Phthalates And Phthalate Alternatives: Effects On Proliferative And Estrogenic Target Genes In Ishikawa Cells, Ranjani Sundar '15, Ping Yin, Serdar E. Bulun
Student Publications & Research
Phthalates are used as plasticizers in many of the products found in medical, household, and industrial applications. Much research has not been completed on the effects of these phthalates as potential endocrine disrupting chemicals (EDCs). As these chemicals are ingested, the mechanism by which they affect the reproductive system is largely unknown. The purpose of this study was to observe how 2 phthalates, Di-n-butyl phthalate (DBP) and Diisononyl phthalate (DINP), and 2 phthalate alternatives, Dioctyl terephthalate (DOTP) and BHT (butylated hydroxytoluene)affect uterine cells in comparison to a vehicle treatment and 17β-Estradiol treatment. Changes in expression of mRNA were observed using …
Life History Tradeoffs And The Costs Of Reproduction In Arizona Bark Scorpions (Centruroides Sculpturatus), Michael Marie Webber
Life History Tradeoffs And The Costs Of Reproduction In Arizona Bark Scorpions (Centruroides Sculpturatus), Michael Marie Webber
UNLV Theses, Dissertations, Professional Papers, and Capstones
Tradeoffs in life history evolution result from conflicts in the time and energy that can be simultaneously invested in activities such as growth, reproduction, and survival. Reproduction is an energetically costly activity for organisms, and is known to elicit alterations in the daily activity patterns of individuals. I investigated reproductive tradeoffs in the Arizona Bark Scorpion,Centruroides sculpturatus. To better understand the nature of tradeoffs in this species, I examined the influence of reproductive state on the predatory efficiency and thermal preference of reproductive females. Further, I examined the influence of reproductive activities (i.e. gestation) on the ability of reproductive females …
The Grass Shrimp, Palaemonetes, Pugio: Hypoxic Influences On Embryonic Development, Christensen C. Javier
The Grass Shrimp, Palaemonetes, Pugio: Hypoxic Influences On Embryonic Development, Christensen C. Javier
UNLV Theses, Dissertations, Professional Papers, and Capstones
Grass shrimp,Palaemonetes pugio, can survive in brackish waters and estuarine ecosystems despite the frequent oscillations and fluctuations in salinity, temperature and oxygen. AdultP. pugiohave the ability to osmoregulate (Romney and Reiber 2011), change cardiac parameters to tolerate temperatures (not yet published, Mika and Reiber) and oxyregulate (Guadagnoli and Reiber 2013). Manipulation of cardiac parameters allows for these methods of regulation. However, cardiac contraction and internal convection of oxygen do not occur until later stages of embryonic development. Studies focused on these morphological and physiological advantages may provide further understanding of the regulatory mechanisms within grass shrimp embryos, larvae and adults. …

Absence Of Manganese Superoxide Dismutase Delays P53-Induced Tumor Formation., Adam J. Case, Frederick E. Domann
Absence Of Manganese Superoxide Dismutase Delays P53-Induced Tumor Formation., Adam J. Case, Frederick E. Domann
Journal Articles: Cellular & Integrative Physiology
BACKGROUND: Manganese superoxide dismutase (MnSOD) is a mitochondrial antioxidant enzyme that is down-regulated in a majority of cancers. Due to this observation, as well as MnSOD's potent antioxidant enzymatic activity, MnSOD has been suggested as a tumor suppressor for over 30 years. However, testing this postulate has proven difficult due to the early post-natal lethality of the MnSOD constitutive knock-out mouse. We have previously used a conditional tissue-specific MnSOD knock-out mouse to study the effects of MnSOD loss on the development of various cell types, but long-term cancer development studies have not been performed. We hypothesized the complete loss of …

Fat mass is linked mechanistically to the cardiovascular system through leptin, a 16 kDa protein produced primarily by adipocytes. In addition to increasing blood pressure via hypothalamic-sympathetic pathways, leptin stimulates monocyte migration, cytokine secretion, and other functions that contribute to atherosclerotic plaque development. These functions are also characteristics of CD16-positive monocytes that have been implicated in the clinical progression of atherosclerosis. This investigation sought to determine if leptin promoted the development of such CD16-positive monocytes. Cells from 45 healthy men and women with age ranging from 20 to 59 years were analyzed. Circulating numbers of CD14++16++ monocytes, which are primary …

Over-Expressed Copper/Zinc Superoxide Dismutase Localizes To Mitochondria In Neurons Inhibiting The Angiotensin Ii-Mediated Increase In Mitochondrial Superoxide, Shumin Li, Adam J. Case, Rui-Fang Yang, Harold D. Schultz, Matthew C. Zimmerman
Journal Articles: Cellular & Integrative Physiology
Angiotensin II (AngII) is the main effector peptide of the renin-angiotensin system (RAS), and contributes to the pathogenesis of cardiovascular disease by exerting its effects on an array of different cell types, including central neurons. AngII intra-neuronal signaling is mediated, at least in part, by reactive oxygen species, particularly superoxide (O2 (•-)). Recently, it has been discovered that mitochondria are a major subcellular source of AngII-induced O2 (•-). We have previously reported that over-expression of manganese superoxide dismutase (MnSOD), a mitochondrial matrix-localized O2 (•-) scavenging enzyme, inhibits AngII intra-neuronal signaling. Interestingly, over-expression of copper/zinc superoxide dismutase (CuZnSOD), which is believed …

Cross education is the phenomenon by which the untrained limb will experience a gain in strength following a unilateral resistance training program. However, little is known as to the underlying adaptation occurring in the untrained limb. Purpose: To examine the effect of dynamic unilateral resistance training on the strength and neuromuscular adaptations of both the trained and untrained legs. Methods: Eight previously untrained males (22.38±2.92 y, 1.73±0.08 m, 75.26±14.53 kg) completed a four-week unilateral resistance training program, while another eight untrained males (24.00±4.57 y, 1.84±0.05 m, 94.21±16.14 kg) served as controls. Isometric leg extension strength, leg press 1 repetition maximum …
Neuromuscular Fatigue Following A Singles Badminton Match, Zengyuan Lin
Neuromuscular Fatigue Following A Singles Badminton Match, Zengyuan Lin
Theses: Doctorates and Masters
A typical badminton singles match involves numerous intense and high impact movements. Lunges were accounted for approximately 15% of overall movements and were believed to presumably induce significant muscle damage following a match. However, no previous study has investigated changes in knee extensor muscle function after a badminton match. The present study investigated changes in knee extensor neuromuscular function and muscle soreness after a simulated 1-h badminton singles match in relation to the number of lunges performed in the match.
Ten state-level male badminton players were recruited (n=10), with each player played a total of eight simulated 1-h matches under the …
Pain Assessment And Possible Mechanism Of Delayed Onset Muscle Soreness, Wing Yin Lau
Pain Assessment And Possible Mechanism Of Delayed Onset Muscle Soreness, Wing Yin Lau
Theses: Doctorates and Masters
Muscle pain is felt during exercise or daily activities for several days after performing unaccustomed exercise, which is referred to as delayed onset muscle soreness (DOMS). Many people experience DOMS, but its underlying mechanisms are not fully understood. One of the challenges in the investigation of DOMS is its subjective nature, which makes the assessment ambiguous, thus establishing a standardised protocol is necessary. The present thesis scrutinised muscle pain assessments (Study 1, Study 2), developed a new assessment of muscle pain focusing on muscle fascia (Study 3), and investigated why DOMS is reduced after the second than the first bout …

Isokinetic Fatigue Characteristics For The Leg Extensors Versus Flexors, Elias M. Carrillo, Matt S. Stock, Jacob A. Mota, Kendra D. Olinghouse, Alexander S. Drusch, Brennan J. Thompson
Kinesiology and Health Science Faculty Publications
Maximal isokinetic muscle actions are often used in research studies to examine fatigability and even estimate muscle fiber-type. However, the majority of previous investigations have examined these topics for the leg extensors (i.e., quadriceps), and we are unaware of investigations that have specifically assessed the fatigue characteristics for the flexors (i.e., hamstrings). The purpose of this study was to compare the percent decline values for the leg extensors versus flexors for 50 and 100 repeated, maximal concentric isokinetic muscle actions. Fifteen healthy men (mean ± SD age = 23 ± 3 years; body mass = 94.1 ± 11.9 kg) with …

The effects of a single bout of resistance exercise (RE) in conjunction with peri-exercise branched chain amino acid (BCAA) and carbohydrate (CHO) ingestion on skeletal muscle signaling markers indicative of muscle protein synthesis (MPS) were determined. It was hypothesized that CHO + BCAA would elicit a more profound effect on these signaling markers compared to CHO. Twenty-seven males were randomly assigned to CHO, CHO + BCAA, or placebo (PLC) groups. Four sets of leg presses and leg extensions were performed at 80% 1RM. Supplements were ingested 30 min and immediately prior to and after RE. Venous blood and muscle biopsy …

The Effect Of Moderate Altitude On High Intensity Running Performance During A Game With Collegiate Female Soccer Players, Jonathan Bohner
The Effect Of Moderate Altitude On High Intensity Running Performance During A Game With Collegiate Female Soccer Players, Jonathan Bohner
Electronic Theses and Dissertations
Competition often requires teams that reside at sea level to compete against opponents whose residence is at a moderate altitude. This may pose a potential competitive disadvantage considering that moderate altitude may cause decrements in VO2max, distance covered, and time to exhaustion in endurance athletes. The purpose of this study was to examine the effect of altitude on game performance measures. Six NCAA Division I female soccer players (20.33 ± 1.21 y; 168 ± 6.45 cm; 62.5 ± 6.03 kg) were retrospectively examined. Comparisons were made between two competitions that were played at sea level (SL) within two weeks of …

An Analysis Of Abnormal Electrocardiograms In First And Second Year Medical School Students, Molly Anne Kalish
PCOM Biomedical Studies Student Scholarship
The high stress level placed upon medical school students, particularly during their first and second didactic years, may have a negative effect on their health. Although surveys and subjective questionnaires have been used to evaluate the effects medical school has upon a student’s cardiovascular health, there has been little clinical data obtained to confirm this notion. The aim of this longitudinal study was to demonstrate whether any abnormal cardiovascular parameters, specifically QTc wave interval, cardiac axis vector and blood pressure abnormalities, could be documented to occur in two different classes of medical students during their first two years. Such information …
Transmural Heterogeneity Of Cellular Level Cardiac Contractile Properties In Aging And Heart Failure, Premi Haynes
Transmural Heterogeneity Of Cellular Level Cardiac Contractile Properties In Aging And Heart Failure, Premi Haynes
Theses and Dissertations--Physiology
The left ventricle of the heart relaxes when it fills with blood and contracts to eject blood into circulation to meet the body’s metabolic demands. Dysfunction in either relaxation or contraction of the left ventricle can lead to heart failure. Transmural heterogeneity is thought to contribute to normal ventricular wall motion but it is not well understood how transmural modifications affect the failing left ventricle. The overall hypothesis of this dissertation is that normal left ventricles exhibit transmural heterogeneity in cellular level contractile properties and with aging and heart failure there are region-specific changes in cellular level contractile mechanisms.

ETSU Faculty Works
The purpose of this study was to evaluate the relationship between force production asymmetry and performance of athletes with differing strength levels in males and females. Collegiate athletes (n=129) from various sports were ranked according to isometric mid-thigh pull peak force and the top (‘strong’) and bottom (‘weak’) 25% were used for analysis. Symmetry index (SI) scores were calculated and correlated with their respective force-time characteristics using bivariate correlations. For the weaker males, several negative moderate correlations were observed; however, no statistically significant correlations were observed for the females in either group. These findings indicate that force production asymmetry is …

The barbell deadlift is a large muscle mass exercise that primarily involves extension at the hip and knee joints. Although it is often incorporated into strength training programs with a variety of other exercises (e.g., back squat), no previous studies have examined the effects of deadlift training on strength or power adaptations. The purpose of this investigation was to examine the effects of a ten week barbell deadlift training program on vertical jump height in previously untrained women. Twenty-eight women (mean ± SD age = 22 ± 2 years) were randomly assigned to either the training group (n = 17) …

Architectural and physical performance measurements are commonly implemented to identify various physical capacities in many populations. However, previous research has suggested architectural measures, notably in the leg extensors, are ineffective predictors of vertical jumping (VJ) performance. Given the functional relevance of rapid strength development on explosive dynamic tasks, further research is warranted examining, a) the presence of associations of maximal (e.g., peak torque; PT) and, in particular, explosive (e.g., rate of torque development; RTD) strength-related characteristics with jumping performance in the leg extensors, and b) the extent to which PT and RTD either uniquely, or synergistically contribute to VJ performance. …

Tinnitus and hearing loss are the frequent auditory-related co-morbidities of blast trauma. The etiology of blast-induced tinnitus is also muddled by brain mechanisms associated with emotional and cognitive problems such as anxiety, memory loss, and depression. We set out to develop a realistic and ecologically valid model to address changes of cognitive status and psychological state that are associated with blast- induced tinnitus. In this study, 19 adult rats were randomly divided …

Cervical Remodeling/Ripening At Term And Preterm Delivery: The Same Mechanism Initiated By Different Mediators And Different Effector Cells, Juan Miguel Gonzalez Velez
Wayne State University Dissertations
Premature cervical remodeling/ripening is believed to contribute to preterm delivery (PTD), the leading cause of perinatal morbidity and mortality. Despite considerable research, the causes of term and PTD remain unclear, and there is no effective treatment for PTD. We tested the hypothesis that complement activation plays a role in cervical remodeling and PTD. We studied cervical remodeling at term.
We studied two mouse models of inflammation-induced PTD. The first model was induced by vaginal administration of lipopolysaccharide (LPS)and the second one by administration of progesterone antagonist RU486. Increased cervical C3 deposition and macrophage infiltration and increased serum C3adesArg and C5adesArg …

Improvements in muscular strength require an individual to train with progressively increased external loads over time. While it is well documented that the initial adaptations to strength training occur rapidly, previous researchers have not thoroughly examined the most effective means of inducing these changes. The purpose of this study was to examine the ability to add 2.27 kg to the barbell for 20 consecutive training sessions, and to compare these responses between the sexes. Thirty-four subjects ([mean ± SD age = 23 ± 3 years] men, n = 17; women, n = 17) participated in this study. The subjects were …

Participations in a resistance training program results in both neural adaptations and increases in lean body mass, the latter of which is considered a slow process. The majority of previous investigations that have examined the time course associated with muscular adaptations have utilized exercise machines or single-joint movements. Furthermore, the adaptations associated with resistance training in women are not as well understood as those for men. The purpose of this investigation was to examine the effects of a four week resistance training intervention on changes in leg lean mass in untrained, college-aged women. Thirty-five women (mean ± SD age = …

Anthropometric and physical performance measurements are commonly used for identifying specific physical traits in youth. Laboratory-based tests (e.g., linear velocity transducers), while accurate, provide practical limitations due to high costs and technical necessities. Thus, commonly used field tests may be useful alternatives for assessing fitness/performance characteristics of youth. The purpose of this study was to examine the relationships between anthropometric measures and lower-and upper-body power and strength measures; and to assess the predictive ability of these measures for lower-body vertical jump peak power (PP) output in youth. Thirty-nine pre-adolescent (mean±SD, range: age=7.8±1.7, 5-12 years) children volunteered to participate in this …
