Background: Per- and polyfluoroalkyl substance exposure has been implicated as a factor contributing to the development of many adverse health outcomes across populations of all ages. Despite this, the factors associated with PFAS concentrations in humans are not well understood. Most studies evaluating factors associated with PFAS concentrations consider a small number of variables. Differences in the factors associated with PFAS levels in populations of high exposure versus low or background exposure (i.e., the general population) have not been considered. Heavy metal exposure from anthropogenic pollutants negatively affects many marine organisms, including foraminiferan protists. Foraminifera are often used as bioindicators in benthic environments due to their sensitivity to environmental stress. Primary research has commonly focused on how heavy metals affect foraminiferal assemblages, distribution, ultrastructure, and shell morphology. Multi-chambered calcareous and agglutinated forams are the primary focus of such ecotoxicological studies; however, less is known about how heavy metals affect the physiology and shell microarchitecture of monothalamid agglutinated species. Background: Air pollution leads to more than four million premature deaths worldwide and nearly 200,000 in the US each year. While there is extensive research relating PM2.5 to hospital visits for various health outcomes, there is relatively little research on ultrafine particles (UFPs), a particularly small and dangerous air pollutant. Another important exposure is temperature due to its association with dehydration, heat stress, and cardiovascular-related hospital admissions. The Akwesasne reservation along the St. Lawrence River has experienced significant exposure to several persistent organic pollutants due to rapid industrialization. The Aluminum Company of America, General Motors, and Reynolds Metals operated aluminum foundries that used polychlorinated biphenyls (PCBs) as hydraulic fluids, which contaminated the community environment, the soil, air and water, and community food sources with PCBs. We administered the World Health Organization Disability Assessment Schedule II (WHODAS II) to 119 Akwesasne Mohawk adults residing along the St. Lawrence River in New York. Phthalates have been implicated as reproductive toxicants in animal models and in human populations. This study examined associations between potential exposure sources and urinary phthalate concentrations among couples undergoing infertility treatment. Women (n=56) and their male partners (n=43) undergoing in vitro fertilization (IVF) were enrolled in a prospective cohort investigation of environmental chemicals and IVF outcomes at an academic medical center. On the day of oocyte retrieval, participants provided urine samples and completed questionnaires detailing use of personal care products (PCPs), and consumption of medications, foods, and beverages in the preceding 24-hours. 1,4-dioxane is a volatile organic compound categorized by the United States Environmental Protection agency (EPA, 2017) as a possible human carcinogen potent to cause tumors in humans. Lead (Pb) is ubiquitous in the environment and exposure can have adverse health effects even at low concentrations, particularly in young children. Blood lead levels (BLLs) are the best indicator of recent Pb exposure. Based upon data from the NHANES, the Centers for Disease Control and Prevention’s (CDC) blood Pb (BPb) reference value, defined as the 97.5th percentile of children 1-5 years old, was lowered from 10 μg/dL to 5 μg/dL in 2012. According to more recent NHANES data, the 97.5th percentile has decreased to 3.5 μg/dL. If the CDC recommends a further lowering of the BPb reference value, many …

Exposures to environmental contaminants, including non-essential toxic trace elements, may be associated with in vitro fertilization (IVF) outcomes. Follicular fluid (FF) envelops the developing oocyte, offering a “snapshot” of the oocyte microenvironment. FF may better estimate trace element concentrations associated with IVF outcomes than more commonly employed blood and urine biomarkers of exposure. Hence, FF concentrations of four widespread non-essential toxic trace elements (arsenic, mercury, cadmium, and lead) were investigated in relation to a spectrum of IVF endpoints in n=56 women undergoing IVF. Epitranscriptomic marks in the form of enzyme-catalyzed modifications to RNA nucleosides can be important regulators of translation and play integral roles in the response to stress. Dynamic changes in tRNA modification status can regulate the translation of stress response proteins whose transcripts have distinct codon biases. The epitranscriptomic writer Alkylation repair homolog 8 (ALKBH8) plays a crucial part in the translation of codon biased transcripts, as it regulates stop codon recoding, which is a specialized form of translation used to generate selenoproteins. Human biomonitoring for toxic metals such as lead (Pb) provides information about absorbed dose from all exposure routes, and is critical for identifying exposed individuals and assessing health risks. Blood Pb and bone Pb are the primary validated biomarkers for assessing short-term and historical Pb exposures, respectively, while non-traditional matrices, including keratinized tissues, can provide complementary exposure information. Hard keratinized tissues – including hair, nails, hooves, and horns – grow incrementally, potentially preserving chemical exposure histories over a span of days to years. Polychlorinated biphenyls (PCBs) are a class of persistent organic pollutants linked to changes in cognitive function and thyroid hormone (TH) levels. However, most studies of PCBs and cognitive function were conducted among children. No studies among older adults have prospectively collected PCB and cognitive performance measurements to evaluate the association over time, and no longitudinal studies have evaluated the association between PCBs and THs over time among a non-occupationally exposed population. The paving of the Interoceanic Highway (IOH) brought rapid development to the Madre de Dios (MDD) region in the Peruvian Amazon. Development brings concerns of toxic trace element exposures, and road development has been a major driver of the transition from traditional to calorie-dense processed ‘Western’ diets in lower and middle-income countries. Naphthalene (NA) is an omnipresent air pollutant and respiratory toxicant and a possible human lung carcinogen. NA induces cytotoxicity in airways following bioactivation by CYP (P450) enzymes. The overall goal of this study was to examine the relative contributions of hepatic and pulmonary CYPs to bioactivation, disposition and NA-induced pneumotoxicity in vivo. Substantial differences in the expression and enzymatic activity of P450s occur in various organs in human populations, which could modify susceptibility to NA-induced airway damage. The overall goal of the research conducted in this dissertation is to determine the role of cytochrome (CYP) P450-mediated xenobiotic bioactivation in the development of tobacco smoke-induced lung toxicity. The central hypothesis of this work is that CYP-mediated bioactivation of specific tobacco smoke constituents is an important contributor to the generation of tobacco smoke-induced lung inflammation and injury. This dissertation sought to assess the effect of cesarean delivery (CD) on the risk of developing wheezing or food allergies in early childhood; and whether breastfeeding mediates the association between CD and the outcomes. The use of silver nanoparticles (AgNPs) in a wide variety of consumer products (i.e. toothpastes, food containers, dietary supplements and garments) for their antimicrobial properties can lead to potential oral exposure in humans. To enhance their stability, AgNPs are coated with capping agents such as citrate and polyvinylpyrrolidone (PVP). Fine particulate (PM2.5) air pollution is a major issue in Saudi Arabia. Though the health effects of exposure to PM2.5 have been extensively studied in developed countries of Europe and North America, this research is yet to be done in developing countries of Africa, some parts of Asia and the Middle East including Saudi Arabia. This is the first comprehensive assessment of PM2.5 air pollution in Saudi Arabia describing the major emission sources as well as assessing the association of daily exposures to PM2.5 and its chemical constituents with cardiopulmonary morbidity.

Anthropogenic lead (Pb) pollution is ubiquitous in the environment and a risk factor for both human and wildlife health. Pb exposure has the potential to alter reproductive strategies with respect to mate choice and reproductive output. This could be especially deleterious if these changes disrupt adaptive behavioral and reproductive life history strategies. Therefore, the overall aim of this body of work was to examine the consequences of developmental Pb exposure on reproductive strategies, using Drosophila melanogaster as a model system. Engineered nanomaterials (ENMs) play an increasing role in manufacturing and consumer products. Currently, there is no standard approach to studying ENM toxicity, and a growing body of literature suggests that ENMs may have toxicity differing from similar compounds in bulk or dissolved form. I examined ENMs used in the semiconductor manufacturing process called chemical-mechanical planarization (CMP) for their properties, removal in the wastewater treatment system (WWT), in-vitro toxicity, and location post-inhalation in-vivo. It was found that ENMs in CMP slurries have morphology determined by their elemental composition, but assessment of size and concentration can differ substantially between accepted techniques. Follicular fluid (FF) is in direct contact with granulosa cells that surround a developing oocyte, and high density lipoprotein (HDL) is the sole lipoprotein found in FF. Therefore, we investigated 41 FF HDL particle components, including 26 HDL-cholesterol (HDL-C) subfractions measured for particle size, apolipoproteins, enzymes, and lipophilic micronutrients, in association with embryo quality on 180 women undergoing in vitro fertilization (IVF).
