Investigating The Synergistic Effects Of Two Curcuminoids And Cisplatin On Cancer Cell Migration, Blaine Patty Apr 2018

Investigating The Synergistic Effects Of Two Curcuminoids And Cisplatin On Cancer Cell Migration, Blaine Patty

Mahurin Honors College Capstone Experience/Thesis Projects

Cisplatin is a common chemotherapy drug used to treat various cancers; however, it is relatively ineffective against many cancers, including several types of lung cancer. One approach that could improve cisplatin’s effect is to combine it with another drug that produces a synergistic response greater than either drug alone. Curcumin, a naturally occurring plant compound, has been investigated for synergisms in conjunction with cisplatin chemotherapy, but curcumin use is hampered by its low bioavailability. This project investigated whether two synthetic curcumin analogs, EF-24 and CLEFMA (curcuminoids), which have greater solubility than curcumin, could, when combined with cisplatin, decrease the migration …

Investigating The Synergistic Effects Of Two Curcuminoids And Cisplatin On Cancer Cell Reactive Oxygen Species, Matthew Millay Apr 2018

Investigating The Synergistic Effects Of Two Curcuminoids And Cisplatin On Cancer Cell Reactive Oxygen Species, Matthew Millay

Mahurin Honors College Capstone Experience/Thesis Projects

Cisplatin is an anticancer drug which can cause the production of reactive oxygen species (ROS) that kill cancer cells. Curcumin is a naturally occurring plant compound that can increase ROS levels in cancer cells and enhance the activity of cisplatin against cancer, but it exhibits poor bioavailability. We investigated whether two synthetic curcumin analogs (curcuminoids), EF-24 and CLEFMA, with anti-cancer activity and improved bioavailability, increased cisplatin’s effect against cancer. A spectrophotometric fluorescent ROS assay was used to determine if cisplatin, the curcuminoids or combinations of cisplatin with a curcuminoid affected the level of ROS in the A549 non-small cell lung …

The Combined Effects Of Atrazine And Tetracycline On Primary Producers And Zooplankton In Freshwater Microcosms, Madison R. Kelly Apr 2018

The Combined Effects Of Atrazine And Tetracycline On Primary Producers And Zooplankton In Freshwater Microcosms, Madison R. Kelly

Honors College Theses

Widespread use of agrochemicals such as herbicides and antibiotics increases their likelihood of entering aquatic systems in mixture. Despite different modes of action, atrazine (herbicide) and tetracycline (antibiotic) adversely affect non-target photosynthetic organisms, such algae and macrophytes, with the potential to reduce food availability to higher trophic levels. However, the effects of simultaneous exposure to both contaminants have yet to be determined. I hypothesized that a mixture of atrazine and tetracycline affects freshwater communities differently than each compound alone. A microcosm experiment was conducted to test effects of environmentally relevant concentrations of atrazine (1 μg L-1) and tetracycline …

Evaluating The Aryl Hydrocarbon Receptor As A Target For Pharmacologic Activity Of Repurposed Drugs, Teofilo Borunda Duque Apr 2018

Evaluating The Aryl Hydrocarbon Receptor As A Target For Pharmacologic Activity Of Repurposed Drugs, Teofilo Borunda Duque

Pharmaceutical Sciences ETDs

The discovery of new pharmacologic targets is important for the advancement of pharmacotherapy and identification of new indications for current drugs. The aryl hydrocarbon receptor (AHR) is a physiologic sensor of both chemical environmental pollutants and ligands of natural origin. Given the broad spectrum of ligands that activate the AHR and its relationship with toxicology, the AHR is not thought to be a traditional target for pharmacotherapy. However, multiple studies have shown potential for the AHR as a novel pharmacologic target Therefore, identifying less toxic agents that modulate the AHR may elucidate mechanisms for pharmacological targeting of the AHR. The …

Water Capacity Of Paramo Soils, Rafael Robert Apr 2018

Water Capacity Of Paramo Soils, Rafael Robert

Independent Study Project (ISP) Collection

The pȧramo ecosystems in the high Ecuadorian Andes are grasslands characterized by challenging abiotic factors such as low temperatures and high solar radiation. Another key feature of pȧramos is the humidity and water availability in the ecosystem. Pȧramos are very important to the hydrology of the inter-Andean valley because they are water reserves that distribute fresh water to lower elevations. These hydrologic properties are made possible by the porous volcanic ash soils formed by the volcanic arc along the mountain range. This study is focused in the region of Chimborazo approximately 20 km from Riobamba the capitol city of the …

Barriers To The Elimination Of Plastic In Single Use Beverage Containers In Byron Bay, Nsw, Katherine Senechal

Independent Study Project (ISP) Collection

Plastic debris is a global threat to the natural environment. The accumulation of plastic debris has resulted in the destruction of marine habitat as well as a threat to marine wildlife. Single use plastics, including plastics associated with single use food and beverage containers are contributing to a considerable amount of debris and other litter in the world’s oceans. As awareness of this issue spreads, both large corporations and small businesses are beginning to transition to plastic alternatives for their single use beverage containers. In Byron Bay, Australia, an area of increasing environmental awareness, businesses are beginning to reduce single …

An Inordinate Fondness For Beetles: A Study Of Insect Species Diversity And Abundance In Mazumbai Forest Reserve Versus Nearby Agricultural Areas, Emma Weisner

Independent Study Project (ISP) Collection

This study investigates how human disturbance of ecosystems alters insect diversity and abundance, specifically exploring how insect communities inside Mazumbai Forest Reserve in Tanzania differ from insect communities in agricultural areas near the reserve. Following methods of previous studies on the effect of disturbance on insect populations (Bellamy et al. 2018; McLaughlin & Mineau 1995; Perry et al. 2016), this research utilizes pitfall traps and yellow bowl traps in multiple locations throughout the two study areas to catch insects, which are then identified to their specific order. The collected data support the hypothesis that insect order diversity and abundance vary …

Integration Of Plastic Recycling Within A Smaller Community: Reducing Plastic Pollution In The Environment, Olivia Palenscar

Independent Study Project (ISP) Collection

The purpose of this research is to bring awareness to plastic pollution and find a way integrate plastic recycling workshops in a smaller community. Plastic pollution is a worldwide issue. Plastic is a versatile material and can be used to make almost anything, which is an attractive trait but the amount of plastic being consumed is alarming and only continues to increase. Plastic essentially never goes away because the degradation of plastic can last hundreds of years. The challenge is to find ways that everyone can recycle plastic because over 90% of plastic does not end up being recycled. Most …

Maternal Nicotine Exposure Leads To Augmented Expression Of The Antioxidant Adipose Tissue Triglyceride Lipase Long-Term In The White Adipose Of Female Rat Offspring., Nicole Barra, Taylor Vanduzer, Alison C. Holloway, Daniel B. Hardy Apr 2018

Maternal Nicotine Exposure Leads To Augmented Expression Of The Antioxidant Adipose Tissue Triglyceride Lipase Long-Term In The White Adipose Of Female Rat Offspring., Nicole Barra, Taylor Vanduzer, Alison C. Holloway, Daniel B. Hardy

Physiology and Pharmacology Publications

Globally, approximately 10-25% of women smoke during pregnancy. Since nicotine is highly addictive, women may use nicotine containing products like nicotine replacement therapies for smoking cessation, but the long-term consequences of early life exposure to nicotine remain poorly defined. Our laboratory has previously demonstrated that maternal nicotine exposed (MNE) rat offspring exhibit hypertriglyceridemia due to increased hepatic de novo lipogenesis. Hypertriglyceridemia may also be attributed to impaired white adipose tissue (WAT) lipid storage; however, the effects of MNE on WAT are not completely understood. We hypothesize that nicotine-induced alterations in adipose function (e.g. lipid storage) underlie dyslipidemia in MNE adults. …

Resveratrol And Pinostilbene Confer Neuroprotection Against Aging-Related Deficits Through An Erk1/2-Dependent Mechanism, Erika N. Allen, Sneha Potdar, Victor Tapias, Mayur Parmar, Cassia S. Mizuno, Agnes Rimando, Jane E. Cavanaugh

HPD Articles

Age-related declines in motor function may be due, in part, to an increase in oxidative stress in the aging brain leading to dopamine (DA) neuronal cell death. In this study, we examined the neuroprotective effects of natural antioxidants resveratrol and pinostilbene against age-related DAergic cell death and motor dysfunction using SH-SY5Y neuroblastoma cells and young, middle-aged, and old male C57BL/6 mice. Resveratrol and pinostilbene protected SH-SY5Y cells from a DA-induced decrease in cell viability. Dietary supplementation with resveratrol and pinostilbene inhibited the decline of motor function observed with age. While DA and its metabolites (DOPAC and HVA), dopamine transporter, and …

Individuals with type 2 diabetes (T2D) and dyslipidemia are at an increased risk of cardiovascular disease. Fibrates are a class of drugs prescribed to treat dyslipidemia, but variation in response has been observed. To evaluate common and rare genetic variants that impact lipid responses to fenofibrate in statin‐treated patients with T2D, we examined lipid changes in response to fenofibrate therapy using a genomewide association study (GWAS). Associations were followed‐up using gene expression studies in mice. Common variants in SMAD3 and IPO11 were marginally associated with lipid changes in black subjects (P < 5 × 10‐6). Rare variant and gene expression changes …

Ambiguities In The Efficacy Of Selective Serotonin Reuptake Inhibitors In The Treatment Of Depression, Stephanie Giblin Apr 2018

Ambiguities In The Efficacy Of Selective Serotonin Reuptake Inhibitors In The Treatment Of Depression, Stephanie Giblin

Writing Across the Curriculum

Major depressive disorder (MDD) is the most debilitating mental illness worldwide. Primary treatment of MDD is the administration of selective serotonin reuptake inhibitors (SSRIs). Deemed as the safest psychotropic drug thus far, SSRIs have been shown to reduce depressive symptoms and enhance the quality of life in youth, adult and geriatric populations. However, with rising numbers of individuals taking maintenance doses of SSRIs for five or more years, researchers are beginning to discover severe adverse side effects associated with prolonged treatment. The overall risks of long-term SSRI therapy include tachyphylaxis, tardive dysphoria, structural neurological abnormalities and suicide. SSRIs also have …

Discovery And Effects Of Pharmacological Inhibition Of The E3 Ligase Skp2 By Small Molecule Protein-Protein Interaction Disruptors, John K. Morrow Apr 2018

Discovery And Effects Of Pharmacological Inhibition Of The E3 Ligase Skp2 By Small Molecule Protein-Protein Interaction Disruptors, John K. Morrow

Dissertations and Theses (Open Access)

Skp2 (S-phase kinase-associated protein 2), one component of the SCF E3 ubiquitin ligase complex, directly interacts with Skp1 and indirectly associates with Cullin1 and Rbx1 to bridge the E2 conjugating enzyme with its protein substrate to execute its E3 ligase activity. Skp2 is an Fbox protein (due to it containing an Fbox domain) and it is the rate-limiting component of the SCF complex. Skp2 targets several cell-cycle regulatory proteins for ubiquitination and degradation; most notable and significant for cancer are the cyclin-dependent kinase inhibitor, p27. Skp2 is an oncogene and studies have shown that over-expression of Skp2 leads to increased …

Effects Of Radiation On Neurite Morphology And Cytoskeleton Structure, Melissa Sassman Apr 2018

Effects Of Radiation On Neurite Morphology And Cytoskeleton Structure, Melissa Sassman

Student Scholarship

Long-term manned space exploration to the moon, Mars, and other areas beyond Earth's protective magnetic field poses possible acute and late central nervous systems (CNS) risks. Of particular concern for astronauts is exposure to high atomic number, high energy particles known as HZE particles, a component of galactic cosmic radiation (GCR). Although NASA has radiation safety requirements, the possible effects of GCR and HZE particles on the central nervous systems of astronauts remains unknown. Understanding the risks and effects of galactic cosmic radiation and HZE particles on the central nervous system will allow for safer space exploration.

Additionally, and perhaps …

Signal Detection Of Adverse Drug Reaction Using The Adverse Event Reporting System: Literature Review And Novel Methods, Minh H. Pham

USF Tampa Graduate Theses and Dissertations

One of the objectives of the U.S. Food and Drug Administration is to protect the public health through post-marketing drug safety surveillance, also known as Pharmacovigilance. An inexpensive and efficient method to inspect post-marketing drug safety is to use data mining algorithms on electronic health records to discover associations between drugs and adverse events.

The purpose of this study is two-fold. First, we review the methods and algorithms proposed in the literature for identifying association drug interactions to an adverse event and discuss their advantages and drawbacks. Second, we attempt to adapt some novel methods that have been used in …

Developing Novel Therapeutics For Bacterial Lung Infections, Brandon J. Baer, Ruud Veldhuizen, Cory Yamashita Mar 2018

Developing Novel Therapeutics For Bacterial Lung Infections, Brandon J. Baer, Ruud Veldhuizen, Cory Yamashita

Western Research Forum

Background: Bacterial lung infections are leading causes of death worldwide. Unfortunately, increasing resistance to antibiotics and the inflammation often accompanying these infections are leading to poor outcomes despite antibiotic intervention. Complicating treatment further, the tree-like branching structure of the lung makes drug delivery to distal sites of infection difficult. Our research aims to address these challenges by developing new therapeutics and new tools to improve and assess drug delivery, bacterial killing and inflammation. Our therapy combines host defense peptides, which have been shown to kill antibiotic-resistant bacteria and down regulate inflammation, with a pulmonary vehicle, exogenous surfactant, that can improve …

Killi-Data News (Winter), Tyrone Genade

Killifish Research Review

Valued readers, it is with a heavy heart that I inform you that this is the last issue of Killi-Data News. The good news is that we will be back as Killifish Research Review. The dissolution of Killi-Data International created a prob- lem: how can the newsletter of a defunct organization live on without that organization? But other additional problems were building in the background. The first issue numbered 15 pages. The previous issue was 28 pages. The number of killifish related papers is increasing while time on our end (the editorial team) is running out. It takes a lot …

Killi-Data News (Fall), Tyrone Genade

Killifish Research Review

Many interesting papers have been published over the last three months. The large volume of papers coupled with the start of the new college semester (and the workload it brings) delayed this issue of Killi-Data News. But better late than never—or so I hope! In this issue Richard van der Laan provides an insightful review of the recent Aphanius papers as to their taxonomic implications and questions. The systematic issues he raises show the importance of the Molecular project: we need to get more samples of the various cyprinodontiforme families to resolve unsettled systematic and taxonomic issues. In the Next …

Killifish Research Review

This is the fourth edition, and concluding issue of the first volume, of Killi-Data News and I am happy that it has been well received by readers. At 25 pages this issue is a bit thin- ner than the last but this is because we agreed to make the cut-off for submissions the 1 st of December so we could get this edition out by the New Year. This is an exciting edition full of new species descrip- tions and analyses that will keep taxonomists busy for years to come. Costa has given us two molecular phylogenies on Melanorivulus as …

Agriculture In The Classroom Farm Day Lesson Plan, Tessa Nicole Thompson

Agricultural Education and Communication

Nontraditional educational programs have been proclaimed desirable by many teachers, and proven effective through data collection and feedback (Coombs and Ahmed, 1974). Of the more popular unconventional educational programs, outside learning activities are commonly implemented as they are not only advantageous to students, but to teachers as well (Cengelci, 2013; Knoblock, 2008). Though nontraditional, much thought must be put into creating lesson plans for these educational programs.

El Dorado County’s Agriculture in the Classroom program utilizes nontraditional outdoor learning activities to conduct “Farm Day.” This lesson plan, created for Farm Day, is to teach third graders the importance of composting …
