Tetracycline is an antibiotic that has been prescribed for COVID-19 treatment, raising concerns about antibiotic resistance after long-term use. This study reported fluorescent polyvinylpyrrolidone-passivated iron oxide quantum dots (IO QDs) for detecting tetracycline in biological fluids for the first time. The as-prepared IO QDs have an average size of 2.84 nm and exist a good stability under different conditions. The IO QDs’ tetracycline detection performance could be attributed to a combination of static quenching and inner filter effect. Recently, cannabidiol (CBD), one of the major components of the Cannabis species, has been a focus in the cannabis industry due to its various pharmacological effects. Interestingly, CBD can be converted into several psychoactive cannabinoids, such as 9-tetrahydrocannabinol (Δ9-THC) and its structural isomers, under acidic reaction conditions. In this study, chemical transformation of CBD in ethanol solution was conducted with variation in pH at 2.0, 3.5, and 5.0 by addition of 0.1 M hydrochloric acid (HCl). A metabolomics-based approach to data analysis is required for drug metabolites to be identified quickly. This study developed such an approach based on high-resolution mass spectrometry. Our approach is a two-stage one that combines a time-course experiment with stable isotope tracing. Pioglitazone (PIO) was used to improve glycemic management for type 2 diabetes mellitus. Consequently, PIO was taken as a model drug for identifying metabolites. During Stage I of data analysis, 704 out of 26626 ions exhibited a positive relationship between ion abundance ratio and incubation time in a time-course experiment. Guilu Erxian Jiao (GEJ) is a commonly used nutritional supplement due to its rich content of amino acids. It is also a traditional herbal medicine for improving degenerative joint. This study aimed to investigate the effect and mechanism of GEJ water extract (GEJ-WE) on skeletal muscle in C2C12 myotubes and C57BL/6J mice. Analysis of GEJ-WE were performed by high-performance liquid chromatography fingerprinting with chemical standards. Protein expression, mRNA level, glycogen content, mitochondria activity and ATP level were evaluated by western blots, real-time PCR, PAS staining, MTT and ATP bioluminescence assay, respectively. Lipid oxidations products (LOPs) are reactive mutagenic and carcinogenic species known to be generated in thermally stressed culinary oils. Mapping the evolution of LOPs in culinary oils exposed to standard frying practices – both continuous and discontinuous thermo-oxidation – at 180℃ is vital to our understanding of these processes, and to the development of scientific solutions for their effective suppression. Modifications in the chemical compositions of the thermo-oxidised oils were analysed using a high-resolution proton nuclear magnetic resonance (1H NMR) technique. Red mold rice (RMR) is a traditional Chinese medicine prepared using Monascus fermentation. M. ruber (pilosus) and M. purpureus have a long history of use as food and medicine. As an economically important starter culture, the relationship between the taxonomy of Monascus and production capabilities of secondary metabolites is crucial for the Monascus food industry. In this study, monacolin K, monascin, ankaflavin, and citrinin production by M. purpureus and M. ruber were genomically and chemically investigated. There were few reports about antibiotic residues in egg-containing products. In the study, an effective method for the simultaneous determination of 24 sulfonamide antibiotics in two instant pastries based on a modified QuEChERS sample preparation technique coupled with ultra performance liquid chromatography-tandem mass spectrometry was developed. The results show that the average recoveries of the SAs at 5, 10, and 50 µg·kg-1 levels were 67.6%-103.8%, with relative standard deviations (RSD) of 0.80-9.23%. The limit of detections (LODs) and limit of quantitations (LOQs) were 0.01-0.14 µg∙kg-1 and 0.02-0.45 µg∙kg-1, respectively. This method was suitable for analysis of …

Erinacines derived from Hericium erinaceus have been shown to possess various health benefits including neuroprotective effect against neurodegenerative diseases, yet the underlying mechanism remains unknown. Here we found that erinacine S enhances neurite outgrowth in a cell autonomous fashion. It promotes post-injury axon regeneration of PNS neurons and enhances regeneration on inhibitory substrates of CNS neurons. Using RNA-seq and bioinformatic analyses, erinacine S was found to cause the accumulation of neurosteroids in neurons. ELISA and neurosteroidogenesis inhibitor assays were performed to validate this effect. This research uncovers a previously unknown effect of erinacine S on raising the level of neurosteroids.

The Non-Biological Complex Drug (NBCD) Working Group defines an NBCD as “a medicinal product, not being a biological medicine, where the active substance is not a homo-molecular structure, but consists of different (closely related and often nanoparticulate) structures that cannot be isolated and fully quantitated, characterized and/or described by physicochemical analytical means”. Background: The rise in facemask waste with the uncertainty of deposition data and all its concomitant pressure on management has resulted in new risks and challenges for achieving sustainable development goals (SDGs). The purpose of this mixed methods study was to estimate facemask waste generation and examine management in Bhutan since the COVID-19 pandemic.

The goal is to develop a point-of-care biosensor for the detection of live pathogens contaminating beef products. Biosensing of live pathogens is based on isothermal amplification of nucleic acid on a paper-based device. A colorimetric dye is employed as an indicator of the amplification product for visual result. The assay incorporates a compound Propidium monoazide (PMA) that makes the DNA from dead cells inaccessible for amplification. This approach is especially applicable for pathogens that can enter a viable but non-culturable state (VBNC).

Exposure to certain per-and polyfluoroalkyl substances (PFAS) has been shown to be positively associated with total and/or low-density lipoprotein cholesterol. Examining this association in lipid lowering interventions may provide additional evidence linking PFAS to cardiovascular risk. We examined the relationship of 6 PFAS with cholesterol in a 6-month lifestyle-based intervention. We quantitated PFAS in 350 individuals at baseline and post intervention and examined associations of PFAS with cholesterol before and after intervention. Food frequency questionnaires and GIS analyses were used to investigate PFAS hotspots and possible exposure routes. In the course of maintaining a safe, mission-ready aircraft fleet and operational runways, United States Airmen experience a range of occupational risks. These risks include inhalable hazards generated from oft-used maintenance practices, such as blasting which emits silica from the media abrasives as well as contaminants from the exterior coatings. Additional inhalation hazards range from engine and fuel emissions, chemical agents, radiation, and explosives, to naturally occurring and human-manufactured biopathogens. Numerous dietary quality indices exist to help quantify overall dietary intake and behaviors associated with positive health outcomes. Most indices focus solely on biomedical factors and nutrient or food intake, and exclude the influence of important social and environmental factors associated with dietary intake. Using the Diet Quality Index- International as one sample index to illustrate our proposed holistic conceptual framework, this critical review seeks to elucidate potential adaptations to dietary quality assessment by considering—in parallel—biomedical, environmental, and social factors. The use of plant-based medicine dates back centuries, and cannabis (Cannabis sativa) is one such plant that has been used medicinally and illicitly. Although cannabis contains hundreds of cannabinoids and other natural products, its potential medicinal use was largely ignored by modern researchers due to the legal restrictions and heavy regulations introduced in the 1930s. As restrictions on cannabis access have eased since the 1990s there is renewed interest in the research of cannabinoids and the other components in the cannabis plant. Cilostazol was investigated as a protective agent against lipopolysaccharide (LPS)-associated acute kidney damage in mice. Cilostazol (50 mg/kg/day; p.o) administered for 7 consecutive days before a single LPS dose (2 mg/kg; i.p) hampered serum creatinine and cystatin C as well as renal KIM-1 and NGAL, repressed TLR4 and MYD88 transcription as well as NF-κB p65, IL-1β and MDA content, boosted Nrf-2 mRNA expression, HO-1 activity and GSH content. This was synchronous with an upregulation of p-PI3K and p-Akt expressions. Collectively, cilostazol prevented LPS renal injury which might correspond to modulation of TLR4/NF-κB, Nrf2/HO-1 and PI3K/Akt pathways.

Ceriodaphnia dubia is an EPA-recommended organism used in regulatory toxicity testing of surface waters and commercial chemicals. They are often cultured for extended periods in the laboratory with little knowledge of how it impacts subsequent generations. This thesis investigated if C. dubia cultured for an extended period were representative of three recently-collected field populations by comparing their culturing characteristics and sensitivities to toxicants. Comparative chronic toxicity tests with sodium chloride, the neonicotinoid insecticide thiamethoxam, and mixtures of the two toxicants measured differences in sensitivities among the laboratory and field cultures, specifically in the variability of responses. Tris(1,3-dichloro-2-propyl) phosphate (TDCPP) is an organophosphate flame retardant. The primary TDCPP metabolite, bis(1,3-dichloro-2-propyl) phosphate (BDCPP), is detectable in the urine of over 90 % of Americans. Epidemiological studies show sex-specific associations between urinary BDCPP levels and metabolic syndrome, which is an established risk factor for type 2 diabetes, heart disease, and stroke. We used a mouse model to determine whether TDCPP exposure disrupts glucose homeostasis. Six-week old male and female C57BL/6J mice were given ad libitum access to diets containing vehicle (0.1 % DMSO) and TDCPP resulting in the following treatment groups: 0 mg/kg/day, 0.02 mg/kg/day, 1 mg/kg/day, or 100 …

A variety of molecules can be radiolabeled and delivered to a cancer site for the purposes of diagnostics and therapy. Among the most promising of tumor targeting molecules are peptides and antibodies. These bio-inspired molecules can be designed and synthesized to target and respond to cancer cells based on the properties of those cells. Matrix metalloproteinase (MMP) enzymes are over-expressed by some metastatic cancers, in which they are responsible for the degradation and remodeling of the extracellular matrix. Successful invasive plant management—where invaders are sufficiently reduced and diverse native plant communities recover—remains an elusive goal for land managers. The site- and landscape-scale drivers of variable management outcomes and vegetation recovery are poorly understood due to a lack of rigorous experiments that characterize longer term vegetation trends across contexts. We present the results of a five-year experiment across eight subestuaries of Chesapeake Bay, representing a gradient of watersheds with differing dominant land-use types and anthropogenic impacts, to evaluate invasive and native plant response to herbicide management. This fact sheet explores data from the 2022 Brookings Institution report by Joseph W. Kane, Adie Tomer, Caroline George, and Jamal Russell Black called “Not according to plan: Exploring gaps in city climate planning and the need for regional action." The original report analyzes decarbonization plans for 50 of the largest U.S. cities and comparatively ranks each plan using a point system of 5 categories. This fact sheet focuses on Mountain West cities (Denver, CO; Phoenix, AZ; Las Vegas, NV; Salt Lake, UT; and Albuquerque, NM) included in the original study.
