Use Of Small Molecule Fanconi Anemia Pathway Inhibitors As Sensitizing Agents To Laromustine., Sam W. Marchant

Laromustine is an experimental chemotherapeutic sulfonyl hydrazine prodrug shown in clinical trials to be effective against acute myeloid leukemia. The mechanism of action of laromustine involves interstrand crosslinking, via chloroethylation, and enzyme inhibition, caused by carbamoylation. The work described herein aims to investigate whether inhibition of the replication-dependent interstrand crosslink repair Fanconi Anemia pathway further sensitizes cells to laromustine. By measuring metabolic activity immediately after drug exposure, we find laromustine to be equally as cytotoxic towards Fanconi Anemia deficient and wild type cells. However, through clonogenic assays we show Fanconi Anemia mutations sensitize cells to laromustine’s anti-proliferative effect. Furthermore, we …

Natural habitats of fish are gradually declining due to land use and pollution caused by industrial wastes, intensive agriculture and contamination from sewage systems. The United States has 14,780 wastewater treatment facilities that discharge effluent into navigable waters, and in some cases these discharge waters represent a considerable proportion of the water system flow. Lead (Pb) is ubiquitous in the environment and exposure can have adverse health effects even at low concentrations, particularly in young children. Blood lead levels (BLLs) are the best indicator of recent Pb exposure. Based upon data from the NHANES, the Centers for Disease Control and Prevention’s (CDC) blood Pb (BPb) reference value, defined as the 97.5th percentile of children 1-5 years old, was lowered from 10 μg/dL to 5 μg/dL in 2012. According to more recent NHANES data, the 97.5th percentile has decreased to 3.5 μg/dL. If the CDC recommends a further lowering of the BPb reference value, many …

Objective: The purpose of this study is to evaluate the role of farnesol as a potential neuroprotective agent against 3-nitropropionic acid (3-NP) acid-induced Huntington’s disease (HD) by in vitro, in vivo, and in silico models. 3-NP acid-induced Huntington’s disease in male Wistar rats was used to evaluate the neuroprotective potential of farnesol. Materials and Methods: 3-NP (10 mg/kg/day) was used for the induction of disease, followed by treatment with 50 mg/kg and 100 mg/kg of farnesol for 7 days. Infantile colic (IC) is an important area of current research due to the extreme distress it causes parents and their infants. It is vital that a cause is isolated so that treatment can be found because IC is a risk factor for child abuse. In this paper, two major theories were posed to elucidate the underlying cause of IC—the gastrointestinal model and the neurological model. The gastrointestinal model suggests that IC stems from issues such as an immature gut. The neurological model suggests that infantile migraines are the causative agents of IC. Hashimoto’s Thyroiditis (HT) is one of the most common autoimmune diseases (Hiromatsu et al. 2013 p.13). It primarily affects the thyroid gland. The thyroid gland influences growth and regulates the body’s metabolism by manipulating hormonal levels. Hashimoto’s thyroiditis affects thyroid function through mechanisms that cause the hormone levels to become dysregulated. The standard therapy for Hashimoto’s thyroiditis is hormone replacement. This approach helps most patients by regulating their thyroid hormones, though there are some individuals who fail treatment. A successful cure has not yet been discovered and therefore society is urged to take proper precautions to prevent the spread of the potentially fatal virus. Type 2 diabetes (T2D) is a chronic low-grade inflammatory disease. A T cell cytokine profile (Th17) from PBMCs can distinguish obese T2D from obese non-diabetes subjects. Individual T cell subsets interact with each other and the diverse subsets jointly determine overall inflammation. Cellular metabolism drives cytokine production of CD4+ T cells, and therefore contributes to inflammation in T2D. However, specific changes in metabolism and function of CD4+ T cells during the progression from lean healthy to obese and diabetic stages in people have not been clarified.

Glioblastoma is one of the deadliest cancers, with a median survival of only one year. Even after aggressive treatment consisting of surgical resection, radiation, and chemotherapy, most glioblastoma patients suffer from tumor recurrence within 6-9 months. One reason for treatment failure of anticancer drugs is the blood-brain barrier that protects the brain by impeding xenobiotic uptake from the blood. To this end, efflux transporters at the human blood-brain barrier, such as P-glycoprotein (ABCB1) and Breast Cancer Resistance Protein (ABCG2), prevent many compounds, including anticancer drugs, from entering the brain. Immune checkpoint inhibitors (ICIs), especially those that target programmed cell death protein 1 (PD-1) and programmed cell death ligand-1 (PD-L1), have been shown to provide substantial clinical benefit in many patients with non-small cell lung cancer (NSCLC). While these therapeutic agents can be highly effective in the correct context, the biological systems that malignant cells draft from normal activities of the cell are poorly characterized. Tumor cell-specific expression of PD-L1 is likely important for clinical benefit from PD-1 and PD-L1 inhibitors. Reactive semivolatile contaminants, such as isocyanates, can be particularly difficult to assess in occupational environments. While standard methods exist for isocyanates, there are still significant differences between the results they provide for various occupational environments or processes. This study presents the validation of a laboratory system for the generation of controlled atmospheres of isocyanates. A system consisting of different modules generated airborne methylene diphenyl diisocyanate (MDI) by nebulizing a solution into mixing and exposure chambers with control of flow rate, temperature, and relative humidity. Sampling was performed through an eight-port flow splitter that allowed only very slight within-test variability. MDI …
