Obesity and drug abuse share common neural circuitries including the mesocoticolimbic and striatal dopamine reward system. In the current study, a rat model of diet-induced obesity (DIO) was used to determine striatal dopamine function, impulsivity and motivation as neurobehavioral outcomes and predictors of obesity. For the outcome study, rats were randomly assigned a high-fat (HF) or a low-fat (LF) diet for 8 wk. Following the 8-wk HF-diet exposure, rats were segregated into obesity-prone and obesity-resistant groups based on maximum and minimum body weight gain, respectively, and neurobehavioral outcomes were evaluated. Glutamate is the main excitatory neurotransmitter in the CNS and at the neuromuscular junctions (NMJs) of invertebrate. The characteristic similarities to CNS glutamatergic synapses in vertebrate and the anatomical simplicity of invertebrate NMJs favor the investigation of glutamatergic synaptic functions in this system. This dissertation mainly aimed to physiologically separate two functional vesicle groups, the reserve pool (RP) and readily releasable pool (RRP) within presynaptic nerve terminals of Procambarus Clarkii and Drosophila melanogaster. Song production in songbirds is controlled by parts of the brain known as the song control regions (SCRs). During spring, gonads increase in size, males sing to attract mates, and SCRs become larger. This neuroplasticity is controlled by the change in day length and increased plasma testosterone (T) levels. Plasma T can be reduced by stress through the production of corticosterone (CORT), through the production of beta-endorphin, or through direct effects on the testes via the nervous system. Overproduction of proinflammatory cytokines in the CNS has been implicated as a key contributor to pathophysiology progression in Alzheimer's disease (AD), and extensive studies with animal models have shown that selective suppression of excessive glial proinflammatory cytokines can improve neurologic outcomes. The prior art, therefore, raises the logical postulation that intervention with drugs targeting dysregulated glial proinflammatory cytokine production might be effective disease-modifying therapeutics if used in the appropriate biological time window. : A major function of the brain choroid plexus (CP) is to regulate the exchange of solutes between the blood plasma and the cerebrospinal fluid (CSF) using selective transporters. CSF inorganic phosphate (Pi) concentration is maintained at about one-half that of plasma and is potentially important because of its regulatory, structural, and biochemical functions. Phosphate is critical for ATP and DNA formation, the linked regulation between phosphate and calcium, and as an intracellular buffer. Traumatic brain injury (TBI) often results in disruption of the blood brain barrier (BBB), which is an integral component to maintaining the central nervous system homeostasis. Recently cytosolic calcium levels ([Ca2+]i), observed to elevate following TBI, have been shown to influence endothelial barrier integrity. However, the mechanism by which TBI-induced calcium signaling alters the endothelial barrier remains unknown. In the present study, an in vitro BBB model was utilized to address this issue. Peripheral axotomy causes significant alterations in intrinsic motoneuron activity and excitability. Despite successful reinnervation of peripheral targets after injury, the recovery of motor function is incomplete. Following axotomy in the adult cat, there is an increase in the afterhyperpolarization (AHP) of the action potential in specific motoneuron types. AHP duration can be inversely correlated with the amount of Ih or sag current and further shaped through SK (Small conductance calcium-activated potassium) channel currents (Gustafsson and Pinter, 1985; Kuno, Miyata, and Muñoz-Martinez, 1974a). Stroke is one of the leading causes of death and the top cause of long term disability. Currently there exists no standardized treatment of care for ischemic stroke patients during the days following stroke other than giving aspirin. A drug combination containing fluoxetine, one of two statins, and ascorbic acid was used to try and improve motor function following strokes in rats. Endothelin induced stroke survival surgery was performed on the right cortex of Long Evans and Sprague Dawley rats. We are interested in understanding the development of reciprocal inhibition, an important sensory-motor circuit that prevents co-contraction of antagonist muscles during locomotion. This effect is mediated by a subset of glycinergic neurons termed Ia inhibitory interneurons (IaINs). Transgenic mouse models are useful in helping to identify subsets of neurons in developing animals. In this thesis we first wanted to test the BAC-GlyT2-eGFP mice model by measuring GFP expression overlap in neurons with glycine in late-stage embryos (E17.5) and early postnatal animals (P0/P1). Our laboratory has found increased functional recovery post-stroke with Long Evans rats that received a combination of fluoxetine, simvastatin, and vitamin C. The objectives of this study were to test that treatment and other drug combinations on Sprague Dawley rats, to determine whether Forelimb Asymmetry is a reliable behavioral measure for stroke studies, and to determine the most optimal surgical procedure on Sprague Dawley rats. There were more significant increases in functional recovery observed on the Forelimb Asymmetry than on Montoya Staircase behavioral tests, but there were no significant differences between treatment groups and control. The Orai3 protein forms a Ca2+ channel with a pharmacological profile distinct from its close relative, the store-operated Ca2+ channel Orai1. Though closely related to Orai1, the function of Orai3 in humans is still unclear. This study attempts to contribute to the body of knowledge by undertaking a pharmacological analysis of Orai3 in insect cells. We describe here the creation of a vector capable of expressing the mammalian Orai3 gene on an insect background. Ammonia may increase in the blood and tissues of fishes following feeding or exposure to high concentrations of environmental ammonia arising from the degradation of organic matter in aquatic environments, sewage effluent, and run-off from landfills. Ammonia may be detoxified using the enzyme glutamine synthetase (GS), which catalyzes the conversion of ammonia to glutamine. It has been suggested, however that build-ups of glutamine within the astrocytes of the brain can cause brain swelling by increasing intracellular osmolarity and water uptake by the cells. Emerging data indicate that central neurons participate in diabetic processes by modulating autonomic output from neurons in the dorsal motor nucleus of the vagus (DMV). We tested the hypothesis that synaptic modulation by transient receptor potential vanilloid type 1 (TRPV1) receptors is reduced in the DMV in slices from a murine model of type 1 diabetes. The TRPV1 agonist capsaicin robustly enhanced glutamate release onto DMV neurons by acting at preterminal receptors in slices from intact mice, but failed to do so in slices from diabetic mice. Salmonids, such as Chinook salmon (Oncorhynchus tshawytscha) and steelhead (O. mykiss), are a staple economic, recreational, tribal, and environmental resource, yet many populations are unsustainable. This study was part of a broad scale effort to monitor the impact of downstream migration obstacles on juvenile salmonid health and survival, which is an essential step towards increasing Smolt-to-Adult Return ratios (SARs). Idiopathic pulmonary fibrosis (IPF) is an interstitial lung disease leading to decreased lung volume and eventual respiratory failure. At present, the median post-diagnosis lifespan is between three and six years. Myofibroblasts are collagen-secreting cells essential for wound healing, but also implicated in the fibroproliferation and extra cellular matrix deposition commonly seen in IPF. The nitric oxide (NO) signaling pathway is implicated in protomyofibroblast to myofibroblast transition and regulation. Astrogliosis is induced by neuronal damage and is also a pathological feature of the major aging-related neurodegenerative disorders. The mechanisms that control the cascade of astrogliosis have not been well established. In a previous study, we identified a novel androgen receptor (AR)-interacting protein (p44/WDR77) and found that it plays a critical role in the control of proliferation and differentiation of prostate epithelial cells. In the present study, we found that deletion of the p44 gene in the mouse brain caused accelerated aging with dramatic astrogliosis. Functional plasticity of synaptic networks in the dentate gyrus has been implicated in the development of posttraumatic epilepsy and in cognitive dysfunction after traumatic brain injury, but little is known about potentially pathogenic changes in inhibitory circuits. We examined synaptic inhibition of dentate granule cells and excitability of surviving GABAergic hilar interneurons 8–13 weeks after cortical contusion brain injury in transgenic mice that express enhanced green fluorescent protein in a subpopulation of inhibitory neurons. Whole-cell voltage-clamp recordings in granule cells revealed a reduction in spontaneous and miniature IPSC frequency after head injury; no concurrent change in paired-pulse ratio was found …

The ionotropic glutamate receptors are localized in the pre- and postsynaptic membrane of neurons and mediate the majority of fast excitatory synaptic transmission. These receptors are divided into three subtypes: α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA), kainic acid, and N-methyl-D-aspartic acid (NMDA) receptors. Spatial distribution of synaptic inputs on the dendritic tree of a neuron can have significant influence on neuronal function. Consequently, accurate anatomical reconstructions of neuron morphology and synaptic localization are critical when modeling and predicting physiological responses of individual neurons. Previous studies regarding subventricular zone (SVZ) neural stem cells (NSCs) have either observed global changes in the SVZ niche containing NSCs or an impure NSC population. By imaging wholemount preparations of mouse brains and staining with molecular markers specific for proliferating NSC nuclei, the study was able to give a more accurate depiction of the SVZ NSC population than its predecessors. This study will quantify the percentage of proliferating NSCs at various ages of adult murine brain development in order to elucidate a possible correlation between NSC division kinetics and the declining neurogenic output witnessed with aging.

The study aimed to understand the neuronal regulation of male sexual and agonistic behavior in broiler breeders. First, brain structures associated with sexual and agonistic behavior were identified by mapping Fos expression. The ventromedial subnucleus of medial portion of bed nucleus of the stria teriminalis (BSTM2) was specifically activated by male courtship behavior. The medial preoptic nucleus (POM) and lateral septum (SL) were associated with both sexual and agonistic behaviors. The bed nucleus of the pallial commissure (NCPa) and the paraventricular nucleus (PVN) were closely related to stress. Radial Glia (RG) are a mitotically active population of cells which reside within the ventricular zone at the lateral ventricle and give rise to the pyramidal neurons and astrocytes of the neocortex. Through cellular divisions, RG produce two daughter cells, one which resides in the ventricular zone and becomes another RG while the other is an immature progenitor which migrates away from the ventricle and populates the growing cortex. Lack of linearity and sensitivity, oxygen dependence, biofouling and tissue inflammation hinder the development of implantable biosensors for continuous monitoring of glucose. Herein, we report the development of stacked outer membranes based on LBL/PVA hydrogels that improve sensor sensitivity, linearity, oxygen independence and counter biofouling and inflammation. While the inner LBL membrane affords tunable diffusivity, the outer PVA is capable of releasing anti-inflammatory drugs/tissue response modifying agents to counter acute and chronic inflammation, and to induce neo-angiogenesis at the implant site.
