Cancer cell lines are essential resources for connecting genomic and molecular features with drug response and identifying biomarkers. Therefore, large-scale cancer cell line resources have been growing, but this introduces challenges such as data standardization, consistent annotation, matching identifiers, cross-database integration, and a reproducible computational framework. CellMiner Cross Database (CellMinerCDB) integrates pharmacogenomic datasets from multiple resources and provides a standardized analysis of cell lines across datasets. This thesis examines cancer cell line cross-databases and their importance in pharmacogenomics, with a particular focus on CellMinerCDB. Francisella tularensis is a highly infectious intracellular bacterium and the causative agent of tularemia, with pulmonary infection resulting in severe disease and high mortality if untreated. While protective immunity to F. tularensis has been largely attributed to CD4⁺ T cell–mediated IFN-γ responses, the contribution of γδ T cells to mucosal immunity and vaccine-induced protection remains poorly defined. The objective of this study was to characterize the role of γδ T cells during primary pulmonary infection with F. tularensis live vaccine strain (LVS) and following intranasal immunization with inactivated F. tularensis (iFt) prior to LVS challenge.

Acute myeloid leukemia (AML) is a fatal blood cancer with cytotoxic chemotherapy offering at best 25% 5-year survival. While targeted BCL2 and FLT3 inhibitors venetoclax and gilteritinib are used upfront in the treatment of a subset of adult patients with AML and help to extend the survival of some patients, a curative treatment combination with minimal side effects has yet to be discovered. We find that use of the dual histone acetyltransferase p300/CBP bromodomain inhibitor CCS1477 (inobrodib), together with venetoclax and gilteritinib, virtually eliminates leukemia stem cells in an aggressive preclinical model of DNMT3A/FLT3-mutant AML by impairing pro-oncogenic survival and …

Autoimmune disorders disproportionately affect females, a bias largely attributed to estrogen's modulation of B lymphocyte development and tolerance. Bisphenol A (BPA), a common plastic monomer, shows estrogenic activity and acts as an endocrine disruptor and epigenetic modifier. This thesis examines how BPA may increase autoimmune risk by mimicking estrogen such that it can excite estrogen receptors which rescue autoreactive B lymphocytes from apoptosis. Store-operated calcium entry (SOCE) through calcium release-activated
calcium (CRAC) channels is the dominant pathway for sustained Ca2+ influx in
non-excitable cells and is essential for T cell activation, driving downstream
signaling through the calcineurin-NFAT transcriptional axis. Dysregulation of
SOCE produces multi-systemic disease phenotypes, establishing that precise,
spatiotemporally controlled modulation of this pathway is a fundamental
therapeutic requirement. ML-9 is one of the only characterized small molecules
capable of targeting SOCE upstream at the level of STIM1 localization, but lacks
the selectivity, pharmacokinetic properties, and spatiotemporal control necessary
for therapeutic development.

• Microtubules are composed of of a- and b- tubulin heterodimers, vital for cell structure, shape, and intracellular transport • Microtubules are also useful as they separate chromosomes during cell division • Microplastics (< 5 mm) are considered a health threat and have been found throughout the whole body (1) o In several studies, microplastics have been correlated to higher rates cardiorespiratory issues, endocrine disruption, and cancer (1) • Tubulin-binding medications are widely used as chemotherapeutic agents as they are known for disrupting cell division and targeting tubulin (2,3) o In this study, these medications were used to determine whether microtubules play a role in microplastic uptake § Colchicine typically binds to tubulins and arrests polymerization and growth § Nocodazole disrupts polymerization and prevents spindle formation § Vinblastine inhibits microtubule assembly and disruptions miotic spindle formation.

However, chronic or excessive inflammation can lead to tissue damage and contribute to a wide range of diseases, including neurodegenerative disorders, metabolic syndromes, and immune dysregulation. 3,4 Interleukin-1 beta (IL-1β) is a key pro-inflammatory cytokine that plays a central role in initiating and amplifying the immune response. 1,2 It is primarily produced by activated monocytes and macrophages in response to stimuli such as pathogens or stress signals. Triple Negative Breast Cancer (TNBC) is characterized by the absence of estrogen, progesterone, and HER2 receptors, limiting targeted treatment options. Digitoxin, a drug used to treat heart failure and atrial fibrillation through Na⁺/K⁺-ATPase inhibition, has demonstrated anticancer effects in TNBC cells, including apoptosis via the NF-κB pathway. This study investigates whether the Na⁺/K⁺-ATPase and sodium–calcium exchanger contribute to the mechanism of action of digitoxin in TNBC cells. We hypothesize that these transporters mediate digitoxin-induced cytotoxic effects. The cornea, the foremost anterior portion of the eye, plays a critical role in vision by refracting light and serving as a protective barrier. Its three distinct layers—the outermost epithelial layer, the thick stromal layer, and the innermost endothelial layer work together to maintain corneal transparency and function. The epithelial layer is particularly susceptible to damage, which can compromise the eye’s defense against foreign particles such as bacteria and viruses, potentially leading to infection and blindness. Perfluorooctane sulfonate (PFOS) is a persistent environmental pollutant associated with potential hepatoxic effects and other health risks. Despite its widespread distribution, the mechanisms underlying its toxicities remain to be fully understood. To investigate PFOS toxicology, my study utilized HepG2 and THLE-2 human hepatic cell models to replicate conditions reflecting PFOS accumulation in the liver. Cell viability, cell stress, and cell death assays were conducted to assess the toxicological influence of the chemical on both cell lines. Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants widely detected in drinking water and food sources, resulting in chronic human exposure. Among these compounds, hexafluoropropylene oxide dimer acid (HFPO-DA), commonly known as GenX, has been introduced as a short-chain replacement for legacy PFAS such as perfluorooctanesulfonic acid (PFOS) and perfluorooctanoic acid (PFOA). Cigarette smoke is a plight on human health, being a major contributor to lung diseases. The aryl hydrocarbon receptor (AHR) directs immune regulation and metabolic adaptation in response to environmental toxins. This study investigates transcriptomic changes between wild-type and AHR knockout mouse lung tissue under smoke and air conditions. Single-cell RNA sequencing data were generated by the Butcher Lab and analyzed in Seurat for differential gene expression (DGE) analysis. Schwann cells are the glial cell of the peripheral nervous system. They are dependent on receptor tyrosine kinase-mediated RAS signaling, cell matrix adhesion, and cell-cell contact to proliferate during nerve development and repair. These pathways become deregulated with loss of function of the merlin tumor suppressor encoded by NF2, leading to benign tumors called schwannomas. Patients with NF2-related schwannomatosis (NF2-SWN) develop bilateral vestibular schwannomas that cause hearing loss, tinnitus, and death from brainstem compression if untreated. Severe NF2-SWN causes multiple spinal and peripheral schwannomas and neuropathic pain. Schwannomas require careful monitoring, trial and error use of repurposed anti-cancer …

Objective: Mouthwash is widely used to maintain oral health by reducing the prevalence of pathogenic microbiomes. The RALGEF family of proteins are direct downstream effectors of the RAS oncoprotein. RALGEFs act as guanine nucleotide exchange factors (GEFs) for downstream RAL proteins, thus tying RAS to the regulation of RAL. Genetic studies have demonstrated RAL proteins to be key drivers of RAS-driven transformation and metastasis. Furthermore, this pathway has been implicated in chemoresistance, exocyst-mediated transport, and the modulation of cancer stem cells. Photo-oxidation of spilled crude oil generates polar compounds that can heighten toxicity to marine fish embryos. We investigated the effects of photo-oxidized crude oil on early life stages of Gulf killifish (Fundulus grandis) by exposing embryos under flow-through conditions to water-accommodated fractions of four oil treatments: fresh oil unexposed to either sunlight or weathering referred to as dark control (DC), a UV-light oxidized whole oil (LT), the polar fraction extracted from LT (Pol) and the remnants after extracting the Pol from LT (NP).
