eight 'short day' selections from the Australian Temperate breeding program were compared in a trial planting on a commercial property at Wanneroo from a mid May planting. Thirty-three of the selections were derived from crosses made in 2003 (new Stage 4 selections) and twenty-nine were from crosses made between 1998 and 2002 (Intermediate Stages 5 and 6). In addition, three advanced selections were compared in large plots of 1,000-1 ,500 plants together with three commercial control cultivars, Gaviota, Camino Real and Kiewa. These selections were 'fast tracked' for future commercialisation by being selected for bulking up in September 2004.

Snp-Phage – High Throughput Snp Discovery Pipeline, Lakshmi K. Matukumalli, John J. Grefenstette, David L. Hyten, Ik-Young Choi, Perry B. Cregan, Curt P. Van Tassell

Department of Agronomy and Horticulture: Faculty Publications

Background: Single nucleotide polymorphisms (SNPs) as defined here are single base sequence changes or short insertion/deletions between or within individuals of a given species. As a result of their abundance and the availability of high throughput analysis technologies SNP markers have begun to replace other traditional markers such as restriction fragment length polymorphisms (RFLPs), amplified fragment length polymorphisms (AFLPs) and simple sequence repeats (SSRs or microsatellite) markers for fine mapping and association studies in several species. For SNP discovery from chromatogram data, several bioinformatics programs have to be combined to generate an analysis pipeline. Results have to be stored in …

Evaluation Of The Single Kernel Characterization System (Skcs) For Measurement Of Sorghum Grain Attributes, S. R. Bean, O. K. Chung, M. R. Tuinstra, Jeffrey F. Pedersen, J. Erpelding

Department of Agronomy and Horticulture: Faculty Publications

The single kernel characterization system (SKCS) has been widely used in the wheat industry, and SKCS parameters have been linked to end-use quality in wheat. The SKCS has promise as a tool for evaluating sorghum grain quality. However, the SKCS was designed to analyze wheat, which has a different kernel structure from sorghum. To gain a better understanding of the meaning of SKCS predictions for grain sorghum, individual sorghum grains were measured for length, width, thickness (diameter), and weight by laboratory methods and by the SKCS. SKCS predictions for kernel weight and thickness were highly correlated to laboratory measurements. However, …

Department of Agronomy and Horticulture: Faculty Publications

Background: Single nucleotide polymorphisms (SNP) constitute more than 90% of the genetic variation, and hence can account for most trait differences among individuals in a given species. Polymorphism detection software PolyBayes and PolyPhred give high false positive SNP predictions even with stringent parameter values. We developed a machine learning (ML) method to augment PolyBayes to improve its prediction accuracy. ML methods have also been successfully applied to other bioinformatics problems in predicting genes, promoters, transcription factor binding sites and protein structures. Results: The ML program C4.5 was applied to a set of features in order to build a SNP classifier …

Department of Agronomy and Horticulture: Faculty Publications

Here, we describe two members of the Arabidopsis (Arabidopsis thaliana) Yellow Stripe-Like (YSL) family, AtYSL1 and AtYSL3. The YSL1 and YSL3 proteins are members of the oligopeptide transporter family and are predicted to be integral membrane proteins. YSL1 and YSL3 are similar to the maize (Zea mays) YS1 phytosiderophore transporter (ZmYS1) and the AtYSL2 iron (Fe)-nicotianamine transporter, and are predicted to transport metal-nicotianamine complexes into cells. YSL1 and YSL3 mRNAs are expressed in both root and shoot tissues, and both are regulated in response to the Fe status of the plant. β-Glucuronidase reporter expression, driven by …

Soybean Cultivars Resulted From More Recombination Events Than Unselected Lines In The Same Population, T. R. Stefaniak, D. L. Hyten Jr., V. R. Pantalone, A. Klarer, T. W. Pfeiffer

Department of Agronomy and Horticulture: Faculty Publications

The selection of superior adapted cultivars has contributed to the doubling of soybean [Glycine max (L.) Merr.] yields in the USA since 1930. Genetic variation was required for this selection to be effective. The objective of this study was to evaluate the importance of homologous chromosome meiotic recombination in the creation of soybean cultivars. A set of 10 adapted high-yielding cultivars selected from the cross ‘Williams’ × ‘Essex’ was com-pared with a set of 156 random recombinant inbred lines (RILs) from the same population. Crossover events were identified using 143 simple sequence repeat (SSR) markers span-ning all 20 soybean …

Impacts Of Genetic Bottlenecks On Soybean Genome Diversity, D. L. Hyten, Qijian Song, Youlin Zhu, Ik-Young Choi, Randall L. Nelson, Jose M. Costa, James E. Specht, Randy C. Shoemaker, Perry B. Cregan

Department of Agronomy and Horticulture: Faculty Publications

Soybean has undergone several genetic bottlenecks. These include domestication in Asia to produce numerous Asian landraces, introduction of relatively few landraces to North America, and then selective breeding over the past 75 years. It is presumed that these three human-mediated events have reduced genetic diversity. We sequenced 111 fragments from 102 genes in four soybean populations representing the populations before and after genetic bottlenecks. We show that soybean has lost many rare sequence variants and has undergone numerous allele frequency changes throughout its history. Although soybean genetic diversity has been eroded by human selection after domestication, it is notable that …

Department of Agronomy and Horticulture: Faculty Publications

In previous works, it has been shown, by using ethylene inhibitors and precursors, that ethylene could participate in the regulation of the enhanced ferric reductase activity of Fe-deficient Strategy I plants. However, it was not known whether ethylene regulates the ferric reductase gene expression or other aspects related to this activity. This paper is a study of the effects of ethylene inhibitors and precursors on the expression of the genes encoding the ferric reductases and iron transporters of Arabidopsis thaliana (FRO2 and IRT1) and Lycopersicon esculentum (= Solanum lycopersicum) (FRO1 and IRT1) plants. The effects …

Key Grazing Levels (Foo Targets) For Pasture And Sheep Management, Department Of Agriculture, Western Australia

Animal production and livestock published reports

Food On Offer (FOO) is the amount of pasture in front of an animal, and is measured in kilograms Dry Matter per hectare. Managing pastures to key grazing levels - or FOO targets - can result in improved production of grazing systems in Mediterranean environments which are characterized by slow winter growth, peak spring growth and dry summers. These targets vary according to what production response is required.

This information sheet illustrates key target FOO for sheep grazing annual pastures during autumn and spring.

Establishing Effects Of Application Of Swine Manure On Crop Leaf Damage To Growing Crops, Charles Shapiro, University Of Nebraska-Lincoln

Department of Agronomy and Horticulture: Faculty Publications

Swine manure from a below barn storage facility was applied to growing corn and soybeans to determine phytotoxic effects. The manure had an electrical conductivity of around 20 mmhos/cm and was diluted for treatment at full, half, quarter and none. The manure mixtures were applied at a rate of 0.5 in/acre using a procedure that wet the foliage three times in a 15-minute time span. Manure mixtures were applied at V7 and V14 for corn and V3 and R1 for soybeans. Phytotoxic effects were shown at the high rate for both crops, but soybeans were killed at the early application …

Topcrop W.A. State Focus 2002-2003 Wheat Protein, Alison Slade, Jeremy Lemon

Bulletins 4000 -

The State Focus program is supported by the Grains Research and Development Corporation (GRDC) in conjunction with TOPCROP to provide grower groups with a new learning concept. The concept was developed in Victoria and has been adopted by South Australia and Western Australia.

Wheat protein was selected as the Western Australian State Focus in response to a wide-ranging consultation of grower groups, researchers and the TOPCROP West management group. As crop yield potential increases through matching varieties to growing season length, weed and disease control, and timely sowing systems, the crop’s demand for nitrogen increases. Unless the demand is matched …

Soybean [Glycine max (L.) Merr.] is a versatile crop due to its multitude of uses as a high protein meal and vegetable oil. Soybean seed traits such as seed protein and oil concentration and seed size are important quantitative traits. The objective of this study was to identify representative protein, oil, and seed size quantitative trait loci (QTL) in soybean. A recombinant inbred line (RIL) population consisting of 131 F6-derived lines was created from two prominent ancestors of North American soy-beans (‘Essex’ and ‘Williams’) and the RILs were grown in six environments. One hundred simple sequence repeat (SSR) markers …

Department of Agronomy and Horticulture: Faculty Publications

In sorghum, ingress of Cochliobolus heterostrophus stimulates the synthesis of 3-deoxyanthocyanidins that act as phytoalexins. Apigeninidin and luteolinidin are two major phytoalexins induced in the first 24 h after infection. In an attempt to understand genetic regulation of the biosynthesis of sorghum phytoalexins, we isolated a differentially expressed partial cDNA. Characterization and comparison showed that this cDNA sequence corresponds to a putative flavonoid 3-hydroxylase. Full length sequence characterization allowed us to establish that the sorghum putative f3h cDNA encodes a peptide of 517 amino acids that has domains conserved among cytochrome P450 proteins functioning in …

Altering FA content in soybean [Glycine max (L.) Merr.] oil for improved functionality is a research goal of many soybean breeders. Several of the genes that alter palmitic, stearic, oleic, linoleic, and linolenic acids are modifier genes with small effects, causing these FA traits to act as quantitative traits. The objective of this study was to identify modifier FA quantitative trait loci (QTL) in soybean. A recombinant inbred line population was created from two prominent ancestors of currently avail-able U.S. cultivars (Essex and Williams) and grown in five environments. One hundred simple se-quence repeat markers spaced throughout the genome …

Predicting Phenological Development In Winter Wheat, Qingwu Xue, Albert Weiss, P. Stephen Baenziger Jan 2004

Predicting Phenological Development In Winter Wheat, Qingwu Xue, Albert Weiss, P. Stephen Baenziger

Department of Agronomy and Horticulture: Faculty Publications

Accurate prediction of phenological development is important in the winter wheat Triticum aestivum agroecosystem. From a practical perspective, applications of pesticides and fertilizers are carried out at specific phenological stages. In crop-simulation modeling, the prediction of yield components (kernel number and kernel weight) and wheat-grain yield relies on accurate prediction of phenology. In this study, a nonlinear multiplicative model by Wang & Engel (WE) for predicting phenological development in differing winter wheat cultivars was evaluated using data from a 3 yr field experiment. In the vegetative phase (emergence to anthesis) the daily development rate (r) was simulated based on the …

Chitosanase May Enhance Anti-Fungal Defense Responses In Transgenic Tobacco, Bill L. Hendrix Jan 2003

Chitosanase May Enhance Anti-Fungal Defense Responses In Transgenic Tobacco, Bill L. Hendrix

Inquiry: The University of Arkansas Undergraduate Research Journal

Fungicides are expensive, dangerous, and can be harmful to the environment, but they are often necessary for profitable farming operations. New technologies may soon allow farmers to replace these chemicals with genetically engineered plants producing antifungal enzymes that degrade fungal cell walls. To explore this option, a Paenbacillus chitosanase gene was cloned, sequenced, and modified for plant expression. The modified gene was delivered to tobacco (Nicotiana tabacum L cv. Xanthine) leaf disks via Agrobacterium tumenfaciens-mediated transformation. The putative GMOs were tested for transgene integration, transcription, and translation. Confirmed transformants were then screened for enhanced responses to a Rhizoctonia solani cell …

Forage Yield And Economic Losses Associated With The Brown-Midrib Trait In Sudangrass, M. D. Casler, Jeffrey F. Pedersen, D. J. Undersander Jan 2003

Forage Yield And Economic Losses Associated With The Brown-Midrib Trait In Sudangrass, M. D. Casler, Jeffrey F. Pedersen, D. J. Undersander

Department of Agronomy and Horticulture: Faculty Publications

Brown-midrib genes increase digestibility due to reduced lignification in sudangrass, Sorghum bicolor subsp. drummondii (Nees ex Steud.) de Wet & Harlan. Brown-midrib lines are known to be low in forage yield potential, but this reduction in forage yield has not been previously quantified. The objectives of this study were to quantify the increase in forage quality and decrease in forage yield and to provide an economic assessment of this dichotomy. Piper and Greenleaf (normal leaves) were compared with their brown-midrib counterparts and four highly selected brown-midrib (FG) lines at two locations for 2 yr. Brown-midrib lines averaged 9.0% lower in …
