Within a group, individuals establish their preferred distance from each other, or social space, a form of social behaviour. The resulting distance depends on the exchange of social cues from others that needs to be perceived and integrated within the organism’s neural circuitry. In humans, social spacing can be impaired in neuropsychiatric disorders such as autism and schizophrenia. Ion channels are essential for neural function, playing a variety of necessary cellular roles including excitability, maintaining ion gradients, and volume control. While the role of ion channels in neurons is well-defined, much less is known about excitability in neural progenitor cells. Recently it has emerged that these neural precursors may be affected by channelopathies, indicating a critical role of ion channels in neural development. Nicotiana glauca is a medicinal plant used by traditional healers as antibacterial, antifungal, antiviral and anti-inflammatory medicines. The leaves of N. glauca are very rich in indolic alkaloids which give it a larvicidal power which allows us to use it as a bio-insecticide.In the present study we were perfomed in the direct (mortality) and indirect (food attractiveness and pupation) toxic effects of the aqueous extract of N. glauca on the mortality and feeding behavior of the fruit fly Drosophila melanogaster. Phosphoinositides (PI) are lipid molecules reversibly phosphorylated by kinases at the 3’, 4’ and 5’ -OH groups of their myoinositol ring, producing seven PI species. PIs act in a dynamic and acute manner at the synapse, facilitated by multiple kinases that synthesize their respective PI species at the precise location and time. There are three phosphatidylinositol 4-kinase (PI4K) genes that encode three different PI4K enzymes (PI4KII, PI4KIIIβ (fwd) and PI4KIIIα) to synthesize PI4P (converted to PIP2), serving vital functions at the synapse. Plants are rich in bioactive chemical secondary metabolites and have proven insecticidal activity by killing or repelling insects. In this work, we aim to evaluate the direct and delayed effects of ethanolic plant extracts on the vinegar fly (Drosophila melanogaster). The treatment was performed by ingestion on second instar larvae (L2) to evaluate the impact of the ethanolic extract on development for 15 days and subsequently on the feeding behavior of the larvae. Molecular chaperones maintain protein homeostasis (proteostasis) by ensuring the proper folding of polypeptides. Loss of proteostasis has been linked to the onset of numerous neurodegenerative disorders including Alzheimer’s, Parkinson’s, and Huntington’s disease. Hsp110 is a member of the Hsp70 class of molecular chaperones and acts as a nucleotide exchange factor (NEF) for Hsp70, the preeminent Hsp70-family protein folding chaperone. Hsp110 promotes rapid cycling of ADP for ATP, allowing Hsp70 to properly fold nascent or unfolded polypeptides in iterative cycles. Previous studies in Drosophila melanogaster have established that overexposure to light damages photoreceptors and that photoreceptor degeneration contributes to shortened lifespans. However, the role of diet and circadian rhythms in the eye has not yet been elucidated. Many vital processes in the eye are regulated by circadian rhythms, and circadian misalignment has been proven as a driver of aging. To successfully navigate the complex visual world, animals must extract relevant information from the deluge of light-carried signals that arrive at their eyes. Early vision filters are passive, energy-saving gates that block out irrelevant signals. The remaining incoming signals are then subject to active filtering by visual attention systems which are energetically expensive, especially for smaller animals, which are subject to similar survival challenges as larger animals.

The presence of neuronal cytoplasmic inclusions (NCIs) composed of RNA-binding proteins (RBPs) and neurofilaments is considered to be ALS’s neuropathological hallmark. RGNEF has been previously shown to interact with TDP-43 and to have a regulatory effect on the expression levels of NEFL mRNA and NFL protein in vitro. Here, I examined the mechanism of the RGNEF N-terminus, leucine-rich domain (LeuR) domain’s interaction with TDP-43. Intellectual disability (ID) is a neurodevelopmental disorder associated with many epigenetic regulators and chromatin modifying enzymes like histone lysine methyltransferases (KMTs) and demethylases (KDMs). Here, I systematically investigate the role of 7 KDMs: Su(var)3-3, KDM2, Lid, CG2982, UTX, KDM4B, JHDM2, and 1 KMT: trr in the context of learning and memory using Drosophila melanogaster. Genetic knockdown of each gene in the mushroom body (MB) of flies are tested for short- and long-term memory impairment using courtship conditioning. Nociception is the perception of and response to harmful stimuli. Nociception is essential for minimizing tissue damage, but aberrant nociceptive pathways can result in chronic pain. Chronic pain in the U.S. is commonly managed with wide-acting opioids, and precisely defining the components of nociceptive pathways could uncover novel targets for pain therapies. Gene transcription required for long-term memory requires the modification of histones. However, there are still many uncertainties about the identity and spatial expression of genes regulated by histone modifications during memory related processes. In this project I examined the role of Drosophila melanogaster methyltransferases Set1 and trx in courtship memory. Genetic knockdown of Set1 and trx in the mushroom body (MB) revealed that Set1 was necessary for short- and long-term memory, while trx was only required for long-term memory. The SWI/SNF complex is an evolutionarily conserved ATP-dependent chromatin remodelling complex that has been implicated in the aetiology of intellectual disability (ID). Among the dominant ID genes, the SWI/SNF complex is the most highly enriched protein complex. However, its role in the nervous system is not yet understood. I systematically investigated the role of this complex in the development of the Drosophila mushroom body (MB), a complex brain structure required for learning and memory. This thesis addresses neuroscience research focusing on the brain’s mechanisms underlying behavioral choice, or prioritization, and decision-making. The research has been conducted with Drosophila melanogaster, the fruit fly – a good model from both the behavioral and neural perspectives. This project specifically observes the co-regulation of sleep with two other behaviors – courtship and oviposition. The overlap between the sleep and courtship circuits in the brain should provide a good model for behavioral prioritization, and the interaction between sleep and ovipositional preference should provide a model for understanding the effects of sleep on decision-making. If eusociality evolved through modification of pre-social mechanisms for regulating personal reproduction, then even insects like Drosophila may be vulnerable to latent effects of 'queen' pheromone. Here, I test if male fruit flies respond to a eusocial queen bee pheromone. I found that male flies were attracted to queen bee pheromone, and pheromone-treated males raised the intensity of their courting towards conspecific females. These novel observations from Drosophila suggest that male flies have the capacity to respond to queen pheromone in a manner that is comparable to the native response from male (drone) bees. Recent studies have linked neuropsychiatric disorders to older parents. These disorders often include changes in social behaviours like the social spacing between neighbouring individuals, which can be modeled in organisms such as Drosophila melanogaster. I investigated the effects of aging on the social space between neighbouring D. melanogaster and how aging impacts the next generation. To achieve this, I used the social space assay and found that individuals become less social with age and that this effect is passed on to the first generation only. Understanding the biophysical basis of animal magnetoreception has been one of the greatest challenges in sensory biology. Recently it was discovered that the light-dependent magnetic sense of Drosophila melanogaster is mediated by the ultraviolet (UV)-A/blue light photoreceptor cryptochrome (Cry). Here we show, using a transgenic approach, that the photoreceptive, Drosophila-like type 1 Cry and the transcriptionally repressive, vertebrate-like type 2 Cry of the monarch butterfly (Danaus plexippus) can both function in the magnetoreception system of Drosophila and require UV-A/blue light (wavelength below 420 nm) to do so. Appropriate termination of the phototransduction cascade is critical for photoreceptors to achieve high temporal resolution and to prevent excessive Ca(2+)-induced cell toxicity. Using a genetic screen to identify defective photoresponse mutants in Drosophila, we isolated and identified a novel Galpha(q) mutant allele, which has defects in both activation and deactivation. We revealed that G(q) modulates the termination of the light response and that metarhodopsin/G(q) interaction affects subsequent arrestin-rhodopsin (Arr2-Rh1) binding, which mediates the deactivation of metarhodopsin. We further showed that the Galpha(q) mutant undergoes light-dependent retinal degeneration, which is due to the slow accumulation of stable Arr2-Rh1 complexes. Our study …

Forward genetic screens using chemical mutagens have been successful in defining the function of thousands of genes in eukaryotic model organisms. The main drawback of this strategy is the time-consuming identification of the molecular lesions causative of the phenotypes of interest. With whole-genome sequencing (WGS), it is now possible to sequence hundreds of strains, but determining which mutations are causative among thousands of polymorphisms remains challenging. The steroid hormone ecdysone plays vital roles during Drosophila development. Pulses of 20E during Drosophila life cycle function as temporal cues, signaling the onset of metamorphic processes, including the stage specific programmed cell death of larval tissues. Ecdysone is the critical developmental cue orchestrating the metamorphic reformation of CNS, resulting in the formation of adult-specific neural circuitry. Ecdysone signaling is transduced by a heterodimeric receptor complex formed between two nuclear receptors: EcR and Ultraspiracle (USP). Mushroom bodies (MBs) are the site of multi modal sensory integration critical for associative conditioning in Drosophila. They have been central to research on the structure function relationship in the brain for over one hundred years due to their unique shape and readily accessible physiology.
