In this thesis, delivery systems for siRNA delivery are introduced with special attention to non-viral vectors. Many successful vectors used in vivo were reviewed. Our work focused on the effect of different architectures of oligospermine polmers on their suitability for siRNA delivery in lung cancer cells. Different archituctures showed different polyplex structures and variable transfection efficiencies. Moreover, we presented a review on nanoimprint lithography techniques with an outlook on possible biological applications in the field of gene and drug delivery.

Complex engineered nanoparticles (CENPs) are being used on various applications. Their properties are different from those of neat nanoparticles. The dissertation explores these differences from four aspects: 1) Modify carbon nanomaterials’ inert surfaces and investigate the effect on thermal and rheological behavior of their dispersions; 2) Generate self-assembly bi-layer structure of oxide nanoparticles via surface modification; 3) Study interaction between lysozyme and different surface-charged ceria nanoparticles; 4) Investigate the biodistribution and transformations of CENPs in biological media.

The objective of this study was to determine the residual pro-or anti-oxidant effects in rat brain 30 days after systemic administration of a 5 nm citrate-stabilized ceria dispersion. A ∼4% aqueous ceria dispersion was iv-infused (0 or 85 mg/kg) into rats which were terminated 30 days later. Ceria concentration, localization, and chemical speciation in the brain was assessed by inductively coupled plasma mass spectrometry (ICP-MS), light and electron microscopy (EM), and electron energy loss spectroscopy (EELS), respectively. Because of its large surface area and easy access for both delivery and monitoring, the skin is an attractive target for gene therapy for cutaneous diseases, vaccinations and several metabolic disorders. The critical factors for DNA delivery to the skin by electroporation (EP) are effective expression levels and minimal or no tissue damage. Here, we evaluated the non-invasive multielectrode array (MEA) for gene electrotransfer. For these studies we utilized a guinea pig model, which has been shown to have a similar thickness and structure to human skin. The causes of urolithiasis include such influences as diet, metabolic disorders, and genetic factors which have been documented as sources that aggravate urinary calculi depositions and aggregations, and, implicitly, as causes of urolithiasis. This study endeavors to detail the scientific mechanisms involved in calcium oxalate calculi formation, and, more importantly, their inhibition under growth conditions imposed by the traditional medicinal approach using the herbal extract, Larrea tridentata. The calculi were synthesized without and with Larrea tridentata infusion by employing the single diffusion gel technique. The University of Nebraska and AgBios, Inc. launched the AgBiosafety website (http://agbiosaftzy.unl.edu) with the purpose of educating the public about agricultural biotechnology risk and safezy issues through science-based content. This article discusses the creation ofthe website, its components, and data gathered from usage statistics and a web-delivered survey. Also included is a discussion of the results ofdata gathered and recommendations for future web-based educational efforts in biotechnology safety and risk assessment.

Polypeptides, linear macromolecules, are formed from amino acid residues by linkage of peptide bonds. Proteins are polypeptides too, with more complex conformations contributing to specific functionalities. Disulfide bonds are very important to maintain the structure and functions of proteins, which will form between two cysteine (Cys) residues under oxidizing circumstance.

Cys containing polypeptides are designed and synthesized by F-moc (9-Fluorenylmethyloxycarbonyl) chemistry. The skin is a major exposure route for many potentially toxic chemicals. It is, therefore, important to be able to predict the permeability of compounds through skin under a variety of conditions. Available skin permeability databases are often limited in scope and not conducive to developing effective models. This sparseness and ambiguity of available data prompted the use of fuzzy set theory to model and predict skin permeability. Using a previously published database containing 140 compounds, a rule-based Takagi–Sugeno fuzzy model is shown to predict skin permeability of compounds using octanol-water partition coefficient, molecular weight, and temperature as inputs. Thin wall microcapsules were formed via Layer-by-Layer Self-Assembly of alternate adsorption of oppositely charged polyelectrolyte on microcores. After the core dissolution, empty polymeric shells with 20–25 nm thick walls were obtained. These microcapsules were loaded with Myoglobin, Hemoglobin and Glucose Oxidase by opening capsule pores at low pH and closing them at higher pH. The native structure of the enzyme was not affected due to different treatments. Biocompatible nanoshells were also prepared for encasing DNA. An operating submarine creates a unique air quality mixture of compounds that result from a combination of human metabolism, construction materials, materials brought onboard and compounds created through the interaction of ship systems. A comprehensive study of submarine atmospheres is ongoing during deployments of U.S. Navy nuclear submarines. As part of the overall effort, a paired air sampling comparison field validation was conducted to compare the air sampling effectiveness of passive diffusive monitors compared to more traditional active air sampling methods when sampling for acrolein, formaldehyde and ozone in the enclosed submarine atmosphere. The toxicity of common compounds of lead, cadmium and zinc was evaluated in waters similar to that found in the world's largest lead producing area in Missouri. Static, acute toxicity tests were performed using fathead minnows (Pimephales promelas) and water fleas (Daphnia magna) respectively. Test organisms were subjected to varying amounts of sulfide, carbonate, chloride and sulfate salts of lead, zinc and cadmium mixed in hard, alkaline waters typical to this region. Median lethal concentrations were calculated using nominal versus measured metal concentrations. A dissolved oxygen probe and an ion specific electrode were used to study the lead-induced metabolic inhibition in a toluene-enriched microbial population. Predicted toxicity values were compared to the actual toxicity responses using a free metal ion toxicity model (TM) which linked metabolic inhibition with lead activity. Experimentally derived values for the model parameters (lead activity and a lead distribution coefficient) were used in the TM. It was postulated that cellular metabolism is disrupted by the conformational changes to the cell's plasma membrane produced by lead ion adsorption. This study refines risk analysis procedures for trichloroethylene (TCE) using a physiologically based pharmacokinetic (PBPK) model in conjunction with the Monte Carlo method. The Monte Carlo method is used to generate random sets of model parameters, based on the mean, variance, and distribution types. The procedure generates a range of exposure values for human excess lifetime cancer risk of lxl0 (exp-6), based on the upper and lower bounds and the mean of a 95% confidence interval. This thesis assesses the risk of the health liabilities from exposure to toxic metals found in the composted material of Air Force municipal solid waste (MSW). The goal is to determine the probability that the composted MSW could be a health hazard if it were used as a soil amendment. The research limited the assessment of the exposure risk to heavy metals found in raw MSW and its resulting compost. This study describes morphological changes in the lung at several time intervals following a single injection of 1-Nitronaphthalene (100 mg/kg intraperitoneal) in male Sprague-Dawley rats using transmission and scanning electron microscopy. Dilation of the smooth endoplasmic reticulum and mitochondrial distension in the nonciliated bronchiolar (Clara) cells was apparent as early as 1 hour after injection, while adjacent ciliated cell showed minimal changes. At 6 hours, when respiratory distress was apparent in the animals, Clara and ciliated cells exhibited chromatin clumping, loss of mitochondrial cristae and cytoplasmic vacuolization. This research project was undertaken to understand the process of desorption/removal of organic compounds from soil matrices under the influence of temperature and gas purging. The project was divided in two parts :

A. Plug deposition experiments: Columns packed with soil matrices were purged with an inert gas under isothermal conditions. Organic compounds were then introduced as a plug on the front of the column by on-line injection. To anticipate my remarks, the answer to the title question is “No, we cannot guarantee the safety of genetically engineered organisms released into the environment.” Indeed, it is a tenet of the scientific method that nothing can be proved, only disproved. Degradation of aniline and nitrobenzene in aqueuous solutions was studied. Five reaction processes were utilized, including ultraviolet light, ozone, O2/UV, O3/O2/UV, and H2O2/UV. A 30 liter semi-batch gas sparged reactor was used for the development of reaction rate and mass transfer parameters in these systems. The results were used to model a continuous stirred tank reactor (CSTR) for the O3/O2/UV case. Reasonable agreement was obtained for steady state exit concentrations between the experimental data and those predicted from the model.

Reactions of dilute mixtures of dichlorobenzene in hydrogen and in helium were studied in a tubular reactor at a pressure of 1 atm, residence times of 0.04 to 2.7 seconds, and temperature range of 825 to 1000° C. Product distributions at various reaction conditions are measured with a flame ionization detector gas chromatograph, and qualitative analysis of batch samples was also performed by GC/Mass Spectrometry. The major products were observed as the thermodynamically stable species, HC1, Cis), benzene and chlorobenzene. Edgeboro Landfill, a site used for the disposal of municipal and industrial refuse, is one of the largest landfills in New Jersey. The landfill has been in operation since 1954, serving the solid waste disposal needs of local townships and a major portion of Middlessex and the surrounding counties. The landfill was not equipped with a liner and a leachate collection system at the time of start up. The leachate collection line around the landfill, on-site leachate treatment by three stabilization ponds and subsurface cut-off wall were provided to minimize the discharge of leachate into adjacent waters in 1979. The purpose of this initial laboratory scale research project was to study and demonstrate the feasibility in removal of selected toxic organic compounds from soil by thermal desorption and gas purge. Applying heat and a flow of inert gas to the soil increases the volatilization and effects removal of organic compounds from soil. The work was done in two parts: First, organic solvents were injected into columns containing real and simulated soil matrices.
