Chemistry & Biochemistry Faculty Publications
Nanomaterials possess unusually high surface area-to-volume ratios and surface-determined physicochemical properties. It is essential to understand their surface-dependent toxicity in order to rationally design biocompatible nanomaterials for a wide variety of applications. In this study, we have functionalized the surfaces of silver nanoparticles (Ag NPs, 11.7 ±+2.7 nm in diameter) with three biocompatible peptides (CALNNK, CALNNS, CALNNE) to prepare positively (Ag-CALNNK NPs+ζ), negatively (Ag-CALNNS NPs−2ζ), and more negatively charged NPs (Ag-CALNNE NPs−4ζ), respectively. Each peptide differs in a single amino acid at its C-terminus, which minimizes the effects of peptide sequences and serves as …

Physiologically-Based Pharmacokinetic Modeling Of Acetaminophen Metabolism And Toxicity, David M. Ng, Ali Navid
Physiologically-Based Pharmacokinetic Modeling Of Acetaminophen Metabolism And Toxicity, David M. Ng, Ali Navid
STAR Program Research Presentations
Acetaminophen is a common analgesic and antipyretic. Metabolism of acetaminophen and acetaminophen-induced liver necrosis are predicted using physiologically-based pharmacokinetic (PBPK) modeling. Pharmacokinetic means the model determines where the drug is distributed in the body over time. Physiologically-based means the anatomy and physiology of the human body is reflected in the structure and functioning of the model. Acetaminophen is usually safe and effective when taken as recommended, but consumption at higher levels may lead to liver damage. Additionally, other factors such as alcoholic liver disease, smoking, and malnutrition affect the maximum safe dose of acetaminophen.

Cationic Amphiphilic Drug-Induced Autophagosome Accumulation Is Due To Autophagosome Sequestration Within Vimentin Intermediate Filament Networks Resulting In Prolonged Autophagosome Half-Life, Miriam Devorah Kleinman
Wayne State University Dissertations
Accumulations of autophagosomes and non-esterified cholesterol are
observed in several cell lines derived from lysosomal storage diseases,
including Niemann Pick Type C (NPC). The relationship between
autophagosome accumulation and lysosomal non-esterified cholesterol is
unclear. Exposure of murine hepatoma 1c1c7 cultures to the cationic
amphiphilic drugs (CADs) U18666A, imipramine and clozapine caused
lysosomal non-esterified cholesterol and autophagosome accumulation.
Measurement of LC3-II conversion in the presence of lysosomal inhibitors
bafilomycin A1 and NH4Cl, degradation of long-lived proteins, and
colocalization of GFP-LC3 and LAMP1 indicated an increase in
autophagosome synthesis without compensatory increase in clearance.
Autophagosome synthesis was blocked using 3-MA to monitor …

Classification Of Plants According To Their Heavy Metal Content Around North Mara Gold Mine, Tanzania: Implication For Phytoremediation, N. Mganga, Mlk Manoko, Z. K. Rulangaranga
Tanzania Journal of Science
Plants like other living organisms respond differently under different environmental conditions. An elevated level of heavy metals is one of the stresses which results into three classes of plants depending on their heavy metal content. The classes of plant species according to their accumulated heavy metals around North Mara Gold Mine were not known. To study such classes, a line transect of 700m long was established opposite the gold mine wastes. A total of eight sampling points were systematically established each after every 100m in that transect. Fifteen plant species were sampled; at least one species per sampling point. Approximately …
Compensatory Renal Hypertrophy, Mitochondria And Redox Status, Bavneet Benipal
Compensatory Renal Hypertrophy, Mitochondria And Redox Status, Bavneet Benipal
Wayne State University Dissertations
A reduction in functional renal mass occurs in humans during aging and severe kidney damage from diseases, injuries, infections and congenital conditions and after nephrectomy. Nephrectomy, or surgical removal of a kidney or a section of a kidney, is performed for treatment of unilateral secondary renal cancer, infections and for kidney transplantation. As a result, the remaining renal tissue undergoes compensatory growth due primarily to hypertrophy, in which both the size and functional capacity of the remaining kidney are increased. Renal compensatory hypertrophy is associated with a series of physiological, morphological and biochemical changes that also have toxicological implications.

Tanzania Journal of Science
The paper discusses sources, concentrations and physicochemical parameters of heavy metals of environmental concern in the wetlands of Lake Victoria basin in Tanzania. The metals investigated include copper (Cu), lead (Pb), zinc (Zn), cadmium (Cd), chromium (Cr), nickel (Ni), arsenic (As), manganese (Mn), cobalt (Co) and mercury (Hg). These metals are released into the wetlands via weathering and erosion of rock formations, mining operations and industrial activities, discharge of municipal and domestic waste, and use of agrochemicals. The levels of these heavy metals in soil, sediment, water, and biota differ. Wetlands impacted by gold mining activities in the South Lake …
Smoking, Anemia, And Risk Of Oral Clefts In Utah, Melinda Michelle Moss
Smoking, Anemia, And Risk Of Oral Clefts In Utah, Melinda Michelle Moss
All Graduate Theses and Dissertations, Spring 1920 to Summer 2023
Cigarette smoke contains sufficient carbon monoxide to induce maternal and fetal hypoxia. Hypoxia is a known teratogen, and consequently maternal smoking has been the focus of many studies on adverse birth outcomes, including cleft lip and palate. Current literature of epidemiological studies on smoking and clefts suggests a modest but statistically significant increase in risk of clefting associated with maternal smoking. A biological condition that may also contribute to hypoxia is anemia. Data from the Utah Child and Family Health Study was used to assess the effects of hypoxia-inducing conditions, maternal smoking, anemia, and their interaction, on the risk of …

Heavy Metal Contamination Of Mangrove Sediments And The Associated Biota In Dar Es Salaam, Tanzania, Sd Mremi, Jf Machiwa
Tanzania Journal of Science
Mangrove wetlands are efficient in trapping pollutants that may have detrimental effects on mangrove dependent food chains. Mangrove ecosystems that are within urban areas are likely to suffer more from chemical pollution than those in rural areas. Heavy metals in mangrove plant parts, sediments and crabs from mbweni, msimbazi and mtoni mangrove ecosystems in Dar es salaam were analysed using an atomic absorption spectrophotometer, in order to assess the impact of heavy metal pollution on mangrove biota. Sediment samples from msimbazi and mtoni mangrove areas which are located within the city, had higher levels of pb, zn and cu than …

Heavy Metals And Organic Pollutants In Sediments Of Dar Es Salaam Harbour Prior To Dredging In 1999, J F Machiwa
Tanzania Journal of Science
Heavy metals and organic contaminants were analysed in the sediments of the inner area of the Dar es Salaam harbour. Complementary analytical parameters, such as, the organic carbon content and the silt/clay fraction of the sediments showed good positive correlation (r = 0.64). Stations that had high content of fine grain material in the sediment also indicated relatively high level of organic carbon and pollutants. The Florida criteria (MacDonald 1993) for assessment of pollution of tropical marine sediments was adopted in oredr to evaluate the extent of pollution in Dar es Salaam harbour sediments. The Florida criteria is one of …

In an effort to elucidate the mechanism by which indomethacin (IN) attenuates the stimulatory effect of estradiol (E$\sb2$) on rabbit splenic red pulp macrophages (RPM), thirty-nine female New Zealand White rabbits were divided into 10 groups: ovariectomized (OVX), OVX/IN at 0.1 and 5.0 mg/kg body weight (bw)/day; sham OVX (SOVX), SOVX/IN at 0.1 and 5.0 mg/kg bw/day; OVX/25 mg E2, OVX/25 mg E$\sb2$/IN at 0.1 and 5.0 mg/kg bw/day; intact Control. Quantitative changes in RPM population in response to the treatments were measured using a 0 to 4 histologic grading scale. Estradiol treatment resulted in increased RPM grade when compared …
Covalent Binding And Other Mechanisms Of Primaquine Toxicity, Lucille S. K. Chau Yau
Covalent Binding And Other Mechanisms Of Primaquine Toxicity, Lucille S. K. Chau Yau
Loma Linda University Electronic Theses, Dissertations & Projects
A dose-dependent covalent binding of primaquine to mouse liver microsomal protein was found in both in vitro and in vivo studies. The binding in vitro was linear during the first 90 minutes of incubation and dependent on the concentration of mouse liver microsomal protein. The in vitro covalent binding was markedly enhanced by cofactors. Pretreatment of mice with phenobarbital, a cytochrome P450 inducer, increased the microsomal cytochrome P450 content and the in vitro covalent binding significantly. Binding was markedly inhibited when microsomal metabolism inhibitors (SK&F-525A, metyrapone and piperonyl butoxide), and sulfhydryl-contai ni ng compounds were added to the incubation system. …
