Cranial nerves provide afferent pathways that enable bottom-up modulation of central autonomic and cognitive neural pathways. Because these pathways project to brainstem nuclei such as the nucleus tractus solitarius and engage neuromodulatory systems, including the locus coeruleus, electrical stimulation offers a mechanistically grounded approach to influencing brain function. This thesis tested whether active transcutaneous auricular vagus nerve stimulation (taVNS) produces physiological responses indicative of parasympathetic engagement, defined as decreases in heart rate, increases in heart rate variability, and decreases in respiration relative to a dose-matched earlobe control condition. Breast cancer survivors (BCS) often report noticeable cognitive difficulties despite showing limited impairment on standardized neuropsychological tests. Some have theorized that this is simply an insensitivity of standardized testing to subtle cognitive changes (test-insensitivity hypothesis), whereas others have suggested there are early neural changes that are detected by patients but precede detection by these tests (early signal hypothesis) or that when they achieve normal performance, it is through saliently effortful compensatory processes (compensation hypothesis). Trait measures of negative emotionality have sometimes been shown to play a role in shaping threat learning, but findings are mixed. For example, Johnson et al (2023) showed that the inhibitory and prospective subscales of the Intolerance for Uncertainty Scale (IUS) were associated with diminished and enhanced threat-safety discrimination, respectively, during the acquisition phase of a Pavlovian fear conditioning task. This finding was subsequently replicated in an independent sample (Kinney-Petrucha, 2024, unpublished). Music is a type of aesthetic stimulus that unfolds dynamically over time. Combining approaches from previous research on musical preferences and research on spatial integration as a driver of visual aesthetic appreciation, we investigated how the integration of musical percepts over time influences listener enjoyment and the level of agreement between the enjoyment ratings of each listener. We used a DNN tool to quantify temporal dependencies in 32 music clips and also computed complexity scores for them. This study investigated whether adopting an out-of-body perspective in virtual reality (VR) reduces subjective and physiological emotional responses to vicarious pain stimuli compared to inhabiting a first-person embodied avatar. Twenty-four participants experienced two counterbalanced VR conditions: (1) Avatar Embodiment (AV), in which they inhabited a photorealistic avatar from a first-person perspective, and (2) a Virtual Out-of-Body Experience (vOBE), in which they observed their avatar from a displaced third-person perspective. Perceptual illusions arising from multisensory integration reveal how the brain constructs subjective experience from sensory input, and provide a powerful means of allowing the investigation of neural processes that track perceptual interpretation rather than physical stimulation alone. While visual illusions induced by auditory or tactile stimuli have been fairly well studied, the neural mechanisms underlying visually-induced illusory touch remain unclear. This study investigated whether illusory tactile perception engages the somatosensory cortex in a manner similar to veridical touch. Prediction error (PE) is the discrepancy between expected and actual outcomes. It is a central mechanism in associative learning particularly within fear-conditioning paradigms. Identifying physiological markers of PE in aversive contexts has important implications for refining learning models and enhancing clinical interventions for anxiety-related disorders. This study examined skin conductance response (SCR) as a potential physiological index of PE by analyzing both anticipatory and omission-related responses across multiple learning phases. Sonic Hedgehog (Shh) is best known for its role in development, yet evidence supports Shh signaling plays a functional role in the adult brain. Previous work demonstrated that Shh is expressed by dopamine (DA) neurons in the ventral midbrain and that loss of Shh from these neurons leads to late-onset motor and structural abnormalities resembling Parkinson’s disease. Psychopathy is a complex personality disorder characterized by distinct emotional and interpersonal deficits, and affects approximately 25% of the prison population and 4.5% of nonincarcerated populations. The manifestation of psychopathy can be understood through two distinct variants, each demonstrating unique etiological pathways and clinical presentations. Primary psychopathy is characterized by core interpersonal and affective deficits, including shallow emotions and manipulativeness, while demonstrating reduced autonomic responsivity and lower anxiety levels. Secondary psychopathy presents with higher levels of impulsivity, anxiety, and emotional dysregulation, showing stronger associations with various psychopathologies. Anorexia nervosa (AN) is a severe psychiatric disorder with the highest mortality rate among mental health conditions, yet effective biological treatments remain limited. The activity-based anorexia (ABA) paradigm, which combines food restriction with wheel access to model the hyperactivity and self-starvation cycle characteristic of AN, shows considerable individual variability in vulnerability. Memory interference occurs when previously formed memories disrupt the acquisition or retrieval of new information. A prominent example of proactive interference is latent inhibition (LI), defined as a reduced ability to form or express Pavlovian associations with a stimulus following non-reinforced exposure. The interference-based model suggests that LI arises because the memory that the pre-exposed stimulus is inconsequential interferes with the memory that it predicts a reinforcer.

Fear is a fundamental component of human learning, critical for survival, and extensively studied through Pavlovian conditioning paradigms (LeDoux & Pine, 2016). This thesis examines the interplay between stimulus-level uncertainty and individual differences in intolerance of uncertainty (IU) on threat-safety discrimination learning. Using a within-subject design, participants were conditioned to certain (100% reinforcement), uncertain (50% reinforcement), and safety (0% reinforcement) cues through the pairing of colored visual stimuli with aversive electrical shocks. Discrimination learning was assessed using both implicit (SCR) and explicit (self-reported US expectancy estimates and unpleasantness ratings) measures.

Much of human experience is inherently subjective, such as our personal response to artwork. Traumatic brain injury (TBI) is known to involve damage in the neurovascular unit. TBI may disrupt the brain’s neurovascular coupling–the relationship between cerebral blood flow and neural activity–that can lead to cognitive and functional impairments. This study attempted to non-invasively investigate neurovascular coupling in patients with moderate to severe TBI (msTBI) during the first year post-injury using a combined metric of cerebral blood flow (CBF) and fractional amplitude of low-frequency fluctuations (fALFF), a proxy for spontaneous neural activity. This study investigated the molecular mechanisms underlying chemotherapy-induced cognitive impairment (CICI) following doxorubicin and cyclophosphamide treatment in a rat model, revealing three significant alterations in hippocampal tissue: selective downregulation of heterogeneous nuclear ribonucleoprotein A1 (hnRNPA1) (21% decrease, p=0.0298), compromised astrocytic network integrity evidenced by reduced GFAP-positive area in cornu ammonis 3 (CA3) (24%, pB=0.026) and dentate gyrus (DG) (19%, pB=0.027) regions, and elevated cellular senescence marker p19Arf specifically in DG (69%, pB=0.033) and CA3 (27%, pB=0.024). The lOFC has long been implicated in signaling reward expectancies, with controversy as to whether it signals the economic value of future outcomes, their sensory features to guide behavior or both. This study aims to examine the intricate relationship between language experience and the neural processing of stop consonant speech sounds. Previous research has shown minimal differences in amplitudes and latencies of cortical Auditory Evoked Potentials (AEPs) to speech sounds across listeners from various language backgrounds. It is only towards the later latencies (around the P2 peak at 200 ms), that there have been suggestions of cross-linguistic differences. Ventral tegmental area (VTA) dopamine neurons signal and participate in reward-related learning. Specifically, dopamine is postulated to encode reward-related environmental stimuli to compute reward prediction errors (RPEs). It is through the computation and maintenance of RPEs that learning occurs. However, little is known about the neural mechanisms that underlie how dopamine neurons compute RPEs and facilitate reward-related learning. Ensemble perception is a process that allows our sensory systems to rapidly extract summary information about the stimuli in the environment. For example, we are able to get a sense of the average number of items in a group of similar items (Burr & Ross, 2008; Halberda, Sires, & Feigenson, 2006) or the average size of a group of similar shapes of different sizes (Ariely, 2001). Locomotion, a fundamental behavior, is classically assessed using simple metrics of velocity and total distance moved in an open field. This behavior however offers rich data about the organization and breadth of actions an animal can take while only facing limitations due to the bio-mechanical constraints of the organism. Importantly, these actions are not biased towards a particular task-solution space imposed by many behavioral paradigms. Here, to analyze motor complexity in freely ambulating mice, we developed a method for the automatic and unbiased identification of locomotor bouts which we call Bout Finder (BF).
