PRECS student projects
The ability of soybean cyst nematodes (SCN. Heterodera glycines) to lay dormant as eggs within a cyst for up to 11 years, has made this parasite a principal target for soybean crop pest management. Research on SCN hatching will improve understanding of SCN biology will uncover new mechanisms for their control. This poster summarizes three experiments using hatching stimulants, soybean root exudate (SRE) zinc chloride, testing whether it affects post hatch development.

In 2013, Castro et al., produced the first linkage map for primocane-fruiting blackberries using the cross ‘Prime-Jim®’ x ‘Arapaho.’ This mapping population has been maintained since the original experiment with the hopes of conducting future studies. Further research was determined to be needed on the population to better characterize additional phenotypic traits, and since nine years had passed since the seedlings were established, there was a need to assess the continued genetic purity of the population using molecular methods. Soya bean (Glycine max (L.) Merr.) is sought after for both its oil and protein components. Genetic approaches to add value to either component are ongoing efforts in soya bean breeding and molecular biology programmes. The former is the primary vegetable oil consumed in the world. Tan spot, caused by the ascomycete fungus, Pyrenophora tritici-repentis (Ptr), is a major foliar fungal disease of wheat worldwide. To date, Ptr isolates have been grouped into eight races based on the production of three host selective effectors (toxins), Ptr ToxA, ToxB, and ToxC, which are associated with necrosis and chlorosis symptoms on wheat differential lines. Kualitas benih akan mempengaruhi dinamika pertumbuhan tanaman. Tujuan penelitian ini adalah untuk mengkaji respons benih terdeteriorasi terhadap aplikasi berbagai pelapis benih, baik berupa insektisida maupun pelapis hayati berupa suspensi mikroorganisma yang dicerminkan dengan peningkatan laju tumbuh tanaman. Penelitian merupakan percobaan petak terbagi dua faktor, yaitu tingkat deteriorasi (D) yang terdiri dari benih berkualitas baik (DB = 84 %) dan benih terdeteriorasi (DB = 75 %). Diaphorina citri (Hemipteran: Psyllidae), the Asian citrus psyllid, is a phloem-feeding, invasive species endemic to southern Asia that was first identified in the United States in Florida in 1998. Since introduction, D. citri has spread across the major citrus growing regions of the United States. This is of great concern to the citrus industry because D. citri is the primary vector of Candidatus Liberibacter asiaticus the causal agent of Haunglongbing (HLB). Maize was domesticated from teosinte in Mexico some 7,000 to 10,000 years ago and quickly spread through the Americas. It has become one of the most important crops at a local and global level. Two types, Northern Flint corn and Southern Dent corns provided the basis of the genetic background of modern maize hybrids. The development of hybrids, first double-cross and later single-cross hybrids, along with a transition to high input farming provided huge yield increases, which have continued to improve with improving technology.

California is currently facing a historic drought, and this has led many farmers in the state to severely cut back on irrigation. Optimal use of water for irrigation requires a comprehensive understanding of how plants respond physiologically to water stress (Chapter 1). By monitoring water requirements in crops and managing irrigation to meet those requirements, growers can significantly reduce water use (Chapter 2). The soybean cyst nematode (SCN), Heterodera glycines Ichinohe, is an endoparasitic nematode and one of the major pests of soybean (Glycine max L.) in the United State and all over the world where soybean is grown. Piratebush (Buckleya distichophylla (Nutt.) Torr.) is a rare, hemiparasitic shrub with the only extant populations in western North Carolina, northeastern Tennessee, and southwestern Virginia. The preferred natural hosts of piratebush, Carolina and eastern hemlocks, have seen sharp declines over the last decade due to the invasive hemlock woolly adelgid. Virginia pine, another important host of piratebush, is also susceptible to disease, specifically Cronartium appalachianum, a rust fungus for which piratebush is the secondary host. Use of biological control offers a cost effective and environmentally safe method for controlling plant diseases. Biocontrol agents that can colonize roots and live endophytically within plant tissue should allow for effective disease control. The goal of this research was to develop protocols to isolate putative Streptomyces species from potato stem and tuber tissue due to their ability to produce inhibitory compounds which could potentially reduce diseases caused by Streptomyces scabies. Streptomyces species are ubiquitous soil bacteria that are promising as biological control agents due to their prolific antibiotic production that can inhibit soil-borne plant pathogens. This includes Streptomyces scabies, which causes scab disease on underground storage organs. The goal of this research was to test two Streptomyces isolates that have known inhibitory and biocontrol abilities against S. scabies in a potato (Solanum tuberosum) agricultural system for their effectiveness in biocontrol of this pathogen in a radish (Raphanus sativus) greenhouse assay. Despite a continuous rise in consumption of coffee over the past 60 years and recent studies showing positive benefits linked to human health, intensive coffee farming practices have been associated with environmental damage, risks to human health, and reductions in biodiversity. In contrast, organic farming has become an increasingly popular alternative, with both environmental and health benefits. The oomycete Phytophthora parasitica has a worldwide distribution and is an economically important pathogen of more than 100 species4. RNA-seq analysis showed that one gene, PPTG_16698 has the 5th highest level of expression of all transport proteins in the zoospore stage, and is highly conserved throughout Phytophthora species. This project attempts to characterize the important biological role that PPTG_16698 plays in P. parasitica and other oomycetes. The Dectes stem borer, Dectes texanus Leconte, has caused significant economic damage to soybean fields in south central Nebraska during recent years. Most economic injury occurs when soybean plants become susceptible to late season lodging due to larval girdling. Developing a comprehensive management plan for Dectes stem borer in Nebraska is limited by lack of knowledge of seasonal activity and effective sampling plans to monitor adult populations. Bacterial wilt, caused by Curtobacterium flaccumfaciens pv. flaccumfaciens, was first recognized and described as a new dry bean disease near Redfield, South Dakota, after the 1921 growing season on the farm of the Office of Forage Investigations. The grower on this farm informed the federal investigators that he believed the disease to be the same one that killed 90% of his crop in 1920. For the 1921 season, he saved and planted the seed harvested from the previous year, resulting in an estimated 25% loss.

The low rainfall areas of West Asia are characterized by pastoral production system primarily based on small ruminants (sheep and goats). Rangelands serve as the main source of feed. With increasing population, the demand for livestock products has increased. As a consequence, the stocking rates have soared, increasing the pressure on rangelands. Rangeland degradation has therefore been increasing threatening the sustainability of livestock production system and the livelihood of the people dependent on it. The value of traditional versus improved management systems for grassland is discussed. The set target is the persistence of the ecological, economic and social multifunctionality of grasslands and the increase of food output from these areas without increasing parallel requirements for arable crop or energy inputs. Redesign of agricultural systems according to ecological principles has been proposed for the development of sustainable systems. We review a wide variety of ecologically-based crop production practices that focus on forage crops in farming systems and discuss their potential role in enhancing the profitability, environmental sustainability and resilience. Crop-livestock systems that most closely mimic natural systems through appropriate integration of diverse components appear to offer the greatest potential benefits. These systems are more energy efficient and combine high productivity with low ecological footprint. There are many dimensions to the concept of seed quality. Viable seed which will not germinate when provided with all the requirements for germination is dormant. Forage grasses mostly exhibit non-deep physiological dormancy (PD), while physical dormancy (PY) is common in forage legumes where imbibition is prevented by the seed coat’s impermeability to water (hard seed). Methods for breaking PD and PY to allow germination testing and/or crop establishment are discussed.

Seed quality is defined as (i) maintaining genetic purity using seed certification principles (generation system; isolation; previous cropping history); (ii) achieving high levels of physical purity (especially low weed seed levels) by in field weed control and seed cleaning; (iii) achieving high levels of seed germination at harvest and maintaining germination through the seed supply chain. Seed production systems must not only produce high quality seed but also achieve economic seed yields. The profitable and sustainable management of livestock production from grassland systems is challenging and it can be difficult to develop a research structure that addresses farmer’s needs and has acceptable rigour and on-ground impact. This paper describes the attributes of research, development and extension (RD&E) programs that are required for a successful partnership between researchers, farmers and advisors. Insights are provided from the EverGraze program that designed, tested and implemented farming systems based on perennial pastures across southern Australia.
