The dinoflagellate Karenia brevis, blooms annually in the Gulf of Mexico, producing a suite of neurotoxins known as the brevetoxins. The cellular toxin content of K. brevis, however, is highly variable between or even within strains. I investigated biochemical differences between high (KbHT) and low (KbLT) toxin producing cultures both derived from the Wilson strain, related to energy-dependent quenching (qE) by photosystem II, and the content of reduced thiols of the proteome. By characterizing the xanthophyll content of the two strains I was able to determine that KbLT performs qE inconsistently. To investigate the …

The coffee ring effect (CRE) phenomenon is originated from the nonuniform solvent evaporation of a sessile droplet deposited onto the flat substrate. Once the evaporation starts, the droplet suspended particles move outwards by the radial flow and concentrate in the edge region of the evaporating droplet, resulting in the formation of the so-called CRE stains. In this work we have expanded the applications of the CRE from separation of particles and macromolecules to small molecules, in particular, coupled to surface-enhanced Raman spectroscopy (SERS). Ochratoxin A (OTA) is a potent mycotoxin found in a wide range of agricultural products that has been linked to mitochondrial damage and renal disease. The standard methods for OTA analysis currently rely on the use of high-performance liquid chromatography (HPLC) coupled to fluorescence detection or mass spectrometry. Toward a highthroughput analysis of OTA, a single-stranded DNA aptamer, modified with a fluorophore, coupled to a complementary sequence, modified with a FRET-based quencher that dissociates in the presence of the target toxin, is proposed. This study analyses the spatial and temporal variability of chlorophyll-a in Chesapeake Bay; assesses the performance of Ocean Color 3M (OC3M) algorithm; and develops a novel algorithm to estimate chlorophyll-a for coastal shallow water. The OC3M algorithm yields an accurate estimate of chlorophyll-a concentration for deep ocean water (RMSE=0.016), but it failed to perform well in the coastal water system (RMSE=23.17) of Chesapeake Bay. A novel algorithm was developed which utilizes green and red bands of the Moderate Resolution Imaging Spectroradiometer (MODIS) sensor. The novel algorithm derived the chlorophyll-a concentration more accurately in Chesapeake Bay (RMSE=4.20) than the OC3M algorithm. …

Research in the multidisciplinary science of ecotoxicology is crucial to assess injuries to ecosystem resources from chemical spills or other stressors used to support environmental decision-making. Established guidelines recommend the use of non-standard native species in toxicity investigations. This work focused on the use of native species for aquatic toxicity assessment to make more relevant conclusions on the potential for adverse biological effects to occur as a result to single chemical exposures or exposures to a complex mixture like oil. Estrogens play a major role in the initiation and progression of breast cancer. Elevated lifetime exposure to estrogens is associated with an increased risk of developing breast cancer. Degraded pollinator habitat may have far-reaching consequences for recovery of imperiled flowering plant populations. Studies indicate that broad-spectrum insecticides used in mosquito abatement are detrimental to non-target invertebrates, including pollinators. A decline in efficient pollinators can reduce plant fitness by decreasing reproductive output and constraining genetic diversity, a challenge for rare plants.
In 2015, I monitored flower visitation and fruit set of three imperiled plant species throughout protected areas on three islands in The Lower Florida Keys. An ecological risk assessment was conducted on the risk to fish of chemical contaminants detected in the habitat of Albula vulpes in South Florida, to evaluate whether contaminants may be a driver of declines in the recreational bonefish fishery. All available contaminant detection data from Biscayne Bay, Florida Bay, and the Florida Keys were compared to federal and state guidelines for aquatic health to identify Contaminants of Potential Ecological Concern (COPECS). The contamination of the environment, accidental or intentional, in particular with chemical toxins such as industrial chemicals and chemical warfare agents has increased public fear. There is a critical requirement for the continuous detection of toxins present at very low levels in the environment. The central Everglades serve as a Wildlife Management Area and as a Water Conservation Area for the Miami-Fort Lauderdale metropolitan area. It is also home to over 22 nonnative freshwater fish species and carries restrictive fish consumption guidelines for Mercury. In my study, boat anglers and canal bank anglers were personally interviewed in the field, to research their awareness and perspectives of these potential environmental and health threats. The study found 78% of anglers were aware of the presence of nonnative fish species, but favored native fish species, and that 69% were aware of mercury advisories, but did not eat …

Since the U.S.-led invasion of Iraq in 2003, epidemics of birth defects and cancers are rising in many Iraqi cities. In 2012, the World Health Organization (WHO) and the Iraqi Ministry of Health (MoH) undertook a large-scale epidemiological study to determine the prevalence of birth defects in the Iraqi population. A report which appeared on the WHO website in September 2013, claims that "The rates for spontaneous abortion, stillbirths and congenital birth defects found in the [Iraq] study are consistent with or even lower than international estimates." This thesis analyses buckwheat as a cover crop in Florida. The study was designed to demonstrate: soil enrichment with nutrients, mycorrhizal arbuscular fungi interactions, growth in different soil types, temperature limitations in Florida, and economic benefits for farmers. Buckwheat was planted at the FIU organic garden (Miami, FL) in early November and harvested in middle December. After incorporation of buckwheat residues, soil analyses indicated the ability of buckwheat to enrich soil with major nutrients, in particular, phosphorus. Symbiosis with arbuscular mycorrhizal fungi increased inorganic phosphorus uptake and plant growth. Regression analysis on aboveground buckwheat biomass weight and soil characteristics showed …

The purpose of this study is to investigate supervisory support as a moderator of the effects of role conflict and role ambiguity on emotional exhaustion and job satisfaction. This study also examines the moderating role of supervisory support on the relationship between emotional exhaustion and job satisfaction. Data were collected from a sample of frontline hotel employees in Northern Cyprus. The aforementioned relationships were tested based on hierarchical multiple regression analysis. The results demonstrate that supervisory support mitigates the impact of role conflict on emotional exhaustion and further reveal that supervisory support reduces the effect of emotional exhaustion on job …
