The direct incorporation of carboxylated carbon nanotubes (f-CNTs) into hydrophobic drug particles during their formation via anti-solvent precipitation is presented. The approach is tested using two drugs namely antifungal agent Griseofulvin (GF) and antibiotic Sulfamethoxazole (SMZ) that have very different aqueous solubility. It is observed that the f-CNTs dispersed in the water serve as nucleating sites for crystallization and are readily incorporated into the drug particles without altering crystal structure or other properties. The results show that the hydrophilic f-CNTs dramatically enhance dissolution rate for both drugs. The inherent heterogeneity of tumor tissue presents a major challenge to nanoparticle-medicated drug delivery. This heterogeneity spans from the molecular to the cellular (cell types) and to the tissue (vasculature, extra-cellular matrix) scales. Here we employ computational modeling to evaluate therapeutic response as a function of vascular-induced tumor tissue heterogeneity. Using data with three-layered gold nanoparticles loaded with cisplatin, nanotherapy is simulated with different levels of tissue heterogeneity, and the treatment response is measured in terms of tumor regression. The results show that tumor vascular density non-trivially influences the nanoparticle uptake and washout, and the associated tissue response. The drug …

The aim of this study was to determine the levels of heavy metals: lead, cadmium, copper, zinc, arsenic and mercury in facial cosmetics (lipstick, lip glossy, facial powder, foundation, eyeliner, eye shadow and mascara) which were purchased randomly in Dar es Salaam, Tanzania. The levels of lead, cadmium, copper and zinc were determined using Atomic Absorption Spectrometry (AAS). The levels of arsenic were determined using Hydride Generation Atomic Absorption Spectrometry (HGAAS), and levels of mercury were determined using Cold Vapour Atomic Absorption Spectrometry. This paper aims to shed light on the dissonance caused by the superimposition of Dominant Human Systems on Natural Systems. I highlight the synthetic nature of Dominant Human Systems as egoic and linguistic phenomenon manufactured by a mere portion of the human population, which renders them inherently oppressive unto peoples and landscapes whose wisdom were barred from the design process. The study aimed at assessing the levels of heavy metals and cyanide in water and sediments in villages surrounding Buzwagi gold mine, both during wet and dry seasons. A total of 56 samples of water (28 from each season) and 44 samples of sediments (22 from each season) were collected from the wells in the area and were analyzed for total cyanide and heavy metals. The total cyanide was analyzed spectrophotometrically whilst the heavy metals were analyzed by an Inductively Coupled Plasma (ICP). Status Of Selected Heavy Metals Dispersion From Top Soil In And Around Automobile Workshop Areas In Zanzibar Municipality, Tanzania, John Am Mahugija, Hawa M Sheikh

The distributions of selected heavy metals in top soil in areas with and without automobile workshops in Zanzibar municipality were investigated. Sampling was done during dry and rainy seasons. The samples were digested with aqua regia (HCl and HNO3, 3:1) and analysed by Atomic Absorption Spectrophotometry. The concentrations of zinc, copper and lead in the samples ranged 0.27–419.2, 0.10–167 and 0.02–271 mg/kg dry weight, respectively. The concentrations were higher in areas with automobile workshops than those without. The concentrations of the heavy metals were highest at point sources and decreased with increase in distance during the dry season. The concentrations …

The Human Cytochrome P450 (CYP) Allele Nomenclature Database, a critical resource to the pharmacogenetics and genomics communities, will be transitioning to the Pharmacogene Variation (PharmVar) Consortium. In this report we provide a summary of the current database, provide an overview of the PharmVar consortium and highlight the PharmVar database which will serve as the new home for pharmacogene nomenclature.

This is largely because modern medicine is often expensive, inaccessible, or has undesirable side effects. This study investigates use of traditional medicine in Morogoro Urban district where people do have access to hospitals. In particular, this study aims to assess the utilization and accessibility the plant species Securinega virosa for medicinal use by identifying and interviewing the actors involved in the supply chain from harvester to consumer. “Kratom” is the common name for the botanical mitragyna speciosa. It is a tree native to Southeast Asia in which leaves contain the psychoactive alkaloids mitragynine and 7-hydroxymitragynine. Kratom is often ingested as teas, chewed, or smoked. It acts as a stimulant in small doses and as an opioid in large doses. Overdoses can result in vomiting, seizures, and death. Recently the Drug Enforcement Agency (DEA) placed Kratom on Schedule 1, but, due to public outcry, it was almost immediately removed.

Currently the quantitation of opiates at the Erie County Medical Examiner’s Office Toxicology Laboratory is conducted through whole blood analysis. The objective of this thesis project was to determine if the analysis of opiates could be conducted through liver analysis, and if the analysis of opiates would provide a more accurate quantitation compared to the blood analysis. The quantitation of opiates was conducted from the livers of sixty-four possible overdose cases that were brought into the Erie County Medical Examiner’s Office between 2013 and 2015. Effective verification for compliance with water quality standards in uranium mining in Tanzania requires data sensitive to monitor heavy metal concentration in water around the Mkuju River Uranium Project before mining commences. The area susceptible for pollution by the project was estimated using AERMOD dispersion model and found to cover about 1300 km2. Thirty one surface and groundwater samples were collected and analysed for heavy metals and physicochemical properties using ICP-MS and standards techniques, respectively. Calpain mediated cleavage of CDK5 natural precursor p35 causes a stable complex formation of CDK5/p25, which leads to hyperphosphorylation of tau. Thus inhibition of this complex is a viable target for numerous acute and chronic neurodegenerative diseases involving tau protein, including Alzheimer’s disease. Since CDK5 has the highest sequence homology with its mitotic counterpart CDK2, our primary goal was to design selective CDK5/p25 inhibitors targeting neurodegeneration. Antimicrobial resistance has been an exponentially growing problem since the discovery of antibiotics. Antibiotics have been misused for many years and this misuse has grown into a real problem for the medical community. While there are countless safeguards to prevent infection by a resistant strain of bacteria, there are still many plagued by it and must be treated with sometimes dangerous antibiotics. Melittin, along with many other peptides, contain potent antimicrobial properties, but are also toxic toward enthrocytes. The control of the secondary structure of peptides provides the key to adjusting their activity.

Parkinson's disease (PD) is the second most common neurodegenerative disorder after Alzheimer's disease, affecting approximately 0.3% of the total U.S. population, and its prevalence increases with age. Two neuropathological hallmarks of PD are the loss of dopaminergic neurons in the substantia nigra pars compacta, a region in the midbrain involved in initiating and sustaining movement, and the presence of cytosolic inclusions called Lewy bodies (LBs) in various brain regions. Nanomaterials possess unusually high surface area-to-volume ratios and surface-determined physicochemical properties. It is essential to understand their surface-dependent toxicity in order to rationally design biocompatible nanomaterials for a wide variety of applications. In this study, we have functionalized the surfaces of silver nanoparticles (Ag NPs, 11.7 ±+2.7 nm in diameter) with three biocompatible peptides (CALNNK, CALNNS, CALNNE) to prepare positively (Ag-CALNNK NPs+ζ), negatively (Ag-CALNNS NPs−2ζ), and more negatively charged NPs (Ag-CALNNE NPs−4ζ), respectively. The scope of this work focuses on computationally modeling compounds with protein structures. While the impetus of drug discovery is the innovation of new therapeutic molecules, it also involves distinguishing molecules that would not be an effective drug. This can be achieved by inventing new tools or by refining old tools. Virtual screening (VS, also called docking), the computational modeling of a molecule in a receptor structure, is a staple in predicting a molecule's affinity for an intended target. The β2 adrenergic receptor (β2AR) regulates smooth muscle relaxation in the vasculature and airways. Long- and Short-acting β-agonists (LABAs/SABAs) are widely used in treatment of chronic obstructive pulmonary disorder (COPD) and asthma. Despite their widespread clinical use we do not understand well the dominant β2AR regulatory pathways that are stimulated during therapy and bring about tachyphylaxis, which is the loss of drug effects. Thus, an understanding of how the β2AR responds to various β-agonists is crucial to their rational use.
