Journal of Dentistry Indonesia
Objective: Mouthwash is widely used to maintain oral health by reducing the prevalence of pathogenic microbiomes. Nonetheless, chlorhexidine, a common active ingredient, is associated with side effects including teeth staining, mucosal peeling, mouth and tongue numbness. This has prompted interest in natural alternatives. This study aimed to identify the cytotoxicity and anti-inflammatory properties of 1’S-1’-acetoxychavicol acetate (ACA) isolated from Alpinia conchigera rhizome against human primary gingival fibroblast and keratinocyte (HPGF and HPGK). Methods: Cytotoxicity of ACA (1.57 mg/mL) against HPGF and HPGK was assessed using the MTT assay. Meanwhile, the anti-inflammatory property of ACA was analyzed based on …

AdipoRon is the first synthetic analog of endogenous adiponectin, which is an adipose tissue derived hormone. As it is characterized with pharmacological properties like adiponectin through binding and activating AdipoR1 and AdipoR2 receptors this makes it a target for therapeutic treatments for a multitude of disorders. In this study AdipoRon effect on HT29 and HCT116 colon cancer cells were investigated. Cytotoxic activity of AdipoRon was observed, with a significant inhibition rate in HT29 and HCT116 cells. Moreover, there was a significant decrease in the number of HCT116 colonies by clonogenicity test. In addition, cell migration assay showed significant decrease in …

Fluid Properties Of Solvents And Oils Used In In Situ Forming Microparticles, Sai Myo Thu Rein, Torsak Intaraphairot
The Thai Journal of Pharmaceutical Sciences
In situ forming microparticle (ISM) has been recently used as the injectable drug delivery system prepared from polymeric non-aqueous phase in oil emulsion. Solvents and oils used in ISM including N-methyl pyrrolidone (NMP), 2-pyrrolidone (PYR), dimethyl sulfoxide (DMSO), glycofurol, triacetin, olive oil, camellia oil and isopropyl myristate (IPM) were evaluated for pH, density, viscosity, flow time, surface tension, contact angle, color distribution, injectability, antimicrobial activities and cytotoxicity. The most viscous camellia oil and olive oil showed the highest flow time with lowest color diffusion and lower injectability; thus, they were suitable for performing as external phase of ISM to prolong …

Vitex glabrata (VG) or Khai-noa fruit extracts were prepared from three methods; squeezing extract (SE), maceration extract (ME), and ethanol extraction (EE) to evaluate their effects in vitro. By the (1,1-diphenyl-2-picrylhydrazyl) scavenging assay, all extracts revealed very weak antioxidant. Results of (3-(4, 5-Dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide, a tetrazole) assay showed the increase in cell viability of SE at 100 and 500 μg/ml on TK6 cells after 24 and 48 h treatments (P < 0.01) without affecting Caco-2 cells. Conversely, a dose- and time-dependent cytotoxicity of all extracts revealed at 1000-2000 μg/ml on both cells (P < 0.01). SE showed cytotoxicity on PC-3 cells and anticancer activity when confirmed by clonogenic assay in a dose-dependent manner. SE also showed cytotoxicity on HT-29 cells after 24 h treatment but the cells could recurrent in the clonogenic assay. No effect was observed on MCF-7 cells after SE alone, SE co-treated with MMC treatment, and in the clonogenic assay. Moreover, HT-29 cells revealed a dose-dependent decrease in viability when treated with SE co-treated with MMC by 24 h. Hence, SE can selectively promote cell viability on TK6 and anticancer activity on PC-3. These results suggest that the different potential of SE depends on different cell lines.

Arylamine N-acetyltransferases, NAT1 and NAT2, catalyze the detoxification and/or activation of drugs and aromatic amine carcinogens. Single nucleotide polymorphisms or SNPs result in different human NAT2 genotypes thus dividing the population into rapid, intermediate, and slow acetylators. We hypothesize allelic variants of NAT2 genotype will decrease levels of N-acetylation, cytotoxicity, oxidative stress, DNA adduct formation, and mutagenesis compared to the reference allele NAT2*4. Cryopreserved human hepatocytes expressing different NAT2 genotypes and NER-deficient Chinese hamster ovary (CHO) cells transfected with human CYP1A2 and human NAT2*4, NAT2*5B or NAT2*7B have been used to investigate N-acetylation of different xenobiotics. In …

Preparation, Evaluation And Metabolites Study In Rats Of Novel Isoginkgetin-Loaded Tpgs/Soluplus Mixed Nanomicelles, Xue Feng, Yu-Ting Chen, Lu-Ya Li, Yu-Peng Sun, Hai-Rong Wang, Lan-Tong Zhang
Journal of Food and Drug Analysis
At present, cancer is one of the most lethal diseases in the world, and researchers are committed to developing effective anticancer drugs. Isoginkgetin (IGG) is a kind of biflavone with the potential to treat cancer due to the features of altering the cell cycle and inhibiting tumor cell infiltration. However, its solubility, absorbability and bioavailability are poor, so in this study, IGG was prepared into mixed nanomicelles and evaluated in vitro and in vivo. After condition optimization, IGG-loaded TPGS/soluplus mixed nanomicelles with particle size of 62.34 ± 1.10 nm, entrapment efficiency of 96.92 ± 0.66% and drug loading of 2.42 …

The sterol ergosterol and ergosta-4,22-diene-3β,7α-diol, together with 4-hydroxybenzaldehyde were isolated from the mushroom species Polyporus molluscensis, Cantharellus isabelinus, C. symoensii and a Podaxis species. Their structures were established on the basis of spectroscopic data. The ethanol extracts of mushroom samples of Agaricus sp. and Termitomyces letestui also showed cytotoxicity against the brine shrimp larvae.
