Fgf4 And Wnt5a/Pcp Signaling Promote Limb Outgrowth By Polarizing Limb Mesenchyme, Keri Lynn Low

The focus of this study was to elucidate the molecular and cellular mechanisms whereby fibroblast growth factors (FGFs) mediate outgrowth of the limb. Specifically, we examined the epistatic relationship between FGF and Wnt/Planar cell polarity (PCP) signaling in establishing cell polarity as a mechanism for outgrowth. By implanting beads into embryonic limbs and lateral plate mesoderm, we established that FGF activates Wnt5a in a gradient fashion. Once it was established that Wnt5a was expressed at the right time and place to turn on PCP signaling, we investigated the ability of Wnt5a to influence cell migration and/or cell polarity. Background: Cartilage disorders affect millions of people in the United States alone, with effects ranging from poor skeletal development and joint pain to shortened lifespan and perinatal lethality. Many of these disorders have their root in defects of collagen, type XI collagen being among the most important. A mouse model of such a type XI collagen defect is the chondrodysplasia (cho) mutant. Mice homozygous for this null mutation in the Col11a1 gene do not express the α1 chain of type XI collagen. This results in a functional knockout of type XI collagen, leading to insufficient skeletal development and perinatal lethality. …

Pka As An Upstream Kinase For Lkb1/Strad/Mo25, Seth Taylor Herway

The LKB1/STRAD/MO25 complex (LSMK) has been identified as the major upstream kinase for AMP-activated protein kinase (AMPK). PKA phosphorylates LKB1 at the Ser428 residue in humans and Ser431 residue in mice. We investigated PKA as an upstream kinase for LSMK. LKB1 that had been incubated with PKA prior to incubation with AMPK experienced up to a 51% increase in AMPK Kinase activity compared to LKB1 alone (p < 0.05). When blocked with a PKA Inhibitor, the kinase effect of PKA on LKB1 was eliminated. Rat epitrochlearis muscle tissue incubated with epinephrine experienced no increase in AMPK activity compared with controls indicating that epinephrine does not cause AMPK activity in this type of tissue. In conclusion, phosphorylation by PKA can increase the AMPKK activity of LKB1-STRAD-MO25 in vitro. Because LKB1 has been found to be constitutively active, it is postulated that phosphorylation by PKA may act to enhance LKB1-AMPK interaction and thus achieve its effect.

Skeletal muscle contraction results in the phosphorylation and activation of the AMP-activated protein kinase (AMPK) by an upstream kinase, AMPKK. The LKB1-STRAD-MO25 complex is the major AMPKK in skeletal muscle; however, LKB1-STRAD-MO25 activity is not increased by muscle contraction. This relationship suggests that phosphorylation of AMPK by LKB1-STRAD-MO25 during skeletal muscle contraction may be regulated by allosteric mechanisms. In this study we tested an array of metabolites including glucose-6-phosphate (G6P), fructose-6-phosphate (F6P), fructose 1,6-bisphosphate (F1,6-P2), 3-phosphoglycerate (3PG), glucose-1-phosphate (G1P), glucose-1,6-bisphosphate (G1,6-P2), adenosine diphosphate (ADP), carnitine (Carn), acetyl-carnitine (Acarn), inosine monophosphate (IMP), inosine, and ammonia for allosteric regulation. Proton channels are essential for many of the processes of life. The influenza A viral protein M2 is responsible for sensing the conditions necessary for viral RNA release. The proton-translocating FoF1 ATPase (ATP synthase) uses a proton gradient to drive adenosine triphosphate (ATP) synthesis. We have directly measured proton uptake in vesicles containing reconstituted M2 or FO by monitoring external pH after addition of valinomycin to vesicles with 100-fold diluted external [K+]. This proton flux assay was utilized to quantify proton flux through single M2 and Fo channels. Contrary to previous reports, proton uptake by M2 was not significantly altered …

In vertebrate embryos, the ophthalmic trigeminal (opV) placode contributes sensory neurons to the trigeminal ganglion during development of the peripheral nervous system. FGFR4 is expressed transiently in the trigeminal placode as cells undergo an epithelial-mesenchymal transition and begin to migrate toward the condensing ganglion. Because of the restricted spatiotemporal expression of FGFR4 in the opV placode, it was hypothesized that FGFR4 is necessary for the process of epithelial-mesenchymal transition in the opV placode. Using electroporation, an FGFR4 inhibitory gene construct was introduced into 6—10 somite stage chick embryos. This secreted inhibitory form of the FGFR4 gene blocked endogenous FGFR4 signaling, …

Pax3 has been used as a valuable marker in research aimed at understanding tissue interactions involved in trigeminal ophthalmic (opV) placode development. While Pax3 expression coincides with opV neuron specification, the function of Pax3 in these cells has not previously been investigated. Splotch mutant mice (which lack Pax3) have a reduced trigeminal ganglion; however it is not clear whether this reduction is due to neural crest or placode cells. In this experiment, we used four male llamas that were three to four years of age from the Khara race. They were divided into two ad libitum feeding groups. The digestible energy (ED) was determined using samples that were sent to the laboratory. These samples were derived from a conventional in vivo assay using the method of total fecal collection in order to determine the difference between energy consumed and energy excreted. Isoflavones, the most abundant phytoestrogens in soy foods, are structurally similar to 17beta-estradiol. Few studies have examined the nociception and stress hormone responses after consumption of soy isoflavones. Methods: In this study, ovariectomized (OVX) female Long-Evans rats were fed either an isoflavone-rich diet (Phyto-600) or an isoflavone-free diet (Phyto-free). We examined the effects of soy isoflavones on metabolism by measuring body weights, food/water intake, adipose tissue weights as well as serum leptin levels. Phytoestrogens derived from soy foods (or isoflavones) have received prevalent usage due to their 'health benefits' of decreasing: a) age-related diseases, b) hormone-dependent cancers and c) postmenopausal symptoms. However, little is known about the influence of dietary phytoestrogens on regulatory behaviors, such as food and water intake, metabolic hormones and neuroendocrine parameters. Acetyl-CoA carboxylase (ACC) catalyzes the formation of malnoyl-CoA, which in turn controls the rate of fatty acid metabolism. ACC beta or 2 has been shown to be localized on the mitochondria in close proximity to carnintine palmitoyl transferase 1 (CPT-1), the enzyme responsible for the influx of acyl-CoA into the matrix where beta oxidation takes place. CPT-1 is inhibited by malonyl-CoA produced by ACC. It has been well documented that AMP activated kinase (AMPK) when activated phosphorylates and inactivates ACC. Secretory phospholipase A2 (sPLA2) is an important part of apoptosis and disposal of damaged and dying cells. However, healthy cells are not susceptible to attack by sPLA2. Recent studies have focused on membrane properties necessary to induce susceptibility in both artificial and biological membranes. Hydrolysis of phospholipids by sPLA2 requires at least two preliminary steps: first, adsorption of the enzyme to the cellular membrane, and second, movement of a phospholipid into the active site of the enzyme. We determined the effects of susceptibility on each of the two steps and determined the contributions changing the equilibrium constants have on susceptibility. …

Background- It is well established that the growth of the prostate gland is a hormone-dependent phenomenon involving both androgenic and estrogenic control. Proliferation of prostate cells is, at least in part, under control of estrogen receptor beta (ER-beta). Phytoestrogens bind ER-beta with high affinity and therefore may have antiproliferative effects in the prostate. Methods- The prostates of male Long-Evans rats fed a diet high in phytoestrogens (Phyto-600) or very low levels of phytoestrogens (Phyto-free) were analyzed to determine the impact of dietary phytoestrogens on prostate weight and androgen receptor (AR) expression in the prostate. In learning and memory tasks, requiring visual spatial memory (VSM), males exhibit higher performance levels compared to females (a difference attributed to sex steroid hormonal influences). Based upon the results from our companion investigation, this study examined the influence of prenatal sex steroid hormone manipulations on VSM in adulthood, as assessed in the radial arm maze. Additionally, the influence of dietary soy phytoestrogens (i.e., the presence of high or low estrogen-like compounds present in the animal's diet) on VSM was examined in combination with the prenatal hormonal manipulations. In learning and memory tasks, requiring visual spatial memory (VSM), males exhibit superior performance to females (a difference attributed to the hormonal influence of estrogen). This study examined the influence of phytoestrogens (estrogen-like plant compounds) on VSM, utilizing radial arm-maze methods to examine varying aspects of memory. Additionally, brain phytoestrogen, calbindin (CALB), and cyclooxygenase-2 (COX-2) levels were determined. Results: Female rats receiving lifelong exposure to a high-phytoestrogen containing diet (Phyto-600) acquired the maze faster than females fed a phytoestrogen-free diet (Phyto-free); in males the opposite diet effect was identified. Macroinvertebrate communities were examined on 4 winter dates over a 4-yr period in 2 high-altitude Rocky Mountain streams to document overwintering assemblages potentially experiencing spring acid pulses. Taxa richness values were comparable to other published lists for alpine and montane stream systems despite the fact that most literature reflected summer collections. Mean benthic density ranged from 1406 to 19,734 organisms/m2, and drift rates ranged from 0 to 1740 organisms/100 m3. Benthic collections showed higher taxa richness than drift collections while the Ephemeroptera and Plecoptera occurred in greater proportions in drift than in benthos. Species of Artemisia (subgenus Tridentatae) dominate much of western North America. The genetic variation that allows this broad ecological adaptation is facilitated by hybridization and polyploidization. Three separate studies were performed in this group using randomly amplified polymorphic DNA (RAPD). Fifty-seven 10-mer primers generated nearly 400 markers from genomic DNA obtained from leaf tissue. From 1990 to 1992 we surveyed for Flammulated Owls (Otus flammeolus) in 3 areas in Idaho: Salmon National Forest (SNF), Payette National Forest and adjacent Hells Canyon National Recreation Area (PNF-HCNRA), and Nez Perce National Forest (NPNF). We also collected and summarized information on all historic and modern records of Flammulated Owls in Idaho. Flammulated Owls were detected on 65% of 68 routes (2–16 km in length) surveyed at densities ranging from 0.04 to 1.25 singing males/40 ha. Owls were detected on survey routes as early as 10 May and as late as 23 July. Mean percent canopy …

Totals of 101 native Yellowstone cutthroat (Oncorhynchus clarki bouvieri), 27 introduced lake trout (Salvelinus namaycush), and 40 introduced longnose sucker (Catostomus catostomus) from Yellowstone Lake, Wyoming, USA, were examined for eye flukes. Metacercariae of the trematode fluke Diplostomum were in vitreous humor and/or lens of 94% of Yellowstone cutthroat trout, 92% of lake trout, and 78% of longnose sucker. Longnose sucker had 7% prevalence of infection in both lens and vitreous humor of metacercariae, while Yellowstone cutthroat trout had 3% and lake trout 8%. Pinus aristata Engelm. forest vegetation in Colorado was studied to determine vegetation composition and the relationship between vegetation and environment. Species percent cover, tree size class, and environmental variables were recorded for 49 plots. Previously collected data for 4 plots from New Mexico were included. Environmental variables included latitude, longitude, elevation, slope, aspect, topographic position, parent material, percent rock cover, mean rock size, litter depth, estimated plot age class, and evidence of anthropogenic disturbance. One species of trematode (Glypthelmins quieta), 1 species of cestode (Distoichometra bufonis), and 5 species of nematodes (Aplectana incerta, A. itzocanensis, Rhabdias americanus, Physaloptera sp., and Physocephalus sp.) were found. The greatest prevalence (41%) and mean intensity (231.7) were recorded for Aplectana incerta in Bufo woodhousii. It appears hybrids harbor fewer parasites than either parent species.

The western spruce budworm (Choristoneura occidentalis Freeman) prefers to feed on flushing buds and current-year needles of Douglas-fir (Pseudotsuga menziesii [Mirb.] Franco). Budworm larvae will not typically consume older age classes of needles unless all current-year foliage is depleted. We tested the following null hypotheses: (1) budworm larvae can feed on foliage with a wide range of qualities (i.e., current 1-, 2-, or 3-year-old needles) without measurable effects on fitness; and (2) budworm adults do not show any oviposition preference linked to the age of the foliage they fed on as larvae. Congress is currently evaluating the wilderness status of Bureau of Land Management (BLM) public lands in Utah. Wilderness areas play many important roles, and one critical role is the conservation of biological diversity. We propose that objectives for conserving biodiversity on BLM lands in Utah be to (1) ensure the long-term population viability of native animal and plant species, (2) maintain the critical ecological and evolutionary processes upon which these species depend, and (3) preserve the full range of communities, successional stages, and environmental gradients. Pollinator preferences were observed for the six species of section Erythranthe of the genus Mimulus using greenhouse-grown plants placed in a meadow in the Red Butte Canyon Natural Area, Salt Lake County, Utah. The principal pollinators were hummingbirds and bumble bees. Hummingbirds preferred the species with the most reflexed tubular flowers regardless of color, whereas bumblebees preferred pink. lavender, or yellow flowers to red flowers regardless of shape. Results for the six species were confirmed by observations of F2 hybrid recombinant plants selected such that flower color could be held constant and flower shape varied and vice versa.

Red Butte Canyon is a protected, near pristine canyon entering Salt Lake Valley, Utah. It contains a well-developed riparian zone and a perennial stream; hillside vegetation ranges from grasslands on the lower limits to Douglas-fir and aspen stands at the upper elevations. In this paper we describe the history of human impact, natural history aspects of climate, geology, and ecology, and faunal and floral information for key species in the canyon. The role and importance of Research Natural Areas is discussed, particularly with respect to the need to protect Red Butte Canyon—one of the few remaining undisturbed riparian ecosystems in …
