A better understanding of how proteasome activity is regulated can facilitate the search for proteasome enhancement strategies for disease treatment. A cell culture study shows cAMP-dependent protein kinase (PKA) activates 26S proteasomes by phosphorylating Ser14 of RPN6 (pS14-RPN6), but this discovery and its physiological significance remain to be established in vivo. To test the hypothesis that pS14-RPN6 mediates the activation of proteasomes by PKA and reduces proteotoxicity in animals, two knock-in mouse models with Ser14 of endogenous Rpn6 mutated to either Ala (S14A) or Asp (S14D) to respectively block or mimic pS14-Rpn6 were created. Sex Identification Of Mytilus Edulis Following 26 Day Treatment To Ee2 And Kz, Helen C. Poynton, K. Garrett Evensen

Data and Datasets

Blue mussel M. edulis were exposed for 26 days to 50 ng/L 17alpha-ethinylestradiol or 30 ug/L ketoconazole for 26 days in four replicate tanks. Prior to exposure, mussel sex was identified using the VERL-VCL RT-qPCR assay on hemolymph collected from each mussel. Following exposure, mantle tissue was collected from each treated mussel and the VERL-VCL RT-qPCR assay was used to identify the sex of the mussels.

To continue to meet the demands of increasing food production and environmental sustainability, new strategies are needed to enhance the efficiency of finishing cattle production. A review of the literature identified several nutritional and physiological constraints that limit the ability for cattle to digest starch and absorb glucose from the small intestine. Comparative sequencing analyses revealed a missing region of the sucrase-isomaltase protein which could affect the capacity for starch digestion in cattle. Post-flaking sampling and handling recommendations were generated based on a series of experiments to improve estimates of starch availability and flaking consistency for commercial feedlots and laboratories. …

Mining has left massive environmental and physical scars across the landscape. Aquatic and riparian landscapes in particular have been significantly impacted by traditional mining practices. Waste products left over from hard-rock mining leech heavy metals onto the landscape and these metals spread from headwater streams to major waterways (Lottermoser 2010). Cardiac voltage-gated sodium channels (NaV1.5) are essential to generate sodium current (INa) and regulate cardiac electrical conduction. Mutations in NaV1.5 can lead to cardiac disorders such as Brugada syndrome and Long QT Syndrome type 3 (LQTS3), potentially resulting in sudden cardiac death. Mutations p.I239V and p.M1487K have been identified in LQTS3 patients. The p.I239V mutation was found in a patient with mild symptoms and in family members who experienced sudden cardiac death, while the p.M1487K mutation is associated with more severe symptoms. Thyroid hormone (TH) is crucial for many biological processes, including the development of the auditory system. Appropriate thyroid hormone signaling is important for the development of the cochlea, leading to the successful onset of hearing. More specifically, TH influences the timing of remodeling of the greater epithelial ridge (GER), a transient cochlear structure that secretes proteins that contribute to the formation of the tectorial membrane. Developmental hypothyroidism results in delayed GER regression, whereas excess TH results in premature remodeling. Both conditions result in deafness. Behavioral health organizations often face the challenge of high employee turnover rates, which can critically affect their ability to treat mental illnesses successfully. High counselor turnover at one such organization, BHOX, threatens the provision of timely access to quality mental care. This qualitative case study examined counselor turnover and its impact on the quality of behavioral health services. Commercial running power (RP) meters may provide endurance runners and coaches with improved precision for monitoring running intensity. Cyclists rely upon the relationship between cycling power and metabolic demand to regulate physiological intensity during training and racing, independent of changes in the physical environment (e.g., terrain, wind). Chronic systemic inflammation, known as inflammaging is considered a hallmark of aging and associated diseases. While adipose tissue has long been considered only as a caloric reservoir regulating systemic energy homeostasis, research in the past couple of decades substantiate its endocrine function to secrete an array of inflammatory mediators and hormones, which have physiological effects on multiple organ systems, and contribute to inflammaging. Among the earliest changes to occur in Alzheimer’s disease, metabolic dysfunction and chronic neuroinflammation are now known to be major driving forces in disease progression. The paradigm of ‘immunometabolism’ seeks to bridge these two facets, positing that metabolic transformations are indispensable in determining the response of immune cells, such as microglia – the brain’s resident immune population. Proinflammatory stimulation of microglia leads to a shift away from mitochondrial respiration towards a dramatic upregulation of the glycolysis pathway for energy production. Influenza-A virus (IAV) infects yearly an estimated one billion people worldwide, resulting in 300,000–650,000 deaths. Preventive vaccination programs and antiviral medications represent the mainstay of therapy, but with unacceptably high morbidity and mortality rates, new targeted therapeutic approaches are urgently needed. Since inflammatory processes are commonly associated with measurable changes in the cell membrane potential (Em), we investigated whether Em hyperpolarization via TREK-1 (K2P 2.1) K þ channel activation can protect against influenza-A virus (IAV)-induced pneumonia. Previous work has examined the temporal and behavioral aspects of proximate fitness on life history outcomes particularly regarding highly perturbed environments (i.e., climate and land use change, resource extraction, agricultural erosion, etc.). Metabolic-associated fatty liver disease (MAFLD) affects 1 in 3 people in the US and is a healthcare burden on the order of billions of dollars. MAFLD is characterized by pathological lipid accumulation in the liver, excess production of triglycerides and cholesterol, and export of LDL particles that raise serum lipid levels. These metabolic perturbations can lead to the development of liver fibrosis, chronic inflammation, and insulin resistance. Diagnosing MAFLD prior to fibrosis is crucial for successful intervention and prevention of advanced liver disease and other metabolic complications. It has been established that older runners experience a reduction in strength and power. These changes may contribute to different running biomechanics from younger runners. It has recently become evident that the bacterial stringent response is regulated by a triphosphate alarmone (pGpp) as well as the canonical tetra- and pentaphosphate alarmones ppGpp and pppGpp [together, (p)ppGpp]. Often dismissed in the past as an artifact or degradation product, pGpp has been confirmed as a deliberate endpoint of multiple synthetic pathways utilizing GMP, (p)ppGpp, or GDP/GTP as precursors. Tracing how free-ranging organisms interact with their environment to maintain water balance is a difficult topic to study for logistical and methodological reasons. We use a novel combination of triple-oxygen stable isotope analyses of water extracted from plasma (δ16O, δ17O, δ18O) and bulk tissue carbon (δ13C) and nitrogen (δ15N) isotopes of feathers and blood to estimate the proportional contribution of marine resources, seawater, and metabolic water used by two species of unique songbirds (genus Cinclodes) to maintain their water balance in a seasonal coastal environment. Social context alters cognitive performance, but the mechanisms for this remain unclear. One possibility is that stressors associated with social contexts mediate changes in cognitive performance by influencing attention. We conducted two experiments to investigate how social context affects performance on an attention task in capuchin monkeys (Cebus [Sapajus] apella). We additionally validated and measured physiological stress through salivary cortisol. In experiment one, capuchins were exposed to three conditions where they were either isolated or paired with an affiliative or agonistic social partner separated by a barrier. Nestlings allocate energy to support their rapid growth and developmental period; thus, their challenges and the subsequent energetic demands can be much different than an adult. The rise in e-cigarette use is likely due to the overwhelming evidence regarding the addictive properties of nicotine and the damage caused by traditional tobacco cigarettes. I investigated the effects of diet and sex on zebrafish bone metabolism. Zebrafish were subjected to either a high-calorie or low-calorie diet over a five-week period. After this diet, zebrafish scales were used to measure alkaline phosphatase (ALP) and tartrate-resistant acid phosphatase (TRAP) enzyme activity with a fluorescent substrate. The enzyme activities were markers of the osteoblasts (ALP) and the osteoclasts (TRAP). These data were compared among sex and diet of the zebrafish using a repeated measures ANOVA statistical test. Neurodegenerative diseases such as multiple sclerosis affect millions of people around the globe, and promising treatments to counter its progress have been studied extensively. In our study, we used Dawley rats and tested them using the Montoya Staircase test to evaluate their ability to use their forelimbs. After training, surgery was performed using lysolecithin to induce focal demyelination on the rats. Abnormal glycosylation can impact cardiac proteins, affecting signaling, contraction, and metabolism, which leads to compromised cardiac function. O-GlcNAcylation, the only form of intracellular glycosylation, involves adding a single GlcNAc molecule to proteins via enzymes: O-GlcNAc Transferase (OGT) for addition and O-GlcNAcase (OGA) for removal. The Ednie/Bennett lab has previously demonstrated that in both chronic (OGT KO) and acute (OGT inhibition) models of reduced cardiomyocyte O-GlcNAcylation, a disruption of Ca2+ homeostasis occurs. This study delved into the cardiac voltage-gated sodium channel (NaV 1.5) in these models. Cilia are broadly categorized into primary and motile cilia. The intraflagellar transport(IFT) system, composed of two large multimeric IFT complexes, is universally present in all cilia and facilitates bidirectional cargo transport. While the role of IFT in primary cilia and eukaryotic motile cilia has been demonstrated, the association between IFT and mammalian motile ciliogenesis remains largely unknown. Aim 1 of the present study is to uncover the role of IFT in mammalian motile ciliogenesis. This experiment will aim to explore the heterodimerization of Dopamine (D2) and Adenosine (A2A) receptors in cells when bound to different ligands. Borroto-Escuela et al. (2018) investigated the dimerization of the A2AR-D2R in rats when administered with cocaine, and they found that disrupting the A2AR-D2R complex restores the cocaine reward system in rats. This was a strong influence as to why we chose to investigate A2AR-D2R dimerization in Chinese hamster ovary (CHO) cells.
