การแบ่งกลุ่มย่อยตามลักษณะทางคลินิกรูปแบบใหม่โดยอาศัยปัญญาประดิษฐ์ ร่วมกับการวิเคราะห์โอมิกส์แบบบูรณาการหลายระดับ เพื่อระบุกลุ่มตัวแทนสารบ่งชี้ทางชีวภาพสำหรับภาวะออทิซึมสเปกตรัม, กัลยภัสสร์ หัสเสม

โรคออทิซึมสเปกตรัมเป็นความผิดปกติเกี่ยวกับพัฒนาการในช่วงเริ่มแรกของระบบประสาทและสมองซึ่งมีอาการทางคลินิกที่หลากหลายมาก ปัจจุบันยังไม่มีสารบ่งชี้เพื่อช่วยในการวินิจฉัยและแบ่งผู้ป่วยเป็นกลุ่มย่อย แต่การวินิจฉัยที่รวดเร็วเป็นสิ่งสำคัญที่จะช่วยให้ผู้ป่วยออทิซึมสเปกตรัมสามารถได้รับการเรียนรู้และพัฒนาพฤติกรรมที่จำเป็นในการดำรงชีวิตได้ การวินิจฉัยโรคออทิซึมสเปกตรัมตามมาตรฐานสากลใช้แบบสอบถามที่มีความซับซ้อนและใช้เวลามาก ซึ่งในปัจจุบันมีการนำเอาเทคโนโลยีปัญญาประดิษฐ์มาประยุกต์ใช้ในการวิเคราะห์ข้อมูลขนาดใหญ่ที่มีความซับซ้อน อย่างไรก็ตามการนำเอาปัญญาประดิษฐ์มาใช้วิเคราะห์อาการทางคลินิกของผู้ป่วยออทิซึมสเปกตรัมยังไม่แพร่หลายนัก ในการศึกษานี้ผู้วิจัยต้องการนำเอาปัญญาประดิษฐ์มาใช้แบ่งผู้ป่วยออทิซึมสเปกตรัมจำนวน 85 ราย ออกเป็นกลุ่มย่อยตามอาการทางคลินิกจากข้อมูลแบบทดสอบในการวินิจฉัยผู้ป่วยออทิซึมสเปกตรัม ADI-R จำนวน 123 ข้อ จากนั้นได้ทำการสร้างโมเดลในการจัดกลุ่มและเปรียบเทียบประสิทธิภาพรวมถึงจำนวนข้อคำถามที่ใช้ในแต่ละโมเดล นอกจากนี้ผู้วิจัยได้ศึกษาหาสารบ่งชี้ทางชีวภาพที่เป็นตัวแทนของโรคออทิซึมสเปกตรัมกลุ่มย่อยจากลักษณะอาการทางคลินิกโดยอาศัยการบูรณาการโอมิกส์หลายระดับ จึงได้ดำเนินการวิเคราะห์รูปแบบทรานสคริปโตม และวิเคราะห์รูปแบบโปรตีโอมของผู้ป่วยออทิซึมสเปกตรัมโดยใช้เซลล์เพาะเลี้ยงชนิดลิมโฟบลาสต์เป็นต้นแบบ โดยใช้เทคนิคการแยกโปรตีนด้วยกระแสไฟฟ้า 2 มิติ (2D-GE) จากนั้นจะทำการระบุโปรตีนด้วยเทคนิค LC-MS/MS เพื่อหาโปรตีนที่ถูกรบกวนการทำงานอย่างมีนัยสำคัญ และบูรณาการข้อมูลเพื่อศึกษาความสอดคล้องและสัมพันธ์กันระหว่างการแสดงออกของโปรตีนและยีน และทำการยืนยันระดับการแสดงออกที่ผิดปกติไปของยีนด้วยเทคนิค Real-time qPCR และโปรตีนด้วยเทคนิค Western blot ในการศึกษาครั้งนี้จากผลการวิเคราะห์ข้อมูลลักษณะอาการทางคลินิกของผู้ป่วยออทิซึมสเปกตรัมจากแบบทดสอบ ADI-R 123 ข้อ พบว่าวิธี K-means clustering ที่แบ่งผู้ป่วยออกเป็น 4 กลุ่มย่อยนั้นให้ประสิทธิภาพในการแบ่งดีที่สุด และเมื่อทำการสร้างโมเดลในการจัดกลุ่มผู้ป่วยตามผลการแบ่งกลุ่มดังกล่าว พบว่าอัลกอริทึม Random forest ที่มีจำนวนต้นไม้ 10 ต้น โดยกระบวนการคัดเลือกข้อมูลแบบ Forward selection ให้ผลการทำนายถูกต้องมากที่สุด (100%) โดยใช้ข้อคำถาม ADI-R เพียง 5 ข้อ ในการแบ่งผู้ป่วยออกเป็นกลุ่มย่อย และจากการวิเคราะห์รูปแบบโปรตีโอมจากเซลล์เพาะเลี้ยงชนิดลิมโฟบลาสต์ของผู้ป่วยออทิซึมสเปกตรัมและออทิซึมสเปกตรัมกลุ่มย่อยด้วยเทคนิค 2D-GE ผู้วิจัยได้ทำการคัดเลือกจุดโปรตีนที่มีการแสดงออกผิดปกติอย่างมีนัยสำคัญและมีค่าสัดส่วนความเข้มแตกต่างกันอย่างน้อยสองเท่ามา 19 จุด เพื่อทำการระบุชนิดของโปรตีนด้วยเทคนิค LC-MS/MS โดยพบว่าจากโปรตีนทั้งหมด HNRNPA2B1 มีระดับการแสดงออกเพิ่มขึ้นในผู้ป่วยออทิซึมสเปกตรัมกลุ่มย่อยที่ไม่มีความผิดปกติด้านภาษารุนแรง โดยที่โปรตีน HNRNPA2B1 MDH2 DLD และ MSN มีระดับการแสดงออกลดลงในผู้ป่วยออทิซึมสเปกตรัมกลุ่มย่อยที่มีความผิดปกติด้านภาษารุนแรง ดังนั้นจากการศึกษาวิจัยในครั้งนี้ได้แสดงให้เห็นว่าการใช้เทคโนโลยีปัญญาประดิษฐ์ในการแบ่งผู้ป่วยออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยตามลักษณะอาการทางคลินิกนั้น สามารถลดความหลากหลายตามลักษณะอาการที่แสดงออกของโรคออทิซึมสเปกตรัมในการวิเคราะห์รูปแบบทรานสคริปโตมและโปรตีโอม รวมถึงการค้นหาโปรตีนที่จำเพาะต่อผู้ป่วยออทิซึมสเปกตรัมกลุ่มย่อย ได้อย่างมีประสิทธิภาพมากขึ้น ซึ่งโปรตีนเหล่านี้อาจใช้เป็นโปรตีนบ่งชี้ในการศึกษาเชิงลึกในประชากรผู้ป่วย เพื่อพัฒนาเป็นสารบ่งชีทางชีวภาพ (Biomarker) หรือศึกษากลไกพยาธิสภาพในระดับชีวโมเลกุลของโรคออทิซึมสเปกตรัมได้ต่อไปในอนาคต

I tested the hypothesis that recombinant leptin protein can be introduced to zebrafish in vivo through non-invasive soaking in a solution containing the protein. One way to study various molecules’ effects in vivo is through intraperitoneal or intracerebroventricular injections during the embryonic or larval stage, which is invasive, difficult to administer, and can have a high mortality rate. 48 hours post fertilization (hpf) zebrafish were soaked in a His-tagged recombinant leptin protein solution at 10 nM and 100 nM concentrations (produced by Genscript). After soaking, zebrafish larvae were washed extensively to remove all recombinant protein on their exterior before homogenization. …

To study melanoma mechanism, screening the amount of melanin production and invasion study are major keys for drug development. In this work, we developed paper-based devices for culturing melanoma cells. Through the development process, we obtained the optimal parameters, which were applied for 3 different projects; 1) Anti-melanogenic effect screening, 2) Impedance-based E-screen assay, and 3) A membrane insert for real-time invasion assay. World’s population of older people is rising, dietary supplements promoting healthy lifespan are needed. Moreover, Alzheimer’s disease (AD), an age-related degenerative disease, becomes a public health problem in aging society. Recent reports using cell culture models of AD suggest that amyloid precursor protein (APP), a protein causally related to AD, plays an important role as an inhibitor of neurite outgrowth. Medicinal herbs with neurite outgrowth stimulatory effect may help to prevent and cure AD. Growth factors and hormones often mediate their biological effects through series of protein-protein interaction between cytoplasmic signaling molecules or transcription factors. Interfering of these protein-protein interactions has been shown to reduce or inhibit biological and growth responses to various growth factors and hormones. We previously demonstrated that progesterone receptor (PR) contains a polyproline domain (PPD) which directly interact to Src homology 3 (SH3) domain-containing molecules and expression of PR-PPD inhibits EGFR-mediated NSCLC cell proliferation. หนึ่งในสาเหตุสำคัญของการเกิดโรคความเสื่อมของระบบประสาทนั้นเกิดได้จากภาวะเครียดออกซิเดชันภายในเซลล์ประสาท โดยพบว่าสารสื่อประสาทชนิดกลูตาเมตเมื่อมีระดับสูงผิดปกติสามารถเหนี่ยวนำให้เกิดอนุมูลอิสระและภาวะเครียดออกซิเดชันนำไปสู่การสูญเสียและการตายของเซลล์ประสาทได้ โดยเฉพาะการตายของเซลล์จากการเกิดกระบวนการออโตฟาจีมากเกินไป ซึ่งเป็นกระบวนการที่มีบทบาทสำคัญในการควบคุมการตายของเซลล์ผ่านวิถีอื่นๆ รางจืด (Thunbergia laurifolia) เป็นพืชสมุนไพรไทยมีสรรพคุณในการถอนพิษ ต้านการอักเสบ และ ต้านอนุมูลอิสระได้ดี จุดประสงค์ในการวิจัยครั้งนี้ คือ ศึกษาคุณสมบัติการต้านอนุมูลอิสระของสารสกัดหยาบจากใบรางจืด และการปกป้องการตายของเซลล์ประสาทผ่านวิถีออโตฟาจี จากการเหนี่ยวนำภาวะเครียดออกซิเดชันด้วยกลูตาเมตโดยทดสอบในเซลล์ประสาทส่วนฮิปโปแคมปัสของหนูชนิด HT-22 ใบรางจืดถูกนำมาสกัดด้วยเอทานอล จากผลการทดสอบพบว่าสารสกัดหยาบจากใบรางจืดมีความสามารถในการต้านอนุมูลอิสระ และมีปริมาณสารประกอบ ฟลาโวนอยด์และฟีนอลิกสูง โดยจากการวิเคราะห์ด้วยเครื่อง LC-MS และเปรียบเทียบผลที่ได้กับฐานข้อมูล METLIN-(CA, USA) พบสารออกฤทธิ์สำคัญที่ยังไม่มีรายงานในสารสกัดเอทานอลของใบรางจืด ได้แก่ Apigenin 7-O-glucoside และ 7-Hydroxycoumarin ยิ่งไปกว่านั้นสารสกัดหยาบจากใบรางจืดยังสามารถลดการตายของเซลล์ประสาทจากการเหนี่ยวนำความเป็นพิษด้วยกลูตาเมต โดยสามารถยับยั้งการเกิดอนุมูลอิสระภายในเซลล์ และเพิ่มการแสดงออกของยีนต้านอนุมูลอิสระที่ทำหน้าที่สำคัญในกระบวนการต้านอนุมูลอิสระในร่างกาย ได้แก่ superoxide dismutase 1 และ 2 (SOD1 and SOD2) catalase (CAT) และ glutathione peroxidase (GPx) ซึ่งสัมพันธ์กับการลดการเกิดกระบวนการออโตฟาจีภายในเซลล์และเพิ่มปริมาณ ไมโตคอนเดรียโปรตีน โดยพบว่าสารสกัดหยาบจากใบรางจืดสามารถลดอัตราส่วนการแสดงออกของโปรตีน LC3B-II/LC3B-I และเพิ่มการแสดงออกของไมโตคอนเดรียโปรตีน TOM20 เมื่อทำการตรวจหาตำแหน่งที่จำเพาะต่อการแสดงออกของโปรตีนในกระบวนการออโตฟาจี (LC3B) และไมโตคอนเดรียด้วยกล้องจุลทรรศน์ชนิดคอนโฟคอล พบว่า สารสกัดหยาบจากใบรางจืดสามารถลดการเกิด LC3-puncta ซึ่งเป็นลักษณะเด่นของการเกิดกระบวนการออโตฟาจี นอกจากนี้สารสกัดหยาบจากใบรางจืดยังลดการ colocalization ของ LC3B และไมโตคอนเดรีย จากผลการศึกษานี้เราพบสารออกฤทธิ์สำคัญใหม่ที่ยังไม่มีรายงานมาก่อนของใบรางจืด โดยสารออกฤทธิ์นี้มีความสามารถในการปกป้องเซลล์ประสาทชนิด HT-22 จากภาวะเครียดออกซิเดชันและเป็นปัจจัยสำคัญทำให้สารสกัดจากใบรางจืดมีความสามารถลดการตายของเซลล์ประสาทผ่านวิถีออโตฟาจีได้ ซึ่งอาจสามารถนำไปพัฒนาเป็นสมุนไพรทางเลือกหรือยาเพื่อใช้ในการป้องกันหรือรักษาการเกิดโรคความเสื่อมของระบบประสาทได้ในอนาคต

Exosomes are small, cargo-containing vesicles secreted by cells to facilitate intercellular communication. Of exosome cargos, microRNAs are especially interesting because of their involvement in gene regulation. Recently, our lab has shown that exosomes and their microRNA cargo are absorbed through the diet and elicit effects exogenously. Human and animal milk contain exosomes, which may have implications in infant and adult nutrition. There is evidence that bovine milk exosomes enhance growth of murine C2C12 myotube cell cultures, but whether this translates to muscle in vivo is unknown. Ulcerative colitis (UC) is a disabling inflammatory bowel disease. UC is characterized by chronic, relapsing inflammation of the colon and rectum. Current treatments such as monoclonal antibodies against TNF-α, IL-6, IL-12/p40, adhesion molecules, Janus kinases (JAK) inhibitors have side effects or lose their effects over time. Alternative approaches with fewer side effects for patients are needed. Montmorency tart cherries (Prunus cerasus) are a good source of anti-inflammatory flavonoids. Cholangiocarcinoma (CCA), a malignancy transformed from cholangiocytes in the bile ducts is more common in Asia and has the highest incidence rate in Thailand. CCA is an aggressive malignancy which has high mortality, high recurrence rate and poor prognosis due to late diagnosis and lack of effective treatment, therefore identification of novel therapeutic targets could lead to the development of more efficient therapy. CCA is associated with chronic inflammation that could upregulate Toll-like receptor 3 (TLR3) in CCA cells. การปลอมปนของสารสเตียรอยด์ชนิดเดกซาเมทาโซนและเพรดนิโซโลนในผลิตภัณฑ์สมุนไพรเป็นปัญหาสำคัญที่ก่อผลเสียร้ายแรงต่อสุขภาพของผู้ใช้ ในหลาย ๆ ประเทศทั่วโลก โดยเฉพาะอย่างยิ่งในประเทศกำลังพัฒนา งานวิจัยนี้ ประสบความสำเร็จในการพัฒนาอุปกรณ์สำหรับแยกและตรวจวัดปริมาณการปลอมปนของสารสเตียรอยด์ดังกล่าว โดยใช้อุปกรณ์ตรวจวิเคราะห์ฐานกระดาษและใช้หลักการทางเคมีไฟฟ้า (electrochemical paper-based analytical device หรือ ePAD) ซึ่งประกอบด้วยกระดาษ Whatman SG81 ตลับพลาสติกจากการพิมพ์สามมิติ และขั้วไฟฟ้าชนิด screen printed electrode โดยกระดาษ Whatman SG81 ใช้สำหรับแยกสารสเตียรอยด์ชนิดเดกซาเมทาโซนและเพรดนิโซโลน โดยใช้วัฏภาคเคลื่อนที่เป็นสารละลาย 60% ethyl acetate ใน cyclohexane ซึ่งผลการศึกษาพบว่าสามารถแยกสารสเตียรอยด์ดังกล่าวออกจากสิ่งเจือปนต่าง ๆ ได้ภายใน 7 นาที สารสเตียรอยด์ที่แยกออกจากกันบนกระดาษ นำมาวัดปริมาณด้วยเทคนิคเคมีไฟฟ้าชนิด differential pulse voltammetry โดยใช้ตลับพลาสติกจากการพิมพ์สามมิติที่ถูกออกแบบทำให้สารละลายอิเล็กโทรไลต์ถูกขังอยู่บนกระดาษ ทำให้สามารถตรวจวัดสัญญาณทางเคมีไฟฟ้าได้คงที่ ซึ่งอุปกรณ์ที่พัฒนาขึ้นสามารถตรวจวัดปริมาณปลอมปนของสารสเตียรอยด์ทั้งชนิดเดกซาเมทาโซนและ เพรดนิโซโลนได้ในช่วงความเข้มข้นระหว่าง 10 ถึง 500 µg/mL โดยมีค่าสัมประสิทธิ์สหสัมพันธ์เท่ากับ 0.988 และ 0.994 ตามลำดับ มีขีดจำกัดในการตรวจวิเคราะห์เชิงคุณภาพเท่ากับ 3.59 µg/mL และ 6.00 µg/mL ตามลำดับ และขีดจำกัดในการตรวจวิเคราะห์เชิงปริมาณมีค่าเท่ากับ 11.98 µg/mL และ 20.02 µg/mL ตามลำดับ การตรวจวัดปริมาณปลอมปนของสารสเตียรอยด์ทั้งสองชนิดในสิ่งตัวอย่างผลิตภัณฑ์สมุนไพรจริงด้วยวิธีที่พัฒนาขึ้นมีค่าสอดคล้องกับผลที่ตรวจด้วยวิธี HPLC และเป็นวิธีที่ใช้งานง่าย มีต้นทุนต่ำ รวดเร็ว และมีศักยภาพนำไปใช้ตรวจหาสิ่งตัวอย่างผลิตภัณฑ์สมุนไพรที่สงสัยว่ามีการปลอมปนของสารสเตียรอยด์ ในระดับภาคสนาม

A paper-based analytical devices (PADs) provides a great benefit in medical diagnostics, especially in the developing world. The first part of this dissertation describes the effect of sample viscosity on PADs for the measurement of pH and nitrite concentration in human saliva. Preliminary study found that viscosity had a significant effect on the calibration curve of nitrite and a consistent curve could be generated after added washing buffer. Nitrite measurement in artificial saliva with viscosity 1.87-5.10 mPa-s was tested on the proposed device and also showed a good correlation with the standard spectrophotometry method (p=0.8484, paired sample t-test, n=20). Human immunodeficiency virus type-1 (HIV-1) can cause acquired immunodeficiency syndrome (AIDS), a world serious public health problem. HIV-1 protease (HIV-1 PR) and reverse transcriptase (HIV-1 RT) are necessary target enzymes which have been used for antiretroviral drugs development. Herein, Lignosus rhinocerus (LR), Auricularia polytricha (AP) and Astraeus spp. (AH) crude extracts were screened for inhibitory activities on both enzymes. Crude hexane extract of AP (APH) exhibited significant inhibition on both HIV-1 PR and RT activities not only in an in vitro non-cell based but also cell based assays. วิทยานิพนธ์นี้เป็นการนำเสนองานวิจัย การพัฒนาวิธีการใหม่เพื่อควบคุมและเฝ้าระวังโรคติดเชื้อ โดยประกอบด้วยเนื้อหาสำคัญสองส่วน ได้แก่ (i) การพัฒนาแบบจำลองเซลล์ลำไส้ของมนุษย์ที่เพาะเลี้ยงบนกระดาษที่มีการดัดแปรพื้นผิว เพื่อใช้ประโยชน์ในการศึกษากลไกการบุกรุกของเชื้อจุลชีพก่อโรคในระบบทางเดินอาหาร และ (ii) การพัฒนาต้นแบบไบโอเซนเซอร์แบบไร้สายเพื่อการตรวจติดตามการเจริญของเชื้อจุลชีพก่อโรค โดยในส่วนแรก เซลล์ลำไส้ของมนุษย์ HT-29 จำนวน 200,000 เซลล์ ถูกเพาะเลี้ยงในบริเวณชอบน้ำของกระดาษกรอง Whatman No.1 โดยกระดาษถูกดัดแปรพื้นผิวด้วยสารแมทริกซ์นอกเซลล์ (Extracellular matrix: ECM) ชนิดต่าง ๆ ได้แก่ Matrigel, Collagen-1 และ Laminin พื้นที่เพาะเลี้ยงเซลล์มีขนาด 12.56 ตารางมิลลิเมตร ภายใต้สภาวะที่เหมาะสม เซลล์ HT-29 ที่ถูกเพาะเลี้ยงบนกระดาษ มีชีวิตรอดได้ถึง 28 วัน และแสดงลักษณะของเซลล์ที่เจริญเต็มที่ โดยมีการแสดงออกของวิลไล (villi) ที่ผิวเซลล์ด้านบน (apical) และมีการแสดงออกของโปรตีน ZO-1 ซึ่งแสดงถึงเซลล์มีการสร้างไทต์จังก์ชันเกิดขึ้น จากการใช้สารเชื่อมพันธะสำหรับเชื่อมพันธะ ECM แต่ละชนิด พบว่าเมื่อใช้สารเจนิพินในการเชื่อมพันธะ เซลล์มีชีวิตรอดทั้งหมด (100 -112.8%) ในขณะที่มีเพียง 1.1 - 67.3 % ของเซลล์รอดชีวิต หากใช้สารกลูตารัลดีไฮด์ในการเชื่อมพันธะ แต่การใช้สารเชื่อมพันธะทั้งสองชนิด ทำให้เซลล์ไม่สามารถแบ่งตัวมีรูปร่างแบบสามมิติได้ เมื่อสังเกตด้วย SEM เซลล์มีลักษณะแบน การดัดแปรพื้นผิวกระดาษด้วยสาร ECM ชนิด Matrigel ส่งผลให้เซลล์มีชีวิตรอดมากที่สุดและสาร ECM ชนิด Laminin ทำให้เซลล์เกิดการแปรสภาพ (cell differentiation) ได้ดีที่สุด งานวิจัยส่วนที่ 2 เป็นการพัฒนาไบโอเซนเซอร์ไร้สายสำหรับติดตามการเจริญของเชื้อจุลชีพก่อโรค ได้แก่ เชื้อ Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Streptococcus gordonii และ Candida albicans พบว่าเชื้อดังกล่าวสามารถสร้างไบโอฟิล์มบนผิวของขั้วไฟฟ้าคาร์บอนอสัณฐาน (glassy carbon electrode) และสามารถให้ค่าศักย์ไฟฟ้าที่เป็นลบได้ …

Pancreatic ductal adenocarcinoma (PDAC) is among the most lethal of all cancers with a 5-year survival rate of only 8.2%. This is because PDAC is diagnosed in its advanced stages and is characterized by radio and chemotherapy resistance. Aggressiveness of PDAC tumors is attributed to its high metabolic phenotype, which is characterized by increased glycolysis rate and lactate secretion, while oxidative metabolism is reduced. These metabolic features are required to fulfill the biosynthetic demands of proliferating PDAC cells. Women with lung NETs have significantly better survival rates as compared to men suggesting the involvement of sex steroids and their receptors in the progression of lung NETs. Recent data suggested that progesterone receptor (PR) may play a role in the survival of lung NET patients. PR exists as two major isoforms, PRA and PRB. ในปัจจุบัน ออทิซึมสเปกตรัม (Autism spectrum disorder, ASD) ยังคงเป็นโรคที่ยังคงไม่ทราบกลไกระดับโมเลกุล แน่ชัด ยีนและโปรตีนที่เคยมีการรายงานการแสดงออกผิดปกติไปในผู้ป่วยออทิซึมสเปกตรัมมีความสามารถในการทำซ้ำ (Reproducibility) ต่ำหรืออาจไม่มีเลย ซึ่งน่าจะมาจากความหลากหลายของอาการแสดงของผู้ป่วย จากการศึกษาล่าสุด แสดงให้เห็นว่าการจัดกลุ่มออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยตามความรุนแรงของลักษณะทางคลินิกสามารถช่วยในการระบุยีนและกลไกที่ผิดปกติไปในกลุ่มย่อยของผู้ป่วย อย่างไรก็ตาม การแบ่งกลุ่มผู้ป่วยเป็นกลุ่มย่อยตามความรุนแรงของลักษณะทางคลินิกใน ออทิซึมสเปกตรัมยังไม่มีการใช้กันอย่างแพร่หลายในการค้นหาโปรตีน Biomarker ที่เกี่ยวข้องกับออทิซึมสเปกตรัม ในการศึกษาครั้งนี้ ผู้วิจัยทำการวิเคราะห์โดยการแบ่งกลุ่มผู้ป่วยจำนวน 85 รายออกเป็นกลุ่มย่อยทางคลินิกตามคะแนน ADI-R และทำการวิเคราะห์รูปแบบการแสดงออกของยีนในออทิซึมสเปกตรัมในแต่ละกลุ่มย่อยเปรียบเทียบกับกลุ่มควบคุมคนปกติที่อายุใกล้เคียงกันและเพศเดียวกันกับผู้ป่วย (Sex/age-matched control) ซึ่งสามารถระบุยีนที่มีการแสดงออกผิดปกติ (Differentially expressed genes, DEGs) ในออทิซึมสเปกตรัม นอกจากนี้ ผู้วิจัยได้ทำการวิเคราะห์รูปแบบการแสดงออกของโปรตีโอมจากเซลล์ไลน์เม็ดเลือดขาว (Lymphoblastoid cell lines,LCLs) ด้วยเทคนิค Two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) ร่วมกับ Nano-liquid chromatography-tandem mass spectrometry (Nano-LC-MS/MS) รายชื่อของโปรตีนที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญที่ได้จากการวิเคราะห์ถูกนำมาเปรียบเทียบกับ DEGs และผลการวิเคราะห์ Meta-analysis ของโปรตีนและโปรตีโอมในการศึกษาก่อนหน้า หน้าที่ทางพยาธิสภาพที่มีความเกี่ยวข้องกับโปรตีนถูกวิเคราะห์โดยโปรแกรม Ingenuity Pathway Analysis (IPA) และได้ทำการคัดเลือกโปรตีนที่น่าสนใจมาทำการตรวจสอบโดยใช้ Western blot analysis จากการศึกษาครั้งนี้พบว่าผลการวิเคราะห์การแบ่งกลุ่มผู้ป่วยออกเป็นกลุ่มย่อยทางคลินิกตามคะแนน ADI-R สามารถแบ่งผู้ป่วยออกเป็น 4 กลุ่มย่อย ซึ่งพบว่าหนึ่งในกลุ่มนั้น เป็นกลุ่มออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษารุนแรง จากผลวิเคราะห์รูปแบบ ทรานสคริปโตมของเซลล์ไลน์เม็ดเลือดขาว พบว่ามีจุดโปรตีนจำนวน 82 จุดที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญในกลุ่มออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษารุนแรง และได้ทำการคัดเลือก 18 จุดโปรตีนจากทั้งหมดมาวิเคราะห์ชนิดของโปรตีนโดยเทคนิค Nano-LC-MS/MS ซึ่งพบว่ามีความเกี่ยวข้องกับการทำงานของระบบประสาทและกระบวนการอักเสบ จากการศึกษานี้พบว่าจากโปรตีนทั้งหมด การแสดงออกของ Isocitrate dehydrogenase (IDH2) และ Diazepam binding inhibitor (DBI) ลดลงในออทิซึมสเปกตรัม ซึ่งโปรตีนเหล่านี้ยังพบการแสดงออกผิดปกติในการวิเคราะห์ Meta-analysis ของทรานสคริปโตมและโปรตีโอมร่วมด้วย การแบ่งกลุ่มออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยทางคลินิกตามด้วยการวิเคราะห์ทรานสคริปโตมและโปรตีโอมแบบบูรณาการร่วมกัน ทำให้การศึกษานี้สามารถระบุโปรตีนที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญในกลุ่มย่อยของออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษาอย่างรุนแรง ซึ่งโปรตีนเหล่านี้อาจทำหน้าที่เป็นตัวแทนในการช่วยระบุกลไกทางโมเลกุลของออทิซึมสเปกตรัมในอนาคต ตลอดจน อาจพัฒนาไปเป็น …

กระบวนการสร้างนิวไรท์ เป็นกระบวนการที่สำคัญในการเจริญพัฒนา การซ่อมแซมส่วนที่เสียหาย รวมถึงการสื่อสารระหว่างเซลล์ของเซลล์ประสาท ซึ่งหากกระบวนการการสร้างนิวไรท์เกิดความผิดปกติสามารถก่อให้เกิดโรคความผิดปกติของระบบประสาท เช่น โรคอัลไซเมอร์ โรคพาร์กินสัน และโรคฮันติงตัน เป็นต้น ในการศึกษาวิจัยนี้ศึกษาฤทธิ์การต้านสารอนุมูลอิสระ และการกระตุ้นการสร้างนิวไรท์จากสารสกัดเมล็ดของหมามุ่ยไทย และหมามุ่ยอินเดียในเซลล์ประสาทเพาะเลี้ยงชนิด Neuro2a การศึกษาคุณสมบัติในการต้านสารอนุมูลอิสระโดยวิธี DPPH และ ABTS assays นอกจากนั้นทำการศึกษาอัตราการรอดชีวิตของเซลล์ประสาทโดยวิธี MTT พร้อมกับศึกษาการแสดงออกของยีนที่เกี่ยวข้องกับการกระตุ้นการงอกของนิวไรท์ ประกอบด้วย Ten-4 และ GAP-43 โดยวิธี Real-time PCR ซึ่งจากการทดสอบพบว่าสารสกัดจากเมล็ดหมามุ่ยไทย และเมล็ดหมามุ่ยอินเดียด้วยตัวทำละลายเอทานอลมีความสามารถในการต้านสารอนุมูลอิสระ และพบว่าสารสกัดจากเมล็ดหมามุ่ยไทย และเมล็ดหมามุ่ยอินเดียไม่เป็นพิษต่อเซลล์ประสาท จากนั้นทำการศึกษาฤทธิ์ และกลไกของสารสกัดจากสมุนไพรต่อการกระตุ้นการงอกของนิวไรท์ โดยพบว่าสารสกัดจากเมล็ดหมามุ่ยไทยด้วยตัวทำละลายเอทานอล สามารถกระตุ้นการสร้างนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน Ten-4 และ GAP-43 ในขณะที่สารสกัดจากเมล็ดของหมามุ่ยอินเดียด้วยตัวทำละลายเฮกเซนกระตุ้นการสร้างนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน Ten-4 อีกทั้งศึกษาผลการยับยั้งการแสดงออกของยีน Ten-4 ต่อการกระตุ้นการสร้างนิวไรท์ของสารสกัดสมุนไพร ซึ่งพบว่ามีการสร้างนิวไรท์ และความยาวของนิวไรท์ลดลง จากการศึกษาสามารถบ่งชี้ได้ว่าสารสกัดจากเมล็ดหมามุ่ยไทย และหมามุ่ยอินเดียสามารถปกป้องเซลล์ประสาทจากสารอนุมูลอิสระได้ และสามารถกระตุ้นการงอกของนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน GAP-43 และ Ten-4 ซึ่งเป็นอีกทางเลือกหนึ่งในการพัฒนายาการกระตุ้นการเจริญพัฒนาของเซลล์ประสาทสำหรับป้องกัน หรือรักษาโรคความผิดปกติของระบบประสาท

โรคออทิซึมสเปกตรัม (Autism Spectrum Disorder, ASD) คือ โรคทางพัฒนาการระบบประสาทที่พบในเพศชายมากกว่าเพศหญิงโดยไม่ทราบสาเหตุ สาเหตุการเกิดโรคออทิซึมสเปกตรัมยังไม่เป็นที่ทราบแน่ชัดแต่การได้รับบิสฟีนอลเอขณะตั้งท้องมีความสัมพันธ์ต่อการเกิดโรคออทิซึมสเปกตรัมในลูกหนู อย่างไรก็ตามยังไม่มีการศึกษาแน่ชัดว่าการได้รับบิสฟีนอลเอขณะตั้งท้องส่งผลต่อการแสดงออกของยีนที่เกี่ยวข้องกับโรคออทิซึมสเปกตรัมหรือไม่ โดยเฉพาะเมื่อวิเคราะห์แบบแยกเพศ ในการศึกษาครั้งนี้ ทางคณะผู้วิจัยจึงทำการศึกษาผลของการได้รับบิสฟีนอลเอในขณะตั้งท้องต่อการแสดงออกของยีนในสมองและความเกี่ยวข้องกับโรคออทิซึมสเปกตรัมรวมไปถึงหน้าที่ของเซลล์ระบบประสาท ในการทำนายว่าการได้รับบิสฟีนอลเอสามารถเปลี่ยนแปลงการแสดงออกของยีนและมีความสัมพันธ์กับโรคออทิซึมสเปกตรัมหรือไม่ ทางผู้วิจัยได้ทำการวิเคราะห์แบบ meta-analysis โดยใช้ข้อมูลการแสดงออกของยีนที่ผิดปกติจากการได้รับบิสฟีนอลเอที่ได้ตีพิมพ์ไปแล้วและใช้การวิเคราะห์แบบ hypergeometric distribution หาความสัมพันธ์ระหว่างกลุ่มยีนดังกล่าวกับรายชื่อยีนจากฐานข้อมูลโรคออทิซึมสเปกตรัมต่าง ๆ สำหรับการศึกษาถึงผลของการได้รับบิสฟีนอลเอขณะตั้งท้องต่อการแสดงออกของยีนที่มีความเกี่ยวข้องกับโรคออทิซึมสเปกตรัมนั้น คณะผู้วิจัยได้นำสมองส่วนฮิปโปแคมปัสที่แยกได้จากลูกหนูที่แม่ได้รับบิสฟีนอลขณะตั้งท้องมาวิเคราะห์ทางทรานสคริปโตมิกส์ด้วยเทคนิค RNA-seq และใช้โปรแกรม Ingenuity Pathway Analysis วิเคราะห์อินเตอร์แอกโตมและบทบาทหน้าที่ทางชีวภาพของกลุ่มยีนที่มีการแสดงออกผิดปกติ ยีนที่มีความสัมพันธ์กับโรคออทิซึมสเปกตรัมถูกคัดเลือกเพื่อยืนยันด้วยเทคนิค quantitative RT-PCR นอกจากนี้เซลล์ระบบประสาทที่แยกได้จากลูกหนูถูกนำมาเพาะเลี้ยงเพื่อศึกษาความยาวของสายใยประสาท การศึกษาพบว่า จากการวิเคราะห์ทาง meta-analysis ยีนที่มีการแสดงออกผิดปกติจากการได้รับบิสฟีนอลเอมีความสัมพันธ์อย่างมีนัยสำคัญกับหน้าที่และโรคออทิซึมสเปกตรัม การวิเคราะห์ RNA-seq ในสมองส่วนฮิปโปแคมปัสที่แยกได้จากลูกหนูที่แม่ได้รับบิสฟีนอลเอขณะตั้งท้องพบว่ายีนที่มีการแสดงออกผิดปกติมีความสัมพันธ์กับโรคออทิซึมสเปกตรัม เป็นที่น่าสนใจอย่างยิ่งว่ากลุ่มยีนที่เคยมีการรายงานว่ามีความผิดปกติในโรคออทิซึมสเปกตรัม ได้แก่ Auts2 Foxp2 และ Smarcc2 แสดงออกผิดปกติอย่างจำเพาะต่อเพศ การวิเคราะห์เชิงอินเตอร์แอกโตมพบว่ายีนที่มีการแสดงออกผิดปกติจากการได้รับบิสฟีนอลเอมีความเกี่ยวข้องอย่างมีนัยสำคัญกับบทบาทหน้าที่ทางระบบประสาทที่เคยมีรายงานในผู้ป่วยออทิซึมสเปกตรัม การวิเคราะห์ความยาวของสายใยประสาทพบว่าการได้รับบิสฟีนอลเอส่งเสริมความยาวของสายใยประสาทและการแตกแขนงของสายใยประสาทแต่ลดขนาดของตัวเซลล์อย่างมีนัยสำคัญทางสถิติ จากการศึกษาครั้งนี้พบว่าการได้รับบิสฟีนอลเอขณะตั้งท้องส่งผลต่อการแสดงออกที่ผิดปกติของยีนในสมองส่วนฮิปโปแคมปัสและกลุ่มยีนดังกล่าวมีความเกี่ยวข้องกับโรคออทิซึมสเปกตรัมและแสดงออกจำเพาะในแต่ละเพศประกอบกับการได้รับบิสฟีนอลเอขณะตั้งท้องส่งเสริมความยาวของสายใยประสาท การศึกษานี้แสดงให้เห็นว่าการได้รับบิสฟีนอลเอขณะตั้งครรภ์อาจเป็นสาเหตุหรือเพิ่มความเสี่ยงต่อโรคออทิซึมสเปกตรัมและอาจจะเป็นหลักฐานที่อธิบายถึงความชุกของโรคนี้ในเพศชายที่มากกว่าเพศหญิงซึ่งควรศึกษาเพิ่มเติมในอนาคต

The Nod-like receptor 3 (NLRP3) inflammasome is an intracellular sensor that sets off the innate immune system in response to microbial-derived and endogenous metabolic danger signals. We previously reported that γ-tocotrienol (γT3) attenuated adipose tissue inflammation and insulin resistance in diet-induced obesity, but the underlying mechanism remained elusive. Pyruvate kinases serve an important role in regulating glycolysis pathways. Different pyruvate kinase isozymes can dictate which of several different products will be formed during the final step of glycolysis. Being able to reliably copy active pyruvate kinases (PK) would allow study of the different isozymes, with the goal of eventually creating plants that are more nutritionally beneficial for humans. This study focused on improving the protocol for replicating each of two isolated PK isozymes from the model plant Arabidopsis thaliana.
