This thesis examines the disproportionate burden of lead exposure carried by low income, POC communities. The systemic nature of this problem is a symptom of a longstanding legacy of environmental injustice in the United States. Decades of federal neglect are reflected in the higher statistics of lead exposure and poisoning in predominantly black communities. While it is understood that lead exposure poses a serious threat to physical health and early cognitive development, there is a discouraging lack of urgency to remove the toxin from non-wealthy communities. Pharmaceutical contaminants are increasingly prevalent in aquatic environments, yet their sublethal effects on ecologically relevant behaviors remain poorly understood, particularly in estuarine systems. Selective serotonin reuptake inhibitors (SSRIs), such as fluoxetine, are of particular concern due to the conserved role of serotonin in regulating behavior across vertebrates. Here, we investigated how acute exposure to environmentally relevant concentrations of fluoxetine influences anti-predator behavior in Gulf killifish (Fundulus grandis), a key estuarine fish commonly found in saltmarsh habitats. Brown Bullhead Ameiurus nebulosus, Yellow Bullhead Ameiurus natalis, and Bluegill Lepomis macrochirus are unique in their stark differences. Bullhead are scaleless bottom feeders that can tolerate high pollution and low O2 conditions. In contrast, Bluegill are scaled, aggressive nesters and are less tolerant of high pollutants or low-oxygen conditions. Among these pollutants include a class of chemicals called neonicotinoids (hereafter referred to as neonics). This study examines the effects of acute exposure to crystal violet (CV) on the freshwater zooplankton Daphnia pulex in response to growing concerns over synthetic dyes from industrial runoff in aquatic ecosystems. The primary objective is to assess CV's impact on D. pulex's heart rate, mobility, morphology, and lethality via a series of acute exposure tests. 48-h acute toxicity (0-2000 μg/L), 48-h immobilization (0-180 μg/L), and 9-h heart rate (0-2,000 μg/L) tests were conducted using third-generation daphnids, followed by stereomicroscopic examination and statistical analysis. Our work was focused on the impact of 5, 10, 15, 20, 25 and 30 mg/kg of Imidacloprid the nicotinergic neuronal pathway inhibitor (N-ACh receptors) on the activities of mitochondrial detoxification enzymes (Catalase, SOD, GST and GPx) and acetylcholine esterase (AchE) in forebrain region of Wistar rats. Despite the opioid crisis taking headlines, the abuse of cocaine and amphetamines remains rampant and laboratory instruments must be able to effectively and reliably handle the overwhelming case load. In this study, an Agilent 6475 Triple Quadrupole (QQQ) LCMS, recently acquired by the Tarrant County Medical Examiner’s Office (TCMEO) in Fort Worth, Texas, was validated for identifying and quantifying CocAmps. Results show that the instrument was fit-for-purpose and that the instrument passed all validation tests. Reproducibility was ≤ 20% RSD for 12 QC replicates, 6 of blood and 6 of urine. We are facing an aging crisis, with 20% of the U.S. population projected to be geriatric (65+) by 2030 and live another 40+ years. Age-related diseases accompany a growing geriatric population, emphasizing the need to understand their etiology. Environmental pollutants compound this crisis by: 1) geriatrics are more susceptible, exacerbating age-related diseases and comorbidities, and 2) they accelerate biological aging, inducing age-related diseases at younger ages. Ciguatera poisoning (CP) is one of the most prominent seafood-borne illnesses worldwide, affecting between 25,000–500,000 people annually. Ciguatera is caused by the ingestion of seafood contaminated with algal-derived ciguatoxins (CTX), the precursors of which are produced by benthic and toxigenic dinoflagellates in the genera Gambierdiscus and Fukuyoa. These compounds enter marine food webs through herbivory and are subsequently transferred to higher trophic levels, undergoing complex metabolic biotransformations, which produce structurally related CTX derivatives. Consumption of CTX induces a variety of severe neurological, gastrointestinal, and/or cardiovascular symptoms beginning within hours and lasting for weeks to months following exposure. Acute myeloid leukemia (AML) is a commonly diagnosed cancer in smokers. When current or former smokers have AML, they have worse survival compared to never smoking patients. This has been observed clinically for decades, but then it is unknown how smoking leads to worsened AML survival. Smoking causes oxidative stress and altered DNA methylation that persists for decades in peripheral blood mononuclear cells, but these changes from smoking have not been evaluated in the context of AML. Methamphetamine (MAMP) is a highly addictive illicit drug typically abused for its nervous system stimulating effects. Conversely, methamphetamine has therapeutic use treating attention deficit hyperactivity disorder, controlling appetite and assisting with weight loss, and is available as the pure l-isomer in over-the-counter (OTC) nasal inhalers due to its decongestant activity. Because l-methamphetamine (l-MAMP) is available in OTC form, forensic guidelines require a sample to contain greater than 20% d-methamphetamine (d-MAMP) when classifying results as consistent with illicit MAMP use. Hexavalent Chromium [Cr(VI)] is a known human lung carcinogen and general health hazard. The mechanism of carcinogenesis remains poorly understood, but chromosome instability (CIN) is the major theory in its carcinogenic mechanism. Homologous recombination (HR) repair is a DNA repair pathway that prevents CIN by repairing DNA double-strand breaks. RAD51, a key mediator protein of HR repair, along with the RAD51 paralogs (RAD51B, C, D, XRCC2, and 3) are required for HR repair. Although these drugs are widely utilized, little is known about the cellular transport of platinum drugs. It is hypothesized that platinum utilizes transporters for other metals to enter the cell. The exposome encompasses the concept that an individual’s health is a combination of genetic, environmental, and occupational exposures. A worker’s exposomal profile could potentially influence their susceptibility to hazardous occupational exposures. Over 35% of adults in the United States have metabolic dysfunction (MetDys), yet interactions between MetDys and hazardous occupational exposures are generally unknown. MetDys is associated with HFWD-consumption and obesity and induces a chronic low grade systemic inflammation and alters serum adipokine profiles. To estimate the extent of PCB detections in Los Angeles County schools, I applied a mathematical model to estimate the range of PCB detections in schools built between 1950 and 1984. Cell-penetrating peptides containing arginine as positively charged residues and tryptophan or diphenylalanine as hydrophobic residues were synthesized. The synthesis was accomplished through the Fmoc solid-phase peptide synthesis in the presence of HBTU and DIPEA. The side-chain protected linear peptides were cleaved from the resin and cyclized in the presence of DIC and HOAt in the solution phase overnight. MALDI-TOF mass spectrometry was used to characterize the peptides.

The cytotoxicity of the synthesized peptides was determined in CCRF-CEM (human, lymphoblast peripheral blood), and HEK-293 (human, embryonic epithelial kidney healthy) cells using the MTS assay. A concentration of 10 µM was found …

Atmospheric carbon dioxide is rising at an accelerated rated due to increased anthropogenic activities. Metals have also been a noted problem; however, little research has addressed combined exposure of both pollutants to sensitive, calcifying organisms in freshwater habitats. This study examined copper accumulation (over 7 d) and activity of the antioxidant enzymes, catalase and glutathione peroxidase (over 2 d), in the freshwater common pond snail, Lymnaea stagnalis, after exposure to ambient and increased (2000 µatm) CO2 and copper (control, 5, and 20 µg/L). Seasonality is a consistent component of aquatic ecosystems yet most fish biological and ecotoxicological studies commonly employ field sampling protocols focused during the warm open water season with minimal emphasis placed on winter sampling, especially for north-temperate latitude ecosystems. Such strategies limit our understanding of poikilotherm biology and ecology during the overwintering seasons. Here, I investigated seasonal changes in yellow perch (Perca flavescens) biology, ecology and ecotoxicology over a one-year period in Lake Manganese. Nanotechnology takes advantage of cellular biology’s natural nanoscale operations by interacting with biomolecules differently than soluble or bulk materials, often altering normal cellular processes such as metabolism or growth. To gain a better understanding of how copper nanoparticles hybridized on cellulose fibers called carboxymethyl cellulose (CMC) affected growth of Saccharomyces cerevisiae, the mechanisms of toxicity were explored. Multiple methodologies covering genetics, proteomics, metallomics, and metabolomics were used during this investigation. Agricultural herbicides enter aquatic environments after rain events where they affect microscopic aquatic plants (phytoplankton) and animals (zooplankton) that form the base of aquatic food webs. Atrazine, an herbicide with low solubility in water, is often mixed with surfactants such as alkyl polyglucoside (APG) to improve effectiveness. Although APG has low-toxicity, a potential drawback to increased atrazine solubility is greater adverse effects on aquatic organisms. I hypothesized that atrazine and APG decrease phytoplankton abundance more than atrazine alone. Thiamethoxam with mefenoxam is the most widely used neonicotinoid insecticide/fungicide mixture applied to soybean (Glycine max [L]) as seed treatments. Based on the systemic nature of thiamethoxam and mefenoxam, residues of this insecticide/fungicide mixture may be present in soybean vegetative and floral tissue and negatively impact beneficial insects. Although neonicotinoids are often applied in combination with systemic fungicides, the research on ecological risks of neonicotinoids has been focused on the analysis of these compounds without considering their interaction with other agrochemicals. Alcoholic liver disease (ALD) is characterized by gut dysbiosis and hepatic and intestinal inflammation. Gut microbiota is critically maintained by antimicrobial peptides (AMPs) expressed in the epithelial and immune cells. AMPs are also identified as immune-regulators in various diseases. Previous studies demonstrated that deficiency of AMPs REG3 lectins exacerbated alcoholic steatohepatitis. LL-37 is the sole member of human cathelicidin AMP family, named CRAMP in mouse. Despite of the extensive studies in infectious diseases and in gut-microbiota-related inflammatory bowel disease, the role of LL-37/CRAMP in ALD is yet unknown. This paper aims to shed light on the dissonance caused by the superimposition of Dominant Human Systems on Natural Systems. I highlight the synthetic nature of Dominant Human Systems as egoic and linguistic phenomenon manufactured by a mere portion of the human population, which renders them inherently oppressive unto peoples and landscapes whose wisdom were barred from the design process. Widespread use of agrochemicals such as herbicides and antibiotics increases their likelihood of entering aquatic systems in mixture. Despite different modes of action, atrazine (herbicide) and tetracycline (antibiotic) adversely affect non-target photosynthetic organisms, such algae and macrophytes, with the potential to reduce food availability to higher trophic levels. However, the effects of simultaneous exposure to both contaminants have yet to be determined. I hypothesized that a mixture of atrazine and tetracycline affects freshwater communities differently than each compound alone.
