The mediodorsal thalamus (MD) is thought to be key component of the network that processes chemosensory information to guide our consummatory choices. Previous studies show that the mediodorsal thalamus receives projections from both the piriform cortex (PC) and gustatory cortex (GC), suggesting that it may process chemosensory information from both areas. Although the mediodorsal thalamus has been shown to respond to odors detected by sniffing, it remains unknown how its neurons represent experienced odors, tastes, and odor-taste mixtures originating from the mouth. Importantly, humans and animals with mediodorsal thalamic lesions do not suffer from anosmia, but experience deficits in odor …

Pediatric spinal cord injury (SCI) disrupts the efferent and afferent flow of the developing brain, leading to devastating functional impairments below the injury site, yet our understanding of its impact on the brain remains limited. This study examines supraspinal reorganization in children with SCI using electrophysiology and neuroimaging techniques to understand the relationship between residual spinal transmission and supraspinal reorganization. Chapter 2 discusses the development of a child-centric approach using ‘learn, play, and practice’ to foster a trusting relationship with each child and increase compliance with experimental protocols. The diversification of animal communication systems is driven by many interacting factors. Unintended receivers play an important part in this process, yet little is known about their role in signal evolution. Flies of the genus Ormia are parasitoids of crickets and rely on acoustic cues to locate hosts. In Hawaii, selection imposed by Ormia ochracea has led to recent and rapid diversification in the songs of their host. Here, we compare neural and behavioral responses of Hawaiian flies to those of an ancestral population to understand the role of parasitoid sensory and behavioral variation in the evolution of host songs. …

Circadian rhythms dictate the timing of both once-in-a-lifetime adult emergence (eclosion) and daily locomotor activity rhythms in the flesh fly S. crassipalpis. Light cycles are considered the primary environmental time cue (zeitgeber), but the life history of S. crassipalpis suggests that temperature cycles (thermocycles) may also play a key role. This work evaluates the efficacy of thermocycling as a zeitgeber in S. crassipalpis. We found that shifting both light and temperature cycles of sufficient amplitude affect the phasing of eclosion and locomotor activity, but result in different patterns. Schizophrenia is a troubling and severe mental illness that is only incompletely treated by currently available drugs. New drug development is hindered by a scarcity of functionally relevant pharmacodynamic biomarkers that are translatable across preclinical and human subjects. Although psychosis is a major feature of schizophrenia, cognitive and negative symptoms determine the long-term functional outcomes for patients. Stimulus-evoked neural synchrony at gamma (~ 40 Hz) frequency plays an important role in the processing and integration of sensory information. In the Unites States, approximately 1.5 million people currently have a spinal cord injury and suffer permanent sensory and motor loss due to the disruption of the spinal cord. Due to the significant morbidity, it is vital to understand the functional impact of disrupting neural descending pathways that modulate spinal neurons involved in intermuscular coordination critical for gait behaviors. Lewy body disorders are a group of neurodegenerative conditions characterized by the pathological misfolding and aggregation of the abundant protein, α-synuclein. The most common Lewy body disorders are Parkinson’s disease and dementia with Lewy bodies. Apart from ageing, male sex is a major risk factor for Lewy body disorders, as men are at ~1.5-fold higher risk for these diseases than women. Yet, preclinical studies on Lewy body disorders rarely examine sex as a biological variable, and the mechanisms underlying sex-skewedness in disease risk remain undetermined.

Major depressive disorder (MDD) is a leading cause of disability worldwide, with a lifetime prevalence rate of approximately 20%. Inadequate pharmacological treatment methods for MDD are a significant debilitating factor. Patient estimates suggest that the treatment resistance rate for pharmacological interventions is over 30%. Postmortem analyses of human tissue of individuals diagnosed with MDD have shown an increase in Poly (ADP-ribose) polymerase 1 (PARP-1) mRNA gene expression in prefrontal cortical white matter when compared to psychiatrically normal brain tissue. Hypocretin/Orexin (HCRT) neurotransmission facilitates drug-seeking behavior. HCRT neurotransmission at HCRT-receptors 1 and 2 (HCRT-R1 and -R2, respectively) is implicated in addiction. During the shift to alcohol-dependency, adaptations in neurotransmitter systems occur in reward- and stress-related brain regions. Specifically, neurotransmission systems in the central amygdala (CeA) are modulated by alcohol drinking/exposure. Therefore, this study investigated Hcrtr1 and Hcrtr2 mRNA expression in the CeA of alcohol-dependent rats and in non-dependent controls during acute alcohol withdrawal. Imbalances between inhibitory, GABAergic neurons, and excitatory, glutamatergic neurons, in this region are essential to brain circuity during social responses and are thought to be involved with behaviors associated with autism spectrum disorder (ASD). Alzheimer’s disease (AD) is symptomized by amyloid-beta plaques in the brain and accounts for more than 65 percent of dementia cases. Vitamin B12 (cobalamin) deficiency can result in similar cognitive impairment and roughly 15% of the elderly are vitamin B12 deficient. Vitamin B12 deficiency results in the accumulation of toxic methylmalonic acid and homocysteine. Hyperhomocysteinemia is a strong risk factor for AD. To test if vitamin B12 deficiency stimulates amyloid-beta toxicity, Caenorhabditis elegans expressing amyloid-beta in muscle were fed either vitamin B12-deficient OP50-1 or vitamin B12-rich HT115(DE3) E. coli bacteria. Vps54 is a subunit of the Golgi-associated retrograde protein (GARP) complex, which is involved in tethering endosome-derived vesicles to the trans-Golgi network (TGN). The “wobbler” mouse is the phenotypic result of a destabilizing point mutation in Vps54. This mutation causes neurodegeneration and is subsequently used as a model for human motor neuron disease. Presently, it is unclear how disruption of GARP complex function leads to motor neuron degeneration. The neuroinflammatory response to peripheral nerve injury is associated with chronic pain and significant changes in the expression profiles of RNAs in neurons, glia and infiltrating immune cells: a neuro-immune triad. Chronic constriction injury (CCI) of the rat sciatic nerve provides an opportunity to mimic neuropathic injury and quantitatively assess behavior and differential gene expression in individual animals. Olfactory sensory neurons (OSNs) are contained within the olfactory epithelium (OE) and are responsible for detecting odorant molecules in the air. The exposure of OSNs to the external environment is necessary for their function, but it also leaves them exposed to potentially harmful elements and thus results in a high turnover rate. Loneliness, the perception of unmet social needs, has been shown to relate to recollection-based recognition deficits, but the relationship between loneliness and recognition memory (i.e., recollection and familiarity) has not been thoroughly examined. The current study hypothesized that more lonely individuals would have lower recognition memory performance, specifically recollection, with smaller ERP parietal old-new effects than less lonely individuals. Forty participants, grouped into less (n = 13) and more (n = 9) lonely groups based on their R-UCLA responses, completed an associative memory task. EEG was used to assess recognition memory effects. The thalamic reticular nucleus (TRN) is an interface between thalamus and cortex that regulates thalamocortical rhythms and modulates sensory processing. Thalamocortical axon collaterals provide feedforward excitatory input onto GABAergic TRN neurons, which in turn convey feedback inhibition to dorsal thalamus. Here we used the mouse visual system to study the organization, pattern of innervation and functional responses between TRN and the dorsal lateral geniculate nucleus (dLGN). Schizophrenia is highly comorbid with other psychiatric disorders such as Alcohol Use Disorder (AUD) with a prevalence rate of 27% - 65%, which is significantly higher than AUD exhibited by the general population (6%). Endocranial morphology provides evidence of sensory ecology and sociality of extinct vertebrates. The Earliest Pliocene Gray Fossil Site (GFS) of NE Tennessee features a conspicuous dominance of skeletal elements belonging to the dwarf tapir, Tapirus polkensis. Numerous individuals in one fossil locality often suggests gregarious behavior, but sociality in T. polkensis contradicts behavior documented for extant Tapirus species. I test T. polkensis for variation in sensory and social ecology using computed tomography and 3D digital endocasts from an ontogenetic sequence. We explored the immune neuropathology underlying multi-day relief from neuropathic pain in a rat model initiated at the sciatic nerve by using a nanoemulsion-based nanomedicine as a biological probe. The nanomedicine is theranostic: both therapeutic (containing celecoxib drug) and diagnostic (containing near-infrared fluorescent (NIRF) dye) and is small enough to be phagocytosed by circulating monocytes. A model of neuropathic pain is initiated by tying four 1mm spaced knots around the sciatic nerve with chromic gut suture, which results in neuroinflammation, and a resultant pain-like behavior manifests. Chronic pain includes debilitating conditions affecting large populations of people throughout the world. These diseases are further complicated by comorbid psychological disorders which make chronic pain conditions difficult to treat. Nonpharmacological treatment options have more recently become an important mainstay of effective therapies and are currently being used to supplement or replace traditional pharmacological treatment methods. Exercise-based therapies in particular are of interest due to their demonstrated success in reducing pain symptoms as well as improving psychological outcomes. Phototransduction, transmission of visual information down the optic nerve incurs delays on the order of 50 – 100ms. This implies that the neuronal representation of a moving object should lag behind the object’s actual position. However, studies have demonstrated that the visual system compensates for neuronal delays using a predictive mechanism called phase advancing, which shifts the population response toward the leading edge of a moving object’s retinal image. Cleft palate is a common birth defect. Etiologic mechanisms of palate cleft include defects in palate morphogenesis, mandibular growth, or spontaneous fetal mouth movement. Cleft palate linked to deficient fetal mouth movement has been demonstrated directly only in a single experimental model of loss of neurotransmission. Here, using retinoid deficient mouse embryos, we demonstrate directly for the first time that deficient fetal mouth movement and cleft palate occurs as a result of mis-patterned development of pharyngeal peripheral nerves and cartilages.
