Disruption of conditioned avoidance response (CAR) in rodents is one trademark feature of many antipsychotic drugs. In adult rats, repeated olanzapine (OLZ) treatment causes an enhanced disruption of avoidance response (sensitization), whereas repeated clozapine (CLZ) treatment causes a decreased disruption (tolerance). The present study addressed (1) whether OLZ sensitization and CLZ tolerance can be induced in adolescent rats, and (2) the extent to which OLZ sensitization and CLZ tolerance induced in adolescence persists into adulthood. Spea is a genus of toad-like, arid adapted frogs distributed throughout much of the western U.S. and northern Mexico. Two species, (S. Bombifrons and S. Multiplicata) are syntopic throughout most of the northern Chihuahuan Desert, a region that is experiencing rapid urbanization. For this study, I examined 936 male advertisement calls from urban and rural populations of S. multiplicata, and a rural population of S. bombifrons in west Texas and south-central New Mexico. The following study investigated the extent to which time-dependent memory consolidation facilities the generalization of person-specific traits (individuated targets) to other, familiar social group members (familiar targets) Sixty-Nine (N = 69) participants learned to distinguish between two arbitrary groups, one positive and one negative. Participants learned the negative or positive information about a subset of the group members (individuated targets) and no individuating information about the familiar targets. Participants returned either without a time-delay containing sleep (2-6 hours after learning) or a time-delay with sleep (48 hours after learning). The evaluative oddball is an implicit measure for detecting (concealed) attitudes. In evaluative oddball studies, low occurrence stimuli such as negative pictures are presented among high occurrence context stimuli such as positive pictures. Late positive potentials (LPPs) of the event-related potential (ERP) are larger to evaluatively incongruent stimuli such as negatives compared to congruent stimuli such as positives with the context (e.g., positives). In prior evaluative oddball paradigms, this evaluative congruity effect of the LPP was reduced when participants concealed compared to truthfully reported attitudes. Nerve regeneration techniques have been studied in great depth in recent years. Peripheral nerve injuries, in particular, can be caused by accidents, falls, athletic injuries, etc. Current methods of repair include direct suturing of the 2 injured nerve ends or the use of autografts, when a nerve section is taken from another area of the body. Both methods have a series of limitations which have led researchers to study alternative methods of repair. Cognitive and communicative deficits associated with traumatic brain injury are an active area of research. However, no research to date has reported on the functionality of motor speech following a concussion. A common tool used to evaluate the motor speech status of potentially impaired individuals is the measurement of the diadochokinetic speech rate. The purpose of this study is to investigate diadochokinetic speech rates in individuals who experience a sports-related concussion. Background: Lamotrigine is an anticonvulsant drug indicated for the maintenance treatment of bipolar I disorder and for various types of epilepsy. An orally disintegrating tablet (ODT) of lamotrigine was developed to provide a formulation option that might increase patient convenience and satisfaction. Methods: Subjects with mood disorders who reported difficulty swallowing currently prescribed lamotrigine immediate-release medication (baseline) were enrolled and treated with lamotrigine ODT for three weeks (end of treatment). Subject satisfaction and convenience were measured using the Treatment Satisfaction Questionnaire for Medication (TSQM). In the U.S., about 200,000 people are currently living with spinal cord injury (SCI). An estimated of 50%-70% of all SCI cases occurs in the range of ages between 15-35 years old. The destructive neurotrauma results in the majority of adult disability, even after patients suffering with SCI survived from the acute death. Chronic intermittent ethanol exposure (CIE) is associated with degeneration of hippocampal neurons. The present study used hippocampal cultures to examine the loss of NeuN immunoreactivity, a relaible marker or neuronal density, after 1, 2, or 3 cycles of 5 days EtOH exposure (50 mM), followed by a 24-hour period of EWD or continuous EtOH exposure. NeuN immunoreactivity was decreased by 13%, 19%, and 16% in the CA1, CA3, and dentate gyrus after 3 cycles of CIE respectively; thionine staining confirmed significant cellular losses within each hippocampal subregion. The ZNF9 gene on chromosome 3 encodes the cellular nucleic acid binding protein (CNBP), a ubiquitously expressed, 177 amino acid (≈19.5kDa) protein that is highly conserved among vertebrates. The function of the protein is largely unknown, however an expansion in the first intron of the protein results in myotonic dystrophy type 2 (DM2), a multisystemic disease featuring cardiac arrhythmia, muscle wasting, cataracts, and a range of neuropathologies. Remarkably, we recently discovered that CNBP is involved in regulating the activity of β-secretase, the enzyme that produces the first cleavage event in the generation of the amyloid-β peptide (Aβ). Glutamate is the main excitatory neurotransmitter in the CNS and at the neuromuscular junctions (NMJs) of invertebrate. The characteristic similarities to CNS glutamatergic synapses in vertebrate and the anatomical simplicity of invertebrate NMJs favor the investigation of glutamatergic synaptic functions in this system. This dissertation mainly aimed to physiologically separate two functional vesicle groups, the reserve pool (RP) and readily releasable pool (RRP) within presynaptic nerve terminals of Procambarus Clarkii and Drosophila melanogaster. Alzheimer’s disease (AD) is a progressive neurodegenerative dementia characterized by amyloid plaque accumulation, synapse/dendrite loss, and cognitive impairment. Transgenic mice expressing mutant forms of amyloid-β precursor protein (AβPP) and presenilin-1 (PS1) recapitulate several aspects of this disease and provide a useful model system for studying elements of AD progression. AβPP/PS1 mice have been previously shown to exhibit behavioral deficits and amyloid plaque deposition between 4–9 months of age. We crossed AβPP/PS1 animals with mice of a mixed genetic background (C57BL/6 × 129/SvJ) and investigated the development of AD-like features in the resulting outcrossed mice. Rats learn to prefer flavors associated with postingestive effects of nutrients. The physiological signals underlying this postingestive reward are unknown. We have previously shown that rats readily learn to prefer a flavor that was consumed early in a multi-flavored meal when glucose is infused intragastrically (IG), suggesting rapid postingestive reward onset. The present experiments investigate the timing of postingestive fat reward, by providing distinctive flavors in the first and second halves of meals accompanied by IG fat infusion. We previously observed that mental manipulation of the pitch level or temporal organization of melodies results in functional activation in the human intraparietal sulcus (IPS), a region also associated with visuospatial transformation and numerical calculation. Two outstanding questions about these musical transformations are whether pitch and time depend on separate or common processing in IPS, and whether IPS recruitment in melodic tasks varies depending upon the degree of transformation required (as it does in mental rotation). Glioblastoma multiforme (GBM) is a debilitating malignant brain tumor with expected patient survival of less than a year and limited responsiveness to most treatments, often requiring biopsy for diagnosis and invasive surgery for treatment. We propose a DNA computer system, consisting of input, computation, and output components, for diagnosis and treatment. The input component will detect the presence of three GBM biomarkers: vascular endothelial growth factor (VEGF), caveolin-1α (CAV), and B2 receptors. The computation component will include indicator segments for each of these genes, and ensure that output is only released if all the biomarkers are present. The output component …

Post-traumatic stress disorder (PTSD), an anxiety disorder precipitated by exposure to extreme emotional and/or physical stress, is characterized by persistent, intrusive memories of the precipitating trauma. Thus, the pathogenesis of PTSD has been conceptualized as involving a deficit in consolidation mechanisms underlying the extinction of fear memory. The mechanisms underlying this deficit have not been elucidated. In addition to intrusive memories, patients with PTSD display heightened sensitivity of the hypothalamic pituitary adrenocortical (HPA) axis to glucocorticoid negative feedback. As glucocorticoids are important modulators of memory consolidation, increased sensitivity to HPA negative feedback, by blunting glucocorticoid responses, may contribute to memory-related …

The works presented in this dissertation provide insights into the mechanisms of chemotherapy-induced cognitive impairment (CICI or “ChemoBrain”) and take steps toward outlining a preventive strategy. CICI is now widely recognized as a complication of cancer chemotherapy experienced by a large percentage of cancer survivors. Approximately fifty percent of existing FDA-approved anti-cancer drugs generate reactive oxygen species (ROS). Doxorubicin (Dox), a prototypical ROS-generating chemotherapeutic agent, produces the reactive superoxide radical anion (O2-•) in vivo. Dox treatment results in oxidation of plasma proteins, including ApoA-I, leading to TNF-α-mediated oxidative stress in plasma and brain. TNF-α elevation in brain …

In recent years, there have been many studies looking at estrogen therapy (ET) as a treatment for Alzheimer's disease. Literature review suggests a close relationship between estrogen and apolipoprotein E (apoE) in the central nervous system. The mechanism underlying the interaction of apoE genotype with estrogen in the nervous system is not yet known. Therefore, the objectives of this research are 1) to determine the effects of estradiol treatment on basal cell proliferation and neuronal differentiation and 2) to identify the estrogen receptor subtype (ERα and ERβ) that mediates the increase of apoE levels, neurite outgrowth and neuronal numbers.

This study is a preliminary investigation of the validity of using time-based measures to quantify sustained attention in children ages 6-12. Problems with sustained attention negatively affect childhood learning and development. The prevalence of disorders known to impact sustained attention performance continue to rise in the United States. Currently, commercially available, objective measures of sustained attention use normative comparisons that provide limited information about the effect such problems have on child performance in natural settings.
