The Impact Of Diet-Induced Obesity On Cerebrovasculature Structure And Function, Riley M. Feagle Apr 2025

The Impact Of Diet-Induced Obesity On Cerebrovasculature Structure And Function, Riley M. Feagle

MUSC Theses and Dissertations

Adequate cerebral perfusion is necessary to help the brain maintain a constant and steady state of nutrient delivery. Since the brain is highly metabolic, changes in the vasculature structure can impact perfusion levels and, therefore, reduce the ability of the brain to receive the support necessary for full functionality. Vascular dysfunction is a common comorbidity among obese individuals. Given the strong association between obesity and cognitive dysfunction, understanding the correlation between reduced cerebral perfusion and altered cerebrovascular structure is crucial. The overall hypothesis of this thesis originated after ample preliminary data, and states that diet-induced obesity reduces cerebral perfusion and …

Influences Of Sex, Alcohol, And Stress On Conditioned Fear Responses, Heyam K. Mayberry Jan 2025

Influences Of Sex, Alcohol, And Stress On Conditioned Fear Responses, Heyam K. Mayberry

MUSC Theses and Dissertations

Learned fear responses are acquired through the identification and association of threatening stimuli with aversive outcomes. Fear responding and the proper maintenance of defensive behaviors is integral to survival. However, without the homeostasis of such systems, fear responses can become maladaptive and lead to dysregulated behavioral responding. Psychological conditions such as alcohol use disorder (AUD) and post-traumatic stress disorder (PTSD) often lead to impairments in fear learning, deficits in fear memory, and heightened fear responses. While there are clear sex differences in the prevalence of both AUD and PTSD, development of either disorder increases the risk for comorbid AUD/PTSD, leading …

Complement Propagates Visual System Pathology After Traumatic Brain Injury, Davis Marchant Borucki Jun 2024

Complement Propagates Visual System Pathology After Traumatic Brain Injury, Davis Marchant Borucki

MUSC Theses and Dissertations

Background: Traumatic brain injury is associated with the development of visual system disorders, including deficits in visual acuity. Visual deficits can present with delay and worsen over time, suggesting that ongoing neuroinflammatory changes contribute to their development and progression. Complement activation is strongly associated with the neuroinflammatory response after TBI and has been shown to contribute to neuronal and synaptic loss. However, the contributions of complement activation after TBI to vision loss remains unclear.

Methods: We employed an open skull controlled cortical impact model of traumatic brain injury in adult male mice to study how TBI affects vision. Animals were …

A Multi-Modal Imaging Analysis Of Inter-Community Hub Nodes In Subjective Cognitive Decline Linking Longitudinal Hub Function Disruption To White Matter Integrity Kurtosis, Duncan Nowling Oct 2023

A Multi-Modal Imaging Analysis Of Inter-Community Hub Nodes In Subjective Cognitive Decline Linking Longitudinal Hub Function Disruption To White Matter Integrity Kurtosis, Duncan Nowling

MUSC Theses and Dissertations

Subjective Cognitive Decline (SCD) has garnered much interest as a potential identifiable preclinical stage and indicator of risk for cognitive decline in Alzheimer’s Disease and related dementias (ADRD). Identification of individuals in this stage though is difficult, as they present with objectively normal cognitive evaluation scores, relying instead upon self-report of concern about decline in cognitive abilities. The use of non-invasive in-vivo imaging methods like BOLD functional imaging and diffusion tensor have allowed for complex mapping of both the functional and structural network features unique to this condition. This study furthers this network biomarker map of SCD by investigating the …

Activity Dynamics Of The Nucleus Accumbens Neurons During Natural And Drug Reward Seeking, Reda M. Chalhoub Jun 2023

Activity Dynamics Of The Nucleus Accumbens Neurons During Natural And Drug Reward Seeking, Reda M. Chalhoub

MUSC Theses and Dissertations

The nucleus accumbens core (NAc) represents a major neural substrate of reward encoding that has been found to play a role in differentially regulating natural and drug rewards, including cocaine. Its constituent D1- and D2- receptor expressing medium spiny neurons (MSN) are alternatively hypothesized to have either opposing effects on reward seeking or a shared role in forming encoding “ensembles” – a sparsely activated population of neurons that encode reward associations- that coordinate behavioral responding to rewards and cues predicting reward availability. Previous methods used to study these neuronal populations failed to temporal quantify the cell-type specific activity in reward …

The Role Of Complement In Stroke And Traumatic Brain Injury, Christine Couch May 2023

The Role Of Complement In Stroke And Traumatic Brain Injury, Christine Couch

MUSC Theses and Dissertations

Brain and neural injury are a non-specific disease category that includes traumatic brain injury (TBI) and stroke. Both TBI and stroke are common, costly, and leading causes of severe disability in adults. Both stroke and TBI are responsible for substantial disability in working age adults, with stroke being the second leading cause of death worldwide [1] and TBI a major cause of disability in people younger than their 40's [2]. The immune response after brain injury is multifactorial and involves both local and systemic events at the cellular and molecular level. The complement system is a component of both the …
