Parkinson's disease (PD) is a progressive neurodegenerative disorder marked by dopaminergic neuronal loss,oxidative stress, neuroinflammation, and a-synuclein aggregation. Despite advances, current treatments only offersymptomatic relief without altering disease progression. This review aims to evaluate the therapeutic potential of plant-derived phytochemicals in PD through evidence from both in vivo and in vitro experimental models. Key findingsindicate that rodent models (e.g., MPTP, rotenone) and neuronal cell lines (e.g., SH-SY5Y) help replicate PD pathologyand mechanistic insights. Phytochemicals such as baicalein, morin, ferulic acid, and bacopa demonstrate neuroprotectiveeffects via antioxidant, anti-inflammatory, and mitochondrial-stabilizing pathways. The high content of unsaturated fatty acids in avocado oil (Persea americana) has prompted in-depth exploration across research subjects. This study uses bibliometric analysis to assess global research trends on avocado oil and its fatty acid composition from 2004 to 2024. The search string retrieved 255 articles from the Scopus database and then analyzed using R BibliometriX and VOSviewer software. These tools analyze descriptive structures and collaborative patterns and create network data maps. For comparison, we separated the data into two research periods, namely 2004-2013 and 2014-2024. Introduction: Meridianin C (MC) is a marine-derived indole alkaloid that has demonstrated kinase inhibitory and anti-tumor activities. Despite its diverse biological properties, no previous reports have systematically evaluated the in vivo quantitative analysis of MC and its metabolites. Methods: In this study, MC was synthesized following a previously reported procedure with slight modifications. A sensitive, accurate, and reliable ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC–MS/MS) method was developed to simultaneously detect MC and its five major metabolites (MC-1-N-O-GluA, MC-1-N-O-SO3H, MC-2′-N-O-GluA, MC-2′-N-O-SO3H, and MC-O-GluA-didehydration) in rat plasma. Traumatic brain injury (TBI) is a leading cause of long-term neurological impairment, with aging significantly increasing vulnerability and worsening outcomes. Older individuals experience greater cognitive and motor deficits after TBI and often show diminished responses to therapeutic interventions. Both aging and TBI independently promote heightened neuroinflammation and cognitive dysfunction, increasing susceptibility to poor recovery. Sleep is a fundamental biological need, and sufficient sleep is required for physical and mental well-being. Sleep is regulated by two biological mechanisms: sleep-wake homeostasis and circadian rhythm. Circadian rhythms are near 24-h rhythms in behavior and physiology that are generated by endogenous mechanisms. In mammals, the suprachiasmatic nucleus (SCN) is the central circadian pacemaker, which is known to receive a significant amount of serotonergic input that likely modulates sleep rhythms. Many preoperative and postoperative cataract patients struggle with comprehending the use of prescription medication as directed. Language barriers and low health literacy levels are major factors contributing to improper use of prescriptions. To increase patients comprehension, the Fort Lauderdale Eye Institute employed an educational intervention consisting of a live presentation and an instructional video. Results found that 44% of patients were hesitant to ask questions to clinical staff, 32% felt overwhelmed, and nearly 70% lacked confidence in using their prescribed eye drops. Rett syndrome (RTT) is a rare neurodevelopmental disorder caused by mutations in the gene coding for methyl-CpG binding protein 2 (MeCP2). Currently, there is no cure for RTT and few effective treatment options to address the social, cognitive, motor, and respiratory symptoms that patients face. With the goal of identifying novel targets, our lab performed RNA-sequencing experiments in human autopsy samples and found decreased levels of the M1 muscarinic acetylcholine receptor. Pseudolysin (PLN) protein is a major virulent factor in Pseudomonas aeruginosa infection. PLN belongs to the family of zinc-dependent metalloproteases. It is naturally inhibited by a smaller protein called streptomyces metalloprotease inhibitor (SMPI), secreted by Streptomyces nigrescens. Previous and current studies demonstrate that the interaction between PLN and SMPI offers unique mechanistic insights for drug development against infections caused by P. aeruginosa. Ofloxacin, a second-generation fluoroquinolone, is a relatively rare but documented cause of fixed drug eruption (FDE), a localized cutaneous hypersensitivity reaction marked by recurrent, well-demarcated erythematous patches or macules that may develop into blisters upon re-exposure to the offending agent. The underlying mechanism is immune-mediated, involving T-cell activation, cytokine release, and keratinocyte apoptosis. Auditory processing begins in the cochlea and ascends through brainstem relays to the cortex, where sound features with increasing complexity are encoded. The 40 Hz auditory steady-state response (ASSR) reflects induced gamma synchrony shaped by local cortical circuit dynamics. Disrupted 40 Hz ASSR is a consistent finding in schizophrenia, linked to N-methyl-D-aspartate receptor (NMDAR) hypofunction in inhibitory interneurons, making it a promising translational biomarker. Synthetic cathinones are a widely abused class of new psychoactive substances that pose significant challenges in forensic toxicology owing to their complex metabolic pathways and lack of verified reference standards. This study investigated the in vitro metabolism of nine methylenedioxy-substituted synthetic cathinones, including methylone and eight structural isomers, which were categorized into three groups: Group 1 (dimethylone, ethylone, and butylone), Group 2 (dibutylone, pentylone, and eutylone), and Group 3 (N-ethylpentylone and dipentylone). Chronic atrophic gastritis (CAG) is precancerosis with limited treatment options. This study investigated the therapeutic effects of velvet antler (VA), a traditional natural medicine, on CAG, with a focus on ferroptosis. In vivo experiments demonstrated the efficacy of VA in alleviating CAG, and network pharmacology analysis revealed its involvement in related terms, including inflammation, oxidative stress, and cell proliferation. Analyses further confirmed the modulation of ferroptosis by VA, particularly via the Keap1-Nrf2/HO-1/GPX4 pathway. Jiao Sanxian (JSX) consists of three traditional Chinese medicine including charred hawthorn, malt and Liu Shen Qu, they are also from Chinese medicine homologous food. It is commonly used to relieve functional dyspepsia (FD). The present study aims to explore the mechanism of JSX (granule), with the addition of prebiotics for the treatment of FD using mice model. The network pharmacology and molecular docking were used to forecast therapeutic targets for JSX. Attention Deficit Hyperactivity Disorder (ADHD) is a common neurodevelopmental disorder in children, characterized by inattention, hyperactivity, and impulsivity. The American Academy of Pediatrics (AAP) endorses stimulant medications as the first-line treatment option. When combined with behavioral therapy, this pharmacologic class can improve focus, executive functioning, and the overall quality of life. While stimulant prescriptions can have a positive effect when used as directed, there is concern for the potential of provider overprescription and patient misuse related to these medications. Background: Methotrexate (MTX) is the cornerstone disease-modifying anti-rheumatic drug for the treatment of Juvenile Idiopathic Arthritis (JIA). While most patients receiving MTX have a favorable outcome, approximately 30% do not respond, and several experience toxic events, suggesting interindividual difference. Furthermore, evaluation of MTX response takes several months, with the risk of missing the early “window of opportunity” for treatment. Predicting MTX response prior to administration would greatly benefit these patients and their treating physicians by saving valuable time during the early stages of the disease onset. Acute monocytic leukemia (monocytic AML) is a subtype of AML marked by a proliferation of abnormal monoblasts. This subtype represents approximately 10% of AML cases. The prognosis of monocytic AML is poor, with a 5-year survival of ~30%. Most patients are diagnosed at an age when they are unlikely to survive first-line non-targeted cytotoxic chemotherapy as a bridge to hematopoietic stem cell transplant (HSCT). Even patients who achieve remission commonly relapse. This population of patients would greatly benefit from precision-targeted therapies but currently there are none approved for monocytic AML.

Opioid use disorder occurs alongside impaired risk-related decision-making, but the underlying neural correlates are unclear. We developed an approach-avoidance conflict task using a modified conditioned place preference procedure to study neural signals of risky opioid seeking in the prefrontal cortex, a region implicated in executive decision-making. Following morphine conditioned place preference, rats underwent a conflict test in which fear-inducing cat odor was introduced in the previously drug-paired side of the apparatus. While the saline-exposed control group avoided cat odor, the morphine group included two subsets of rats that either maintained a preference for the paired side despite the presence of …

Substance use disorders (SUD) in the United States are steadily increasing. Potent and dangerous substances such as methamphetamine (MA) and fentanyl (FEN) are often co-used as a polysubstance (POLY), either intentionally or unknowingly as FEN is a common adulterant. SUD is characterized by physiological and psychological dependence involving the negative reinforcement of anhedonia and withdrawal which drive hypermotivated drug-taking behaviors. Current theory suggests that recruitment of hypocretin/orexin (HCRT) neuropeptide in reward and stress associated brain regions contributes to drug dependence. Pyridostigmine bromide (PB) is an organophosphate (OP) nerve agent prophylactic used to protect warfighters against chemical warfare agents. OPs inhibit the neural enzyme acetylcholinesterase (AChE) in the brain. This leads to the buildup of acetylcholine, causing respiratory failure, seizures, and death. PB functions by inhibiting ~30% of peripheral AChE temporarily, protecting the enzymes so that the body is able to restore sufficient cholinergic function post-exposure. PB does not cross the blood-brain barrier, though it may still indirectly affect toxicity in the brain due to repeated AChE inhibition throughout treatment, or by inhibition of non-target ChE’s in the blood. Serotonin 5-HT receptors were one of the first serotonin receptors to be pharmacologically characterized. In mammals, they are expressed throughout the body in nearly every cell and tissue type, with the highest density in cortical layer V of the brain. They are involved in several aspects of normal physiological processes and behaviors and have been implicated in the etiology of neuropsychiatric diseases such as schizophrenia. Atypical antipsychotics have targeted blockade of 5-HT receptors as part of their therapeutic mechanism. Fear-related disorders are often treated with exposure therapy, a technique based on the concept of extinction. However, this type of therapy is ineffective for many individuals, so finding a pharmacological adjunct that augments the extinction of fear could lead to better treatment outcomes. In recent preclinical work, investigators have shown that psychedelics can accelerate the extinction of fear, potentially through their impact on neurotrophic signaling. We previously found that psilocybin exerted sex-dependent effects on the extinction of conditioned fear to an isolated cue (i.e., a tone). Specifically, psilocybin enhanced the extinction of cue fear in male rats but impaired it …
