Investigation Of Mirnas Expression In A Citron-Kinase Mutant Model Of Microcephaly, Shan Parikh

Mutation of Citron-Kinase (Cit-K) in rodents causes substantial reductions in the number of neurons generated in the CNS and results in a primary microcephaly-like phenotype. Evidence from drosophila genetics has further established a genetic link between Cit-K and a protein Argonaut 1 (AGO1), which is required for proper functioning of the miRNA machinery (2). Experiments characterizing the role of miRNAs in the developing cortex demonstrate the requirement of miRNAs for differentiation of neural progenitor cells starting at embryonic day 12.5 (3). Together, this evidence links the role of miRNAs to neurogenesis and thus this relationship warrants further investigation. Here miRNA …

Characterizing And Treating The Neuropathology Of Tuberous Sclerosis Complex In The Mouse, Sharon W. Way

Tuberous sclerosis complex (TSC) is a multisystem, autosomal dominant disorder affecting approximately 1 in 6000 births. Developmental brain abnormalities cause substantial morbidity and mortality and often lead to neurological disease including epilepsy, cognitive disabilities, and autism. TSC is caused by inactivating mutations in either TSC1 or TSC2, whose protein products are known inhibitors of mTORC1, an important kinase regulating translation and cell growth. Nonetheless, neither the pathophysiology of the neurological manifestations of TSC nor the extent of mTORC1 involvement in the development of these lesions is known. Murine models would greatly advance the study of this debilitating disorder. This thesis …

The development of the brain and its underlying circuitry is dependent on the formation of trillions of chemical synapses, which are highly specialized contacts that regulate the flow of information from one neuron to the next. It is through these synaptic connections that neurons wire together into networks capable of performing specific tasks, and activity-dependent changes in their structural and physiological state is one way that the brain is thought to adapt and store information. Locus Of Control And The Age Difference In Free Recall From Episodic Memory, Paul Amrhein, Judith K. Bond, Derek Hamilton

The authors investigated the relation of locus of control (LOC) to age differences in free-recall memory performance. Older and younger participants completed P. C. Duttweiler's (1984) Internal Control Index (ICI) and subsequently performed free-recall memory tasks. Compared with the younger participants, the older participants exhibited poorer recall with more intrusions and uncorrected repetition errors as well as reduced categorical clustering. For the older participants with less internal LOC, recall proportion and item-pair associative recall clustering were lower than for the older participants with more internal LOC. By contrast, the younger participants did not exhibit any LOC effects in their recall …

The study of socioeconomic status (SES) and the brain finds itself in a circumstance unusual for Cognitive Neuroscience: large numbers of questions with both practical and scientific importance exist, but they are currently under-researched and ripe for investigation. This review aims to highlight these questions, to outline their potential significance, and to suggest routes by which they might be approached. Although remarkably few neural studies have been carried out so far, there exists a large literature of previous behavioural work. This behavioural research provides an invaluable guide for future neuroimaging work, but also poses an important challenge for it: how …

The extent to which a non-sedative dose of chlordiazepoxide (CDP) is able to modify the behavioral responses toward a predator odor was assessed in juvenile rats. Play behavior was suppressed and defensive behaviors were enhanced in the presence of a collar previously worn by a cat, when tested 24 hours later in the same context as that where the exposure occurred, and when tested in a context different than that in which the exposure occurred for up to 3 hours after exposure. In Vitro Guidance Of Retinal Axons By A Tectal Lamina-Specific Glycoprotein Nel, Yulan Jiang, Hiroya Obama, Soh-Leh Kuan, Ritsuko Nakamura, Chizu Nakamoto, Zhufeng Ouyang, Masaru Nakamoto

Nel is a glycoprotein containing five chordin-like and six epidermal growth factor-like domains and is strongly expressed in the nervous system. In this study, we have examined expression patterns and in vitro functions of Nel in the chicken retinotectal system. We have found that in the developing tectum, expression of Nel is localized in specific laminae that retinal axons normally do not enter, including the border between the retinorecipient and non-retinorecipient laminae. Nel-binding activity is detected on retinal axons both in vivo and in vitro, suggesting that retinal axons express a receptor for Nel. In vitro, Nel inhibits retinal axon …

The extent to which social variables may modulate the fear associated with a predator cue was assessed in juvenile rats. Cat odor reduced play to a comparable extent in both socially housed and isolate-housed rats, although socially housed rats exhibited more risk assessment during extinction. Rats that had played previously in the context used for assessing fear hid slightly less when exposed to cat odor than those rats that had not played previously in the testing context. However, no other differences were found between these two groups suggesting that prior social experience with the testing context has minimal effects on …

Previous studies documented significant behavioral changes in the offspring of cocaine-exposed mothers. We now explore the hypothesis that maternal cocaine exposure could alter the fetal epigenetic machinery sufficiently to cause lasting neurochemical and functional changes in the offspring. Pregnant CD1 mice were administered either saline or 20 mg/kg cocaine twice daily on gestational days 8–19. Male pups from each of ten litters of the cocaine and control groups were analyzed at 3 (P3) or 30 (P30) days postnatum. Effects Of Neonatal Handling On Play Behavior And Fear Towards A Predator Odor In Juvenile Rats, Stephen M. Siviy

The effects of brief daily separation, also known as "handling," during the first 2 weeks of life on play behavior and fearfulness toward a predatory odor were assessed in juvenile rats. Handled rats were more playful than nonhandled control rats, and while handling had no effect on the direct response of these rats toward a predatory odor, handled rats did not exhibit a conditioned suppression of play when tested later in the same context where they had been exposed to the predatory odor.
